Novel biomimetic composite was prepared by the reaction of collagen and poly(γ‐benzyl L ‐glutamate)‐co‐poly(glutamic acid) (PBLG‐co‐PGA), which were crosslinked by non‐toxic crosslinking reagents 1‐ethyl‐(dimethylaminopropyl) carbodiimide (EDC) and N‐hydroxysuccinimide (NHS). The composite was characterized by FTIR and DSC. FTIR results confirmed that the collagen in the composite was successfully crosslinked with PBLG‐co‐PGA. DSC results showed that the composites possessed higher shrinkage temperature and higher thermal stability than the collagen. The water absorption test showed that the water absorbency of the composites increased with the increase in PBLG‐co‐PGA content in the composite. The studies of collagenase degradation and the tensile strength showed that the biostability and the tensile strength of the composites were significantly improved in comparison with that of the collagen. According to the investigations of cell adherent ratio and cell proliferation in vitro, the composite possessed good biocompatibility. Copyright © 2007 John Wiley & Sons, Ltd.  相似文献

The hydrolytic degradability and erythromycin release from stoichiometric ionic complexes of biotechnological poly(beta,L-malic acid)s and poly(gamma,D-glutamic acid)s with alkyltrimethylammonium surfactants were investigated. The influence of pH, temperature and antibiotic load on hydrolysis rate was examined. It was found that poly(malic acid) complexes degraded by a surface erosion mechanism at a higher rate than poly(glutamic acid) complexes, which eroded in bulk. Erythromycin was lodged in the paraffinic subphase of the complexes and upon aging it was delivered according to a sigmoidal profile that appeared to be independent on the antibiotic load.  相似文献

The micro construction of poly(epsilon-caprolactone) (PCL) and poly(L-lactic acid) (PLLA) blend films fabricated by solution casting under microwave irradiation was investigated by selective enzymatic degradation and scanning electron microscopy (SEM). The results were totally different from the blends obtained by conventional methods. The blend was more homogeneous and the PCL continuous phase more compact as no spherulites and tiny zone separation were observed from the film surface and no PCL network was observed inside the film, and the degradation of a PCL plank by Pseudomonas lipase was significantly retarded. The distributed PLLA micro spheres were enlarged and amorphous. The thermal behavior of the blend by microwave heating revealed that PCL and PLLA underwent a melting process, which induced the variations of the PCL phase and PLLA spheres. The weight loss caused by degradation of the PCL/PLLA blend obtained by conventional methods (B50c) is greater than that of the blend obtained by microwave methods (B50m), which reflects the change in morphology from a loose PCL network (B50c) to a dense PCL plank (B50m).  相似文献

Comb-like ionic complexes, nATMA . PG(DL)GA, were prepared from microbial poly(gamma-glutamic acid), with a nearly racemic configuration, and alkyltrimethylammoniun bromides, with linear alkyl chains containing an even number of carbon atoms, n, ranging from 12 to 22. The complexes had a nearly stoichiometric composition, displayed thermal stability up to temperatures above 200 degrees C and were insoluble in water but soluble in organic solvents. In the solid state, they were arranged in a regularly layered structure with the alkyl side chain crystallized for n > or = 18. Heating above melting entailed a contraction in the interlayer distance which varied from 1 to 10% depending on the value of n. Comparison with data reported on similar complexes obtained from nearly enantiomerically pure poly(gamma-glutamic acid) revealed an overall behavior very similar for the two series but with specific significant differences concerning side chain crystallinity and dimensional response to temperature.  相似文献

