Amphiphilic core–shell nanostructures containing 19F stable isotopic labels located regioselectively within the core domain were prepared by a combination of atom transfer radical polymerization (ATRP), supramolecular assembly, and condensation‐based crosslinking. Homopolymers and diblock copolymers containing 4‐fluorostyrene and methyl acrylate were prepared by ATRP, hydrolyzed, assembled into micelles, and converted into shell‐crosslinked nanoparticles (SCKs) by covalent stabilization of the acrylic acid residues in the shell. The ATRP‐based polymerizations, producing the homopolymers and diblock copolymers, were initiated by (1‐bromoethyl)benzene in the presence of CuBr metal and employed N,N,N,N,N″‐pentamethyldiethylenetriamine as the coordinating ligand for controlled polymerizations at 75–90 °C for 1–3 h. Number‐average molecular weights ranged from 2000 to 60,000 Da, and molecular weight distributions, generally less than 1.1 and 1.2, were achieved for the homopolymers and diblock copolymers, respectively. Methyl acrylate conversions as high as 70% were possible, without observable chain–chain coupling reactions or molecular weight distribution broadening, when bromoalkyl‐terminated poly(4‐fluorostyrene) was used as the macroinitiator. Poly(4‐fluorostyrene), incorporated as the second segment in the diblock copolymer synthesis, was initiated from a bromoalkyl‐terminated poly(methyl acrylate) macroinitiator. After hydrolysis of the poly(methyl acrylate) block segments, micelles were formed from the resulting amphiphilic block copolymers in aqueous solutions and were then stabilized by covalent intramicellar crosslinking throughout the poly(acrylic acid) shells to yield SCKs. The SCK nanostructures on solid substrates were visualized by atomic force microscopy and transmission electron microscopy. Dynamic light scattering was used to probe the effects of crosslinking on the resulting hydrodynamic diameters of nanoparticles in aqueous and buffered solutions. The presence of fluorine atoms in the diblock copolymers and resulting SCK nanostructures allowed for characterization by 19F NMR in addition to 1H NMR, 13C NMR, and IR spectroscopy. © 2001 John Wiley & Sons, Inc. The general approach involving the organization of polymers into micellar assemblies followed by stabilization through covalent intramicellar crosslinking of the assemblies has emerged as a powerful method for the production of well‐defined nanostructured materials, having an amphiphilic core‐shell morphology. When the covalent crosslinks are limited to the chain segments that compose the polymer micelle shell, then shell crosslinked knedel‐like (SCK) nanostructures result. The shell composition dictates the interactions of the SCKs with external agents, forms a barrier layer over the core domain, and provides robust character to the nanoparticle. Because of the stability that the crosslinked shell provides, the core domain can be of dramatically different compositions and properties—glassy, fluidlike, and crystalline polymer chains have been employed for the core material and the effects that each contributes to the overall nanostructure properties have been examined. Most notably, the shell crosslinks allow for complete removal of the core to generate hollow (solvent‐filled) nanoscale cagelike structures. © 2000 John Wiley & Sons, Inc. A method of one‐stage soap‐free emulsion polymerization to synthesize narrowly dispersed core‐shell microspheres is proposed. Following this method, core‐shell microspheres of poly(styrene‐co‐4‐vinylpyridine), poly(styrene‐co‐methyl acrylic acid), and poly[styrene‐co‐2‐(acetoacetoxy)ethyl methacrylate‐co‐methyl acrylic acid] are synthesized by one‐stage soap‐free emulsion polymerization of a mixture of one or two hydrophobic monomers and a suitable hydrophilic monomer in water. The effect of the molar ratio of the hydrophobic monomer to the hydrophilic one on