Geometric and micro-structure design, tribology properties of beetle joints were experimentally studied, which aimed to enlighten ideas for the joint design of MEMS. The observation by using SEM and microscopy suggested that beetle’s joints consist of a concave surface matched with a convex surface. The heads of the beetles, rubbing with flat glass, were tested in fresh and dried statuses and compared with sapphire ball with flat glass. Frictional coefficient of the joint material on glass was significantly lower than that of the sapphire sphere on glass. The material of the joint cuticle for convex surface is rather stiff (the elastic modulus 4.5 Gpa) and smooth. The surface is hydrophobic (the contact angle of distilled water was 88.3°). It is suggested here that the high stiffness of the joint material and hydrophobicity of the joint surface are parts of the mechanism minimizing friction in insect joints.  相似文献

The evolution of a myrmecophilous lifestyle in beetles is often associated with morphological alterations. In particular, the antennae of all members of the myrmecophilous ground beetle tribe Paussini are greatly modified, with flagellomeres flattened or crassate, frequently reduced in number from 9 to 5 or even 1 single “antennal club”. The enhanced glandular function of the antennal club has been recently described by scanning (SEM) and transmission (TEM) electron microscopy in Paussus favieri Fairmaire, 1851, where the antenna has become a complex glandular organ, supplying rewarding substances to the ants. In the present work, the antennal sensilla of P. favieri are investigated by SEM, TEM and focused ion beam (FIB/SEM) technology. Most sensilla of scape and antennal club are highly modified mechanoreceptors (i.e. multipointed, fringed, branched, brush-like, sickle-shaped), singly or grouped in tufts (“antennal symphilous organs”). These “trichomes”, here assigned to 8 different morphotypes of sensilla chaetica (Ch.1–Ch.8), show a variable number of basal pores (present also at the base of the taste sensilla Ch.9), which spread dense substances of unknown chemical composition on the seta. Although hygro-, thermo- and chemoreceptors are reduced in number as compared with non-myrmecophilous relatives, and mainly relegated to the apex of the antennal club, their diversity is comparable to that of other carabid beetles: two types of sensilla trichodea (Tr.1–Tr.2); three types of basiconica (Ba.1–Ba.3); one type of campaniformia (Ca); one type of coeloconica (Co) and one type of Böhm sensilla (Bo). Contrary to the hypothesis that Paussus species lack a Johston's organ, a non-connective chordotonal organ composed of 9 groups of scolopidia has been found inside the pedicel. A comparison between sensilla of P. favieri and those of other non-myrmecophilous and myrmecophilous ground beetle species is provided.  相似文献

Iridoids, belonging to a group of cyclopentanoid monoterpenoids, are secreted by many species of leaf beetles as a defense against predators. Using chemically modified precursors of iridoid biosynthesis, it has been shown that some leaf beetle larvae can synthesize these iridoids de novo as well as sequester plant-produced molecules. Stable isotope techniques can provide useful methods for studying terpenoid biosynthesis without disturbing the natural conditions much. Two terpenoid biosynthesis pathways (mevalonic acid (MVA) pathway and methylerythritol-4-phosphate (MEP) pathway) may lead to different delta13C signatures of the products. Our results from natural abundance 13C and 13C-labelled iridoid precursors in Gastrophysa viridula and Phaedon cochleariae suggested that the two leaf beetle species use only de novo synthesis of their defensive iridoids. We observed that the isotope signature of the leaf-beetle-produced iridoids (via the MVA pathway) resembled that of the MEP-derived monoterpenoids from plants. Owing to this close similarity in the natural 13C abundances in the plant and insect compounds, a determination of iridoid-origin in leaf beetle secretion may only be possible by use of isotopically labelled compounds.  相似文献