The naturally occurring edible biopolymer poly(gamma-glutamic acid) (gamma-PGA) is shown to be an efficient chelating agent of vanadium(IV). The structure of poly(gamma-glutamic acid)oxovanadium(IV) (VO-gamma-PGA) complex in solution has been analyzed by electron spin resonance and UV-visible absorption spectra. The equatorial coordination sphere of vanadium(IV) is proposed to be [2 x carboxylate (2O)-VO-(OH2)2]. The binding isotherm is determined for suspensions of gamma-PGA in vanadium(IV) oxide sulfate (VS) solutions of different concentrations, and the data have been adjusted to fit the modified Langmuir equation. The maximum amount of vanadium bound per gram of gamma-PGA is estimated to be 141 mmol . g(-1) with a binding constant of 22 L . g(-1) at pH 3.  相似文献

In this work, electrically conductive polyaniline (PAni) doped with camphorsulfonic acid (CPSA) is blended with poly(L-lactide-co-epsilon-caprolactone) (PLCL), and then electrospun to prepare uniform nanofibers. The CPSA-PAni/PLCL nanofibers show a smooth fiber structure without coarse lumps or beads and consistent fiber diameters (which range from 100 to 700 nm) even with an increase in the amount of CPSA-PAni (from 0 to 30 wt.-%). However, the elongation at break decreases from 391.54 +/- 9.20% to 207.85 +/- 6.74% when 30% of CPSA-PAni is incorporated. Analysis of the surface of the nanofibers demonstrates the presence of homogeneously blended CPSA-PAni. Most importantly, a four-point probe analysis reveals that electrical properties are maintained in the nanofibers where the conductivity is significantly increased from 0.0015 to 0.0138 S x cm(-1) when the nanofibers are prepared with 30% CPSA-PAni. The cell adhesion tests using human dermal fibroblasts, NIH-3T3 fibroblasts, and C2C12 myoblasts demonstrate significantly higher adhesion on the CPSA-PAni/PLCL nanofibers than pure PLCL nanofibers. In addition, the growth of NIH-3T3 fibroblasts is enhanced under the stimulation of various direct current flows. The CPSA-PAni/PLCL nanofibers with electrically conductive properties may potentially be used as a platform substrate to study the effect of electrical signals on cell activities and to direct desirable cell function for tissue engineering applications.  相似文献

A series of activated urethane‐type derivatives of γ‐benzyl‐L ‐glutamate were synthesized, and their potential as monomers for polypeptide synthesis was investigated. The derivatives of the focus of this work were a series of N‐aryloxycarbonyl‐γ‐benzyl‐L ‐glutamate 1 , of which aryl groups were phenyl, 4‐chlorophenyl, and 4‐nitrophenyl. These urethanes 1 were reactive in polar solvents such as dimethylsulfoxide, N,N‐dimethylformamide (DMF), and N,N‐dimethylacetamide (DMAc), and were efficiently converted into poly(γ‐benzyl‐L ‐glutamate) (poly(BLG)) under mild conditions; at 60 °C without addition of any catalyst. Among the three urethanes, that having 4‐nitrophenoxycarbonyl group 1c was the most reactive to give poly(BLG) efficiently, as was expected from the highly electron deficient nature of the nitrophenoxycarbonyl group. On the other hand, the urethane 1a having phenoxycarbonyl group was also efficiently converted into poly(BLG), in spite of the intrinsically less electrophilicity of the phenoxycarbonyl group. In addition, the successful formation of poly(BLG) by the reaction of 1a favored its diluted concentration (0.1 M) much more than 2.0 M, the optimum initial concentration for 1c . 