the size, the core thickness, and the shell thickness of the core‐shell microspheres is discussed. The molar ratio of the hydrophobic and hydrophilic monomers and the hydrophilicity of the resultant oligomers of the hydrophilic monomer are optimized to synthesize narrowly dispersed core‐shell microspheres. A possible mechanism of one‐stage soap‐free emulsion polymerization to synthesize core‐shell microspheres is suggested and coagglutination of the oligomers of the hydrophilic monomers on the hydrophobic core is considered to be the key to form core‐shell microspheres. © 2008 Wiley Periodicals, Inc. An easy and novel approach to the synthesis of functionalized nanostructured polymeric particles is reported. The surfactant‐free emulsion polymerization of methyl methacrylate in the presence of the crosslinking reagent 2‐ethyl‐2‐(hydroxy methyl)‐1,3‐propanediol trimethacrylate was used to in situ crosslink colloid micelles to produce stable, crosslinked polymeric particles (diameter size ~ 100–300 nm). A functionalized methacrylate monomer, 2‐methacryloxyethyl‐2′‐bromoisobutyrate, containing a dormant atom transfer radical polymerization (ATRP) living free‐radical initiator, which is termed an inimer (initiator/monomer), was added to the solution during the polymerization to functionalize the surface of the particles with ATRP initiator groups. The surface‐initiated ATRP of different monomers was then carried out to produce core–shell‐type polymeric nanostructures. This versatile technique can be easily employed for the design of a wide variety of polymeric shells surrounding a crosslinked core while keeping good control over the sizes of the nanostructures. A novel primary amine‐containing monomer, 1‐(3′‐aminopropyl)‐4‐acrylamido‐1,2,3‐triazole hydrochloride (APAT), was prepared from N‐propargylacrylamide and 3‐azidopropylamine hydrochloride via copper‐catalyzed Huisgen 1,3‐dipolar cycloaddition (click reaction). Poly(N‐isopropylacrylamide)‐b‐poly(1‐(3′‐aminopropyl)‐4‐acrylamido‐1,2,3‐triazole hydrochloride), PNIPAM‐b‐PAPAT, was then synthesized via consecutive reversible addition‐fragmentation chain transfer polymerizations of N‐isopropylacrylamide and APAT. In aqueous solution, the obtained thermoresponsive double hydrophilic block copolymer dissolves molecularly at room temperature and self‐assembles into micelles with PNIPAM cores and PAPAT shells at elevated temperature. Because of the presence of highly reactive primary amine moieties in PAPAT block, two types of covalently stabilized nanoparticles namely core crosslinked and shell crosslinked micelles with ‘inverted’ core‐shell nanostructures were facilely prepared upon the addition of glutaric dialdehyde at 25 and 50 °C, respectively. In addition, the obtained structure‐fixed micelles were incorporated with gold nanoparticles via in situ reduction of preferentially loaded HAuCl4. High resolution transmission electron microscopy revealed that gold nanoparticles can be selectively loaded into the crosslinked cores or shells, depending on the micelle templates employed. © 2008 Wiley Periodicals, Inc. Amphiphilic tris(2,2′‐bipyridine)ruthenium‐cored star‐shaped polymers consisting of one polystyrene block and two poly(N‐isopropylacrylamide) blocks were prepared by the “arm‐first” method in which RAFT polymerization and nonconvalent ligand–metal complexation were employed. The prepared amphiphilic star‐shaped metallopolymers are able to form micelles in water. The size and distribution of the micelles were studied by dynamic light scattering and transmission electron microscopy techniques. Preliminary studies indicate that the polymer concentration and the hydrophilic poly(N‐isopropylacrylamide) block length can affect the morphologies of the formed metal‐interfaced core–shell micelles in water. © 2007 Wiley Periodicals, Inc. Double hydrophilic diblock