We describe a simple method for the simultaneous determination of organolead and organomanganese compounds in seawater samples. It is based on ultrasound-assisted emulsification microextraction. Trimethyllead, triethyllead, tetraethyllead, cyclopentadienylmanganese tricarbonyl and its methyl derivative were separated and determined using gas chromatography and mass spectrometry. Trimethyllead and triethyllead were derivatized with sodium tetraphenylborate before being submitted to the preconcentration step. Detection limits ranged from 7.0 to 41 ng L?1 depending on the compound. Recoveries ranged from 84 to 118 %, depending on the compound and the sample analyzed. Seawater samples were collected at different sites of the Cartagena Bay and none of the target analytes were found at levels above the corresponding detection limits.

A selective and low organic-solvent-consuming method of sample preparation combined with high-performance liquid chromatography with diode-array detection is introduced for analysis of phthalic acid esters in edible oils. Sample treatment involves initial liquid–liquid partitioning with acetonitrile, then QuEChERS cleanup by dispersive solid-phase extraction with primary secondary amine as sorbent. Preconcentration of the analytes is performed by ionic-liquid-based dispersive liquid–liquid microextraction, with the cleaned-up extract as disperser solvent and 1-hexyl-3-methylimidazolium hexafluorophosphate as extraction solvent. Under the optimized conditions, correlation coefficients (r) were 0.998–0.999 and standard errors (S y/x ) were 2.67–3.37?×?103 for calibration curves in the range 50–1000 ng g?1. Detection limits, at a signal-to-noise ratio of 3, ranged from 6 to 9 ng g?1. Intra-day and inter-day repeatability, expressed as relative standard deviation, were in the ranges 1.0–6.9 % and 2.4–9.4 %, respectively. Recovery varied between 84 % and 106 %. The developed method was successfully used for analysis of the analytes in 28 edible oils. The dibutyl phthalate content of four of the 28 samples (14 %) exceeded the specific migration limit established by domestic and international regulations.

We investigate the performance of highly anisotropic nanostructured thin film ultrathin-layer chromatography (UTLC) media with porosity and architecture engineered using the glancing-angle deposition (GLAD) process. Our anisotropic structures resemble nanoblades, producing channel-like features that partially decouple analyte migration from development direction, offering new separation behaviours. Here we study GLAD-UTLC plate performance in terms of migration distance, plate number, retention factor and a figure of merit specific to GLAD-UTLC, track deviation angle. Migration distances increase with porosity by a factor of two for all feature orientations (up to a maximum of 22 mm) over the range of porosities considered in this study. Plate numbers approaching 1100 are observed for GLAD-UTLC plates when the nanoblade features are aligned with the development direction. We present a theoretical model describing mobile phase flow in anisotropic GLAD-UTLC media, and find good agreement with experimental results. Our plates provide channel features that reduce transverse spot broadening while providing the wide pores required for rapid migration and high separation performance. These improvements may enable a greater number of parallel separations on miniaturized GLAD-UTLC plate formats. Their small sizes should also make them compatible with the Office Chromatography concept in which office peripherals (inkjet printers and flatbed scanners) replace conventional TLC instruments. Equipped with a better understanding of the unique GLAD-UTLC elution behaviours, we expect to further improve performance in the future.  相似文献

Ultrathin-layer chromatography (UTLC) differs from high-performance thin-layer chromatography (HPTLC) and from thin-layer chromatography (TLC) in two basis things: the layer thickness, and the migration distances of the analytes. UTLC has a monolithic or a nanostructured stationary silica gel phase bound directly to the glass plates. Layer thickness in UTLC is 10 μm, instead of 100–250 μm in HPTLC. Migration distances are in the range of 1–3 cm for UTLC, instead of 8–10 cm for HPTLC. Therefore, the major advantages of UTLC over HPTLC and TLC are the shorter development times and higher separation efficiency and sensitivity. Moreover, separations on UTLC plates require smaller reagent and sample volumes. However, the UTLC plates are very difficult to manage with the TLC and HPTLC equipment currently available. Therefore, the next challenge in this area is the development of an inexpensive solution with appropriate instrumentation (sensitive optical scanners and sample application systems). UTLC had been used for separations of many compounds, e.g., pharmaceutically active ingredients, pesticides, plasticisers, natural products, and other chemical substances.  相似文献

