Ultrasound-targeted microbubble destruction (UTMD) technique can be potentially used for non-viral delivery of gene therapy. Targeting wild-type p53 (wtp53) tumor suppressor gene may provide a clinically promising treatment for patients with ovarian cancer. However, UTMD mediated gene therapy typically uses non-targeted microbubbles with suboptimal gene transfection efficiency. We synthesized a targeted microbubble agent for UTMD mediated wtp53 gene therapy in ovarian cancer cells. Lipid microbubbles were conjugated with a Luteinizing Hormone–Releasing Hormone analog (LHRHa) via an avidin–biotin linkage to target the ovarian cancer A2780/DDP cells that express LHRH receptors. The microbubbles were mixed with the pEGFP-N1-wtp53 plasmid. Upon exposure to 1 MHz pulsed ultrasound beam (0.5 W/cm2) for 30 s, the wtp53 gene was transfected to the ovarian cancer cells. The transfection efficiency was (43.90 ± 6.19)%. The expression of wtp53 mRNA after transfection was (97.08 ± 12.18)%. The cell apoptosis rate after gene therapy was (39.67 ± 5.95)%. In comparison with the other treatment groups, ultrasound mediation of targeted microbubbles yielded higher transfection efficiency and higher cell apoptosis rate (p < 0.05). Our experiment verifies the hypothesis that ultrasound mediation of targeted microbubbles will enhance the gene transfection efficiency in ovarian cancer cells.  相似文献

In this study, the synthesis of Ce0.8Sm0.2O1.9 (SDC) solid electrolyte by the ultrasound assisted co-precipitation method was accomplished to explore the effects of ultrasound power, ultrasound pulse ratio and probe type upon the ionic conductivity of SDC as well as the lattice parameter, the microstructure and the density. Fine powders of uniform crystallite sizes (average 11.70 ± 0.62 nm) were obtained, needing lower sintering temperature. The SDC powders were successfully sintered to a relative density of over 95% at 1200 °C (5 °C min?1) for 6 h. The micrograph of SDC pellets showed non-agglomerated and well-developed grains with average size of about 200 nm. X-ray diffraction analysis showed that the lattice parameter increased with increasing acoustic intensity and reached a maximum for the 14.94 W cm?2. Further, a linear relationship was detected between the lattice parameter and the ionic conductivity, inspiring a dopant like effect of US on the electrolyte properties. The highest ionic conductivity as σ800°C = 3.07 × 10?2 S cm?1 with an activation energy Ea = 0.871 kJ mol?1 was obtained with pulsed ultrasound for an acoustic intensity of 14.94 W cm?2, using 19 mm probe and 8:2 pulse ratio.  相似文献

Suspension of nanocrystalline cellulose (NCC) produced from bleached cotton by controlled sulphuric acid hydrolysis was treated with low frequency ultrasound at 20 kHz and 60% amplitude for 0, 1, 2, 5 and 10 min and the effects of sonication on the properties of both the cellulose nanocrystals and their aqueous suspensions were investigated. Furthermore, a series of nanocellulose films were manufactured from the suspensions that were sonicated for different periods of time and tested. Laser diffraction analysis and transmission electron microscopy proved that sonication not only disintegrated the large NCC aggregates (Dv50 14.7 μm) to individual nanowhiskers with an average length and width of 171 ± 57 and 17 ± 4 nm, respectively, but also degraded the nanocrystals and yielded shorter and thinner particles (118 ± 45 and 13 ± 3 nm, respectively) at 10-min sonication. The ultrasound-assisted disintegration to nano-sized cellulose whiskers decreased the optical haze of suspensions from 98.4% to 52.8% with increasing time from 0 to 10 min, respectively. Sonication of the suspensions significantly contributed to the preparation of films with low haze (high transparency) and excellent tensile properties. With the increasing duration of sonication, the haze decreased and the tensile strength rose gradually. Irrespectively of sonication, however, all films had an outstanding oxygen transmission rate in a range of 5.5–6.9 cm3/m2 day, and a poor thermal stability.  相似文献

