Trichostatin A (TSA), originally developed as an antifungal agent, is one of potent histone deacetylase (HDAC) inhibitors, which are known to cause growth arrest and apoptosis induction of transformed cells, including urinary bladder, breast, prostate, ovary, and colon cancers. However, the effect of HDAC inhibitors on human non-small cell lung cancer cells is not clearly known yet. Herein, we demonstrated that treatment of TSA resulted in a significant decrease of the viability of H157 cells in a dose-dependent manner, which was revealed as apoptosis accompanying with nuclear fragmentation and an increase in sub-G0/G1 fraction. In addition, it induced the expression of Fas/FasL, which further triggered the activation of caspase-8. Catalytic activation of caspase-9 and decreased expression of anti-apoptotic Bcl-2 and Bcl-XL proteins were observed in TSA-treated cells. Catalytic activation of caspase-3 by TSA was further confirmed by cleavage of pro-caspase-3 and intracellular substrates, including poly (ADP-ribose) polymerase (PARP) and inhibitor of caspase-activated deoxyribonuclease (ICAD). In addition, a characteristic phenomenon of mitochondrial dysfunction, including mitochondrial membrane potential transition and release of mitochondrial cytochrome c into the cytosol was apparent in TSA-treated cells. Taken together, our data indicate that inhibition of HDAC by TSA induces the apoptosis of H157 cells through signaling cascade of Fas/FasL-mediated extrinsic and mitochondria-mediated intrinsic caspases pathway.  相似文献

Garlic (Allium sativum L.; Liliaceae) has been widely demonstrated in the role of cancer prevention, but the specific compound in garlic corresponding to this effect and its mechanisms are not clearly known. Allicin is one of the organic sulphur compounds derived from garlic. In the present study we investigated the anti-proliferative and pro-apoptotic activities of allicin in murine T-lymphocytes (EL-4) and the mechanism of inducing apoptosis in?vitro. The results showed that allicin was effective in inhibiting the proliferation of EL-4 cells in?vitro in a concentration-dependent manner. Further, allicin could induce the formation of apoptotic bodies, nuclear condensation, DNA spallation, and even activated the expression of caspase-3, -12 and cytochrome C (cyt C). Finally, allicin up-regulated the ratio of Bax/Bcl-2 and induced a mitochondrion membrane potential (MMP) decrease. Allicin induced apoptosis in EL-4 cells in a time- and concentration-dependent manner, in which the mitochondrial pathway might play a central role.  相似文献

Using tetra-n-butyl titanate and magnesium nitrate as raw materials, Dy3+ and Pr3+ ions in the matrix of magnesium titanate (MgTiO3) was successfully synthesized by a modified solid-state reaction. The mixtures to achieve a solid-state reaction were heated in porcelain crucibles at 600?°C for 2?h, 900?°C for 6?h, and 1000.0?°C for 2?h. The reaction products obtained in an air atmosphere were characterized by X-ray powder diffractions. The optimization of reaction conditions were carried out by thermal gravimetry and differential thermal analysis methods. Surface and elemental analyses were performed by using on SEM instrument. The excitation and emission spectra were recorded by photoluminescence spectrophotometer.  相似文献

Phosphatase-inert peptidomimetics containing phosphonate pSer analogue have been developed as valuable biological tools for probing and regulating pSer-dependent protein-protein interactions (PPIs) in cellular context. Herein, we report a facile and efficient synthesis route of Fmoc-protected phosphonate pSer mimetic and also present the application of this building block in the solid-phase synthesis of a phosphatase-resistant substrate peptide of 14-3-3 ζ, retaining 14-3-3 ζ binding efficacy similar to the parent pSer-containing peptide.  相似文献

Spectroscopic properties and local structure of rare-earth ions in Ge–Ga–S glasses with the addition of alkali halides were investigated. The intensity of the 1.31-μm emission from Dy3+ (6F11/2·6H9/2 → 6H15/2) increased sharply when the appropriate amount of alkali halides was added, at the expense of the 1.75-μm emission intensity (6H11/2 →6H15/2). The lifetimes of the 1.31-μm emission level also increased as much as 35 times from 38 μs for Ge–Ga–S glass (0.1 at.% Dy3+) to 1320 μs for glass containing 10 mol% of CsBr. These enhancements occurred only when the ratio of MX(M = K,Cs and X = Br, I)/Ga was equal to or larger than unity. Phonon side band (PSB) showed that the several local phonon modes, with the frequencies around 100 cm–1, were coupled to 4f electrons of Eu3+. The nearest neighbors of Eu3+ ions are composed of halogen ions that are part of well-structured complex such as EuCl3, tetrahedral 〚GaS3/2Cl〛 subunit and/or Ga2Cl6. A small amount of As was added to increase the resistivity against the recrystallization during re-heating. The best composition for practical usage was 0.7 〚Ge0.25As0.10S0.65〛–0.15 GaS3/2–0.15 CsBr. This glass also exhibited high resistance against the attack of liquid water and is therefore a potential material for efficient fiber-optic amplifiers.  相似文献