The paper presents the results of investigations on new procedures of determination of selected cleaning additives in diesel fuel. Two procedures: one-step analysis using gas chromatography with flame ionization detection (GC-FID) or mass spectrometry (GC-MS) and a two-step procedure in which normal-phase high-performance liquid chromatography (NP-HPLC) was used for preliminary separation of the additives, were compared. The additive fraction was collected using either simple elution or eluent backflush. Final determinations were performed by GC-FID and GC-MS. The studies revealed that it was impossible to determine the investigated analytes by one-step procedures, i.e. by using solely HPLC or GC. On the other hand, the use of a two-step procedure ensures reproducible results of determinations, and the limits of quantitation are, depending on the method of fraction collection by HPLC, from 1.4–2.2 ppm (GC-MS in SIM mode) to 9.6–24.0 ppm (GC-FID). Precision and accuracy of the developed procedures are compared, and possible determination errors and shortcomings discussed.

An efficient method for analyzing illegal and medicinal drugs in whole blood using fully automated sample preparation and short ultra-high-performance liquid chromatography–tandem mass spectrometry (MS/MS) run time is presented. A selection of 31 drugs, including amphetamines, cocaine, opioids, and benzodiazepines, was used. In order to increase the efficiency of routine analysis, a robotic system based on automated liquid handling and capable of handling all unit operation for sample preparation was built on a Freedom Evo 200 platform with several add-ons from Tecan and third-party vendors. Solid-phase extraction was performed using Strata X-C plates. Extraction time for 96 samples was less than 3 h. Chromatography was performed using an ACQUITY UPLC system (Waters Corporation, Milford, USA). Analytes were separated on a 100 mm?×?2.1 mm, 1.7 μm Acquity UPLC CSH C18 column using a 6.5 min 0.1 % ammonia (25 %) in water/0.1 % ammonia (25 %) in methanol gradient and quantified by MS/MS (Waters Quattro Premier XE) in multiple-reaction monitoring mode. Full validation, including linearity, precision and trueness, matrix effect, ion suppression/enhancement of co-eluting analytes, recovery, and specificity, was performed. The method was employed successfully in the laboratory and used for routine analysis of forensic material. In combination with tetrahydrocannabinol analysis, the method covered 96 % of cases involving driving under the influence of drugs. The manual labor involved in preparing blood samples, solvents, etc., was reduced to a half an hour per batch. The automated sample preparation setup also minimized human exposure to hazardous materials, provided highly improved ergonomics, and eliminated manual pipetting.

A new method was developed for the determination of fungicides in red wine using ultrasound-assisted ionic liquid dispersive liquid–phase microextraction followed by high-performance liquid chromatography. The ionic liquid, 1-hexyl-3-methylimidazolium hexafluorophosphate (IL) was quickly disrupted by ultrasonication and dispersed in wine as fine droplets. At this stage, the analytes were extracted into the fine droplets of IL. After centrifugation, the concentration of the enriched fungicides in the sedimented phase was determined. Extraction conditions including the type of extraction solvent, the extraction solvent volume, ultrasonication time, centrifugation time and sample pH were optimized. The performance of the method was studied in terms of linearity, precision, and recovery. Quantitative recoveries (>70%) except for pyrimethanil were obtained, and method precision was also satisfactory (RSD?<?10%). Enrichment factors range from 100 to 200, and the limits of detection are at the low μg per liter level for most of the target compounds.