1H NMR spectroscopic analyses of the reactions in situ revealed that the predominant pathway from 1 to poly(BLG) involved the intramolecular cyclization of 1 into the corresponding N‐carboxyanhydride, with release of phenol and its successive ring‐opening polymerization with release of carbon dioxide. © 2008 Wiley Periodicals, Inc. AB block copolymers composed of hydrophilic poly(ethylene glycol) (PEG) and hydrophobic poly(amino acid) with a carboxyl group at the end of PEG were synthesized with α‐carboxylic sodium‐ω‐amino‐PEG as a macroinitiator for the ring‐opening polymerization of N‐carboxy anhydride. Characterizations by 1H NMR, IR, and gel permeation chromatography were carried out to confirm that the diblock copolymers were formed. In aqueous media this copolymer formed self‐associated polymer micelles that have a carboxyl group on the surface. The carboxyl groups located at the outer shell of the polymeric micelle were expected to combine with ligands to target specific cell populations. Phase equilibrium studies for semiconcentrated solutions of rodlike poly(γ‐benzyl L ‐glutamate) having oligo(ethylene glycol) as side chains (PBLG‐g‐OEG) have been investigated. The phase‐boundary concentrations in isotropic and anisotropic phases for N,N‐dimethylformamide (DMF) solution of PBLG‐g‐OEG with short side chains (PBLG2‐g‐380) are higher than those for solution of PBLG‐g‐OEG with long side chains (PBLG2‐g‐770). The lattice theory and the scaled particle theory for nematic solution, which don't distinguish the molecular architecture of the rodlike polymer, cannot explain this experimental result. Repulsive interaction between rodlike polymers by means of the attached side chains is proposed for the molecular orientation of PBLG‐g‐OEG in anisotropic state in order to describe the experimental result. Ternary phase diagrams of PBLG‐g‐OEG/poly(ethylene glycol) (PEG)/DMF show that the miscibility of rodlike PBLG‐g‐OEG and coiled PEG is most enhanced in the system of PBLG2‐g‐770, which has longest and largest amount of side chains. Single‐walled carbon nanotubes (SWCNTs) have been functionalized with poly(γ‐benzyl‐L ‐glutamate) (PBLG) by ring‐opening polymerizations of γ‐benzyl‐L ‐glutamic acid‐based N‐carboxylanhydrides (NCA‐BLG) using amino‐functionalized SWCNTs (SWCNT‐NH2) as initiators. The SWCNT functionalization has been verified by FTIR spectroscopy and transmission electron microscopy. The FTIR study reveals that surface‐attached PBLGs adopt random‐coil conformations in contrast to the physically absorbed or bulk PBLGs, which exhibit α‐helical conformations. Raman spectroscopic analysis reveals a significant alteration of the electronic structure of SWCNTs as a result of PBLG functionalization. The PBLG‐functionalized SWCNTs (SWCNT‐PBLG) exhibit enhanced solubility in DMF. Stable DMF solutions of SWCNT‐PBLG/PBLG with a maximum SWCNTs concentration of 259 mg L?1 can be readily obtained. SWCNT‐PBLG/PBLG solid composites have been characterized by differential scanning calorimetry, thermogravimetric analysis, wide/small‐angle X‐ray scattering (W/SAXS), scanning electron microscopy, and polarized optical microscopy for their thermal or morphological properties. Microfibers containing SWCNT‐PBLG and PBLG can also be prepared via electrospinning. WAXS characterization reveals that SWCNTs are evenly distributed among PBLG rods in solution and in the solid state where PBLGs form a short‐range nematic phase interspersed with amorphous domains. © 2010 Wiley Periodicals, Inc. Poly(3‐hexylthiophene)‐b‐poly(γ‐benzyl‐L ‐glutamate) (P3HT‐b‐PBLG) rod–rod diblock copolymer was synthesized by a ring‐opening polymerization of γ‐benzyl‐L ‐glutamate‐N‐carboxyanhydride using a benzylamine‐terminated regioregular P3HT macroinitiator. The opto‐electronic properties of the diblock copolymer have been investigated. The P3HT precursor and the P3HT‐b‐PBLG have similar UV–Vis spectra both in solution and solid state, indicating that the presence of PBLG block does not decrease the effective conjugation length of the semiconducting polythiophene segment. The copolymer displays solvatochromic behavior in THF/water mixtures. The morphology of the diblock copolymer depends upon the solvent used for film casting and annealing results in morphological changes for both films deposited from chloroform and trichlorobenzene.

