Polythiophene derivative films have been synthesized on electrochemically roughened silver, gold, and copper electrodes by anodic oxidation of the corresponding monomers. Surface‐enhanced Raman scattering (SERS) analyses of these coatings led to high‐quality spectra with high signal‐to‐noise ratios. In contrast with platinum, the use of SERS‐active metals allowed the observation of important changes in the positions, widths, and relative intensities of the Raman bands during the polymer doping‐dedoping process. In situ SERS investigations revealed that the modifications in the spectral features, when the polymer oxidation degree is progressively increased, are due to a transition from the aromatic to the quinoid structure and to an increase of structural defects along the polymer chains. Moreover, in the case of soluble polyalkylthiophene films, SERS analyses were also carried out using colloidal silver solutions. Despite the very low polymer concentration and the mild experimental conditions used in these experiments, a large amplification of the Raman signal took place. Two other methods for obtaining polybithiophene–silver composite films are reported. In these cases, thanks to the silver particles, the polymer displays a SERS effect, which greatly improves the signal‐to‐noise ratio of the Raman spectra, thus allowing a much better vibrational analysis of both doped and undoped states.  相似文献

A near‐infrared surface‐enhanced Raman spectroscopy (NIR‐SERS) method was employed for oxyheamoglobin (OxyHb) detection to develop a simple blood test for liver cancer detection. Polyvinyl alcohol protected silver nanofilm (PVA‐Ag nanofilm) used as the NIR‐SERS active substrate to enhance the Raman scattering signals of OxyHb. High quality NIR‐SERS spectrum from OxyHb adsorbed on PVA‐Ag nanofilm can be obtained within 16 s using a portable Raman spectrometer. NIR‐SERS measurements were performed on OxyHb samples of healthy volunteers (control subjects, n = 30), patients (n = 40) with confirmed liver cancer (stage I, II and III) and the liver cancer patients after surgery (n = 30). Meanwhile, the tentative assignments of the Raman bands in the measured NIR‐SERS spectra were performed, and the results suggested cancer specific changes on molecule level, including a decrease in the relative concentrations and the percentage of aromatic amino acids of OxyHb, changes of the vibration modes of the CaHm group and pyrrole ring of OxyHb of liver cancer patients. In this paper, principal component analysis (PCA) combined with independent sample T test analysis of the measured NIR‐SERS spectra separated the spectral features of the two groups into two distinct clusters with the sensitivity of 95.0% and the specificity of 85.7%. Meanwhile, the recovery situations of the liver cancer patients after surgery were also assessed using the method of discriminant analysis‐predicting group membership based on PCA. The results show that 26.7% surgeried liver cancer patients were distinguished as the normal subjects and 63.3% were distinguished into the cancer. Our study demonstrated great potentials for developing NIR‐SERS OxyHb analysis into a novel clinical tool for non‐invasive detection of liver cancers. Ethyl carbamate (EC), a potentially toxic compound, is found in alcoholic beverages and fermented foodstuff. A combined experimental and theoretical study of Raman on EC is reported in this work for the first time. The Raman bands observed for EC in solid phase are characteristic for the carbonyl group, C―C, C―H and N―H stretching and deformation vibrations. These spectral features coupled with a pKa study allowed establishing the neutral species of EC present in the aqueous solutions experimentally tested at different concentrations. In addition, by performing a density functional theory study in the gas phase, the calculated geometry, the harmonic vibrational modes, and the Raman scattering activities of EC were found to be in good agreement with our experimental data and helped establish the surface‐enhanced Raman scattering (SERS) behavior and EC adsorption geometry on the silver surfaces. The Raman peak at 1006 cm−1, assigned to the υs(CC) + ω(CH) modes, the strongest and best reproducible peak in the SERS spectra, was used for a quantitative evaluation of EC. The limit of detection, which corresponds to a signal‐to‐noise ratio equal to 3, was found to be 2 × 10−7 M (17.8 µg l−1). SERS spectra obtained by using hydroxylamine hydrochloride‐reduced silver nanoparticles provide a fast and reproducible qualitative and quantitative determination of EC in aqueous solution. Local pH environment has been considered to be a potential biomarker for tumor diagnosis because solid tumors contain highly acidic environments. A pH‐sensing nanoprobe based on surface‐enhanced Raman scattering (SERS) using