The preferential solvation parameters of methocarbamol in dioxane + water, ethanol + water, methanol + water and propylene glycol + water mixtures are derived from their thermodynamic properties by using the inverse Kirkwood–Buff integrals (IKBI) method. This drug is sensitive to solvation effects, being the preferential solvation parameter δx1,3, negative in water-rich and co-solvent-rich mixtures, but positive in mixtures with similar proportions of solvents, except in methanol + water mixtures, where positive values are found in all the methanol-rich mixtures. It is conjecturable that the hydrophobic hydration around the non-polar groups in water-rich mixtures plays a relevant role. Otherwise, in mixtures of similar solvent compositions, the drug is mainly solvated by co-solvent, probably due to the basic behaviour of the co-solvents; whereas, in co-solvent-rich mixtures, the preferential solvation by water could be due to the acidic behaviour of water. Nevertheless, the specific solute–solvent interactions present in the different binary systems remain unclear.  相似文献

Preferential solvation parameters of etoricoxib in several aqueous cosolvent mixtures were calculated from solubilities and other thermodynamic properties by using the IKBI method. Cosolvents studied were as follows: 1,4-dioxane, N,N-dimethylacetamide, 1,4-butanediol, N,N-dimethylformamide, ethanol and dimethyl sulfoxide. Etoricoxib exhibits solvation effects, being the preferential solvation parameter δx1,3, negative in water-rich and cosolvent-rich mixtures but positive in mixtures with similar proportions of both solvents. It is conjecturable that the hydrophobic hydration in water-rich mixtures plays a relevant role in drug solvation. In mixtures of similar solvent proportions where etoricoxib is preferentially solvated by the cosolvents, the drug could be acting as Lewis acid with the more basic cosolvents. Finally, in cosolvent-rich mixtures the preferential solvation by water could be due to the more acidic behaviour of water. Nevertheless, the specific solute–solvent interactions in the different binary systems remain unclear because no relation between preferential solvation magnitude and cosolvent polarities has been observed.  相似文献

Standard partial molar volumes of 1:1 salts in aqueous mixtures of ethanol (EtOH), dimethyl sulfoxide (DMSO) and acetonitrile (MeCN) at 298.15 K were obtained from the literature. In such mixtures there is evidence that preferential solvation occurs in the solvent shell around the ion where electrostriction takes place. Specifically, the anions are better solvated by the water whereas the cations are generally solvated by both the water and the nonaqueous component of the mixtures to various extents. There are no clear-cut criteria for how the measured volumes are to be apportioned between the ions in such mixtures. Various solvation models were used to estimate the volumes of the salts by calculation of the electrostriction around the ions. Only the taking into account of the preferential solvation of the ions in the solvation shell yielded calculated results of the standard partial molar volumes of the salts in agreement with the experimental data.  相似文献

Molecular dynamics simulations are used to examine the local solvation structure of single octane and perfluorooctane molecules in liquid water, methanol, acetonitrile, and aqueous mixtures of methanol and acetonitrile. The motivation is to obtain baseline information about the solvation of perfluorooctane by liquids used as the mobile phase in liquid chromatography and how it differs from the solvation of octane. While octane is uniformly solvated by both water and the second component, perfluorooctane is solvated by methanol and acetonitrile with the exclusion of water from the first solvation layer when the solvent is a mixture.  相似文献

Configurational-bias Monte Carlo simulations in the isobaric-isothermal ensemble (T = 323 K and p = 10 atm) were carried out to probe structural properties of an isolated n-octadecane chain solvated in water, methanol, water-rich, or methanol-rich mixtures and, for comparison, of an isolated chain in the gas phase and for neat liquid n-octadecane. The united-atom version of the TraPPE (transferable potentials for phase equilibria) force field was used to represent n-octadecane and methanol and the TIP-4P model was used for water. In all six environments, broad conformational distributions are observed and the n-octadecane chains are found to predominantly adopt extended, but not all-trans conformations. In addition, a small fraction of more collapsed conformations in which the chain ends approach each other is observed for aqueous hydration, the water-rich solvent mixture and the gas phase, but the simulation data do not support a simple two-state picture with folded and unfolded basins of attraction. For chains in these three "poor" solvent environments, the dihedral angles near the center of the chain show an enhancement of the gauche population. The ensemble of water-solvated chains with end-to-end contacts is preferentially found in a U-shaped conformation rather than a more globular state. An analysis of the local solvation structures in the water-methanol mixtures shows, as expected, an enrichment of the methyl group of methanol near the methylene and methyl segments of the n-octadecane chain. Interestingly, these local bead fractions are enhanced by factors of 2.5 and 1.5 for methyl and methylene segments reflecting the more hydrophobic nature of the former segments.  相似文献

