We report a simple twisted intramolecular charge transfer (TICT) chromogenic chemosensor for rapid and selective detection of Hg(2+) and Cu(2+). The sensor was composed of an electron-acceptor 4-fluoro moiety and an electron-donor 7-mercapto-2,1,3-benzoxadiazole species where the S together with the 1-N provided the soft binding unit. Upon Hg(2+) and Cu(2+) complexation, remarkable but different absorbance spectra shifts were obtained in CH(3)CN-H(2)O mixed buffer solution at pH 7.6, which can be easily used for naked-eye detection. The sensor formed a stable 2:1 complex with Cu(2+), and both 2:1 and 3:1 complexes with Hg(2+). While alkali-, alkaline earth- and other heavy and transition metal ions such as Na(+), Mg(2+), Mn(2+), Co(2+), Ni(2+), Ag(+), Zn(2+), Pb(2+) and Cd(2+) did not cause any significant spectral changes of the sensor. This finding is not only a supplement to the detecting methods for Hg(2+) and Cu(2+), but also adds new merits to the chemistry of 4,7-substituted 2,1,3-benzoxadiazoles.  相似文献

We describe ZRL1, a turn-on colorimetric and red fluorescent zinc ion sensor. The Zn(2+)-promoted ring opening of the rhodamine spirolactam ring in ZRL1 evokes a 220-fold fluorescence turn-on response. In aqueous media, ZRL1 turn-on luminescence is highly selective for Zn(2+) ions, with no significant response to other competitive cations, including Na(+), K(+), Ca(2+), Mg(2+), Mn(2+), Fe(2+), Co(2+), Ni(2+), Cu(2+), Cd(2+), or Hg(2+). In addition to these characteristics, preliminary results indicate that ZRL1 can be delivered to living cells and can be used to monitor changes in intracellular Zn(2+) levels.  相似文献

A newly synthesized imine-based receptor (L) showed remarkable specificity toward the Hg(2+) ion in aqueous media over other metal ions. Coordination of L to Hg(2+) induces a turn-on fluorescence response. This was explained based on the restricted imine isomerization along with PET on coordination to Hg(2+). X-ray structural evidence tends to favor a C-C bond rotation rather than C═N isomerization for adopting a favorable conformation in L for coordination to Hg(2+). This reagent could be used for imaging the accumulation of Hg(2+) ions in HeLa cells.  相似文献

This paper reports the use of fluorescent gold nanoclusters synthesized using bovine serum albumin (Au-BSA) for the sensing of copper ions in live cells. The fluorescence of the clusters was found to be quenched by Cu(2+) enabling its detection in cells. The selectivity of the nanosensor was demonstrated in the presence of several cations excluding Hg(2+). We did not study the effect of Hg(2+) since it was reported earlier. The present study suggests that Cu(2+) induced fluorescence quenching is due to its binding to BSA rather than the fluorescence quenching by metal-metal interaction as in the case of Hg(2+). The Au-BSA showed excellent selectivity to Cu(2+) at various pH conditions. The 'turn off' of fluorescence can be retrieved by a Cu(2+) chelator glycine. Our results showed that gold clusters can be used as a 'turn off' sensor for copper and a 'turn on' sensor for glycine. Under the experimental conditions, the probe showed a response for Cu(2+) over a range of 100 μM to 5 mM with a detection limit of 50 μM. The role of Cu(2+) in the misfolding and disassembly of Prion Protein (PrP) leading to various maladies is well ascertained. The methodology we reported here seems to be useful in supplementing other techniques in predicting disease conditions involving Cu(2+).  相似文献