nanostars under near‐infrared excitation has been developed for potential biomedical applications. To theoretically investigate the effect of protonation state on SERS spectra of p‐mercaptobenzoic acid (pMBA), we used the density functional theory (DFT) with the B3LYP functional to calculate Raman vibrational spectra of pMBA‐Au/Ag complex in both protonated and deprotonated states. Vibrational spectral bands were assigned with DFT calculation and used to investigate SERS spectral changes observed from experiment when varying pH value between five and nine. The SERS peak position of pMBA at ~1580 cm−1 was identified to be a novel pH‐sensing index, which has small but noticeable downshift with pH increase. This phenomenon is confirmed and well‐explained with theoretical simulation. The study demonstrates that SERS is a sensitive tool to monitor minor structural changes due to local pH environment, and DFT calculations can be used to investigate Raman spectra changes associated with minor differences in molecular structure. Raman‐enhancing properties of chitosan (CS)‐coated gold/silver nanostars (Au/AgNSs) were demonstrated by using them as a surface‐enhanced Raman scattering (SERS) probe. Based on the energy‐dispersive X‐ray spectroscopy element distribution maps and highly enhanced SERS spectra, we suggest that the incorporation of silver into the NS tips leads to a stronger SERS behavior. The SERS spectra of the proteins adsorbed on the NS surface greatly differ from their respective Raman spectra in both the band positions and relative intensities, indicating that the protein molecules penetrate through the CS coating layer and interact closely with the NS surface. Raman and SERS spectra of Chlamydia trachomatis protease/proteasomelike activity factor are reported for the first time, demonstrating the potential of these NSs for the development of a diagnosis method for Chlamydia based on SERS. Measurement and interpretation of the excitation wavelength dependence of surface‐enhanced Raman scattering (SERS) spectra of molecules chemisorbed on plasmonic, e.g. Ag nanoparticle (NP) surfaces, are of principal importance for revealing the charge transfer (CT) mechanism contribution to the overall SERS enhancement. SERS spectra, their excitation wavelength dependence in the 445–780‐nm range and factor analysis (FA) were used for the identification of two Ag‐2,2′:6′,2″‐terpyridine (tpy) surface species, denoted Ag+–tpy and Ag(0)–tpy, on Ag NPs in systems with unmodified and/or purposefully modified Ag NPs originating from hydroxylamine hydrochloride‐reduced hydrosols. Ag+–tpy is a spectral analogue of [Ag(tpy)]+ complex cation, and its SERS shows virtually no excitation wavelength dependence. By contrast, SERS of Ag(0)–tpy surface complex generated upon chloride‐induced compact aggregate formation and/or in strongly reducing ambient shows a pronounced excitation wavelength dependence attributed to a CT resonance (the chemical mechanism) contribution to the overall SERS enhancement. Both the resonance (λexc = 532 nm) and off‐resonance (λexc = 780 nm) pure‐component spectra of Ag(0)–tpy obtained by FA are largely similar to surface‐enhanced resonance Raman scattering (λexc = 532 nm in resonance with singlet metal to ligand CT (1 MLCT) transition) and SERS (λexc = 780 nm) spectra of [Fe(tpy)2]2+ complex dication. Although conventional Raman, surface‐enhanced Raman (SERS) and tip‐enhanced Raman spectroscopy (TERS) have been known for a long time, a direct, thorough comparison of these three methods has never been carried out. In this paper, spectra that were obtained by conventional Raman, SERS (on gold and silver substrates) and TERS (in ‘gap mode’ with silver tips and gold substrates) are compared to learn from their differences and similarities. Because the investigation of biological samples by TERS has recently become a hot topic, this work focuses on biologically relevant substances. Starting from the TER spectra of bovine serum albumin as an example for a protein, the dipeptides Phe–Phe and Tyr–Tyr and the tripeptide Tyr–Tyr–Tyr were investigated. The major findings were as follows. (1) We show that the widely used assumption that spectral bands do not shift when comparing SER, TER and conventional Raman spectra (except due to binding to the metal surface in SERS or TERS) is valid. However, band intensity ratios can differ significantly between these three methods. (2) Marker bands can be assigned, which should allow one to identify and localize proteins in complex biological environments in future investigations. Surface‐enhanced Raman scattering (SERS) spectra of hydrogenated amorphous carbon (a C:H) deposited on silver substrates have been recorded with a confocal Raman microscope. When scattered radiation is collected during a short time from an area of a few square