The parasitoid beetle Dastarcus helophoroides (Fairmaire) (Coleoptera: Bothrideridae) is an important parasite of longicorn beetles (Cerambycidae), and has been used in China for the biological control of the Asian longicorn beetle and the Japanese pine sawyer. In this study the antennal morphology and sensillar ultrastructure of D. helophoroides were observed using scanning electron microscopy and transmission electron microscopy. Two types of sensilla trichodea (Tr. 1 and Tr. 2), two types of sensilla basiconica (Ba. 1 and Ba. 2), three types of sensilla chaetica (Ch. 1, Ch. 2 and Ch. 3), and Bohm's bristles were identified according to the morphology and fine structure of each type of sensilla in both sexes. Ultrastructural studies revealed porous structures on the cuticle wall and dendritic branches in the inner lumen of Tr. 1, Tr. 2, Ba. 1, and Ba. 2, thereby suggesting chemoreceptor functions. No difference in shape, structure, sensilla distribution and typology was observed between the sexes. These structures likely have roles in the host locating and habitat searching behavior of adult D. helophoroides, and suggest future studies on the olfaction and host location behavior of D. helophoroides and other coleopteran parasitoids.  相似文献

Carabus lefebvrei Dejean 1826 is an helicophagous Italian endemic ground beetle that lives in central and south Apennines mountain forests, from lower altitudes to about 1500 m. In ground beetles, no morphofunctional data about immune system is available, even though they are well known both taxonomically and ecologically and they have been often used as indicators of the habitat quality due to their specificity to certain habitat types. In the current investigation the cellular population in the hemolymph of adult and third instar larvae of C. lefebvrei has been characterized by means of light and electron microscopy analysis and phagocytosis assays were performed in vivo by injection of 0.9 microm carboxylate-modified polystyrene latex beads in order to identify the hemocyte types involved in phagocytosis. Four morphotypes of circulating hemocytes were found both in larvae and in adults: prohemocytes, granulocytes, oenocytoids and plasmatocytes. After in vivo artificial non-self-challenge treatments, C. lefebvrei showed a non-specific immune response involving phagocytosis performed by plasmatocytes, both in adults and in larvae and by oenocitoids in larvae. In untreated animals, the hemocyte type presenting a firm phagocytic activity, the plasmatocytes, presented a percentage significantly higher in larvae than in adults, and after latex beads injections in larvae there was a tendency of significant difference in plasmatocyte percentage compared to controls injected with phosphate saline buffer. We think that these differences could be correlated with the peculiar morphology (less chitinization) and ecology of larval stages that are more sensitive to pathogens than adults.  相似文献

Total lifetime distribution analysis was employed to obtain fluorescence lifetime profiles of the intrinsic fluorescence ofPseudomonas fluorescens, Escherichia coli, Bacillus subtilis, andStaphylococcus epidermidis. The lifetimes were measured using a multiharmonic Fourier transform phase-modulation fluorometer which can simultaneously measure the phase shift and demodulation at many modulation frequencies. The 364-nm line from an argon-ion laser and the 325- and 442-nm lines from a helium-cadmium laser were used for sample excitation. Broad emission windows were used to capture as much of the bacterial emission as possible for the lifetime measurements. The maximum entropy method was used to recover lifetime profiles from the multifrequency phasemodulation data. At all three excitation wavelengths, the bacteria exhibited three lifetime components, in the ranges of 0.5-1, 2–3, and 4–8 ns. Using 325-nm excitation, a fourth component, in the range of 9–14 ns, was recovered in all of the bacteria; using 364-nm excitation, the fourth component was resolved only in the two Gram-negative bacteria (P. fluorescens andE. coli). Excitation at 364 nm provided the most reproducible lifetime profiles and showed some differences among the four bacteria.  相似文献

DING proteins constitute an intriguing family of phosphate-binding proteins that was identified in a wide range of organisms, from prokaryotes and archae to eukaryotes. Despite their seemingly ubiquitous occurrence in eukaryotes, their encoding genes are missing from sequenced genomes. Such a lack has considerably hampered functional studies. In humans, these proteins have been related to several diseases, like atherosclerosis, kidney stones, inflammation processes and HIV inhibition. The human phosphate binding protein is a human representative of the DING family that was serendipitously discovered from human plasma. An original approach was developed to determine ab initio the complete and exact sequence of this 38 kDa protein by utilizing mass spectrometry and X-ray data in tandem. Taking advantage of this first complete eukaryotic DING sequence, a immunohistochemistry study was undertaken to check the presence of DING proteins in various mice tissues, revealing that these proteins are widely expressed. Finally, the structure of a bacterial representative from Pseudomonas fluorescens was solved at sub-angstrom resolution, allowing the molecular mechanism of the phosphate binding in these high-affinity proteins to be elucidated.  相似文献