To create a novel vector for specifically delivering anticancer therapy to solid tumors, we used diafiltration to synthesize pH‐sensitive polymeric micelles. The micelles, formed from a tetrablock copolymer [poly(ethylene glycol)‐b‐poly(L ‐histidine)‐b‐poly(L ‐lactic acid)‐b‐poly(ethylene glycol)] consisted of a hydrophobic poly(L ‐histidine) (polyHis) and poly(L ‐lactic acid) (PLA) core and a hydrophilic poly(ethylene glycol) (PEG) shell, in which we encapsulated the model anticancer drug doxorubicin (DOX). The robust micelles exhibited a critical micellar concentration (CMC) of 2.1–3.5 µg/ml and an average size of 65–80 nm pH 7.4. Importantly, they showed a pH‐dependent micellar destabilization, due to the concurrent ionization of the polyHis and the rigidity of the PLA in the micellar core. In particular, the molecular weight of PLA block affected the ionization of the micellar core. Depending on the molecular weight of the PLA block, the micelles triggering released DOX at pH 6.8 (i.e. cancer acidic pH) or pH 6.4 (i.e. endosomal pH), making this system a useful tool for specifically treating solid cancers or delivering cytoplasmic cargo in vivo. Ultrafine fibers of a laboratory‐synthesized new biodegradable poly(p‐dioxanone‐co‐L ‐lactide)‐block‐poly(ethylene glycol) copolymer were electrospun from solution and collected as a nonwoven mat. The structure and morphology of the electrospun membrane were investigated by scanning electron microscopy, differential scanning calorimetry (DSC), wide‐angle X‐ray diffraction (WAXD), and a mercury porosimeter. Solutions of the copolymer, ranging in the lactide fraction from 60 to 80 mol % in copolymer composition, were readily electrospun at room temperature from solutions up to 20 wt % in methylene chloride. We demonstrate the ability to control the fiber diameter of the copolymer as a function of solution concentration with dimethylformamide as a cosolvent. DSC and WAXD results showed the relatively poor crystallinity of the electrospun copolymer fiber. Electrospun copolymer membrane was applied for the hydrolytic degradation in phosphate buffer solution (pH = 7.5) at 37 °C. Preliminary results of the hydrolytic degradation demonstrated the degradation rate of the electrospun membrane was slower than that of the corresponding copolymers of cast film. © 2003 Wiley Periodicals, Inc. Well‐defined poly(L ‐lactide)‐b‐poly(ethylene oxide) (PLLA‐b‐PEO) copolymers with different branch arms were synthesized via the controlled ring‐opening polymerization of L ‐lactide followed by a coupling reaction with carboxyl‐terminated poly(ethylene oxide) (PEO); these copolymers included both star‐shaped copolymers having four arms (4sPLLA‐b‐PEO) and six arms (6sPLLA‐b‐PEO) and linear analogues having one arm (LPLLA‐b‐PEO) and two arms (2LPLLA‐b‐PEO). The maximal melting point, cold‐crystallization temperature, and degree of crystallinity (Xc) of the poly(L ‐lactide) (PLLA) block within PLLA‐b‐PEO decreased as the branch arm number increased, whereas Xc of the PEO block within the copolymers inversely increased. This was mainly attributed to the relatively decreasing arm length ratio of PLLA to PEO, which resulted in various PLLA crystallization effects restricting the PEO block. These results indicated that both the PLLA and PEO blocks within the block copolymers mutually influenced each other, and the crystallization of both the PLLA and PEO blocks within the PLLA‐b‐PEO copolymers could be adjusted through both the branch arm number and the arm length of each block. Moreover, the spherulitic growth rate (G) decreased as the branch arm number increased: G6sPLLA‐b‐PEO < G4sPLLA‐b‐PEO < G2LPLLA‐b‐PEO < GLPLLA‐b‐PEO. © 2006 Wiley Periodicals, Inc. A series of pyrenyl‐terminated poly(γ‐benzyl‐l ‐glutamate)s (py‐PBLGs) with controlled polymer molecular weight (MW = 2.3–14.8 kg mol?1) and molecular weight distribution (PDI = 1.17–1.55) have been prepared from 1‐pyrenemethylamine hydrochloride‐mediated ring‐opening polymerization (ROP) of γ‐benzyl‐l ‐glutamic acid based