micrometres, the subsequently measured SERS spectra often exhibit strong temporal changes (fluctuations). In this paper we present examples of spectra for which the intensity maxima of the fluctuating narrow Raman bands are significantly higher than that of the background (the background is usually dominated by two broad Raman bands centred at about 1350 and 1590 cm−1). In a series of successively measured spectra, one can find spectra with noticeably different total integral intensity. This suggests that the results of averaging the spectra revealing strong and weak fluctuations may be different (at least in intensity). The influence of some electrolytes on the SERS spectral fluctuations is also analysed. Our experiments revealed that the efficiencies of quenching of the SERS spectral fluctuations by various electrolytes are significantly different. We suggest that only anions directly interacting with the metal surface quench strong SERS fluctuations, and that the large differences between chloride and perchloride solutions are caused by differences in the strength of interaction of Cl and ClO4 anions with the silver surface. This paper introduces a new robust method for the removal of background tissue fluorescence from Raman spectra. Raman spectra consist of noise, fluorescence and Raman scattering. In order to extract the Raman scattering, both noise and background fluorescence must be removed, ideally without human intervention and preserving the original data. We describe the rationale behind our robust background subtraction method, determine the parameters of the method and validate it using a Raman phantom against other methods currently used. We also statistically compare the methods using the residual mean square (RMS) with a fluorescence‐to‐signal (F/S) ratio ranging from 0.1 to 1000. The method, ‘adaptive minmax’, chooses the subtraction method based on the F/S ratio. It uses multiple fits of different orders to maximize each polynomial fit. The results show that the adaptive minmax method was significantly better than any single polynomial fit across all F/S ratios. In the present study, an application of a silver colloid substrate in order to obtain Fourier‐transform surface‐enhanced Raman (FT‐SER) spectra of natural historical dyes is presented. In detail, we collected a spectral database from solutions of pure dyes and then we carried out extractionless both hydrolysis and non‐hydrolysis FT‐SERS analyses on wool fibers previously dyed in our laboratory and on ancient textiles. The term ‘extractionless’ refers to a method of SERS analysis applied directly on the fiber, thus avoiding the extraction of dyes from textile samples. The combination of a low‐energy source of radiation, as in the FT‐Raman technique, with SER spectroscopy can bring the important advantage of reducing the fluorescence typical of ancient samples and organic dyes. In some historical textile samples, for which SER spectra by use of visible excitation could not be obtained, the FT‐SER spectrum of an iron‐gall dye was recorded without hydrolysis, while, with an HF hydrolysis pre‐treatment on ancient fibers, madder, lac dye and brazilwood were clearly recognized. The Raman and surface‐enhanced Raman scattering (SERS) spectra of l ‐proline (Pro) and trans‐4‐hydroxy‐ l ‐proline (Hyp) were recorded. SERS spectra were obtained on colloidal Ag prepared by reduction with hydroxylamine. Allowing sufficient time for Pro and Hyp to adjust in the colloidal solution resulted fundamentally in obtaining unique and reproducible SERS spectra. Hyp stabilizes on the surface more rapidly than Pro. The spectral analysis indicates that Pro interacts with the Ag surface through the carboxylate group. The interaction of Hyp with the metal surface occurs through the amino, methylene and carboxylate moieties of the molecule. The spectroscopic results are supported by quantum chemical calculations, performed using extended Hückel theory (EHT) of the title compounds interacting with an Ag cluster model. The assignment of the Raman bands was supported by a normal coordinate analysis performed through Becke, three‐parameter, Lee–Yang–Parr/6‐311 G* + calculations. Although many surface‐enhanced Raman scattering (SERS)‐based methods for detecting specific proteins have been studied, simple and direct detection of total protein in liquid using a SERS‐based method remains difficult. In this study, a distinguishable effect on the SERS spectra from pre‐mixture of phosphomolybdic acid (PMA) with protein was found, indicating that PMA could be used as a SERS reporter for total protein detection in a liquid sample. Further experiments confirmed a good linear relationship between a premixed concentration of protein (casein, whey protein or bovine serum albumin) and the SERS intensity of PMA in our SERS system. Using