The buspirone drug is usually present as hydrochloride form of general formula C(21)H(31)N(5)O(2).HCl, and of molecular weight (MW)=421.96. It is an analgesic anxiolytic drug, which does not cause sedative or depression of central nervous system. In the present work it is investigated using electron impact mass spectral (EI-MS) fragmentation at 70 eV, in comparison with thermal analyses (TA) measurements (TG/DTG and DTA) and molecular orbital calculation (MOC). Semi-empirical MO calculation, PM3 procedure, has been carried out on buspirone both as neutral molecule (in TA) and the corresponding positively charged species (in MS). The calculated MOC parameters include bond length, bond order, particle charge distribution on different atoms and heats of formation. The fragmentation pathways of buspirone in EI-MS lead to the formation of important primary and secondary fragment ions. The mechanism of formation of some important daughter ions can be illuminated from comparing with that obtained using electrospray ESIMS/MS mode mass spectrometer through the accurate mass measurement determination. The losses of the intermediate aliphatic part (CH2)4 due to cleavage of N-C bond from both sides is the primary cleavage in both techniques (MS and TA). The PM3 provides a base for fine distinction among sites of initial bond cleavage and subsequent fragmentation of drug molecule in both TA and MS techniques; consequently the choice of the correct pathway of such fragmentation knowing this structural session of bonds can be used to decide the active sites of this drug responsible for its chemical, biological and medical reactivity.  相似文献

Mesalazine (MZ) drug has been used for several decades as a primary treatment for inflammatory bowel diseases. The drug was investigated using thermal analysis (TA) measurements and electron impact mass spectral fragmentation at 70 and 15 eV of electron energy. The optimum molecular geometry and the total energy of the neutral and the positively charged MZ molecules were calculated by density functional theory method with 6-311++G(d,p) basis sets. Stability of the molecules arising from hyperconjugative interactions, charge delocalization, and the natural atomic charges has been analyzed using natural bond orbital analysis. In electron ionization mass spectrometry, the primary rupture is due to successive loss of H2O (OH from carboxyl and H from phenolic OH of the ring) and CO of the acetyl group. Thermogravimetric results have revealed two stages of mass loss at 75.3 and 25.3 % in ranges 225–350 and 350–650 °C, respectively. The first one may be due to successive losses of different groups or molecules with fast rate of decomposition. A comparison between MS and TA helped in selection the proper pathway representing the fragmentation mechanism of this drug.  相似文献

A planar differential mobility spectrometer (DMS) was coupled to a Mini 10 handheld rectilinear ion trap (RIT) mass spectrometer (MS) (total weight 10 kg), and the performance of the instrument was evaluated using illicit drug analysis. Coupling of DMS (which requires a continuous flow of drift gas) with a miniature MS (which operates best using sample introduction via a discontinuous atmospheric pressure interface, DAPI), was achieved with auxiliary pumping using a 5 L/min miniature diaphragm sample pump placed between the two devices. On-line ion mobility filtering showed to be advantageous in reducing the background chemical noise in the analysis of the psychotropic drug diazepam in urine using nanoelectrospray ionization. The combination of a miniature mass spectrometer with simple and rapid gas-phase ion separation by DMS allowed the characteristic fragmentation pattern of diazepam to be distinguished in a simple urine extract at lower limits of detection (50 ng/mL) than that achieved without DMS (200 ng/mL). The additional separation power of DMS facilitated the identification of two drugs of similar molecular weight, morphine (average MW = 285.34) and diazepam (average MW = 284.70), using a miniature mass spectrometer capable of unit resolution. The similarity in the proton affinities of these two compounds resulted in some cross-interference in the MS data due to facile ionization of the neutral form of the compound even when the ionic form had been separated by DMS.  相似文献

Gliclazide (GL, C15H21N3O3S) drug is used as non-insulin-dependant diabetes mellitus. The drug was investigated using thermal analysis (TA) measurements (TG/DTG) and electron impact mass spectral (EI–MS) fragmentation at 70 eV techniques. The mass spectra of GL at different values of ion source temperatures (400, 416, 425, and 440 K) are recorded and investigated. Semiempirical MO calculation, using PM3 procedure, has been carried out on neutral molecule and positively charged species. These calculations included bond length, bond order, bond strain, partial charge distribution, ionization energy, and heats of formation (ΔH f). PM3 procedure provides a basis for fine distinction among sites of initial bond cleavage, which is crucial to the rationalization of subsequent fragmentation of the molecule. The primary fragmentation pathway in both TA and MS (at different values of ion source temperature) is initiated by S–N bond rupture. TA and DTG show one main weight loss at 250.38 °C and four peaks at 271.6, 360.99, 427.93 and 479.17 °C in DTA, which may be attributed to various fragments. Also, the rate constant (K′) of thermal degradation has been tested isothermally at 210 and 600 °C. The calculated rate values are 9.6 × 10−3 and 0.33 × 10−3 s−1, respectively, and discussed. In MS, the effect of ion source temperature on mass spectral fragmentation processes is discussed on the basis of energy considerations using quasi equilibrium theory.  相似文献

