This perspective article mainly focuses on the development and applications of a pseudobond ab initio QM/MM approach to study enzyme reactions. The following aspects of methodology development are discussed: the approaches for the QM/MM covalent boundary problem, an efficient iterative optimization procedure, the methods to determine enzyme reaction paths, and the approaches to calculate free energy change in enzyme reactions. Several applications are described to illustrate the capability of the methods. Finally, future directions are discussed.  相似文献

药物代谢过程是药物在体内产生药效和毒性的主要过程,发展廉价、方便、快速、高通量的体外药物代谢研究方法对新药的开发和设计、给药的方法和剂量、临床药物的检测等都有重要的指导意义. 细胞色素P450酶(CYP450酶)在药物的I相反应中起到关键作用,以电极代替辅酶NADPH提供CYP450酶催化反应过程中需要的两个电子,构建CYP450酶电化学生物传感器可实现药物的初步筛选. 大量研究表明,CYP450酶在电极表面合适的固定方法与电极材料可有效提高传感器的检测性能. 本文主要综述近年来CYP450酶电化学生物传感器的构建及其在药物代谢研究方面的应用,并展望其研发前景.  相似文献

Biosensors are projected to find many applications due to their high selectivity and sensitivity, rapid reaction, economy and ease of handling in field measurements. Even though biosensors for a wide range of environmental pollutants have been extensively reported in the literature, the decision to develop a suitable biosensing system that can be approved by a regulatory perspective for environmental applications is fraught with technical issues. These issues mainly concern the biological recognition element, the physico-chemical transducer and the interfaces between the biological and the physical components, but also aspects of fluidics, electronics, and software for data processing. This article reviews methods together with a process to move biosensor technology from research laboratories to market, focusing as a case in point on challenges and possible opportunities in the development of photosynthetic-based biosensors for environmental applications.  相似文献

This review presents recent research conducted on the development of various protein-polysaccharide conjugates, their functional properties and industrial applications. These conjugates are formed by the glycosylation of food proteins with carbohydrates via the Maillard reaction and are capable of improving the functional properties of proteins. The Maillard reaction facilitates covalent bonding between a reducing group of a carbohydrate and an amino group of a protein under controlled conditions of temperature, time, pH, and relative humidity. There is a great deal of interest in modifying the functional properties of proteins and in the use of novel conjugates for various industrial applications. This review discusses various methods and their implications for preparing and characterising these conjugates. Furthermore, the physicochemical properties of conjugates such as solubility, thermal stability, emulsifying activity, emulsion stabilising properties, gelling and foaming properties are also analysed. A novel processing technology, a spinning disc reactor, could be an alternative process for the production of protein–polysaccharide conjugates, with desirable functionality in different food systems.  相似文献

Biosensing methods utilize the intrinsic selectivity of a biorecognition process to create relatively simple, low cost, analytical alternatives for a variety of research investigations. Here, biosensor applications of the piezoelectric quartz crystal (PQC) are reviewed. The discussion is divided into sections focusing on the development of PQC based analytical techniques, applications in solution phase sensing pertaining to PQC biosensors, and the current state of knowledge in PQC biosensing applications. Immobilization procedures, dip and dry assay techniques, and solution phase sensing methods are considered in detail.  相似文献

2,4-噻唑烷二酮是一种重要的医药中间体,已经被广泛应用于抗高血糖、抗癌、抗菌、抗病毒等方面,具有广阔的市场前景。首先,通过逆合成法对2,4-噻唑烷二酮五元环的构建方式进行系统分析,按照硫源的引入方式将合成方案分为硫脲法、硫氰酸盐法、羰基硫法和罗丹宁法四类,并对2,4-噻唑烷二酮合成过程中的取代、环化、水解反应机制进行探讨。其次,探究了不同合成方法的反应机理,阐明不同反应原料的反应速率、原子经济性,并针对各自的优缺点进行了评价。最后,根据逆合成分析结果提出两种潜在的合成策略,为开发绿色高效、环境友好、适合工业化生产的2,4-噻唑烷二酮及其类似结构药物的合成方法提供理论基础和研究思路。  相似文献