casein as a reference, a PMA‐mediated SERS method was proposed that can quantitatively analyze protein at 2.5–25 µg/ml with a limit of detection of 1.5 µg/ml. The applicability of surface‐enhanced Raman spectroscopy is demonstrated to probe the adsorption behavior of individual molecules on a Ag electrode. High‐quality SERS spectra of (R)‐di‐2‐naphthylprolinol (DNP) were obtained from ultradilute solutions (10−12 M ) on the Ag‐nanoparticle‐modified Ag electrode, which is attributed to the high electromagnetic (EM) effect of the SERS‐active system as well as to the strong adsorption and interaction of DNP molecules with Ag. The stable SERS spectra present remarkable potential dependence, which gives evidence for the behavior of individual DNP molecules on the Ag surface. Based on statistical analysis for the probability of DNP molecules located in ‘hot spots’, we propose an SERS mechanism for individual molecules in the electrode system, in combination with the hot‐spot model and orientation of the probe molecules. While recording SERS spectra of pure β‐carotene at sub‐micromole concentrations for reference purpose, we discovered an unusual spectral response never reported before. In pre‐resonance conditions with the 532‐nm line, SERS of β‐carotene with AgNPs exhibits among the strong υ(CC) mode at 1512 cm−1 unshifted from normal Raman spectrum, additional strong bands at 1649, 1575 and 1387 cm−1 as well as other medium bands not observed in the Raman spectrum of the crystalline powder. Such behavior is explained in terms of selection rules relaxation upon cyclohexene terminal rings of the β‐carotene interaction with the NP surface. AFM images of the SERS system suggested dimers and trimers clustering of the nanoparticles with adsorbed β‐carotene. In light of the new SERS feature the consequences in correct interpretation of the SERS imaging from complex biosystems containing carotenoids are discussed. Relative intensity ratio of the β‐carotene band at 1512 cm−1 and water against concentration allowed a reliable SERS calibration curve for 50 to 500 nmol l−1 concentration range and provided quantitative SERS assessment of the carotenoid content in the sea urchin (Paracentrotus lividus) gonads extracts. Surface‐enhanced Raman spectroscopy (SERS) has the potential to make a significant impact in biology research due to its ability to provide information orthogonal to that obtained by traditional techniques such as mass spectrometry (MS). While SERS has been well studied for its use in chemical applications, detailed investigations with biological molecules are less common. In addition, a clear understanding of how methodology and molecular characteristics impact the intensity, the number of peaks, and the signal‐to‐noise of SERS spectra is largely missing. By varying the concentration and order of addition of the SERS‐enhancer salt (LiCl) with colloidal silver, we were able to evaluate the impact of these variables on peptide spectra using a quantitative measure of spectra quality based on the number of peaks and peak intensity. The LiCl concentration and order of addition that produced the best SERS spectra were applied to a panel of synthetic peptides with a range of charges and isoelectric points (pIs) where the pI was directly correlated with higher spectral quality. Those peptides with moderate to high pIs and spectra quality scores were differentiated from each other using the improved method and a hierarchical clustering algorithm. In addition, the same method and algorithm was applied to a set of highly similar phosphorylated peptides, and it was possible to successfully classify the majority of peptides on the basis of species‐specific peak differences. Liquid chromatography and mass spectrometry were time‐consuming and expensive as the main methods for the drug analysis at present, and the samples must be pretreated. The Raman spectroscopy measurement methods were fast and simple, so the Raman spectroscopy methods for the drug analysis were explored in this paper. An optical fiber nano‐probe coated with gold nanoparticles was fabricated and used with surface‐enhanced Raman spectroscopy (SERS) to measure levofloxacin lactate. The resulting SERS spectra of levofloxacin lactate in mouse blood that was detected by the optical fiber nano‐probe clearly showed the characteristic wave numbers of levofloxacin lactate, indicating that optical fiber nano‐probes can be used with spectral techniques to analyze drugs in vitro or potentially even in vivo. The ability of normal Raman and surface‐enhanced Raman scattering (SERS) to identify and detect bacteria has shown great success in recent studies. The addition of silver nanoparticles to bacterial samples not only results in an enhanced Raman signal, but it also suppresses the native fluorescence associated with biological