copolymer, poly(N,N‐dimethylacrylamide)‐b‐poly(N‐isopropylacrylamide‐co‐3‐azidopropylacrylamide) (PDMA‐b‐P(NIPAM‐co‐AzPAM), containing azide moieties in one of the blocks was synthesized via consecutive reversible addition‐fragmentation chain transfer polymerization. The obtained diblock copolymer molecularly dissolves in aqueous solution at room temperature, and can further supramolecularly self‐assemble into core‐shell nanoparticles consisting of thermoresponsive P(NIPAM‐co‐AzPAM) cores and water‐soluble PDMA coronas above the lower critical solution temperature of P(NIPAM‐co‐AzPAM) block. As the micelle cores contain reactive azide residues, core crosslinking can be facilely achieved upon addition of difunctional propargyl ether via click chemistry. In an alternate approach in which the PDMA‐b‐P(NIPAM‐co‐AzPAM) diblock copolymer was dissolved in a common organic solvent (DMF), the core‐crosslinked (CCL) micelles can be fabricated via “click” crosslinking upon addition of propargyl ether and subsequent dialysis against water. CCL micelles prepared by the latter approach typically possess larger sizes and broader size distributions, compared with that obtained by the former one. In both cases, the obtained (CCL) micelles possess thermoresponsive cores, and the swelling/shrinking of which can be finely tuned with temperature, rendering them as excellent candidates as intelligent drug nanocarriers. Because of the high efficiency and quite mild conditions of click reactions, we expect that this strategy can be generalized for the structural fixation of other self‐assembled nanostructures. © 2007 Wiley Periodicals, Inc. A hetero‐arm star polymer, poly(ethylene glycol)‐poly(N‐isopropylacrylamide)‐poly(L‐lysine) (PEG‐PNIPAM‐PLys), was synthesized by “clicking” the azide group at the junction of PEG‐b‐PNIPAM diblock copolymer with the alkyne end‐group of poly(L‐lysine) (PLys) homopolymer via 1,3‐dipolar cycloaddition. The resultant polymer was characterized by gel permeation chromatography, proton nuclear magnetic resonance, and Fourier transform infrared spectroscopes. Surprisingly, the PNIPAM arm of this hetero‐arm star polymer nearly lose its thermal responsibility. It is found that stable polyelectrolyte complex micelles are formed when mixing the synthesized polymer with poly(acrylic acid) (PAA) in water. The resultant polyelectrolyte complex micelles are core‐shell spheres with the ion‐bonded PLys/PAA chains as core and the PEG and PNIPAM chains as shell. The PNIPAM shell is, as expected, thermally responsive. However, its lower critical solution temperature is shifted to 37.5 °C, presumably because of the existence of hydrophilic components in the micelles. Such star‐like PEG‐PNIPAM‐PLys polymer with different functional arms as well as its complexation with anionic polymers provides an excellent and well‐defined model for the design of nonviral vectors to deliver DNA, RNA, and anionic molecular medicines. © 2009 Wiley Periodicals, Inc. Reversible addition‐fragmentation chain transfer polymerization was employed to synthesize a set of copolymers of styrene (PS) and 2,3,4,5,6‐pentafluorostyrene (PPFS), as well as block copolymers with tert‐butyl acrylate (PtBA)‐b‐PS‐co‐PPFS, with control over molecular weight and polydispersity. It was found that the copolymerization of styrene and PFS allowed for the preparation of gradient copolymers with opposite levels of monomer consumption, depending on the feed ratio. Conversion to amphiphilic block copolymers, PAA‐b‐(PS‐co‐PPFS), by removing the protecting groups was followed by fitting with monomethoxy poly(ethylene glycol) chains. Solution‐state assembly and intramicellar crosslinking afforded shell crosslinked knedel‐like (SCK) block copolymer nanoparticles. These fluorinated nanoparticles (ca. 20 nm diameters) were studied as potential magnetic resonance imaging (MRI) contrast agents based on the 19F‐nuclei; however, it was found that packaging of the hydrophobic