N‐carboxyanhydride (BLG‐NCA). FTIR analysis revealed that the py‐PBLG9 was conformationally heterogeneous with 35.0% α‐helix, 55.6% β‐sheet, and 9.4% random coil conformations in the solid state, whereas the py‐PBLG66 adopts 100% α‐helix conformation. Py‐PBLGs promote the dispersion of SWCNTs in organic solvents and in the PBLG solid through π–π interaction, as evidenced by the Raman spectroscopic studies. WAXD analysis revealed that the SWCNTs significantly affect the ordering of the py‐PBLG self‐assembly: the long range hexagonal packing of py‐PBLG66 rods is notably enhanced by the addition of SWCNTs, whereas the lamellar packing of py‐PGLG9 β‐sheets is weakened. In the hexagonal lattice, the SWCNTs are intercalated parallel to the py‐PBLG66 rods, in contrast to the normal orientation of the SWCNTs with respect to the extended py‐PBLG9 chains in the β‐sheets. The relative packing structure also affects the intermolecular interaction among the PBLGs: SWCNTs promote the interaction among the py‐PBLG9 chains packed in a lamellar structure and weaken the intermolecular interaction among the py‐PBLG66 columnar hexagonal array. © 2013 Wiley Periodicals, Inc. In this study, a novel drug‐carrying micelle composed of methoxy poly(ethylene glycol) (mPEG)‐b‐poly(L‐lactic acid) (PLLA) with gas‐forming carbonate linkage was fabricated. Here, the gas‐forming carbonate linkage was formed by the chemical coupling of the terminal hydroxyl group of the PLLA block and benzyl chloroformate (BC). mPEG‐b‐PLLA‐BC was self‐organized in aqueous solution: the PEG block on the hydrophilic outer shell and the PLLA‐BC block in the hydrophoboic innor core. The cleavage of carbonate linkage by hydrolysis and formation of carbon dioxide nanobubbles in the micellar core enabled an accelerated release of the encapsulated anticancer drug (doxorubicin: DOX) from the mPEG‐b‐PLLA‐BC micelles. The amount of drug (DOX) released from the mPEG‐b‐PLLA‐BC micelle was higher than that from the conventional mPEG‐b‐PLLA micelle, which allowed for increased in vitro toxicity against KB tumor cells. Poly (glycerol sebacate) (PGS) is a thermoset biodegradable elastomer considered as a promising candidate material for nerve applications. However, PGS synthesis is very time and energy consuming. In this study, the PGS pre‐polymer (pPGS) was synthesized using three synthesis times of 3, 5, and 7 hours at 170°C. Fourier transform infrared (FTIR), nuclear magnetic resonance spectroscopy, X‐ray diffraction analysis, and differential scanning calorimetry thermogram were utilized to study the pPGS behavior. Poly (vinyl alcohol) was used as a carrier to fabricate aligned poly (vinyl alcohol)‐poly (glycerol sebacate) (PVA‐PGS) fibers with various ratios (60:40, 50:50, and 40:60) using electrospinning and crosslinked through the thermal crosslinking method. Morphology of the fibers was studied before and after crosslinking using scanning electron microscopy (SEM). FTIR, mechanical properties in the dry and wet state, water contact angle, in vitro degradation, and water uptake behavior of crosslinked scaffolds were also investigated. 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide (MTT) assay, SEM analysis, and 4′, 6‐diamidino‐2‐phenylindole (DAPI) staining were utilized to determine the biocompatibility of scaffolds. The results show the synthesized pPGS in 3 hours at 170°C is the optimized sample in the terms of chemical reaction. All scaffolds have bead‐free and a uniform fiber diameter. The Young's modulus of crosslinked PVA‐PGS (50:50 and 40:60) fibers is shown to be in the expected range for nerve applications. The cell culture studies reveal PVA‐PGS (50:50 and 40:60) fibers could lead to better cell adhesion and proliferation. The results suggest that PVA‐PGS (50:50 and 40:60) is a suitable and promising biodegradable material in the fabrication of scaffolds for nerve regeneration.  相似文献