Cell transfer by contact printing coupled with carbon-substrate-assisted laser desorption/ionization was used to directly profile and image secondary metabolites in trichomes on leaves of the wild tomato Solanum habrochaites. Major specialized metabolites, including acyl sugars, alkaloids, flavonoids, and terpenoid acids, were successfully detected in positive ion mode or negative ion mode, and in some cases in both modes. This simple solvent-free and matrix-free sample preparation for mass spectrometry imaging avoids tedious sample preparation steps, and high-spatial-resolution images were obtained. Metabolite profiles were generated for individual glandular trichomes from a single Solanum habrochaites leaf at a spatial resolution of around 50 μm. Relative quantitative data from imaging experiments were validated by independent liquid chromatography–mass spectrometry analysis of subsamples from fresh plant material. The spatially resolved metabolite profiles of individual glands provided new information about the complexity of biosynthesis of specialized metabolites at the cellular-resolution scale. In addition, this technique offers a scheme capable of high-throughput profiling of metabolites in trichomes and irregularly shaped tissues and spatially discontinuous cells of a given cell type.

As part of increasing research in the field of separation science, there have been many efforts to undertake planar chromatography with more efficient separation and better resolution in the shortest period of time, together with a specificity and a capability to identify more precisely an unknown compound present in a mixture. Ultra-thin layer chromatography (UTLC) is a modern technique which gives separation within 10–30 mm and development in just 1–6 min, with the consumption of less solvent. The stationary phase of UTLC is made up of a silica gel monolithic layer of 10 μm thickness having 3- to 4-nm mesopores and 1- to 2-μm macropores. Glancing angle deposition (GLAD)-UTLC is a modification of UTLC which gives separation within 15 mm distance and in less than 2 min. Anisotropic media of GLAD UTLC gives a unique migration direction effect. UTLC atmospheric pressure–matrix-assisted laser desorption ionizer–mass spectrometery (UTLC-AP-MALDI-MS) is a choice of technique for the identification of an unknown compound in a mixture or an impure form. ULTC-AP-MALDI-MS allows the fast changing of plates, produces more intact protonated molecules, less fragmentation and less entry of chromatographic material, and yielding less complicated spectra than the vacuum condition. Thus, UTLC is a useful technique for very rapidly giving the separation and identification of new components present in mixtures. This review provides a brief overview of UTLC, the stationary phases used for UTLC, and the detection options and applications of UTLC.  相似文献

The aim of this work is to compare simultaneous isotope dilution analysis of organotin and organomercury compounds by gas chromatography–mass spectrometry (GC–MS) and gas chromatography–inductively coupled plasma mass spectrometry (GC–ICP/MS) on certified bivalve samples. These samples were extracted by microwave with tetramethylammonium hydroxide (TMAH). Derivatization with both NaBEt4 and NaBPr4 was evaluated, and analytical performances were compared. Two CRM materials, BCR-710 and CRM-477, were analyzed by both techniques to verify accuracy. A mixed spike containing 201Hg-enriched methylmercury (MeHg), 199Hg-enriched inorganic mercury (iHg), 119Sn-enriched monobutyltin (MBT), dibutyltin (DBT), and tributyltin (TBT) as well as homemade 116Sn-enriched monophenyltin (MPT), diphenyltin (DPT), and triphenyltin (TPT) was used for the isotope dilution analysis of samples. The two techniques studied were compared in terms of classic analytical parameters: linearity, precision or repeatability (i.e., percent relative standard deviation, RSD%), limit of detection (LOD), and limit of quantification (LOQ), showing excellent linearity, precision below 12 % for all analytes, and LOQs of 0.06–1.45 pg for GC–MS and 0.02–0.27 pg for GC–ICP/MS.

We have extracted ten phthalate esters (C1 to C8) using six different micro-scale methods for extraction, and then separated them by capillary liquid chromatography coupled to UV detection. The methods included liquid-liquid extraction, ultrasonic-assisted extraction, microwave-assisted extraction, dispersive liquid-liquidmicroextraction, dispersive liquid-liquid microextraction solidification of floating organic droplets, and cloud point extraction. The linear range of the analytes is from 0.5 to 50 μg mL?1, and the detection limits range from 0.02 to ~0.17 μg mL?1. The precision and accuracy of all intra- and inter-day analyses are <5.5%. We find that dispersive liquid-liquid microextraction solidification of floating organic droplet (DLLME-SFO) is the best method for quantification of most phthalate esters in water samples and cosmetics because of its low limit of detection and high extraction efficiencies.