Since the landmark publications of the first directed aldol addition reaction in 1973, the site, diastereo‐, and enantioselective aldol reaction has been elevated to the rarefied status of being both a named and a strategy‐level reaction (the Mukaiyama directed aldol reaction). The importance of this reaction in the stereoselective synthesis of untold numbers of organic compounds, both natural and unnatural, cannot be overstated. However, its impact on the field extends beyond the impressive applications in synthesis. The directed aldol reaction has served as a fertile proving ground for new concepts and new methods for stereocontrol and catalysis. This Minireview provides a case history of how the challenges of merging site selectivity, diastereoselectivity, enantioselectivity, and catalysis into a unified reaction manifold stimulated the development of Lewis base catalyzed aldol addition reactions. The evolution of this process is chronicled from the authors’ laboratories as well as in those of Professor Teruaki Mukaiyama.  相似文献

This paper is an overview of the theory of reactive scattering, with emphasis on fully quantum mechanical theories that have been developed to describe simple chemical reactions, especially atom-diatom reactions. We also describe related quasiclassical trajectory applications, and in all of this review the emphasis is on methods and applications concerned with state-resolved reaction dynamics. The review first provides an overview of the development of the theory, including a discussion of computational methods based on coupled channel calculations, variational methods, and wave packet methods. Choices of coordinates, including the use of hyperspherical coordinates are discussed, as are basis set and discrete variational representations. The review also summarizes a number of applications that have been performed, especially the two most comprehensively studied systems, H+H2 and F+H2, along with brief discussions of a large number of other systems, including other hydrogen atom transfer reactions, insertion reactions, electronically nonadiabatic reactions, and reactions involving four or more atoms. For each reaction we describe the method used and important new physical insight extracted from the results.  相似文献

The most important topics in the world today are environmental and resource issues. The development of green and clean energy is still one of the great challenges of social sustainable development. Two-dimensional(2D) metal-organic frameworks(MOFs) and derivatives have exceptional potential as high-efficiency electrocatalysts for clean energy technologies. This review summarizes various synthesis strategies and applications of 2D MOFs and derivatives in electrocatalysis. Firstly, we will outline the advantages and uniqueness of 2D MOFs and derivatives, as well as their applicable areas. Secondly, the synthetic strategies of 2D MOFs and derivatives are briefly classified. Each category is summarized and we list classic representative fabrication methods, including specific fabrication methods and mechanisms, corresponding structural characteristics, and insights into the advantages and limitations of the synthesis method. Thirdly, we separately classify and summarize the application of 2D MOFs and derivatives in electrocatalysis, including electrocatalytic water splitting, oxygen reduction reaction(ORR), CO2 reduction reaction(CO2RR), and other electrocatalytic applications. Finally, the development prospects and existing challenges to 2D MOFs and derivatives are discussed.  相似文献

We review high-performance liquid chromatography (HPLC) methods for the determination of two major statins used in clinical treatment – simvastatin and atorvastatin – in various fields of application, including bio-analytical assays, pharmaceutical assays and environmental applications.

Statin molecules are known to be susceptible to interconversion of the lactone and acidic forms, so it is necessary to consider this phenomenon during method development. We highlight liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) methods, as they have become a method of choice in bio-analytical and environmental applications. We compare the methods from the point of view of sensitivity. We discuss selection of the precursor ion for performing selected reaction monitoring (SRM) in MS detection and sample preparation.  相似文献

Miniaturized sample preparation methods designed as the sample pretreatment for liquid phase separations, such as liquid chromatography, capillary electrophoresis and capillary electrochromatography, have been reviewed especially for the on-line coupling of the sample preparation process and the separation process. The development of the desorption interfaces for the effective combining of the sample preparation and subsequent liquid phase separations is briefly described along with the applications of the combined analytical systems to the analysis of complex sample mixtures such as biological and environmental matrices. Novel use of fine polymeric filaments as the extraction medium for microscale liquid phase separation methods are investigated and a comparison is made with other sample preparation techniques. Polymer coating onto the fibrous material is also introduced to further develop microscale sample preparation methods with improved extraction performance. Several other microscale sample preparation methods having a potential compatibility to the liquid phase separations are also described for future applications of these techniques.  相似文献