PurposeTo investigate biexponential apparent diffusion parameters of prostate central gland (CG) cancer, stromal hyperplasia (SH), and glandular hyperplasia (GH) and compare with monoexponential apparent diffusion coefficient (ADC) value for discriminating prostate cancer from benign hyperplasia.Materials and MethodsTwenty-one CG cancer foci, 23 SH and 26 GH nodules in the CG were analyzed in 39 patients (19 with CG cancer, 20 with peripheral zone cancer but no CG cancer) who underwent preoperative conventional DWI (b-value 0, 1000 s/mm2) and a 10 b-value (range 0 to 3000 s/mm2) DWI. All of the cancer and hyperplastic foci on MR images were localized on the basis of histopathologic correlation. The ADC value of the monoexponential DWI, and the fast apparent diffusion coefficient (ADCf), slow apparent diffusion coefficient (ADCs) value and the fraction of ADCf (f) of the biexponential DWI were calculated for all of the lesions. Receiver operating characteristic (ROC) analysis was performed for the differentiation of CG cancer from SH and GH.ResultsThe ADC values (× 10? 3 mm2/s) were 0.87 ± 0.11, 1.06 ± 0.15, and 1.61 ± 0.27 in CG cancer, SH and GH foci, respectively, and differed significantly, yielding areas under the ROC curve (AUCs) of 1.00 and 0.80 for the differentiation of carcinoma from GH and SH, respectively. The ADCf (× 10? 3 mm2/s), ADCs (× 10? 3 mm2/s) and f for cancer were 1.92 ± 0.38, 0.53 ± 0.17, and 47.7 ± 6.1%, respectively, which were lower than the same values for GH (3.43 ± 0.65, 1.12 ± 0.21, 61.1 ± 8.7%) (all p < 0.01). The ADCf and ADCs for cancer were also lower than those for SH (3.11 ± 0.30, 0.79 ± 0.21) (all p < 0.01). The ADCf yielded AUCs (1.00, p > 0.01) that were comparable to those from ADC for the differentiation of cancer from GH, while ADCf yielded higher AUCs (0.92) compared with ADC (p < 0.01) for the differentiation of cancer from SH. ADCs and f revealed AUCs of 0.97 and 0.90, respectively, for the differentiation of cancer from GH, and the ADCs offered relatively lower AUCs (0.68) for differentiating cancer from SH.ConclusionBiexponential DWI could potentially improve the differentiation of prostate cancer in CG, and the ADCf of the biexponential model offers better accuracy than ADC.  相似文献

Bacterial cellulose (BC) film formation could be a critical issue in nanotechnology applications such as biomedical or smart materials products. In this research, purified pretreated BC was subjected to high intensity ultrasound (HIUS) and was investigated for the development of BC films. The morphological, structural and thermal properties of the obtained films were studied by using FE-SEM, AFM, FT-IR, XRD, TGA and DSC characterizations. Results showed that the most favorable purification treatment was the 0.01 M NaOH at 70 °C for 2 h under continuous stirring. The most suitable ultrasound operating conditions were found to be, 1 cm distance of ultrasonic probe from the bottom of the beaker, submerged in cold water bath cooling around 12 ± 2 °C. The power (25 W/cm2), time (30 min), BC concentration (0.1% w/w), amplitude (20 μm) and frequency (20 kHz) were maintained constant.  相似文献