fluorinated polymers into the core domain restricted the mobility of the chains and prohibited 19F NMR spectroscopy when the particles were dispersed in water without an organic cosolvent. Packing of perflouro‐15‐crown‐5‐ether (PFCE) into the polymer micelle was demonstrated with good uptake efficiency; however, it was necessary to swell the core with a good solvent (DMSO) to increase the mobility and observe the 19F NMR signal of the PFCE. © 2009 Wiley Periodicals, Inc. Dendritic copolymers comprising a hydrophobic core and hydrophilic shell with nearly equal numbers of hydroxyl groups in the shell and different densities in the core were prepared by a multi‐step process based on anionic ring‐opening polymerization. The diversity in the core density was obtained by using copolymer stars with poly(tert‐butyl‐glycidylether)‐block‐polyglycidol arms with nearly equal length of hydrophobic blocks and numbers of hydroxyl groups of polyglycidol but different numbers of arms as macroinitiators. The ability of the dendritic copolymers to serve as a nanocontainer for a ruthenium complex Ru(NH3)3Cl3 with anticancer properties was studied. The possibility of improving the water solubility of this poorly soluble drug by loading it onto dendritic copolymers was investigated. The hydroxyl groups of the dendritic copolymers were used for complexation of the ruthenium compound to the shell. The loading efficiency was analyzed by UV–vis spectroscopy. An oligoamide‐ended poly(ethylene glycol) (PEG) with a PEG weight‐average molecular weight of 5000 (PEG‐5000‐oligoamide), with 3,5‐bis‐[2‐(5‐acetylamino‐2‐isobutoxy‐benzoylamino)‐acetylamino]‐benzoyl as the oligoamide, was synthesized. PEG‐5000‐oligoamide aggregated in chloroform or toluene via hydrogen‐bonding interactions among the oligoamide strands as a core and PEG, which was soluble in the solvents, as a shell. When a chloroform solution of PEG‐5000‐oligoamide at a concentration of approximately 0.06 g/L was cast onto a silicon wafer or a mica plate, rapid solvent evaporation induced its self reassembly as nanofibers. © 2005 Wiley Periodicals, Inc. The light‐responsive behavior in solution and in thin films of block copolymers bearing 2‐nitrobenzyl photocleavable esters as side groups is discussed in this article. The polymers were synthesized by grafting 2‐nitrobenzyl moieties onto poly(acrylic acid)‐block‐polystyrene (PAA‐b‐PS) precursor polymers, leading to poly(2‐nitrobenzyl acrylate‐random‐acrylic acid)‐block‐polystyrene (P(NBA‐r‐AA)‐b‐PS) block copolymers. The UV irradiation of the block copolymers in a selective solvent for PS led to the formation of micelles that were used to trap hydrophilic molecules inside their core (light‐induced encapsulation). In addition, thin films consisting of light‐responsive P(NBA‐r‐AA) cylinders surrounded by a PS matrix were achieved by the self‐assembly of P(NBA‐r‐AA)‐b‐PS copolymers onto silicon substrates. Exposing these films to UV irradiation generates nanostructured materials containing carboxylic acids inside the cylindrical nanodomains. Solution property of poly(γ‐benzyl‐L ‐glutamate)‐b‐polyisoprene‐b‐poly(γ‐benzyl‐L ‐glutamate) (GIG copolymer) was studied by using dynamic light scattering and static light scattering for N,N‐dimethylformamide (DMF) solution and DMF/toluene mixed solutions. GIG copolymer proved to aggregate in DMF and under DMF‐rich condition, that is, high‐polar region. The aggregate decreased in size, and completely disappeared under toluene‐rich condition, that is, low‐polar region. The correlation between solubility parameter and aggregate size of GIG copolymer in the DMF/toluene solution systems quantitatively demonstrated how strongly polarity caused by hydrogen bond made an impact on the aggregation behavior. Because the main driving force to the aggregation under DMF‐rich condition originates with polyisoprene (PIP) blocks, the aggregate in DMF is considered