We report on a small (20?×?10 mm) micromachined device for the detection of gases in micro-gas chromatography (GC). It incorporates a micro-discharge across a 20-μm gap, and a remote electrode in the micro cavity that generates an electrical signal corresponding to the photo-ionization of gaseous analytes in a stream of carrier gas. Multi-component mixtures were detected and the results compared to those obtained with a flame ionization detector. The minimum detectable limit is 350 pg.μL?1 of n-octane in air when applying a 1.4 mW discharge. The combination of wet etching of glass (as used for microfluidic channels) with a lift-off process for detector electrodes by a robust batch process results in a universal, non-destructive, and sensitive microdetector for micro-GC.

In this paper, a simple and versatile route was introduced to prepare solid-phase microextraction coatings on the chemically inert titanium wire. Titania nanotube array film can be created on metallic substrates by electrochemical anodization in fluoride-containing electrolytes and subsequently support various secondary reactions to prepare functional surfaces. In the present work, titania nanotube array-coated titanium wire was successfully modified by nanostructured hydroxyapatite by a simple solution-based in situ chemical deposition method. This coating has a high surface-to-volume ratio with a thickness of about 10 μm. Extraction performance of the fiber was assessed on several polycyclic aromatic hydrocarbons in water solutions. The nanohydroxyapatite-coated fiber showed good precision (<7.4 %), low detection limits (1.79–4.89 ng/L), and wide linearity (0.1–200 μg/L) under the selected conditions. The repeatability of fiber to fiber was 1.9–18.2 %. The new solid-phase microextraction fiber has a lifetime of over 150 extractions due to the hydroxyapatite nanoslices uniformly and strongly deposited on the wire surface. The environmental water sample was used to test the reliability of the solid-phase microextraction–gas chromatography method; some analytes were detected and quantified.

A novel stationary phase based on quinolinium ionic liquid-modified silica was prepared and evaluated for high-performance liquid chromatography. The stationary phase was investigated via normal-phase (NP), reversed-phase (RP), and anion-exchange (AE) chromatographic modes, respectively. Polycyclic aromatic hydrocarbons, phthalates, parabens, phenols, anilines, and inorganic anions were used as model analytes in chromatographic separation. Using the newly established column, organic compounds were separated successfully by both NP and RP modes, and inorganic anions were also separated completely by AE mode. The obtained results indicated that the stationary phase could be applied in different chromatographic modes, with multiple-interaction mechanism including van der Waals forces (dipole–dipole, dipole–induced dipole interactions), hydrophobic, ππ stacking, electrostatic forces, hydrogen bonding, anion-exchange interactions, and so on. The column packed with the stationary phase was applied to analyze phthalates and parabens in hexane extracts of plastics. Tap water and bottled water were also analyzed by the column, and nitrate was detected as 20.1 and 13.8 mg L?1, respectively. The results illustrated that the stationary phase was potential in practical applications.

A micellar electrokinetic chromatography method was developed for determination of 15 suspected fragrance allergens and preservatives. The target compounds are widely used as ingredients in many personal care products, and all of them are included in the European Regulation concerning cosmetic products. The method was optimized by using a central composite experimental design and response surface methodology. A modified chromatographic response function was defined to weigh the terms in the response function adequately. After optimization of experimental conditions, a background electrolyte of 100 mM sodium dodecyl sulphate and 24 mM sodium tetraborate and pH 9.0 was selected for the separation of the analytes. The developed methodology was evaluated in terms of linearity, limits of detection and quantification, precision and accuracy, showing appropriate values (i.e., R 2?=?≥0.99 and accuracy of 89–115 %). Finally, applicability of the micellar electrokinetic chromatography method was assessed by successfully quantifying fragrance allergens and preservatives in commercial personal care products. The most commonly found analyte was linalool (48.3 % of samples) followed by benzoic acid (37.6 %). All samples contained at least one of the target compounds, thus confirming the ubiquity of fragrance allergens and preservatives in personal care products.

