Quality consistency of Glycyrrhiza formula granules is essential for guaranteeing clinical efficacy. However, a suitable method to accurately and conveniently evaluate the consistency of the clinical efficacy of Glycyrrhiza formula granules is currently not available. This study established a method for the simultaneous determination of 12 active components in Glycyrrhiza formula granules using ultra-high-performance liquid chromatography coupled with quadrupole-time-of-flight tandem mass spectrometry. The rate of inhibition of cyclooxygenase-2 by different batches of Glycyrrhiza formula granules was determined. Near-infrared spectra were collected for different batches of Glycyrrhiza formula granules to detect their biological activity in the inhibition of cyclooxygenase-2. The quality consistency of the 11 batches of Glycyrrhiza formula granules was evaluated using principal component and correlation analyses. The results showed significant differences in the formula granules of Glycyrrhiza uralensis produced by the different manufacturers. Some differences were also observed among batches of formula granules produced by the same manufacturer. Correlation analysis of the chemical components and cyclooxygenase-2 activity showed that glycyrrhizic acid, liquiritin, and isoliquiritin were the main active components of Glycyrrhiza. Correlation analysis of the near-infrared spectra and cyclooxygenase-2 inhibition activity showed a high correlation between the active components and three characteristic bands: 3383–3995, 4227–4651, and 5315–5878 cm−1. In this study, the main active anti-inflammatory components of Glycyrrhiza granules were screened. Thus, the near-infrared spectrum and characteristic active band of multi-index active components can be used to quickly detect the quality consistency of Glycyrrhiza formula granules, thereby improving the ability to control the quality and consistency of these granules.  相似文献

Rational design of enzymes is a stringent test of our understanding of protein structure and function relationship, which also has numerous potential applications. We present a novel method for enzyme design that can find good candidate protein scaffolds in a protein-ligand database based on vector matching of key residues. Residues in the vicinity of the active site were also compared according to a similarity score between the scaffold protein and the target enzyme. Suitable scaffold proteins were selected, and the side chains of residues around the active sites were rebuilt using a previously developed side-chain packing program. Triose phosphate isomerase (TIM) was used as a validation test for enzyme design. Selected scaffold proteins were found to accommodate the enzyme active sites and successfully form a good transition state complex. This method overcomes the limitations of the current enzyme design methods that use limited number of protein scaffold and based on the position of ligands. As there are a large number of protein scaffolds available in the Protein Data Band, this method should be widely applicable for various types of enzyme design.  相似文献

A novel spectrofluorimetric method for the determination of peroxynitrite is proposed. The method is based on a mimetic enzyme catalyzed reaction with hemoglobin as the catalyst and l-tyrosine as the substrate. A new fluorescent substance is produced that might probably be the coupled dimmer of tyrosine, which, instead of nitryl-tyrosine, is likely to be a new marking substance of ONOO injury in vivo. Kinetics of the reaction is studied and the possible reaction mechanism is also recommended. The proposed method is simple and highly sensitive with a detection limit of 5.00 × 10−8 mol L−1 of peroxynitrite. A liner calibration graph is obtained over the peroxynitrite concentration range 5.60 × 10−7 to 2.10 × 10−5 mol L−1, with a correlation coefficient of 0.9983. Interferences from some amino acids and metal ions normally seen in biological samples, and also some anions structurally similar to ONOO are studied.  相似文献

A new type of atomic absorption spectrometry using the Zeeman effect of sample materials is proposed. A magnetic field was applied to the sample vapor in the direction perpendicular to the propagation of light emitted from an atomic spectral source. Absorption of radiation polarized perpendicular and parallel to the magnetic field was observed alternatively. The absorption difference was proportional to the true atomic absorption, and was not interfered with by any other molecular absorption and light scattering, i.e., background absorption. The background absorption could be monitored at exactly the same wavelength as an atomic absorption line. Suitable magnetic field strength was found for each line of the various elements.  相似文献

Aconitine hydrolysis is deemed to be the guarantee for the safe application of Aconitum phytomedicine. Studies have suggested that hydrolysates of aconitine not only include benzoylaconitine and aconine, but other hydrolysates. Moreover, these hydrolysates maybe have a mutual transformation relationship, which has not been confirmed. Herein, hydrolysates of aconitine and their mutual transformation relationship were studied by the theoretical quantum chemistry, UPLC-Q-TOF-MS, the separation and identification of target products, etc. Then the toxicity of its hydrolysates was evaluated. The results demonstrate that the probability is the same for aconitine hydrolysis to pyroaconitine and benzoylaconitine, but they are difficult to convert to each other. Aconitine hydrolysis has three independent hydrolysis pathways, 1) to indaconitine, 2) to benzoylaconitine, and aconine, 3) to pyroaconitine and to 16-epi-pyroaconine. The result of embryotoxicity evaluation on zebrafish was aconitine > indaconitine > benzoylaconitine > α-pyroaconitine > β- pyroaconitine > aconine > 16-epi-pyroaconine. In conclusion, aconitine have three independent hydrolysis pathways and the hydrolysates of different pathways cannot be transformed into each other. Pyroaconitine is a hydrolysate of aconitine except for benzoylaconitine, and its toxicity is lower than benzoylaconitine. More importantly, it clarifies the long-standing debate and provides scientific evidence for the processing and detoxification of Aconitum phytomedicine.  相似文献

A novel strategy based on self-assembly technology was devised for design of photosensitive material as a ferroelectric liquid crystal (FLC) alignment layer. This development offers new tools for the study and control at the molecular level of the interaction of FLCs with solid surfaces. The photoreactive material was self-assembled to the substrate by covalent bond linkage due to a special chemical adsorption reaction. Through ester bond linkage, a cyano group with strong polarity was introduced to be terminus of the film. Under irradiation of linearly polarised ultraviolet light, an optically anisotropic self-assembled film was easily obtained. The irradiated film was demonstrated to result in homogenous alignment of FLC by optical transmittance measurements and polarising optical microscopy images of a FLC cell at different rotation angles. The alignment quality of the FLC on this self-assembled monolayer film is comparable to that of commercial rubbed polyimide film. Furthermore, it was also found that the fine alignment of the FLC may be related to the smoothness of the self-assembled film surface owing to its polar end.  相似文献

Photostability of an antidepressant agomelatine under the simulated solar radiation was studied. Quantitative and qualitative analysis of this process was performed with the use of UHPLC-DAD system coupled with a high resolution hybrid ESI-Q-TOF mass spectrometer. In contrast to the foregoing studies agomelatine turned out to be relatively photolabile compound. During the experiment six transformation products were formed, and their structures were elucidated on the basis of MS/MS fragmentation spectra. Four of these photoproducts were found to be a result of aromatic and aliphatic hydroxylation. Additionally, identified products were submitted to the in silico toxicity evaluation, which showed that some of them could be more mutagenic or toxic to rodents than the parent compound.  相似文献

在碱性奈件下,西索米星被NBS(N-溴代琥珀酰亚胺)氧化,其能量可激发共存的二氯荧光素(DCF)而产生化学发光,发光强度与西索米星的质量浓度在一定范围内呈良好的线性关系,基于此,结合流动注射技术,建立了一种直接测定西索米星的化学发光新方法。该方法具有较高的灵敏度,检出限为73μg/L(IUPAC),线性范围为0.1~10mg/L,对5.0mg/L西索米星平行测定11次,其相对标准偏差为2.3%。已用于针剂中西索米星的测定。  相似文献