Zn(2+) plays important roles in various biological systems; as a result, the development of tools that can visualize chelatable Zn(2+) has attracted much attention recently. We report here newly synthesized fluorescent sensors for Zn(2+), ZnAF-Rs, whose excitation maximum is shifted by Zn(2+) under physiological conditions. Thus, these sensors enable ratiometric imaging, which is a technique to reduce artifacts by minimizing the influence of extraneous factors on the fluorescence of a probe. Ratiometric measurement can provide precise data, and some probes allow quantitative detection. ZnAF-Rs are the first ratiometric fluorescent sensors for Zn(2+) that enable quantitative analysis under physiological conditions. ZnAF-Rs also possess suitable K(d) for applications, and high selectivity against other biologically relevant cations, especially Ca(2+). Using these probes, changes of intracellular Zn(2+) concentration in cultured cells were monitored successfully. We believe that these probes will be extremely useful in studies on the biological functions of Zn(2+).  相似文献

Nuclear spin-spin coupling constants (1)J(Hg-Hg) in the systems Hg(2)(2+) and Hg(3)(2+) represent the largest coupling constants so far observed in NMR experiments. We have performed a computational study on these ions, on Hg(2)(2+) complexes with 18-crown-6 and 15-crown-5, and on Hg(3)(2+) with solvent molecules and counterions. The results obtained with our recently developed program for the density functional computation of heavy nucleus spin-spin coupling constants are in good agreement with experiments. The data reveal that the bare ions Hg(2)(2+) and Hg(3)(2+) would afford much larger coupling constants than those experimentally observed, with an upper limit of approximately 0.9 MHz for Hg(2)(2+). This limit is much larger than that previously estimated by Hückel theory. It is demonstrated that in solution or due to complexation the experimentally determined values are much smaller than the free ion's coupling constants. With the help of intuitive MO arguments, it is illustrated how the environment strongly reduces the coupling constants in Hg(2)(2+) and Hg(3)(2+). The two-bond coupling constant (2)J(Hg-Hg) in Hg(3)(2+) is also examined.  相似文献

Three fluorescent probes CdABA', CdABA and ZnABA', which are structural isomers of ZnABA, have been designed with N,N-bis(2-pyridylmethyl) ethylenediamine (BPEA) as chelator and 2-aminobenzamide as fluorophore. These probes can be divided into two groups: CdABA, CdABA' for Cd(2+) and ZnABA, ZnABA' for Zn(2+). Although there is little difference in their chemical structures, the two groups of probes exhibit totally different fluorescence properties for preference of Zn(2+) or Cd(2+). In the group of Zn(2+) probes, ZnABA/ZnABA' distinguish Zn(2+) from Cd(2+) with F(Zn)(2+)-F(Cd)(2+) = 1.87-2.00. Upon interchanging the BPEA and carbamoyl groups on the aromatic ring of the fluorophore, the structures of ZnABA/ZnABA' are converted into CdABA/CdABA'. Interestingly, the metal ions selectivity of CdABA/CdABA' was switched to discriminate Cd(2+) from Zn(2+) with F(Cd)(2+)-F(Zn)(2+) = 2.27-2.36, indicating that a small structural modification could lead to a remarkable change of the metal ion selectivity. (1)H NMR titration and ESI mass experiments demonstrated that these fluorescent probers exhibited different coordination modes for Zn(2+) and Cd(2+). With CdABA' as an example, generally, upon addition of Cd(2+), the fluorescence response possesses PET pathway to display no obvious shift of maximum λ(em) in the absence or presence of Cd(2+). However, an ICT pathway could be employed after adding Zn(2+) into the CdABA' solution, resulting in a distinct red-shift of maximal λ(em).  相似文献

The first example of a new Hg(2+)-sensing system based on the structures of complexes is reported. The system uses a combination of a new chiral bidentate ligand and CD spectroscopy. Significant CD spectral changes are observed when Hg(2+) is added, whereas no CD spectral changes are observed in the cases of Li(+), Na(+), K(+), Mg(2+), Ca(2+), Mn(2+), Fe(2+), Co(2+), Ni(2+), Cu(2+), Zn(2+), Rb(+), Ag(+), Cd(2+), La(3+), Gd(3+), and Pb(2+).  相似文献