Ibuprofen (C15H18O2) is an anti-inflammatory drug. It is important to investigate its structure to know the active groups and weak bond responsible for its medical activity. Consequently in the present study, ibuprofen was investigated by mass spectrometry (MS) and thermal analyses (TAs) (TG/DTG and DTA), and confirmed by semi-empirical molecular orbital (MO) calculation using PM3 procedure, on the neutral and positively charged forms of the drug. These calculations included bond order, bond length, and bond strain, and charge distribution, heat of formation, and ionization energy. The mass spectra and thermal analysis fragmentation pathways were proposed and compared to each other to select the most suitable scheme representing the correct fragmentation pathway of the drug in both techniques. From the electron ionization (EI) mass spectra, the primary cleavage site of the charged molecule is because of the rupture of COOH group (the lowest bond order) followed by propyl group loss. The TAs of the drug revealed high response of the drug to the temperature variation with very fast rate. It decomposed in several sequential steps in the temperature range 25–360 °C. The initial thermal decomposition is similar to that obtained by MS fragmentation of the first rupture (COOH), then subsequent one of propyl loss, and finally of ethylene loss. These mass losses appear as endothermic peaks required energy values of −214.83, −895.95, and −211.10 J g−1, respectively. The order of these losses is also related to the values of the MO calculation parameters. Therefore, the comparison between MS and TA helps in the selection of the proper pathway representing the decomposition of this drug to give its metabolites in in vivo system. This comparison is also successfully confirmed by MO calculations.  相似文献

Sertraline (C(17)H(17)Cl(2)N) as an antidepressant drug was investigated using thermal analysis (TA) measurements (TG/DTG and DTA) in comparison with electron impact (EI) mass spectral (MS) fragmentation at 70eV. Semi-empirical MO-calculations, using PM3 procedure, has been carried out on neutral molecule and positively charged species. These calculations included bond length, bond order, bond strain, partial charge distribution and heats of formation (DeltaH(f)). Also, in the present work sertraline-iodine product was prepared and its structure was investigated using elemental analyses, IR, (1)H NMR, (13)C NMR, MS and TA. It was also subjected to molecular orbital calculations (MOC) in order to confirm its fragmentation behavior by both MS and TA in comparison with the sertraline parent drug. In MS of sertraline the initial rupture occurred was CH(3)NH(2)(+) fragment ion via H-rearrangement while in sertraline-iodine product the initial rupture was due to the loss of I(+) and/or HI(+) fragment ions followed by CH(2)NH(+) fragment ion loss. In thermal analyses (TA) the initial rupture in sertraline is due to the loss of C(6)H(3)Cl(2) followed by the loss of CH(3)-NH forming tetraline molecule which thermally decomposed to give C(4)H(8), C(6)H(6) or the loss of H(2) forming naphthalene molecule which thermally sublimated. In sertraline-iodine product as a daughter the initial thermal rupture is due to successive loss of HI and CH(3)NH followed by the loss of C(6)H(5)HI and HCl. Sertraline biological activity increases with the introduction of iodine into its skeleton. The activities of the drug and its daughter are mainly depend upon their fragmentation to give their metabolites in vivo systems, which are very similar to the identified fragments in both MS and TA. The importance of the present work is also due to the decision of the possible mechanism of fragmentation of the drug and its daughter and its confirmation by MOC.  相似文献

For the detection of security-relevant substances at low concentrations in complex matrices, coupling of thermal desorption–single photon ionization–ion trap mass spectrometry (TD-SPI-ITMS) was successfully tested. The main advantage of taking solid samples with a wipe pad followed by thermal desorption is the low detection limit by enhanced vapor pressure. Single photon ionization is a soft ionization technique which reduces the target ion fragmentation and shields bulk components with high ionization energies (IE) like nitrogen yielding to clearly arranged mass spectra with significant high mass peaks. To obtain low false-positive and false-negative rates, especially necessary for security-relevant substances, the ion trap mass spectrometer allows identification of signals with MS/MS studies. In this concept, the soft ionization technique fits well with the MS/MS studies, as peaks with high masses are generated yielding significant MS/MS fragments. For the ionization, photon energies between about 8 eV (155 nm) and 12 eV (103 nm) were generated with electron-beam-pumped rare gas excimer lamps (EBEL). Depending on the rare gas used, light with different photon energy is generated, adapted to the substances of interest. So, even most narcotics, having relatively low IEs, can be ionized with 8.4 eV photons without massive fragmentation. For most explosives, photons with higher energy must be used as their IEs are higher. In this work, a mobile setup with a commercial ion trap mass spectrometer has been developed and tested. Even a first real-scenario measurement campaign was accomplished successfully proving the field-suitability of the system.  相似文献