to be a core‐shell micelle consisting of flexible PIP core surrounded by rigid poly(γ‐benzyl‐L ‐glutamate) (PBLG) shell. The values of dimensionless parameter ρ, defined as the ratio of radius of gyration 〈S21/2 to hydrodynamic radius RH, revealed that a single chain of GIG copolymer had the form of rigid rod with flexibility, that is, once‐broken rod, caused by the incorporation of a flexible PIP chain between two rigid PBLG rods in the DMF/toluene solution system. © 2010 Wiley Periodicals, Inc. Poly[N‐(4‐vinylbenzyl)‐N,N‐dibutylamine hydrochloride] trithiocarbonate, which contains the reactive trithiocarbonate group and the appending surface‐active groups, is used as both surfactant and macromolecular reversible addition‐fragmentation chain transfer (macro‐RAFT) agent in batch emulsion polymerization of styrene. Under the conditions at high monomer content of ~20 wt % and with the molecular weight of the macro‐RAFT agent ranging from 4.0 to 15.0 kg/mol, well‐controlled batch emulsion RAFT polymerization initiated by the hydrophilic 2‐2′‐azobis(2‐methylpropionamidine) dihydrochloride is achieved. The polymerization leads to formation of nano‐sized colloids of the poly[N‐(4‐vinylbenzyl)‐N,N‐dibutylamine hydrochloride]‐b‐ polystyrene‐b‐poly[N‐(4‐vinylbenzyl)‐N,N‐dibutylamine hydrochloride] triblock copolymer. The colloids generally have core‐shell structure, in which the hydrophilic block forms the shell and the hydrophobic block forms the core. The molecular weight of the triblock copolymer linearly increases with increase in the monomer conversion, and the values are well‐consistent with the theoretical ones. In this work, the poly(methyl methacrylate‐co‐methacrylic acid)/poly(methacrylic acid‐co‐N‐isopropylacrylamide) thermosensitive composite semi‐hollow latex particles was synthesized by three processes. The first process was to synthesize the poly(methyl methacrylate‐co‐methacrylic acid) (poly (MMA‐MAA)) copolymer latex particles by the method of soapless emulsion polymerization. The second process was to polymerize methacrylic acid (MAA), N‐isopropylacrylamide (NIPAAm), and crosslinking agent, N,N′‐methylenebisacrylamide, in the presence of poly(MMA‐MAA) latex particles to form the linear poly(methyl methacrylate‐co‐methacrylic acid)/crosslinking poly(methacrylic acid‐co‐N‐isopropylacrylamide) (poly(MMA‐MAA)/poly(MAA‐NIPAAm)) core–shell latex particles with solid structure. In the third process, part of the linear poly(MMA‐MAA) core of core–shell latex particles was dissolved by ammonia to form the poly(MMA‐MAA)/poly(MAA‐NIPAAm) thermosensitive semi‐hollow latex particles. The morphologies of the semi‐hollow latex particles show that there is a hollow zone between the linear poly(MMA‐MAA) core and the crosslinked poly(MAA‐NIPAAm) shell. The crosslinking agent and shell composition significantly influenced the lower critical solution temperature of poly(MMA‐MAA)/poly(MAA‐NIPAAm) semi‐hollow latex particles. Besides, the poly(MMA‐MAA)/poly(MAA‐NIPAAm) thermosensitive semi‐hollow latex particles were used as carriers to load with the model drug, caffeine. The processes of caffeine loaded into the semi‐hollow latex particles appeared four situations, which was different from that of solid latex particles. Nonporous hydrogel microspheres 0.1–1.3 μm in diameter were prepared by the dispersion copolymerization of 1‐vinyl‐2‐pyrrolidone and ethylene dimethacrylate as a crosslinking agent. The crosslinking was evidenced by solid state 13C NMR and elemental analysis. The effect of various parameters including selection of solvent (cyclohexane, butyl acetate), initiator (4,4′‐azobis(4‐cyanopentanoic acid), 2,2′‐azobisisobutyronitrile, dibenzoyl peroxide) and stabilizer on the properties of resulting microspheres has been studied. Dynamic light scattering and photographic examination were used for determination of the diameter and polydispersity of microspheres. Increasing