We used scanning and transmission electron microscopy to study the typology, morphology, distributions, and ultrastructures of the antennal sensilla of Tetrigus lewisi Candèze, a predatory click beetle that feeds on longhorned beetles, such as, Monochamus alternatus (Coleoptera: Cerambycidae). We observed eight types of sensilla on the antennae, including sensilla chaetica (with three subtypes: ch.1, ch.2, ch.3), sensilla basiconica (subtypes: ba.1, ba.2, ba.3), sensilla trichodea (subtypes: tr.1, tr.2), as well as sensilla auricillica, sensilla coeloconica, sensilla campaniformia, sensilla styloconica and Böhm's bristles. Significant sexual dimorphism was found in the antenna morphology, as well as in the density of type 2 sensilla trichodea and type 1 sensilla basiconica. We observed thick cuticular walls on sensilla chaetica, sensilla trichodea and sensilla campaniformia; clear pore structures on sensilla trichodea, sensilla basiconica and sensilla auricillica; and double walls with spoke-channels on sensilla coeloconica. The chemoreception, mechanoreception and thermo-/hygro-reception functions were deduced from fine structures on the cuticular walls and the dendrites of the different sensilla types. We suggest that all these sensilla have important roles in the host location, mating and predatory behavior of T. lewisi.  相似文献

The morphology and ultrastructure of the hindgut fermentation chamber of a melolonthine beetle, Holotrichia parallela, were examined using light and electron microscopy. The results showed that the anterior portion of the hindgut expanded into a characteristic lobe-like structure described as a fermentation chamber. Hematoxylin-eosin staining revealed that the wall of the fermentation chamber was composed of three main layers: the longitudinal muscle layer, the circular muscle layer, and the columnar epithelial cells. Scanning electron microscopy showed that the ultrastructure of the inner surface of the fermentation chamber was subjected to significant changes during larval development. Only some folds and a few cocci attached to the folds were found in the first-instar larvae. In the fermentation chambers of the second-instar larvae, a cuticular intima began to emerge, and the bacteria multiplied to form bacterial groups attached to the intima. A special lobe-like structure was formed in the third-instar larvae, constituted by bacteria and the bacteria-covered cuticular intima. Transmission electron microscopy revealed that the lobe-like structure held large numbers of rod-shaped bacteria. These data suggest that the hindgut fermentation chamber may have an important role in the symbiotic relationship between microbes and their insect hosts.  相似文献

It has been previously hypothesized that the high fractional anisotropy (FA) values in leptomeningeal cortical subcortical white matter (LCSWM) regions of neonatal brain with bacterial meningitis is due to the presence of adhesion molecules in the subarachnoid space, which are responsible for adherence of inflammatory cells over the subarachnoid membrane. The aim of this study was to look for any relationship between FA values in LCSWM regions and various neuroinflammatory molecules (NMs) in cerebrospinal fluid (CSF) measured in neonates with bacterial meningitis. Diffusion tensor imaging was performed on 18 term neonates (median age, 10.5 days) having bacterial meningitis and 10 age-/sex-matched healthy controls. CSF enzyme-linked immunosorbent assay was performed to quantify NMs [soluble intracellular adhesion molecules (sICAM), tumor necrosis factor-alpha (TNF-alpha) and interleukin-1beta (IL-1beta)]. Significantly increased FA values were observed in LCSWM regions of the patients compared to controls. A significant positive correlation was observed between FA values in LCSWM regions and NMs [sICAM (r=0.67, P=.006), TNF-alpha (r=0.69, P=.005) and IL-1beta (r=0.82, P=.000)] in CSF of these patients. No difference in FA values (P=.99) in LCSWM regions was observed between patients with sterile (0.12+/-0.02) and culture-positive CSF study (0.12+/-0.02). FA may be used as noninvasive surrogate marker of NMs in neonatal meningitis in assessing therapeutic response in future.  相似文献