Nonresonance excitation is a universal ion excitation and ejection method in which increased ion kinetic energy is achieved by the combination of an axial dc dipole and the rf trapping fields. The method does not require the applied excitation frequency to match with the secular frequency of the precursor ions to effect collision-induced dissociation (CID) for tandem mass spectrometry applications. Therefore, it is free of the effects of secular frequency changes caused by space-charge and simplifies the optimization of tandem mass spectrometry parameters when combined with gas chromatography-tandem mass spectrometry (GC-MS/MS). Computer simulations show that in contrast to the resonance excitation process, the nonresonance excitation process is able to accelerate thermal ions to kinetic energies in excess of 40 eV in a few microseconds. Based on simulations, we expect that the rapid deposition of energy by this method may allow the study, in ion traps, of high energy decomposition channels of precursor ions with multiple decomposition channels. Furthermore, the method is able to simultaneously excite multiple precursor ions, for example, excite both analyte and its coeluting isotopically labeled internal standard for GC-MS/MS analysis. A GC-MS/MS analysis of 100 pg of n-butylbenzene is demonstrated with a signal-to-noise ratio of 3624, which is over an order of magnitude higher than the signal-to-noise ratio of 345 obtained by full scan gas chromatography-mass spectrometry. In addition, the nonresonance excitation method can be used as a low pass mass filter in the chemical ionization (CI) mode to eject undesired fragment ions that result from direct electron ionization. This new CI method, selected ejection chemical ionization, can produce a CI spectrum without contamination of sample fragment ions from electron ionization.  相似文献

An important problem in mass isotopomer abundance mass spectrometry (MIAMS) is the dependence of measured mass isotopomer abundances on sample concentration. We have evaluated the role of ionization energy on mass isotopomer abundance ratios of methyl palmitate as a function of sample concentration. Ionization energy was varied using electron impact ionization (EI) and metastable atom bombardment (MAB). The latter generates a beam of metastable species capable of ionizing analyte molecules by Penning ionization. We observed that ionization of methyl palmitate by EI (70 eV) showed the greatest molecular ion fragmentation and also showed the greatest dependence of relative isotopomer abundance ratios on sample concentration. Ionization using the 3P2 and 3P0 states of metastable krypton (9.92 and 10.56 eV, respectively) resulted in almost no molecular ion fragmentation, and the isotopomer abundances quantified were essentially independent of sample concentration. Ionization using the 3P2 and 3P0 states of metastable argon (11.55 and 11.72 eV, respectively) showed molecular ion fragmentation intermediate between that of EI and MAB(Kr) and showed an isotopomer concentration dependence which was less severe than that observed with EI but more severe than that observed with MAB(Kr). The observed decrease in the dependence of isotopomer abundance on sample concentration with a decrease in molecular ion fragmentation is consistent with the hypothesis that proton transfer from a fragment cation to a neutral molecule is the gas phase reaction mechanism responsible for the concentration dependence. Alternative explanations, e.g., hydrogen abstraction from a neutral molecule to a molecular cation, is not supported by these results. Moreover, the MAB ionization technique shows potential for eliminating one source of error in MIAMS measurements of methyl palmitate, in particular, and of fatty acids methyl esters, in general.  相似文献