material. In this report, Raman chemical imaging (RCI) was used to analyze individual bacteria and complex mixtures of spores and vegetative cells. RCI uses every pixel or a binned pixel group (BPG) of the Raman camera as an independent Raman spectrograph, allowing collection of spatially resolved Raman spectra. The advantage of this technique resides primarily in the analysis of samples in complex backgrounds without the need for physically isolating or purifying the sample. Using a chemical imaging Raman microscope, we compare normal RCI to SERS‐assisted chemical imaging of mixtures of bacteria. In both cases, we are able to differentiate single bacterium in the Raman microscope's field of view, with a 60‐fold reduction in image acquisition time and a factor of 10 increase in the signal‐to‐noise ratio for SERS chemical imaging over normal RCI. Madder has been used as a textile dye and pigment in works of art since antiquity. Madder dyes from different botanical species are characterized by different series of anthraquinone derivatives. The occurrence of one or more of these compounds in various relative amounts may provide a useful indication of the plant species used to obtain the dye. In this work, surface‐enhanced Raman spectroscopy (SERS) was applied to the analysis of textile fibers dyed with madder from two different botanical species, Oldenlandia umbellata L. and Rubia tinctorum L. The resulting SERS spectra were interpreted in light of the actual composition of the madder dyes as determined by high‐performance liquid chromatography. Interestingly, the SERS spectra were found to exhibit very distinctive spectral features depending on the different anthraquinone derivatives present in the dyes. In particular, the SERS spectrum of O. umbellata L. was found to match the spectrum of alizarin, while the SERS spectrum of R. tinctorum L. was surprisingly dominated by signals due to the less studied anthraquinone compound pseudopurpurin. With this study, we demonstrate that SERS spectra may offer valuable information regarding the major coloring constituents present in different madder species. Moreover, our work shows that not only can SERS be used successfully to differentiate among closely related anthraquinone derivatives, but also that this technique is particularly suitable for the detection of pseudopurpurin, leading to the identification of this compound in a number of works of art. The effectiveness of surface‐enhanced Raman spectroscopy (SERS) spectrocsopy on Ag colloids has been successfully demonstrated for the identification of a yellow dye in two ancient wool threads found in the Royal Tumulus of In Aghelachem, Libyan Sahara, belonging to the Garamantian period (2nd–3rd century A.D.). High‐performance liquid chromatography (HPLC) highlighted the presence of ellagic acid in the extracts from the threads, excluding other chromophores. This result, together with the abundance of malic acid detected by gas chromatography‐mass spectrometry (GC‐MS), suggested the possible use of pomegranate rind or sumac berries as source of the yellow dye, both plants being documented in the Fezzan area during the Garamantian period. HPLC analyses and SERS spectra acquired on the extracts of the ancient threads were therefore compared with those obtained from pomegranate and sumac extracts of the corresponding fruits and reference dyed wool samples, allowing us to identify the yellow dye as deriving from pomegranate (Punica granatum L.). SERS spectra of ellagic acid and dyes extracted from pomegranate rind and sumac berries are reported here for the first time. A methodological improvement is also presented, based on the use of NaClO4 as aggregating agent, that leads to a significant increase of the signal‐to‐noise ratio in the SERS spectra. The substrate‐dependent surface‐enhanced Raman scattering (SERS) of 4‐aminobenzenethiol (4‐ABT) adsorbed on Au surfaces has been investigated. 4‐ABT is one of the very unique adsorbate molecules whose SERS spectral patterns are known to be noticeably dependent on the relative contribution of chemical enhancement mechanism vs electromagnetic enhancement mechanism. The SERS spectral patterns of 4‐ABT adsorbed on gold substrates with various surface morphology have thus been analyzed in terms of the symmetry types of the vibrational modes. Almost invisibly weak b2 type vibrational bands were observed in the SERS spectra of the 4‐ABT adsorbed on Au colloidal sol nanoparticles or commercially available Au micro‐powders because of the weak contribution of the chemical enhancement. However, greatly enhanced b2 vibrational bands were observed in the spectra of the 4‐ABT molecules adsorbed on the synthesized Au(Zn) sponge or the electrochemically roughened Au(ORC) foil caused by the strong contribution of the chemical enhancement mechanism.