This study presents the ultrasound assisted pretreatment of sugarcane bagasse (SCB) using metal salt with hydrogen peroxide for bioethanol production. Among the different metal salts used, maximum holocellulose recovery and delignification were achieved with ultrasound assisted titanium dioxide (TiO2) pretreatment (UATP) system. At optimum conditions (1% H2O2, 4 g SCB dosage, 60 min sonication time, 2:100 M ratio of metal salt and H2O2, 75 °C, 50% ultrasound amplitude and 70% ultrasound duty cycle), 94.98 ± 1.11% holocellulose recovery and 78.72 ± 0.86% delignification were observed. The pretreated SCB was subjected to dilute acid hydrolysis using 0.25% H2SO4 and maximum xylose, glucose and arabinose concentration obtained were 10.94 ± 0.35 g/L, 14.86 ± 0.12 g/L and 2.52 ± 0.27 g/L, respectively. The inhibitors production was found to be very less (0.93 ± 0.11 g/L furfural and 0.76 ± 0.62 g/L acetic acid) and the maximum theoretical yield of glucose and hemicellulose conversion attained were 85.8% and 77%, respectively. The fermentation was carried out using Saccharomyces cerevisiae and at the end of 72 h, 0.468 g bioethanol/g holocellulose was achieved. Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD) analysis of pretreated SCB was made and its morphology was studied using scanning electron microscopy (SEM). The compounds formed during the pretreatment were identified using gas chromatography–mass spectrometry (GC–MS) analysis.  相似文献

In this work, the pulsed electron beam deposition method (PED) is evaluated by studying the properties of ZnO thin films grown on c-cut sapphire substrates. The film composition, structure and surface morphology were investigated by means of Rutherford backscattering spectrometry, X-ray diffraction and atomic force microscopy. Optical absorption, resistivity and Hall effect measurements were performed in order to obtain the optical and electronic properties of the ZnO films. By a fine tuning of the deposition conditions, smooth, dense, stoichiometric and textured hexagonal ZnO films were epitaxially grown on (0001) sapphire at 700 °C with a 30° rotation of the ZnO basal plane with respect to the sapphire substrate. The average transmittance of the films reaches 90% in the visible range with an optical band gap of 3.28 eV. Electrical characterization reveals a high density of charge carrier of 3.4 × 1019 cm?3 along with a mobility of 11.53 cm²/Vs. The electrical and optical properties are discussed and compared to ZnO thin films prepared by the similar and most well-known pulsed laser deposition method.  相似文献

The evaluation of local muscle recruitment during a specific movement can be done indirectly by measuring changes in local blood flow. Intravoxel incoherent motion perfusion imaging exploits some properties of the magnetic resonance to measure locally microvascular perfusion, and seems ideally suited for this task. We studied the selectivity of the increase in intravoxel incoherent motion blood flow related parameter fD* in the muscles of 24 shoulders after two physical exam maneuvers, Jobe and Lift-off test (test order reversed in half of the volunteers) each held 2 min against resistance. After a lift-off, IVIM blood flow-related fD* was increased in the subscapularis (in 10−3 mm2 s−1, 3.24 ± 0.86 vs. rest 1.37 ± 0.58, p < 0.001) and the posterior bundle of deltoid (2.62 ± 1.34 vs. rest 0.77 ± 0.32, p < 0.001). Those increases were selective when compared with other rotator cuff muscles and deltoid bundles respectively. After a Jobe test, increase in fD* was scattered within the rotator cuff muscles, but was selective for the lateral deltoid compared to the other deltoid bundles (anterior, p < 0.001; posterior, p < 0.05). Those results were similar when the testing order was reversed. In conclusion, this study demonstrated a selective increase in local microvascular perfusion after specific muscle testing of the shoulder muscles with IVIM. This technique has the potential to non-invasively characterize perfusion-related musculoskeletal physiological as well as pathological processes.  相似文献