The fragmentation mechanisms of monolignols under various energetic processes are studied with jet-cooled thermal desorption molecular beam (TDMB) mass spectrometry (MS), 25 keV Bi(3)(+) secondary ion MS (SIMS), synchrotron vacuum-ultraviolet secondary neutral MS (VUV-SNMS) and theoretical methods. Experimental and calculated appearance energies of fragments observed in TDMB MS indicate that the coniferyl alcohol photoionization mass spectra contain the molecular parent and several dissociative photoionization products. Similar results obtained for sinapyl alcohol are also discussed briefly. Ionization energies of 7.60 eV ± 0.05 eV for coniferyl alcohol and <7.4 eV for both sinapyl and dihydrosinapyl alcohols are determined. The positive ion SIMS spectrum of coniferyl alcohol shares few characteristic peaks (m/z = 137 and 151) with the TDMB mass spectra, shows extensive fragmentation, and does not exhibit clear molecular parent signals. VUV-SNMS spectra, on the other hand, are dominated by the parent ion and main fragments also present in the TDMB spectra. Molecular fragmentation in VUV-SNMS spectra can be reduced by increasing the extraction delay time. Some features resembling the SIMS spectra are also observed in the desorbed neutral products. The monolignol VUV-SNMS peaks shared with the TDMB mass spectra suggest that dissociative photoionization of ion-sputtered neutral molecules predominate in the VUV-SNMS mass spectra, despite the extra internal energy imparted in the initial ion impact. The potential applications of these results to imaging MS of biomolecules are discussed.  相似文献

Polyurethanes are widely used in the manufacture of commercial products such as foams and paints. During combustion, these polymers can generate isocyanates, which induce adverse health effects. Polymer pyrolysis (Py) hyphenated with mass spectrometry (MS) allows the investigation of polymer thermal degradation over time/temperature. A diphenylmethanediisocyanate (MDI) polyurethane foam was analyzed with electron ionization (EI) and metastable atom bombardment (MAB) ionization at a pyrolysis temperature of 400 °C. The recently introduced MAB ionization source uses discrete energy stored in metastable atoms of gases to ionize the analytes. This characteristic allows modulation of the ionization energy by simply changing the ionization gas. The extensive fragmentation of molecular ions observed using EI 70 eV is not totally eliminated with EI 10 eV. However, only molecular ions are observed with MAB using N2 as the ionization gas. Temperature gradients were used to separate the products generated during the thermal degradation of a 1,6-hexamethylenediisocyanate (HDI) polyurethane paint. The analysis of mass spectra was facilitated owing to a selective desorption of pyrolysis products. Furthermore, changing the MAB ionization gas allows elucidation of the structure of the pyrolysis products by controlling the extent of their fragmentation. During these experiments, isocyanic acid, methyleneisocyanate, ethyleneisocyanate, propylisocyanate and butylisocyanate were detected.  相似文献

Ciprofloxacin (CPF, C17H18FN3O3) drug is used in the treatment of some bacterial infectious diseases. The drug was investigated using thermal analysis (TA) measurements (TG/DTG) and electron impact mass spectral (EI-MS) fragmentation at 70 eV techniques. Furthermore, the drug was characterized and investigated by other spectroscopic tools as IR, UV–Vis, 1H-, and 13C-NMR. The PM3 procedure provides a basis for fine distinction among sites of initial bond cleavage, which is crucial to the rationalization of subsequent fragmentation of the molecule. The mass spectra and thermal analysis fragmentation pathways were proposed and compared to each other to select the most suitable scheme representing the correct fragmentation of this drug. From EI-MS, the main primary cleavage site of the charged molecule is that due to C–COOH bond cleavage with H-rearrangement to skeleton and CO2 loss which can further decompose by piperazine loss. Thermal analysis of the neutral form of the drug reveals the high response of the drug to the temperature variation with very fast rate. Thermal decomposition has carried out in several sequential steps in the temperature range 40–650 °C. The initial thermal decomposition is similar to that obtained by mass spectrometric fragmentation (C–COOH fragment) but differ in that a rearrangement occurs by OH and CO loss. Therefore, comparison between MS and TA helps in selection the proper pathway representing the fragmentation of this drug. This comparison successfully confirmed by MO calculation. Finally, the effect of fluorine atom on the stability of the drug was discussed.  相似文献

The dissociation of field ionized doubly charged organic molecular ions into doubly charged fragment ions and neutral fragments is discussed. The kinetic energy released during the dissociation of the singly and doubly charged molecular ions rules out the possibility of a direct correlation between their mechanisms of formation. Further, the pressure as well as the temperature dependence revealed that the singly charged molecular ions are formed by direct ionization of the neutral molecules, while the doubly charged molecular ions are formed through a second ionization process of the adsorbed molecular ions on the field anode surface.  相似文献