A home-made high sensitive photoacoustic transducer was used to determine the photoacoustic signal in a solution. The photoacoustic spectra of Ho in different solvents were measured and comparedwith its absorption spectrum. The affection factors of photoacousticsensitivity are discussed. The trivalent rare earth ions Ho3+ and Nd3+in an aqueous acetonitrile solution were determined. The detection limit is 5×10-8 mol/L for Ho3+ and 1.0 ×10-7 mol/L for Nd3+, and corresponds to the absorbance of 1.5×10 and 6.3×10-7.respectively.  相似文献

By means of liquid chromatography–tandem mass spectrometry we showed recently, that the chromosomal deletion or inactivation of the methylthioadenosine phosphorylase (MTAP) gene led to the accumulation of 5′-deoxy-5′-(methylthio)adenosine (MTA) in cancer cells. Here, we expanded the method to other key intermediates of the methionine and polyamine pathways to further elucidate the molecular consequences of a lack of MTAP activity. Employing multiple-reaction monitoring, limits of detection and lower limits of quantification in the range of 2.5–100 and 5.0–500 nM, respectively, were achieved according to the guidelines of the FDA, thus enabling the direct measurement of the metabolites in biological samples without prior enrichment and derivatization with an analytical repeatability of 1–3%. Relative standards deviations for quadruplicate 80% methanol extractions of metabolites from cultured tumor cells ranged from 1.1 to 25.5%, while the combined methodological and biological variability in metabolite concentrations in 10 liver biopsies was 11.8–51.4%. The method enabled the demonstration of changes in the concentration of intermediates of the methionine and polyamine metabolism other than MTA in hepatocellular carcinoma specimens lacking the enzyme MTAP compared to normal liver tissue.  相似文献

Green nanotechnology elucidates highly prioritized anticancer activity. We synthesized Copper oxide nanoparticles (CuONPs) using leaves of Azadirachta indica (A. indica) plants and studied the molecular mechanism of cancer cell apoptosis. After their synthesis, with the help of expository tools like Fourier transform infrared spectroscopy (FT-IR), Transmission electron microscopy (TEM), Dynamic light scattering (DLS) and surface zeta potential we confirmed the successful synthesis of CuONPs. Here, crystalline structure of green synthesized CuONPs of 36?±?8?nm size and spherical shape was able to kill MCF-7 and Hela cells, estimated by MTT assay. Successful internalization of Cu+2 ions inside the cell was estimated by the atomic absorption study. Cellular uptake of Cu+2 ions inflicted significant Reactive Oxygen Species (ROS) generation inside the cancer cells, thereby leading to DNA fragmentation as observed by DAPI staining. In in vivo model, CuONPs reduced the breast tumor volume in Balb/C mice and increased the mean survival time through the alteration of pro-inflammatory cytokines level. In case of both in vivo and in vitro models, CuONPs altered the pro-inflammatory cytokine level and pro-apoptotic protein expressions. In future, green synthesized CuONPs might be beneficial for its application as an anticancer drug in in vivo (mice model) and in vitro, though further study is needed on its toxicity.  相似文献

Oxidative stress has been considered as a major cause of cellular injuries in a variety of clinical abnormalities, especially prominent in neural diseases. One of the usable ways to prevent the reactive oxygen species (ROS)-mediated cellular injury is dietary or pharmaceutical augmentation of some free radical scavenger. Water-soluble amino-fullerene is a novel compound that behaves as a free radical scavenger with excellent biology consistent. In the present study, we have synthesized and characterized a novel cystine C60 derivative for the first time, and investigated the effects on hydrogen peroxide-induced oxidative stress and apoptotic death in cultured rat pheochromocytoma (PC12) cells. PC12 cells treated with hydrogen peroxide underwent apoptotic death as determined by MTT, PI/Hoechst 33342 staining and flow cytometry analysis. These results suggested that cystine C60 derivative has the potential to prevent oxidative stress-induced cell death and has no evident toxicity.  相似文献