This review highlights and critically assesses forensic applications in the developing field of ambient ionization mass spectrometry. Ambient ionization methods permit the ionization of samples outside the mass spectrometer in the ordinary atmosphere, with minimal sample preparation. Several ambient ionization methods have been created since 2004 and they utilize different mechanisms to create ions for mass-spectrometric analysis. Forensic applications of these techniques—to the analysis of toxic industrial compounds, chemical warfare agents, illicit drugs and formulations, explosives, foodstuff, inks, fingerprints, and skin—are reviewed. The minimal sample pretreatment needed is illustrated with examples of analysis from complex matrices (e.g., food) on various substrates (e.g., paper). The low limits of detection achieved by most of the ambient ionization methods for compounds of forensic interest readily offer qualitative confirmation of chemical identity; in some cases quantitative data are also available. The forensic applications of ambient ionization methods are a growing research field and there are still many types of applications which remain to be explored, particularly those involving on-site analysis. Aspects of ambient ionization currently undergoing rapid development include molecular imaging and increased detection specificity through simultaneous chemical reaction and ionization by addition of appropriate chemical reagents.  相似文献

纤维素直接催化转化制乙二醇是一条极具吸引力的生物质转化途径,有助于减轻化石能源资源的消耗。综述了从该反应途径的发现到获得高效、高稳定性催化剂的快速发展过程。基于对钨基催化剂的大量研究结果,本文讨论了反应机制,明确了反应路径、催化剂状态、钨物种及加氢催化活性中心各自在串联反应中的作用。围绕该反应过程的工业化应用需要,讨论了有关原生木质纤维素生物质催化转化以及高效反应过程的发展策略。在此基础上,将纤维素催化转化制乙二醇过程与生物质发酵制丙酮-丁醇-乙醇的生物炼制路线进行整合,构建出一个理想的反应过程潜在应用范例。最后,对纤维素催化转化制乙二醇反应过程进行了总结和前景展望.  相似文献

Microreaction technology, which is an interdisciplinary science and engineering area, has been the focus of different fields of research in the past few years. Several microreactors have been developed. Enzymes are a type of catalyst, which are useful in the production of substance in an environmentally friendly way, and they also have high potential for analytical applications. However, not many enzymatic processes have been commercialized, because of problems in stability of the enzymes, cost, and efficiency of the reactions. Thus, there have been demands for innovation in process engineering, particularly for enzymatic reactions, and microreaction devices represent important tools for the development of enzyme processes. In this review, we summarize the recent advances of microchannel reaction technologies especially for enzyme immobilized microreactors. We discuss the manufacturing process of microreaction devices and the advantages of microreactors compared to conventional reaction devices. Fundamental techniques for enzyme immobilized microreactors and important applications of this multidisciplinary technology are also included in our topics.  相似文献

Original kinetic investigation methods allow the estimation of equilibrium characteristics of a process supposed to be governed by only one regulating component reaction.New notions, especially concerning fictitious advancement steps (fictitious because of arbitrary choices) are introduced. Several applications in heterogeneous kinetics are discussed.In particular, an attempt is made to specify the limits of application of these new notions with the purpose to estimate the true advancement steps, even without an experimental equilibrium approach.  相似文献

Thermal analysis methods are used in process development for the investigation of chemical kinetics, for the evaluation of thermal hazards and for the determination of caloric design data. For these applications heat-flow methods are preferably used.This paper gives a survey of methods and instruments, and then presents a new “bench scale heat-flow calorimeter”. The use of this new instruments is illustrated by investigations on the formation of a Grignard compound and on the isomerisation of trimethyl-phosphite.  相似文献