Structure analyses of underivatized neutral lacto oligosaccharides are systematically performed by ultraviolet matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (UV-MALDI TOF MS) and UV-MALDI ion-trap time-of-flight mass spectrometry (ion-trap/TOF MS) acquired in negative-ion mode. Interestingly, their fragmentation significantly differ each other. In postsource decay (PSD) in UV-MALDI TOF MS, cross-ring cleavage at the reducing terminal predominates. On the other hand, glycosyl bond cleavage (C-type fragmentation) takes place preferentially in collision induced dissociation (CID) in UV-MALDI ion-trap/TOF MS. The cross-ring cleavage in PSD similar to that in in-source decay occurs via a prompt reaction path characteristic of the UV-MALDI process itself. The product ion spectra of UV-MALDI ion-trap/TOF MS are similar to the electrospray ionization (ESI) ion-trap or quadrupole/TOF CID product ion spectra. During ion-trap/TOF MS experiments, the deprotonated molecular ions survive for several tens of milliseconds after CID event because the high internal energy chlorinated precursor ions are cooled by collisional cooling in the ion trap. The results obtained suggest that the PSD from the chlorinated precursor ion in UV-MALDI TOF MS might proceed as a two-step reaction; in the first, a high internal energy deprotonated molecular ion is generated as a reaction intermediate during the flight in the drift tube, and in the second, the rapid decomposition from the deprotonated molecular ion takes place.  相似文献

Mechanistic arguments relative to matrix-assisted laser desorption/ionization (MALDI) mass spectrometry (MS) address observations that predominately singly charged ions are detected. However, recently a matrix assisted laser ablation method, laserspray ionization (LSI), was introduced that can use the same sample preparation and laser as MALDI, but produce highly charged ions from proteins. In MALDI, ions are generated from neutral molecules by the photon energy provided to a matrix, while in LSI ions are produced inside a heated inlet tube linking atmospheric pressure and the first vacuum region of the mass spectrometer. Some LSI matrices also produce highly charged ions with MALDI ion sources operated at intermediate pressure or high vacuum. The operational similarity of LSI to MALDI, and the large difference in charge states observed by these methods, provides information of fundamental importance to proposed ionization mechanisms for LSI and MALDI. Here, we present data suggesting that the prompt and delayed ionization reported for vacuum MALDI are both fast processes relative to producing highly charged ions by LSI. The energy supplied to produce these charged clusters/droplets as well as their size and time available for desolvation are determining factors in the charge states of the ions observed. Further, charged droplets/clusters may be a common link for ionization of nonvolatile compounds by a variety of MS ionization methods, including MALDI and LSI.  相似文献

2-(2'-octenyl)succinic acid has been identified in urine samples from children investigated for a possible inherited metabolic disease. Its structural identification has been achieved by gas chromatography/mass spectrometry using both electron ionization and chemical ionization and by tandem mass spectrometry (MS/MS) using fast-atom bombardment and high-resolution electron-ionization analyses of the molecular ion in a complex biological matrix. The localization of the double bond was obtained by interpretation of a unexpected rearrangement reaction occurring after dimethyl disulfide derivatization.  相似文献

The partial ionization cross section for the formation of SF(3) (+) fragment ions following electron impact on SF(6) is known to have a pronounced structure in the cross section curve slightly above 40 eV. We used the mass-analyzed ion kinetic energy (MIKE) scan technique to demonstrate the presence of a channel contributing to the SF(3) (+) partial ionization cross section that we attribute to the Coulomb explosion of doubly charged metastable SF(4) (2+) ions into two singly charged ions SF(3) (+) and F(+), with a threshold energy of about 45.5 eV. Thus the observed unusual shape of the SF(3) (+) partial ionization cross section is the result of two contributions, (i) the direct formation of SF(3) (+) fragment ions via dissociative ionization of SF(6) with a threshold energy of 22 eV and (ii) the Coulomb explosion of metastable SF(4) (2+) ions with a threshold energy of about 45.5 eV. A detailed analysis of the MIKE spectrum reveals an average kinetic energy release of about 5 eV in the Coulomb explosion of the SF(4) (2+) ions with evidence of a second channel corresponding to an average kinetic energy release of about 1.1 eV.  相似文献

Using a Nier-type electron impact ion source in combination with a double focusing two sector field mass spectrometer, partial cross sections for electron impact ionization of acetylene are measured for electron energies up to 1000 eV. Discrimination factors for ions are determined using the deflection field method in combination with a three-dimensional ion trajectory simulation of ions produced in the ion source. Analysis of the ion yield curves obtained by scanning the deflectors allows the assignment of ions with the same mass-to-charge ratio to specific production channels on the basis of their different kinetic energy distributions. This analysis also allows to determine, besides kinetic energy distributions of fragment ions, partial cross sections differential in kinetic energy. Moreover a charge separation reaction, the Coulomb explosion of the doubly charged parent ions C2H2++ into the fragment ions C2H+ and H+, is investigated and its mean kinetic energy release (KER=3.88 eV) is deduced.  相似文献