We report on a lateral flow microarray that combines multi-spot immunochip technology and immunochromatography. It can serve as a tool for the simultaneous detection of multiple analytes. The test zone of the nitrocellulose support comprises a microarray spotted with up to 32 antigens that can capture labeled gold-antibodies after lateral flow. The detection limits and detectable concentration ranges of the assay were characterized. The method was applied to the determination of drugs of abuse (and their metabolites) in urine, specifically of morphine, amphetamine, methamphetamine, and benzoylecgonine. The assay format is rapid (10 min), and has both a low relative standard deviation (< 9 %) and high recoveries (95–114 %). The detection limits (2–20 ng mL–1 for drugs of abuse) are comparable to those of conventional single-analyte strip methods.

Macroporous reversed-phase (mRP) chromatography was successfully used to develop an accurate and precise method for total protein in serum. The limits of detection (0.83 μg, LOD) and quantification (2.51 μg, LOQ) for the mRP method are comparable with those of the widely used micro BCA protein assay. The mRP method can be used to determine the total protein concentration across a wide dynamic range by detecting chromatographic peaks at 215 nm and 280 nm. The method has the added advantage of desalting and denaturing proteins, leading to more complete digestion by trypsin and to better LC–MS–MS identification in shotgun proteomics experiments.

Presence of fatty acid ethyl esters (FAEE), ethyl glucuronide (EtG), and ethyl sulfate (EtS) in meconium, the first neonatal feces, identifies maternal alcohol consumption during pregnancy. Current meconium alcohol marker assays require separate analyses for FAEE and EtG/EtS. We describe development and validation of the first quantitative liquid chromatography tandem mass spectrometry assay for 9 FAEEs, EtG, and EtS in 100 mg meconium. For the first time, these alcohol markers are analyzed in the same meconium aliquot, enabling comparison of the efficiency of gestational ethanol exposure detection. 100 mg meconium was homogenized in methanol and centrifuged. The supernatant was divided, and applied to two different solid phase extraction columns for optimized analyte recovery. Limits of quantification for ethyl laurate, myristate, linolenate, palmitoleate, arachidonate, linoleate, palmitate, oleate, and stearate ranged from 25–50 ng/g, with calibration curves to 2,500–5,000 ng/g. EtG and EtS linear dynamic ranges were 5–1,000 and 2.5–500 ng/g, respectively. Mean bias and between-day imprecision were <15 %. Extraction efficiencies were 51.2–96.5 %. Matrix effects ranged from ?84.7 to 16.0 %, but were compensated for by matched deuterated internal standards when available. All analytes were stable (within ±20 % change from baseline) in 3 authentic positive specimens, analyzed in triplicate, after 3 freeze/thaw cycles (?20 °C). Authentic EtG and EtS also were stable after 12 h at room temperature and 72 h at 4 °C; some FAEE showed instability under these conditions, although there was large inter-subject variability. This novel method accurately detects multiple alcohol meconium markers and enables comparison of markers for maternal alcohol consumption.

We report on a fast, simple and accurate method for the determination of proline in urine samples by employing a nanostructured film of conducting polypyrrole for electrochemically controlled solid-phase microextraction, and ion mobility spectrometry (IMS) for detection. This method has the advantages of simple sample preparation and a sensitivity of IMS to proline that is higher than that for other amino acids. The calibration curve is linear in the range of 0.5–60 μg L?1 (4–521 nmol L?1), and the detection limit is 0.2 μg L?1. The electrochemical potentials for uptake and release were optimized. The method was successfully applied to the clean-up and quantitation of trace amounts of proline in urine samples.

The fourth harmonic emission (200 nm) of a femtosecond Ti:sapphire laser (35 fs) was generated and used in the multiphoton ionization of 49 pesticides in gas chromatography/time-of-flight mass spectrometry. The limit of detection was improved when the ionization source from the third harmonic emission (267 nm) was replaced with the fourth harmonic emission for several pesticide molecules that contained no conjugated double bonds since their absorption bands are located in the far-ultraviolet region. This analytical instrument was used in the analysis of a series of real samples including potatoes, carrots, and cabbage, and a signal suspected to arise from di-allate was observed for the potato sample.