A novel squaraine-based chemosensor SQ-1 has been synthesized, and its sensing behavior toward various metal ions was investigated by UV-vis and fluorescence spectroscopies. In AcOH-H(2)O (40:60, v/v) solution, Hg(2+) ions coordinate with SQ-1 causing a deaggregation which induces a visual color and absorption spectral changes as well as strong fluorescence. In contrast, the addition of other metals (e.g., Pb(2+), Cd(2+), Cu(2+), Zn(2+), Al(3+), Ni(2+), Co(2+), Fe(3+), Ca(2+), K(+), Mg(2+), Na(+), and Ag(+)) does not induce these changes at all. Thus SQ-1 is a specific Hg(2+) sensing agent due to the inducing deaggregation of the dye molecule by Hg(2+).  相似文献

A minimal core based fluorophore was introduced as a selectively fluorescent "turn on" sensor for Zn(2+) ions in aqueous solution. Addition of Zn(2+) ions to the fluorophore generates a significant emission through a 1:1 ligand-to-metal complex. The fluorescence titration experiment of the minimal core based fluorophore with various metal ions shows that the pyromellitic diimide derivative also has the advantage of a high selectivity to Zn(2+) ions over other metals such as Ni(2+), or Co(2+), Cu(2+), Fe(3+), Fe(2+). More than 8 fold increase in the intensity of fluorescence was observed for the Zn(2+)-bound fluorophore compared to Zn-free fluorophore. Due to its small molecular size, the fluorophore was cell-permeable and successfully applied to the detection of Zn(2+) in living cells. With its relatively high sensitivity to Zn(2+) in living cells, the synthesized new fluorophore will be very useful in the studies on various biological functions of Zn(2+).  相似文献

In this paper, we report new rhodamine hydrazone derivatives bearing thiol and carboxylic acid groups as selective fluorescent and colorimetric chemosensors for Hg(2+). The ring-opening process of spirolactam enables the large fluorescent enhancement and colorimetric change upon the addition of Hg(2+). The sample containing Hg(2+) was mixed with one of the chemosensors in a microchannel where the sensor was examined using confocal laser scanning microscopy. A plot of the fluorescent intensities of both chemosensors versus the log concentration of Hg(2+) exhibited a linear response (r(2)=0.95) in the range of 1 nM-1 μM, and the detection limits were 1 nM and 4.2 nM, respectively. Both chemosensors also enable the visualization of Hg(2+) accumulated in the nematode Caenorhabditis elegans previously exposed to nanomolar concentrations of Hg(2+).  相似文献

A water-soluble fluorescent sensor, 1, based on the "receptor-spacer-fluorophore" [2-(2'-aminophenyl)benzoxazole-amide-2-picolylamine] sensor platform, demonstrates the high sensitivity for Zn(2+) with a 25-fold fluorescence enhancement upon chelation to Zn(2+) and also exhibits high selectivity to Zn(2+) over other metal ions. Coupling T base with Hg(2+) to form stable T-Hg(2+)-T complexes represents a new direction in detection of Hg(2+). Here a graphene oxide (GO)-based fluorescence Hg(2+) analysis using DNA duplexes of poly(dT) that allows rapid, sensitive, and selective detection is first reported. The Hg(2+)-induced T(15)-(Hg(2+))(n)-T(15) duplexes make T(15) unable to hybridize with its complementary A(15) labelled with 6'-carboxyfluorescein (FAM-A(15)), which has low fluorescence in the presence of GO. On the contrary, when T(15) hybridizes with FAM-A(15) to form double-stranded DNA because of the absence of Hg(2+), the fluorescence largely remains in the presence of GO. A linear range from 10 nM to 2.0 μM (R(2) = 0.9963) and a detection limit of 0.5 nM for Hg(2+) were obtained under optimal experimental conditions. Other metal ions, such as Al(3+), Ag(+), Ca(2+), Ba(2+), Mg(2+), Zn(2+), Mn(2+), Co(2+), Pb(2+), Ni(2+), Cu(2+), Cd(2+), Cr(3+), Fe(2+), and Fe(3+), had no significant effect on Hg(2+) detection. Moreover, the sensing system was used for the determination of Hg(2+) in river water samples with satisfactory results.  相似文献