An insect antifreeze protein, MpAFP698, from the desert beetle Microdera punctipennis, shares 77% similarity with TmTHP (YL-3) from Tenebrio molitor. The predicted tertiary structure for MpAFP698 was modeled as a regular A-helix with TCT motifs arrayed to form the putative ice-binding surface. This model was validated via site-directed mutagenesis. The results suggest that the desert beetle in central Asia has evolved antifreeze proteins similar to those from North America continent. In an additional study, the recombinant MpAFP698 and its mutants were expressed in Escherichia coli strain BL21 (DE3). The thermal hysteresis activity of MpAFP698 was 1.73 degree C at 1.0 mg/ml. MpAFP698 was also tested by in vitro antifreeze activity assay to evaluate its cryoprotective effect at -20 degree C. MpAFP698 displays high cryoprotective effect on bacteria cells at freezing temperatures.  相似文献

从四川盆地紫色土中分离获得一株革兰氏阳性细菌,对该菌株形态学及16S序列分析,初步鉴定属于芽孢杆菌属。本文主要分析了一定剂量γ辐照前后该菌株对Sr2+的吸附效果。结果显示:实验辐照条件对该菌株吸附Sr2+有一定抑制作用,主要表现为其对Sr2+吸附效率达到90%以上的平衡点出现时间比无辐照情况下延迟了1.5 d出现,生长速率最大值出现延迟了0.5 d。SEM分析发现辐照后菌体出现不规则变形,说明一定剂量γ辐照会对所选细菌产生明显损伤作用。FTIR分析显示,Sr2+与辐照对Strain No.4菌体有一定协同损伤作用,且处于对数生长期的菌体比迟滞生长期更易受辐射损伤。这种损伤主要表现为菌体蛋白质酰胺基团和多糖中糖环、蛋白质分子中甲基等基团对应谱峰变化。  相似文献

The cytotoxic effect of positively charged polystyrene latex nanoparticles (PSL NPs) was compared between planktonic bacterial cells and bacterial biofilms using confocal laser scanning microscopy, atomic force microscopy, and a colony counting method. Pseudomonas fluorescens, which is commonly used in biofilm studies, was employed as the model bacteria. We found that the negatively charged bacterial surface of the planktonic cells was almost completely covered with positively charged PSL NPs, leading to cell death, as indicated by the NP concentration being greater than that required to achieve single layer coverage. In addition, the relationship between surface coverage and cell viability of P. fluorescens cells correlated well with the findings in other bacterial cells (Escherichia coli and Lactococcus lactis). However, most of the bacterial cells that formed the biofilm were viable despite the positively charged PSL NPs being highly toxic to planktonic bacterial cells. This indicated that bacterial cells embedded in the biofilm were protected by self-produced extracellular polymeric substances (EPS) that provide resistance to antibacterial agents. In conclusion, mature biofilms covered with EPS exhibit resistance to NP toxicity as well as antibacterial agents.  相似文献

Investigation of flapping wings of insect are focused on low Reynolds number effect and the unsteady aerodynamic properties. Interaction between flapping wing of insects and the air flow became one of important and fundamental research topics in micro air vehicle. The present work is aim to investigate the flow behavior of flapping wings of tethered scarab beetle. The generation mechanisms of velocity field and vortex formation are visualized with smoke-wire method. Tethered flight of the drone beetle shows the motion with elastic deformation of flapping wing. Measured flapping frequency is about 71 Hz and its frequency is higher than for dragonfly and butterfly. Beetle decreases negative lift by feathering motion in the upstroke process and increase positive lift by effect of wake capture in the downstroke process.  相似文献

Abstract The dinitrogen fixation activity of Azospirillum sp., and Pantoea agglomerans strains was determined by (15)N(2) incorporation after incubation with (15)N(2) labeled air or/and by acetylene reduction. These bacterial strains were able to fix N(2) both in pure culture and in association with wheat plants in hydroponics. Nitrogenase activity of Azospirillum sp., in pure culture was more rapidly inhibited by the addition of NH(4) (+) than NO(3) (-). The N(2) fixation of P. agglomerans decreased only by NH(4) (+) -addition, but was stimulated by NO(3) (-). Nitrogen fixation in association with wheat plants remained unaffected by both N compounds. However, nitrogen derived from the atmosphere (N(dfa)) contributed only very little to the overall nitrogen nutrition of the plants.  相似文献