In this study, the effects of ultrasound with different ultrasonic frequencies on the properties of sodium alginate (ALG) were investigated, which were characterized by the means of the multi-angle laser light scattering photometer analysis (GPC-MALLS), rheological analysis, circular dichroism (CD) spectrometer and scanning electron microscope (SEM). It showed that the molecular weight (Mw) and molecular number (Mn) of the untreated ALG was 1.927 × 105 g/mol and 4.852 × 104 g/mol, respectively. The Mw of the ultrasound treated ALG was gradually increased from 3.50 × 104 g/mol to 7.34 × 104 g/mol while the Mn of ALG was increased and then decreased with the increase of the ultrasonic frequency. The maximum value of Mn was 9.988 × 104 g/mol when the ALG was treated by ultrasound at 40 kHz. It indicated that ultrasound could induce ALG degradation and rearrangement. The number of the large molecules and small molecules of ALG was changed by ultrasound. The value of dn/dc suggested that the ultrasound could enhance the stability of ALG. Furthermore, it was found that ALG treated by ultrasound at 50 kHz tended to be closer to a Newtonian behavior, while the untreated and treated ALG solutions exhibited pseudoplastic behaviours. Moreover, CD spectra demonstrated that ultrasound could be used to improve the strength of the gel by changing the ratio of M/G, which showed that the minimum ratio of M/G of ALG treated at 135 kHz was 1.34. The gel-forming capacity of ALG was correlated with the content of G-blocks. It suggested that ALG treated by ultrasound at 135 kHz was stiffer in the process of forming gels. The morphology results indicated that ultrasound treatment of ALG at 135 kHz increased its hydrophobic interaction and interfacial activity. This study is important to explore the effect of ultrasound on ALG in improving the physical properties of ALG as food additives, enzyme and drug carriers.  相似文献

Pulsed ultrasound was used to disperse a biphasic mixture of CO2/H2O in a 1 dm3 high-pressure reactor at 30 °C/80 bar. A view cell positioned in-line with the sonic vessel allowed observation of a turbid emulsion which lasted approximately 30 min after ceasing sonication. Within the ultrasound reactor, simultaneous CO2-continuous and H2O-continuous environments were identified. The hydrolysis of benzoyl chloride was employed to show that at similar power intensities, comparable initial rates (1.6 ± 0.3 × 10–3 s–1 at 95 W cm–2) were obtained with those reported for a 87 cm3 reactor (1.8 ± 0.2 × 10–3 s–1 at 105 W cm–2), demonstrating the conservation of the physical effects of ultrasound in high-pressure systems (emulsification induced by the action of acoustic forces near an interface). A comparison of benzoyl chloride hydrolysis rates and benzaldehyde mass transport relative to the non-sonicated, ‘silent’ cases confirmed that the application of ultrasound achieved reaction rates which were over 200 times faster, by reducing the mass transport resistance between CO2 and H2O. The versatility of the system was further demonstrated by ultrasound-induced hydrolysis in the presence of the polysorbate surfactant, Tween, which formed a more uniform CO2/H2O emulsion that significantly increased benzoyl chloride hydrolysis rates. Finally, pulse rate was employed as a means of slowing down the rate of hydrolysis, further illustrating how ultrasound can be used as a valuable tool for controlling reactions in CO2/H2O solvent mixtures.  相似文献

We synthesized oxygen and paclitaxel (PTX) loaded lipid microbubbles (OPLMBs) for ultrasound mediated combination therapy in hypoxic ovarian cancer cells. Our experiments successfully demonstrated that ultrasound induced OPLMBs destruction significantly enhanced the local oxygen release. We also demonstrated that OPLMBs in combination with ultrasound (300 kHz, 0.5 W/cm2, 15 s) yielded anti-proliferative activities of 52.8 ± 2.75% and cell apoptosis ratio of 35.25 ± 0.17% in hypoxic cells at 24 h after the treatment, superior to other treatment groups such as PTX only and PTX-loaded MBs (PLMBs) with or without ultrasound mediation. RT-PCR and Western blot tests further confirmed the reduced expression of HIF-1α and MDR-1/P-gp after ultrasound mediation of OPLMBs. Our experiment suggests that ultrasound mediation of oxygen and drug-loaded MBs may be a useful method to overcome chemoresistance in the hypoxic ovarian cancer cells.  相似文献