The solvation and reaction of ethylene radical cation in aqueous solution has been studied with Car-Parrinello molecular dynamics simulations. All ab initio simulations were performed using a system of 56 water and one ethylene molecule. Using a favorable symmetrically solvated radical cation as the starting point of the simulation a fast addition of water (within 90 fs) to the radical cation is observed. The primary addition product is rapidly deprotonated (within 100 fs) to yield the ethanol-2-yl radical. A second simulation was initiated through vertical ionization of neutral hydrated ethylene, representing a significantly less favorable situation for the addition process. No addition of water can be observed in this second simulation over a time span of 1.7 ps. Taken together the two simulations are indicative of a rearrangement of the solvent shell which represents the major part of the overall reaction barrier. Under these circumstances, the reaction rate of an otherwise spontaneous reaction is limited by the intrinsic solvent relaxation time. This interpretation of the reactivity of hydrated radical cations reconciles previously conflicting experimental condensed phase and theoretical gas phase studies.  相似文献

Solid–liquid–liquid equilibrium data of the ternary systems water+LiCl+2-butanol, water+LiCl+2-methyl-1-propanol (i-butanol) and water+LiCl+1-butanol have been experimentally determined at 25°C. The equilibrium diagrams determined show differences between the systems. In the system with 1-butanol, the solid phase of the liquid–liquid–solid region is monohydrated salt. However, in the systems with 2-butanol and 2-methyl-1-propanol it is anhydrous salt. With respect to the liquid+liquid zone, the three diagrams are very similar with an unusual S-shaped solubility curve in the organic branch that can be explained depending on whether the organic solvent takes part in the solvation of ions. The more salt, the more numbers of ions solvated by water and organic solvent and the solubility of water and salt in the organic phase increase notably producing the unusual S-shaped solubility curve.  相似文献

The solubility of lamotrigine (LTG), clonazepam (CZP) and diazepam (DZP) in some {cosolvent (1) + water (2)} mixtures expressed in mole fraction at 298.15 K was calculated from reported solubility values expressed in molarity by using the densities of the saturated solutions. Aqueous binary mixtures of ethanol, propylene glycol and N-methyl-2-pyrrolidone were considered. From mole fraction solubilities and some thermodynamic properties of the solvent mixtures, the preferential solvation of these drugs by both solvents in the mixtures was analysed by using the inverse Kirkwood–Buff integrals. It is observed that LTG, CZP and DZP are preferentially solvated by water in water-rich mixtures in all the three binary systems analysed. In {ethanol (1) + water (2)} mixtures, preferential solvation by water is also observed in ethanol-rich mixtures. Nevertheless, in {propylene glycol (1) + water (2)} and {N-methyl-2-pyrrolidone (1) + water (2)} mixtures preferential solvation by the cosolvent was observed in cosolvent-rich mixtures.  相似文献

We study the solvation of polar molecules in water. The center of water's dipole moment is offset from its steric center. In common water models, the Lennard-Jones center is closer to the negatively charged oxygen than to the positively charged hydrogens. This asymmetry of water's charge sites leads to different hydration free energies of positive versus negative ions of the same size. Here, we explore these hydration effects for some hypothetical neutral solutes, and two real solutes, with molecular dynamics simulations using several different water models. We find that, like ions, polar solutes are solvated differently in water depending on the sign of the partial charges. Solutes having a large negative charge balancing diffuse positive charges are preferentially solvated relative to those having a large positive charge balancing diffuse negative charges. Asymmetries in hydration free energies can be as large as 10 kcal/mol for neutral benzene-sized solutes. These asymmetries are mainly enthalpic, arising primarily from the first solvation shell water structure. Such effects are not readily captured by implicit solvent models, which respond symmetrically with respect to charge.  相似文献