This paper described the optimized design, synthesis and application of a novel rhodamine thiospirolactam derivative as an 'off-on' fluorescent probe for the detection of Hg(2+) in aqueous samples. The 'off-on' fluorescence and color signal change of the probe is based on an Hg(2+)-triggered domino reaction which brings on the opened-ring form of the rhodamine spirolactam to regain the conjugated system of the rhodamine skeleton. In the well designed probe, the thiospirolactam serves as both Hg(2+) binding unit and electron-defect carbon centre, a phenolic hydroxyl with very strong nucleophilicity after deprotonation is chosen as the attacking unit, and a benzene ring is introduced on the linker to afford steric effects, which benefits an efficient nucleophilic reaction, with a high sensitivity towards Hg(2+). It exhibits a stable response for Hg(2+) from 1.0 × 10(-8) to 1.0 × 10(-6) M, with a detection limit of 3.0 × 10(-9) M. The response of the probe to Hg(2+) is highly selective and pH-insensitive, with a fast response time. All these unique features make it particularly favorable for cellular Hg(2+) imaging applications. It has been preliminarily used for highly sensitive monitoring of Hg(2+) levels in living cells with satisfying resolution.  相似文献

We discuss here a quick, simple, economic and ecofriendly method through a completely green route for the selective detection of Hg(2+) in aqueous samples. Here we exploited the ability of chitosan to generate gold nanoparticles and subsequently to act as a stabilizer for the formed nanoparticles. When chitosan stabilized gold nanoparticles (CH-Au NPs) are interacted with Hg(2+) a blue shift for its localized surface plasmon resonance absorbance (LSPR) band is observed. The blue shift is reasoned to be due to the formation of a thin layer of mercury over gold. A concentration as low as 0.01 ppm to a maximum of 100 ppm Hg(2+) can be detected based on this blue shift of the CH-Au NPs. While all other reported methods demand complex reaction steps and costly chemicals, the method we reported here is a simple, rapid and selective approach for the detection of Hg(2+). Our results also show that the CH-Au NPs have excellent selectivity to Hg(2+) over common cations namely, Pb(2+), Cd(2+), Mn(2+), Fe(2+), Ag(1+), Ce(4+), Ni(2+), and Cu(2+).  相似文献

Two tris(2-aminoethyl)amine (tren) based tripodal amide fluoroionophores, 1 and 2, functionalized with quinoline (chelating fluorophore) and naphthalene (non-chelating fluorophore) respectively, are synthesized in good yields. Fluoroionophore 1 shows a selective UV-Vis spectral shift in the case of Hg(2+) in acetonitrile among different metal ions like Li(+), Na(+), Ca(2+), Mg(2+), Cr(2+), Mn(2+), Fe(2+), Co(2+), Ni(2+), Cu(2+), Zn(2+), Cd(2+), Hg(2+), Pb(2+), and Ag(+). On the other hand, fluoroionophore 2 shows no selectivity towards any of the above metal ions in the UV-Vis study. Furthermore, 1 shows a selective chelation induced fluorescence enhancement in the presence of Hg(2+) whereas 2 shows the enhancement of fluorescence with most of the metal ions via a photoinduced charge transfer mechanism. The naked eye detection of Hg(2+) in an acetonitrile solution of 1 shows a greenish fluorescence upon UV light irradiation. The isolated Hg(2+) complex of 1, 3, shows a similar UV-Vis and fluorescence spectral output as observed from in situ spectroscopic studies of 1 in the presence of Hg(2+). Infra-red (IR) and (1)H- NMR studies also reveal the interaction of Hg(2+) with the quinoline nitrogen atoms as well as with the amide functionality.  相似文献