Power ultrasound, which can enhance nucleation rate and crystal growth rate, can also affect the physico-chemical properties of immersion frozen products. In this study, the influence of slow freezing (SF), immersion freezing (IF) and ultrasound-assisted freezing (UAF) on physico-chemical properties and volatile compounds of red radish was investigated. Results showed that ultrasound application significantly improved the freezing rate; the freezing time of ultrasound application at 0.26 W/cm2 was shorten by 14% and 90%, compared to IF and SF, respectively. UAF products showed significant (p < 0.05) reduction in drip loss and phytonutrients (anthocyanins, vitamin C and phenolics) loss. Compared to SF products, IF and UAF products showed better textural preservation and higher calcium content. The radish tissues exhibited better cellular structures under ultrasonic power intensities of 0.17 and 0.26 W/cm2 with less cell separation and disruption. Volatile compound data revealed that radish aromatic profile was also affected in the freezing process.  相似文献

A new coupling of ultrasound device with membrane process has been developed in order to enhance cross-flow ultrafiltration of colloidal suspensions usually involved in several industrial applications included bio and agro industries, water and sludge treatment. In order to reduce mass transfer resistances induced by fouling and concentration polarization, which both are main limitations in membrane separation process continuous ultrasound is applied with the help of a vibrating blade (20 kHz) located in the feed channel all over the membrane surface (8 mm between membrane surface and the blade). Hydrodynamic aspects were also taking into account by the control of the rectangular geometry of the feed channel.Three colloidal suspensions with different kinds of colloidal interaction (attractive, repulsive) were chosen to evaluate the effect of their physico-chemical properties on the filtration. For a 90 W power (20.5 W cm−2) and a continuous flow rate, permeation fluxes are increased for each studied colloidal suspension, without damaging the membrane. The results show that the flux increase depends on the initial structural properties of filtered dispersion in terms of colloidal interaction and spatial organizations. For instance, a Montmorillonite Wyoming–Na clay suspension was filtered at 1.5 × 105 Pa transmembrane pressure. Its permeation flux is increased by a factor 7.1, from 13.6 L m−2 h−1 without ultrasound to 97 L m−2 h−1 with ultrasound.  相似文献

This study investigated the mechanical bioeffects exerted by acoustic droplet vaporization (ADV) under different experimental conditions using vessel phantoms with a 200-μm inner diameter but different stiffness for imitating the microvasculature in various tumors. High-speed microscopy, passive cavitation detection, and ultrasound attenuation measurement were conducted to determine the morphological characteristics of vascular damage and clarify the mechanisms by which the damage was initiated and developed. The results show that phantom erosion was initiated under successive ultrasound exposure (2 MHz, 3 cycles) at above 8-MPa peak negative pressures (PNPs) when ADV occurred with inertial cavitation (IC), producing lesions whose morphological characteristics were dependent on the amount of vaporized droplets. Slight injury occurred at droplet concentrations below (2.6 ± 0.2) × 106 droplets/mL, forming shallow and rugged surfaces on both sides of the vessel walls. Increasing the droplet concentration to up to (2.6 ± 0.2) × 107 droplets/mL gradually suppressed the damage on the distal wall, and turned the rugged surface on the proximal wall into tunnels rapidly elongating in the direction opposite to ultrasound propagation. Increasing the PNP did not increase the maximum tunnel depth after the ADV efficiency reached a plateau (about 71.6 ± 2.7% at 10 MPa). Increasing the pulse duration effectively increased the maximum tunnel depth to more than 10 times the diameter of the vessel even though there was no marked enhancement in IC dose. It can be inferred that substantial bubble generation in single ADV events may simultaneously distort the acoustic pressure distribution. The backward ultrasound reinforcement and forward ultrasound shielding relative to the direction of wave propagation augment the propensity of backward erosion. The results of the present work provide information that is valuable for the prevention or utilization of ADV-mediated mechanical bioeffects in clinical applications.  相似文献