In contrast to the extensive theoretical investigation of the solvation phenomena, the dissolution phenomena have hardly been investigated theoretically. Upon the excitation of hydrated halides, which are important substances in atmospheric chemistry, an excess electron transfers from the anionic precursor (halide anion) to the solvent and is stabilized by the water cluster. This results in the dissociation of hydrated halides into halide radicals and electron-water clusters. Here we demonstrate the charge-transfer-to-solvent (CTTS)-driven femtosecond-scale dissolution dynamics for I-(H2O)n=2-5 clusters using excited state (ES) ab initio molecular dynamics (AIMD) simulations employing the complete-active-space self-consistent-field (CASSCF) method. The equilibrium solubility and preferential solvation of triclocarban in {1,4-dioxane (1) + water (2)} mixtures at 298.15 K was reported. Mole fraction solubility varies continuously from 2.85 × 10–9 in neat water to 2.39 × 10–3 in neat 1,4-dioxane. Solubility behaviour was adequately correlated by means of the Jouyban-Acree model. Based on the inverse Kirkwood-Buff integrals, preferential solvation parameters were calculated. Triclocarban is preferentially solvated by water in water-rich mixtures (0.00 < x1 < 0.18) and also in 1,4-dioxane-rich mixtures (0.78 < x1 < 1.00) but preferentially solvated by 1,4-dioxane in mixtures with similar solvent compositions.  相似文献

Raman spectra of concentrated solutions of LiCl in HCOOH containing mainly ion pairs and single solvated ions prove to be suitable for an approximate experimental estimation of the influence of the ion on the HCOOH molecule. Remarkable changes in the CO, CO and CH bonds of the solvent molecule due to this influence are observed. These observations are shown to be in good agreement with calculated frequency shifts, obtained by semi-empirical molecular orbital methods applied to simple solvation models.  相似文献

The preferential solvation of solutes in mixed solvent systems is an interesting phenomenon that plays important roles in solubility and kinetics. In the present study, solvation of a lithium atom in aqueous ammonia solution has been investigated from first principles molecular dynamics simulations. Solvation of alkali metal atoms, like lithium, in aqueous and ammonia media is particularly interesting because the alkali metal atoms release their valence electrons in these media so as to produce solvated electrons and metal counterions. In the present work, first principles simulations are performed employing the Car-Parrinello molecular dynamics method. Spontaneous ionization of the Li atom is found to occur in the mixed solvent system. From the radial distribution functions, it is found that the Li(+) ion is preferentially solvated by water and the coordination number is mostly four in its first solvation shell and exchange of water molecules between the first and second solvation shells is essentially negligible in the time scale of our simulations. The Li(+) ion and the unbound electron are well separated and screened by the polar solvent molecules. Also the unbound electron is primarily captured by the hydrogens of water molecules. The diffusion rates of Li(+) ion and water molecules in its first solvation shell are found to be rather slow. In the bulk phase, the diffusion of water is found to be slower than that of ammonia molecules because of strong ammonia-water hydrogen bonds that participate in solvating ammonia molecules in the mixture. The ratio of first and second rank orientational correlation functions deviate from 3, which suggests a deviation from the ideal Debye-type orientational diffusion. It is found that the hydrogen bond lifetimes of ammonia-ammonia pairs is very short. However, ammonia-water H-bonds are found to be quite strong when ammonia acts as an acceptor and these hydrogen bonds are found to live longer than even water-water hydrogen bonds.  相似文献

We present a benchmark study of a combined multipole shielding polarizability/reaction field (MSP/RF) approach to the calculation of both specific and bulk solvation effects on nuclear magnetic shielding constants of solvated molecules. The MSP/RF scheme is defined by an expansion of the shielding constants of the solvated molecule in terms of electric field and field gradient property derivatives derived from single molecule ab initio calculations. The solvent electric field and electric field gradient are calculated based on data derived from molecular dynamics simulations, thereby accounting for solute-solvent dynamical effects. The MSP/RF method is benchmarked against polarizable quantum mechanics/molecular mechanics (QM/MM) calculations. The best agreement between the MSP/RF and QM/MM approaches is found by truncating the electric field expansion in the MSP/RF approach at the linear electric field level which is due to the cancelation of errors. In addition, we investigate the sensitivity of the results due to the choice of one-electron basis set in the ab initio calculations of the property derivatives and find that these derivatives are affected by the basis set in a way similar to the shielding constants themselves.  相似文献

The solvation of simple ions in water is studied using molecular dynamics simulations with a polarizable force field. Previous simulations using this potential demonstrated that anions are more favorably solvated in water than cations. The present work is an attempt to explain this result by examining the effects of ions on the surrounding water structure, with particular focus on the first solvation shell and its interactions with the surrounding water. We conclude that while the first solvation shell surrounding cations is frustrated by competition between ion-water and water-water interactions, solvation of anions is compatible with good water-water interactions.  相似文献