Detection of chelatable zinc (Zn(2+)) in biological studies has attracted much attention recently, because chelatable Zn(2+) plays important roles in many biological systems. Lanthanide complexes (Eu(3+), Tb(3+), etc.) have excellent spectroscopic properties for biological applications, such as long luminescence lifetimes of the order of milliseconds, a large Stoke's shift of >200 nm, and high water solubility. Herein, we present the design and synthesis of a novel lanthanide sensor molecule, [Eu-7], for detecting Zn(2+). This europium (Eu(3+)) complex employs a quinolyl ligand as both a chromophore and an acceptor for Zn(2+). Upon addition of Zn(2+) to a solution of [Eu-7], the luminescence of Eu(3+) is strongly enhanced, with high selectivity for Zn(2+) over other biologically relevant metal cations. One of the important advantages of [Eu-7] is that this complex can be excited with longer excitation wavelengths (around 340 nm) as compared with previously reported Zn(2+)-sensitive luminescent lamthanide sensors, whose excitation wavelength is at too high an energy level for biological applications. The usefulness of [Eu-7] for monitoring Zn(2+) changes in living HeLa cells was confirmed. This novel Zn(2+)-selective luminescent lanthanide chemosensor [Eu-7]should be an excellent lead compound for the development of a range of novel luminescent lanthanide chemosensors for biological applications.  相似文献

A novel fluorescent chemodosimeter based on 1,8-naphthyridine exhibits high selectivity to Zn(2+) and Cu(2+). When 1-(7-acetamino-1,8-naphthyridyl)-2-(6-diacetaminopyridyl)ethene was mixed with CuCl2, hydrolysis of the acetamino group catalyzed by Cu(2+) complex was first observed. Resulting from coordination and hydrolysis catalyzed by the corresponding complex of Zn(2+) or Cu(2+), the highly effective fluorescent detection of Zn(2+) and Cu(2+) is realized with Zn(2+)-selective dual-emission and Cu(2+)-selective ON-OFF behavior.  相似文献

A new fluorescent chemosensor based on the calix[4]arene of partial cone conformation possessing a dansyl moiety has been synthesized. The chemosensor demonstrates selective optical recognition of Hg(2+) and Cu(2+) in two contrasting modes. The receptor exhibited ratiometric sensing of Hg(2+) and "ON-OFF" type of fluorescence behavior in the presence of Cu(2+). The compound behaves as a fluorescent molecular switch upon chemical inputs of Hg(2+) and Cu(2+) ions.  相似文献

A coumarin-derived complex, Hg(2)L(2), was reported as a highly sensitive and selective probe for the detection of mercapto biomolecules in aqueous solution. The addition of Cys to a 99% aqueous solution of Hg(2)L(2) resulted in rapid and remarkable fluorescence OFF-ON (emission at 525 nm) due to the ligand-exchange reaction of Cys with L coordinated to Hg(2+). The increased fluorescence can be completely quenched by Hg(2+) and recovered again by the subsequent addition of Cys. Such a fluorescence OFF-ON circle can be repeated at least 10 times by the alterative addition of Cys and Hg(2+) to the solution of Hg(2)L(2), indicating that it can be used as a convertible and reversible probe for the detection of Cys. The interconversion of Hg(2)L(2) and L via the decomplexation/complexation by the modulation of Cys/Hg(2+) was definitely verified from their crystal structures. Other competitive amino acids without a thiol group cannot induce any fluorescence changes, implying that Hg(2)L(2) can selectively determine mercapto biomolecules. Using confocal fluorescence imaging, L/Hg(2)L(2) as a pair of reversible probes can be further applied to track and monitor the self-detoxification process of Hg(2+) ions in SYS5 cells.  相似文献