Treatment and management of food processing waste is a major challenge for food industry. Potato processing industry generates tremendous amount of peel and consider it as zero valued waste. Again, pomace generated after juice extraction from sweet lime pulp is considered as waste and not properly utilized. Whereas these waste could be utilized for the development of biodegradable packaging film to overcome environmental issues. Composite films were prepared with varying proportion of potato peel powder (PP) and sweet lime pomace (SLP) in the ratio of 0:1(A), 0.5:1(B), 1:1(C), 1:0.5(D), 1:0(E) with an ultrasound treatment of 45 min, and 0:1(F), 0.5:1(G), 1:1(H), 1:0.5(I), 1:0(J) with an ultrasound treatment of 60 min. Ultrasound was applied for 45 and 60 min to film forming solutions to break down biopolymer particles small enough to form a film. All the films were analyzed for their barrier and mechanical properties. It was observed that increasing ultrasound treatment times gives better result in film properties and less PP content also gives better film properties, from these observations film G prepared with 0.5:1 (PP:SLP) showed better characteristics among all other films. Water vapor permeability, moisture absorption, water solubility, breakage strength and elongation capacity of G film were reported as 7.25 × 10−9 g/Pa h m, 12.88 ± 0.348%, 38.92 ± 0.702%, 242.01 ± 3.074 g and 7.61 ± 0.824 mm respectively. However, thermal decomposition for film G took place above 200 °C. The film forming solution of selected G film, added with clove essential oil (1.5%) as an antimicrobial agent was wrapped on bread and stored it for 5 days. The film was successful in lowering the weight loss, reducing the hardness and inhibition of surface microbial load from bread sample.  相似文献

The thin-layer drying behavior of the municipal sewage sludge in a laboratory-scale hot air forced convective dryer assisted with air-borne ultrasound was investigated in between 70 and 130 °C hot air temperatures. The drying kinetics in the convective process alone were compared to that for ultrasound-assist process at three ultrasound powers (30, 90, 150 W). The average drying rates within whole drying temperature range at ultrasound powers of 30, 90 and 150 W increased by about 22.6%, 27.8% and 32.2% compared with the convective drying alone (without ultrasound). As the temperature increasing from 70 °C to 130 °C, there were maximum increasing ratios for the effective moisture diffusivities of the sewage sludge in both falling rate periods at ultrasonic power of 30 W in comparison with other two high powers. In between the ultrasound powers of 0 and 30 W, the effect of the power on the drying rate was significant, while its effect was not obvious over 30 W. Therefore, the low ultrasonic power can be just set in the drying process. The values of the apparent activation energy in the first falling rate period were down from 13.52 to 12.78 kJ mol−1, and from 17.21 to 15.10 kJ mol−1 for the second falling rate period with increasing the ultrasonic power from 30 to 150 W. The values of the apparent activation energy in two falling rate periods with the ultrasound-assist were less than that for the hot air convective drying alone.  相似文献

This study aimed at verifying the effects of phonophoresis associated with Arnica montana on the acute phase of an inflammatory muscle lesion. Forty Wistar male rats (300 ± 50 g), of which the Tibialis Anterior muscle was surgically lesioned, were divided into four groups (n = 10 each): control group received no treatment; the ultrasound group (US) was treated in pulsed mode with 1-MHz frequency, 0.5 W/cm2 intensity (spatial and temporal average – SATA), duty cycle of 1:2 (2 ms on, 4 ms off, 50%), time of application 3 min per session, one session per day, for 3 days; the phonophoresis or ultrasound plus arnica (US+A) group was treated with arnica with the same US parameters plus arnica gel; and the arnica group (A) was submitted to massage with arnica gel, also for 3 min, once a day, for 3 days. Treatment started 24 h after the surgical lesion. On the 4th day after lesion creation, animals were sacrificed and sections of the lesioned, inflamed muscle were removed for quantitative (mononuclear and polymorphonuclear cell count) and qualitative histological analysis. Collected data from the 4 groups were statistically analyzed and the significance level set at p < 0.05. Results show higher mononuclear cell density in all three treated groups with no significant difference between them, but values were significantly different (p < 0.0001) when compared to control group’s. As to polymorphonuclear cell density, significant differences were found between control group (p = 0.0134) and US, US+A and A groups; the arnica group presented lesser density of polymorphonuclear cells when compared (p = 0.0134) to the other groups. No significant difference was found between US and US+A groups. While the massage with arnica gel proved to be an effective anti-inflammatory on acute muscle lesion in topic use, these results point to ineffectiveness of Arnica montana phonophoresis, US having seemingly checked or minimized its anti-inflammatory effect.  相似文献