Theoretical simulations are used to investigate the effects of aqueous solvent on the vibrational spectra of model alpha-helices, which are only partly exposed to solvent to mimic alpha-helices in proteins. Infrared absorption (IR) and vibrational circular dichroism (VCD) amide I' spectra for 15-amide alanine alpha-helices are simulated using density functional theory (DFT) calculations combined with the property transfer method. The solvent is modeled by explicit water molecules hydrogen bonded to the solvated amide groups. Simulated spectra for two partially solvated model alpha-helices, one corresponding to a more exposed and the other to a more buried structure, are compared to the fully solvated and unsolvated (gas phase) simulations. The dependence of the amide I spectra on the orientation of the partially solvated helix with respect to the solvent and effects of solvation on the amide I' of 13C isotopically substituted alpha-helices are also investigated. The partial exposure to solvent causes significant broadening of the amide I' bands due to differences in the vibrational frequencies of the explicitly solvated and unsolvated amide groups. The different degree of partial solvation is reflected primarily in the frequency shifts of the unsolvated (buried) amide group vibrations. Depending on which side of the alpha-helix is exposed to solvent, the simulated IR band-shapes exhibit significant changes, from broad and relatively featureless to distinctly split into two maxima. The simulated amide I' VCD band-shapes for the partially solvated alpha-helices parallel the broadening of the IR and exhibit more sign variation, but generally preserve the sign pattern characteristic of the alpha-helical structures and are much less dependent on the alpha-helix orientation with respect to the solvent. The simulated amide I' IR spectra for the model peptides with explicitly hydrogen-bonded water are consistent with the experimental data for small alpha-helical proteins at very low temperatures, but overestimate the effects of solvent on the protein spectra at ambient temperatures, where the peptide-water hydrogen bonds are weakened by thermal motion.  相似文献

We report here infrared spectra of protonated and lithiated valine with varying degrees of hydration in the gas phase and interpret them with the help of DFT calculations at the B3LYP/6-31++G** level. In both the protonated and lithiated species our results clearly indicate that the solvation process is driven first by solvation of the charge site and subsequently by formation of a second solvation shell. The infrared spectra of Val x Li+ (H2O)4 and Val x H+ (H2O)4 are strikingly similar in the region of the spectrum corresponding to hydrogen-bonded stretches of donor water molecules, suggesting that in both cases similar extended water structures are formed once the charge site is solvated. In the case of the lithiated species, our spectra are consistent with a conformation change of the amino acid backbone from syn to anti accompanied by a change in the lithium binding from a NO coordination to OO coordination configuration upon addition of the third water molecule. This change in the mode of metal ion binding was also observed previously by Williams and Lemoff [J. Am. Soc. Mass Spectrom. 2004, 15, 1014-1024] using blackbody infrared radiative dissociation (BIRD). In contrast to the zwitterion formation inferred from results of the BIRD experiments upon addition of a third water molecule, our spectra, which are a more direct probe of structure, show no evidence for zwitterion formation with the addition of up to four water molecules.  相似文献

The finite difference method for solving the Poisson–Boltzmann equation is used to calculate the reaction field acting on a macromolecular solute due to the surrounding water and ions. Comparisons with analytical test cases indicate that the solvation forces can be calculated rapidly and accurately with this method. These forces act to move charged solute atoms towards the solvent where they are better solvated, and to screen interactions between charges. A way of combining such calculations with conventional molecular dynamics force fields is proposed which requires little modification of existing molecular dynamics programs. Simulations on the alanine dipeptide show that solvent forces affect the conformational dynamics by reducing the preference for internal H-bonding forms, increasing the R-alpha helix preference and reducing transition barriers. These solvent effects are similar to previous explicit solvent simulations, but require little more computation than vacuum simulations. The method should scale up with little increase in computational cost to larger molecules such as proteins and nucleic acids.  相似文献

The preferential solvation parameters of ketoprofen (KTP) in ethanol (EtOH) + water and propylene glycol (PG) + water binary solvent mixtures were obtained from their thermodynamic properties by means of the inverse Kirkwood–Buff integrals (IKBI) and quasi-lattice quasi-chemical methods. According to the IKBI method, it is found that KTP is very sensitive to specific solvation effects, so the preferential solvation parameter by co-solvents, δx 1,3, is negative in the water-rich mixtures of both binary systems but positive in the other compositions at temperatures of 293.15, 303.15 and 313.15 K. From this it can be assumed that, in water-rich mixtures, hydrophobic hydration around the aromatic rings and the methyl group, present in the drug, plays a relevant role in the solvation. The bigger amount of drug solvation by the co-solvent in mixtures of similar solvent proportions and in co-solvent-rich mixtures could be due mainly to polarity effects. Moreover, in these mixtures the solute will be acting as a Lewis acid with the co-solvent molecules, because they are more basic than water.  相似文献