Prostate cancer is the most frequently diagnosed malignancy and the second leading cause of cancer mortality among men in the United States. Accumulating evidence suggests that lysophosphatidic acid (LPA) serves as an autocrine/paracrine mediator to affect initiation, progression and metastasis of prostate cancer. In the current study, we demonstrate that LPA stimulates migration and proliferation of highly metastatic human prostate cancer, PC-3M-luc-C6 cells. LPA-induced migration of PC-3M-luc-C6 cells was abrogated by pretreatment of PC-3M-luc-C6 cells with the LPA receptor 1/3 inhibitor Ki16425 or small interfering RNA (siRNA)-mediated silencing of endogenous LPA receptor 1, implicating a key role of the LPA-LPA receptor 1 signaling axis in migration of PC-3M-luc-C6 cells. In addition, LPA treatment resulted in augmented expression levels of Krüppel-like factor 4 (KLF4), and siRNA or short-hairpin RNA (shRNA)-mediated silencing of KLF4 expression resulted in the abolishment of LPA-stimulated migration and proliferation of PC-3M-luc-C6 cells. shRNA-mediated silencing of KLF4 expression resulted in the inhibition of in vivo growth of PC-3M-luc-C6 cells in a xenograft transplantation animal model. Taken together, these results suggest a key role of LPA-induced KLF4 expression in cell migration and proliferation of prostate cancer cells in vitro and in vivo.  相似文献

The 5D3-5D4 cross-relaxation of Tb3+ in α-GdOF has been investigated by analysis of the 5D3 decay curve. It is shown that the relaxation is electric dipole-dipole in character. The relaxation rate has been evaluated in the temperature region between 4.2 K and room temperature. The temperature dependence of the relaxation rate points to the thermal population of a higher-lying 7F6 or 5D3 level.  相似文献

The high-resolution single-photon pulsed-field-ionization zero-kinetic-energy photoelectron spectra of the X?(+) (2)E(3/2)←X?(1)A(1) transition of CH(3)I and CD(3)I have been recorded. The spectral resolution of better than 0.15 cm(-1) enabled the observation of the rotational structure. CH(3)I(+) and CD(3)I(+) are subject to a weak E?e Jahn-Teller effect and strong spin-orbit coupling. The treatment of the rovibronic structure of the photoelectron spectra in the corresponding spin double group, C(3v)(2)(M), including the effects of the spin-orbit interaction and the vibrational angular momentum, allowed the reproduction of the experimentally observed transitions with spectroscopic accuracy. The relevant spin-orbit and linear Jahn-Teller coupling parameters of the X?(+) ground state were derived from the analysis of the spectra of the two isotopomers, and improved values were obtained for the adiabatic ionization energies [E(I)(CH(3)I)/hc=76931.35(20) cm(-1) and E(I)(CD(3)I)/hc=76957.40(20) cm(-1)] and the rotational constants of the cations. Rovibronic photoionization selection rules were derived for transitions connecting neutral states following Hund's-case-(b)-type angular momentum coupling and ionic states following Hund's-case-(a)-type coupling. The selection rules, expressed in terms of the angular momentum projection quantum number P, account for all observed transitions and provide an explanation for the nonobservation of several rotational sub-bands in the mass-analyzed threshold-ionization spectra of CH(3)I and CD(3)I reported recently by Lee et al. Polyphyllin Ι is a steroidal saponin isolated from the rhizoma of Paris polyphylla. In the present study, we aimed to investigate the anticancer effects of polyphyllin Ι in colorectal cancer and to elucidate the potential underlying molecular mechanisms. Using, CCK8 assay, flow cytometry, laser confocal microscope analysis and western blot, the anticancer effects of the polyphyllin Ι were analysed in colorectal cells. Our results indicate that polyphyllin Ι significantly decreased cell viability of HCT 116 cells and induced autophagy. Furthermore, we found that polyphyllin Ι induced autophagy in an ROS-dependent cell death and not related with PI3 K/AKT/mTOR pathway. We also provide evidence that excessive ROS triggered by polyphyllin Ι could induce G2/M phase arrest via regulating cycle proteins expression of cell cycle regulators, such as p21 and cyclinB1. In conclusion, polyphyllin Ι exhibit anticancer effect through ROS-dependent autophagy and induces G2/M arrest in colorectal cancer.  相似文献

The thermodynamics of the extraction of Eu3+ by thenoyltrifluoroacetone (HTTA) and by mixtures of HTTA and bidentate amine bases such as 2,2-dipyridyl (Dipy) and 1,10-phenanthroline (Phen) in benzene from an aqueous phase fixed to 0.1M ionic strength (NaClO4), pH 3.9, has been investigated. Phenanthroline forms both the Eu(TTA)3·Phen and Eu(TTA)3·2Phen species, which are much stronger than the dipyridyl complexes. The diluent effect on the extraction is also studied.  相似文献