Auger electron spectroscopy (AES) and scanning tunnelling microscopy (STM) have been used to investigate the growth behaviour of ultra-thin Ag films on a Mo(110) surface at room temperature. An analysis of AES and STM measurements indicates that three-dimensional (3D) growth of a Ag film is observed. For submonolayer coverage, the growth of Ag is mediated by a two-dimensional step-flow mechanism. During the initial stage of this growth, the first Ag layer nucleates and creates islands (average size of islands is about 180 ± 20 nm2) at Mo step edges. In the monolayer coverage range, the decoration of substrate steps by Ag can be distinguished by the presence of a fractional step of p1 = 0.86 ± 0.6 Å height at the Ag–Mo boundary. As the sample is post-annealed to 700 K, the morphology of the surface changes. Step-flow growth in this case gives rise to a regular Ag nanostripe network attached to Mo(110) step edges. The corrugation profiles reveal the protrusion of silver nanostripes of thicknesses p1 = 0.98 ± 0.16 Å and p2 = 0.39 ± 0.06 Å for submonolayer and monolayer coverage ranges, respectively, above each single step of a Mo terrace morphology.  相似文献

Background and purposeTo assess the sensitivity of non-localized, whole-head 1H-MRS to an individual's serial changes in total-brain NAA, Glx, Cr and Cho concentrations — metabolite metrics often used as surrogate markers in neurological pathologies.Materials and methodsIn this prospective study, four back-to-back (single imaging session) and three serial (successive sessions) non-localizing, ~3 min 1H-MRS (TE/TR/TI = 5/104/940 ms) scans were performed on 18 healthy young volunteers: 9 women, 9 men: 29.9 ± 7.6 [mean ± standard deviation (SD)] years old. These were analyzed by calculating a within-subject coefficient of variation (CV = SD/mean) to assess intra- and inter-scan repeatability and prediction intervals. This study was Health Insurance Portability and Accountability Act compliant. All subjects gave institutional review board-approved written, informed consent.ResultsThe intra-scan CVs for the NAA, Glx, Cr and Cho were: 3.9 ± 1.8%, 7.3 ± 4.6%, 4.0 ± 3.4% and 2.5 ± 1.6%, and the corresponding inter-scan (longitudinal) values were: 7.0 ± 3.1%, 10.6 ± 5.6%, 7.6 ± 3.5% and 7.0 ± 3.9%. This method is shown to have 80% power to detect changes of 14%, 27%, 26% and 19% between two serial measurements in a given individual.ConclusionsSubject to the assumption that in neurological disorders NAA, Glx, Cr and Cho changes represent brain-only pathology and not muscles, bone marrow, adipose tissue or epithelial cells, this approach enables us to quantify them, thereby adding specificity to the assessment of the total disease load. This will facilitate monitoring diffuse pathologies with faster measurement, more extensive (~90% of the brain) spatial coverage and sensitivity than localized 1H-MRS.  相似文献

Phase transformations in squaric acid (H2C4O4) have been investigated by thermogravimetry and differential scanning calorimetry with different heating rates β. The mass loss in TG apparently begins at onset temperatures Tdi=245±5 °C (β=5 °C min?1), 262±5 °C (β=10 °C min?1), and 275±5 °C (β=20 °C min?1). A polymorphic phase transition was recognized as a weak endothermic peak in DSC around 101 °C (Tc+). Further heating with β=10 °C min?1 in DSC revealed deviation of the baseline around 310 °C (Ti), and a large unusual exothermic peak around 355 °C (Tp), which are interpreted as an onset and a peak temperature of thermal decomposition, respectively. The activation energy of the thermal decomposition was obtained by employing relevant models. Thermal decomposition was recognized as a carbonization process, resulting in amorphous carbon.  相似文献