concentration of steric stabilizer in the initial polymerization mixture decreased the particle size. The particle size depended on the molecular weight of polystyrene‐block‐hydrogenated polyisoprene stabilizer, but not on the number of PS and polybutadiene blocks in the styrene–butadiene block copolymer stabilizers. In this research, thermoresponsive copolymer latex particles with an average diameter of about 200–500 nm were prepared via surfactant‐free emulsion polymerization. The thermoresponsive properties of these particles were designed by the addition of hydrophilic monomers [acrylic acid (AA) and sodium acrylate (SA)] to copolymerize with N‐isopropylacrylamide (NIPAAm). The effects of the comonomers and composition on the synthesis mechanism, kinetics, particle size, morphology, and thermoresponsive properties of the copolymer latex were also studied to determine the relationships between the synthesis conditions, the particle morphology, and the thermoresponsive properties. The results showed that the addition of hydrophilic AA or SA affected the mechanism and kinetics of polymerization. The lower critical solution temperature (LCST) of the latex copolymerized with AA rose to a higher temperature. However, because the strong hydrophilic and ionic properties of SA caused a core–shell structure, where NIPAAm was in the inner core and SA was in the outer shell, the LCST of the latex copolymerized with SA was still the same as that of pure poly(N‐isopropylacrylamide) latex. Through reversible addition‐fragmentation chain transfer (RAFT) polymerization of t‐butyl acrylate (tBA) and RAFT copolymerization of 2‐dimethylaminoethyl methacrylate (DMAEMA) with poly(ethylene glycol) methyl ether methacrylate (PEGMEMA), block‐comb copolymer of PtBA‐b‐P(PEGMEMA‐co‐DMAEMA) was prepared. After the self‐assembly of PtBA‐b‐P(PEGMEMA‐co‐DMAEMA) into core‐shell spherical micelles, P(PEGMEMA‐co‐DMAEMA) segments of the shell was crosslinked with 1,2‐bis(2‐iodoethoxy)ethane and the core of PtBA was selectively hydrolysized with trifluoroacetic acid. Thus, zwitterionic shell‐crosslinked micelles with positively charged outer shell and negatively charged inner core were obtained. Dynamic light scattering, transmission electron microscope, Zeta potential measurement, and nuclear magnetic resonance were used to confirm the formation of the zwitterionic shell‐crosslinked micelles. They showed the excellent resistance to the variation of pH value and possessed the positive values throughout the whole range of pH range even if the carboxylic groups of the micelles was much more than ammonium groups. © 2011 Wiley Periodicals, Inc. Radiolabeling studies were employed to investigate the influence of structure on the efficiency of surface functionalization for poly(acrylic acid)-coated shell crosslinked nanoparticles (SCKs) with two types of amine-terminated DOTA chelators. The addition of mixture of polystyrene‐b‐poly(N‐isopropylacrylamide) (PS‐b‐PNIPAM) and polystyrene homopolymer (h‐PS) in tetrahydrofuran dropwise into water leads to nanoparticles with a PS core and a thermally sensitive PNIPAM shell. The effects of the ratio of the homopolymer to copolymer and temperature on the formation and stabilization of the dispersion were investigated by using a combination of static and dynamic laser light scattering. PNIPAM shell continuously collapses as temperature increases in the range 20–40 °C. Such formed particles are stable even at temperatures much higher than lower critical solution temperature (LCST ~ 32 °C) of PNIPAM. Our results reveal that the area occupied per hydrophilic PNIPAM chain on the hydrophobic PS core remains nearly a constant regardless of the amount of h‐PS in the polymer mixture. This clearly indicates that the surface area occupied per hydrophilic group is a critical parameter for stabilizing particles dispersed in water. © 2010 Wiley Periodicals, Inc.
