A new method for identification and quantitative determination of the azole antifungal agents; ketoconazole, bifonazole, fluconazole and itraconazole is described using TLC with densitometric detection. Validation of the method was carried out to confirm its precision (%RSD ranged between 1.31 and 3.45), recovery (99.7–102.4%) and linearity (r  = 0.99287–0.99722) within the concentration range under investigation. The experimental conditions obtained enable the method to be used for both qualitative and quantitative pharmaceutical analysis.  相似文献

Abstract

The rapid microcomputer processing of quantitative TLC scanning data is described. The microcomputer, using BASIC language, is based on the Intel 8080A microprocessor IC and has 64K bytes of RAM. Data is acquired via a 16-bit integrating ADC. A TLC plate with up to 18 samples may be analyzed in as little as 5 minutes. Graphics capabilities include automatic analysis and depiction of peak heights and peak areas. Display of normal or log-normal plots of data for reference compounds and samples with least-squares linear regression is available. Graphics are directly accessible to permit independent plotting of data.  相似文献

Costunolide and dehydrocostus lactone are two guianolide sesquiterpene lactones present in abundance in Saussurea costus, a plant popularly known as `costus' and renowned throughout the world for its medicinal properties. A simple densitometric TLC method for simultaneous quantification of costunolide and dehydrocostus lactone, without derivatization, has been developed as an alternative to gas chromatography. The method enables rapid, sensitive, and reproducible simultaneous analysis of a variety of samples. The method was validated for precision, repeatability, and accuracy in accordance with ICH guidelines. Amounts of costunolide and dehydrocostus lactone in different samples of costus, estimated by use of this method, were in the range 0.46 to 1.00% and 0.81 to 1.21%, respectively. The proposed method is simple, precise, specific, sensitive, and accurate and can be used by the herbal drug industry for routine quality control of the crude drug costus.  相似文献

Eugenol and gallic acid are reported from the flower buds of Syzygium aromaticum (L.) Merr. & Perry (clove). Both the compounds have been shown to give interesting biological activities and hence serve as biomarkers. We report a simple TLC densitometric method for the quantification of eugenol and gallic acid in clove. The method was validated for precision, repeatability and accuracy. The method was found to be precise with RSD of 0.61 and 1.3 (intraday) and 0.96 and 0.24 (interday) for different concentrations of eugenol and gallic acid respectively. Instrumental precision was 0.24 and 0.21 (% CV) for eugenol and gallic acid respectively. Accuracy of the method was checked by conducting recovery study at two different levels for eugenol and gallic acid and the average percentage recoveries were found to be 99.79% and 97.90% respectively. The contents of eugenol and gallic acid in different samples of clove, as estimated by the proposed method, were found to be in the range of 12.9–14.6% and 0.31–0.61% respectively. The proposed HPTLC method for the estimation gallic acid and eugenol was found to be simple, precise, specific, sensitive and accurate and can be used for routine quality control of clove.  相似文献

建立了Qu ECh ERS/超高效液相色谱-四极杆/静电场轨道阱高分辨质谱(UPLC-Q-OrbitrapHRMS)同时检测水产及水产加工品中16种苯并咪唑类和13种硝基咪唑类药物残留的分析方法。样品经乙酸乙酯提取,浓缩近干,用30%(体积分数)甲醇-水溶液定容,乙腈饱和正己烷、氨基(NH2)吸附剂和Mg SO4净化。采用Thermo fisher Accucore RP-MS色谱柱(100 mm×2.1 mm,2.6μm),以0.1%(体积分数)甲酸-乙腈为流动相进行梯度洗脱,使用可加热电喷雾离子源(HESI),全扫描/数据依赖二级扫描(Full MS/dd-MS2,Top N=1)模式检测,外标法定量。结果表明,29种药物在0.5~400μg/L范围内线性关系良好(r2≥0.9967),在5种水产及水产加工品基质中进行3个不同水平的加标实验,平均回收率为60.2%~123.3%,相对标准偏差(n=6)为0.1%~21.7%,方法的定量下限为0.5~5.0μg/kg。该法操作简便、精密度高、准确性好,适用于批量水产及水产加工品中29种药物的快速检测。  相似文献

This paper describes a novel TLC densitometric method for the determination of solasodine in various Solanum species (Family Solanaceae). Solasodine does not give absorption in UV range. Hence, ion pair complex of solasodine and acid dye was spotted on TLC plates. A developing solvent with an organic acid ensured in situ color development of the complex which could be successfully quantified at 461 nm. Linearity was found to be in the range of 79.2–495 ng spot−1 with correlation coefficient of 0.995. This method is reproducible, specific, eliminates post derivatization steps and the problem of background interference. The method was completely validated and applied to determine solasodine content in various herb samples, herb extract and their formulations. Accuracy of the method was found to be 98.54 ± 2.8%. No matrix interference was observed.  相似文献