A blend of two biodegradable and semi‐crystalline polymers, poly (L‐lactic acid) (PLLA; 70 wt%) and poly (butylene succinate‐co‐L‐lactate) (PBSL; 30 wt%), was prepared in the presence of various polyethylene oxide‐polypropylene oxide‐polyethylene oxide (PEO‐PPO‐PEO) triblock copolymer contents (0.5, 1, 2 wt%). Mechanical, thermal properties, and Fourier transform infrared (FTIR) analysis of the blends were investigated. It was found that the addition of copolymer to PLLA/PBSL improved the fracture toughness of the blends as shown by mode I fracture energies. It was supported by morphological analysis where the brittle deformation behavior of PLLA changed to ductile deformation with the presence of elongated fibril structure in the blend with copolymer system. The glass transition temperature (Tg), melting temperature (Tm) of PLLA, and PBSL shift‐closed together indicated that some compatibility exists in the blends. In short, PEO‐PPO‐PEO could be used as compatibilizer to improve the toughness and compatibility of the PLLA/PBSL blends. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

Two types of three‐arm and four‐arm, star‐shaped poly(D,L ‐lactic acid‐alt‐glycolic acid)‐b‐poly(L ‐lactic acid) (D,L ‐PLGA50‐b‐PLLA) were successfully synthesized via the sequential ring‐opening polymerization of D,L ‐3‐methylglycolide (MG) and L ‐lactide (L ‐LA) with a multifunctional initiator, such as trimethylolpropane and pentaerythritol, and stannous octoate (SnOct2) as a catalyst. Star‐shaped, hydroxy‐terminated poly(D,L ‐lactic acid‐alt‐glycolic acid) (D,L ‐PLGA50) obtained from the polymerization of MG was used as a macroinitiator to initiate the block polymerization of L ‐LA with the SnOct2 catalyst in bulk at 130 °C. For the polymerization of L ‐LA with the three‐arm, star‐shaped D,L ‐PLGA50 macroinitiator (number‐average molecular weight = 6800) and the SnOct2 catalyst, the molecular weight of the resulting D,L ‐PLGA50‐b‐PLLA polymer linearly increased from 12,600 to 27,400 with the increasing molar ratio (1:1 to 3:1) of L ‐LA to MG, and the molecular weight distribution was rather narrow (weight‐average molecular weight/number‐average molecular weight = 1.09–1.15). The 1H NMR spectrum of the D,L ‐PLGA50‐b‐PLLA block copolymer showed that the molecular weight and unit composition of the block copolymer were controlled by the molar ratio of L ‐LA to the macroinitiator. The 13C NMR spectrum of the block copolymer clearly showed its diblock structures, that is, D,L ‐PLGA50 as the first block and poly(L ‐lactic acid) as the second block. © 2001 John Wiley & Sons, Inc. Coelectrospun polylactide(PLA)/gelatin (GE) composite fibrous matrixes have been identified to exhibit much improved performances compared to the respective components; however, the reasons for their water contact angles decreasing to zero at proper PLA/GE ratios remain unclear. To get a deep understanding of the phenomenon, PLA and GE were coelectrospun with different PLA/GE ratios in this study. Although the resulting composite fibers were homogeneous in appearance, they were detected different microscopic structures by transmission electron mircroscope (TEM) and via morphological observations after selective removal of either PLA or GE component. Together with the results of degradation study in phosphate buffered solution, a kind of cocontinuous phase separation microstructure could be identified for the PLA(50 wt%)/GE(50 wt%) composite fibers, which also showed the water contact angle of 0°. This value was far lower than those of electrospun PLA (~123°) and GE (~42°) fibrous matrixes. The X‐ray photoelectron spectrometry (XPS) data revealed that the polar side groups of protein macromolecules have moved toward composite fiber surface with solvent evaporation during electrospinning, due to the hydrophobic interaction between PLA and GE. Then the excellent hydrophilicity of PLA(50 wt%)/GE(50 wt%) composite fibers could be suggested as the consequence of: (1) the cocontinuous phase separation structure could provide more interface and void for water molecules penetrating; and (2) the accumulation of polar groups on composite fiber surface significantly increased the surface wettability.
