利用单分子实验技术测量了DNA分子的力学性质.通过生化手段将单根DNA分子连在顺磁小球与玻璃侧壁之间,利用磁铁对顺磁球施加力进而拉伸DNA.图像分析得到小球运动的轨迹以及DNA的长度,利用均分定理可以求得施加的外力.测量发现在外力作用下DNA分子的力学反应与高分子物理学的蠕虫状链模型符合良好.  相似文献

We study the relaxation dynamics of a semiflexible chain by introducing a time-dependent tension. The chain has one of its ends attached to a large bead, and the other end is fixed. We focus on the initial relaxation of the chain that is initially strongly stretched. Using a tension that is self-consistently determined, we obtain the evolution of the end-to-end distance with no free parameters. Our results are in good agreement with single molecule experiments on double stranded DNA.  相似文献

基于米氏散射理论解释了激光照明下玻璃微珠的二次彩虹精细结构的成因,发现折射率的差异将直接影响二次彩虹精细结构的位置.对于实验中玻璃微珠半径变化引起二次彩虹精细结构间距变化的现象亦用米氏散射理论进行了模拟分析和实验研究.利用米氏散射的近似理论——艾里理论对玻璃微珠的折射率进行了测量.在对玻璃微珠二次彩虹精细结构所计算得到的折射率的统计分析基础上,通过校正测量误差后得到了玻璃微珠折射率的准确数据.  相似文献

We present the reduced dynamics of a bead in a Rouse chain which is submerged in a bath containing a driving agent that renders it out-of-equilibrium. We first review the generalized Langevin equation of the middle bead in an equilibrated bath. Thereafter, we introduce two driving forces. Firstly, we add a constant force that is applied to the first bead of the chain. We investigate how the generalized Langevin equation changes due to this perturbation for which the system evolves towards a steady state after some time. Secondly, we consider the case of stochastic active forces which will drive the system to a nonequilibrium state. Including these active forces results in an extra contribution to the second fluctuation–dissipation relation. The form of this active contribution is analysed for the specific case of Gaussian, exponentially correlated active forces. We also discuss the resulting rich dynamics of the middle bead in which various regimes of normal diffusion, subdiffusion and superdiffusion can be present.  相似文献

We have determined the fluorescence yield of stained micro beads, used for calibration purposes in flow cytometry, as function of the irradiance of the exciting laser beam. A rate equation model has been applied to derive the number of fluorescence molecules carried by each micro bead. To derive in situ photo-physical properties of the specific dye, required for the rate equation model, we discuss an approach based on flow cytometric sorting of micro beads, which have passed two laser beams with properly chosen different irradiances, and subsequent observation of single molecule bleaching employing high sensitivity microscopy. The feasibility of our approach is demonstrated presenting first results concerning saturation of fluorescence of beads in flow and single molecule bleaching by high sensitivity microscopy.  相似文献

The adsorption behaviors of glycine on diamond(001) are systematically investigated by first-principles calculations.We have considered all possible adsorption configurations without a surface dangling bond and give a quantitative analysis for the relationship between the deviation of carbon bond angle and adsorption energy. We found that a smaller distortion of carbon covalent bond angle results in a more stable adsorption structure, and the most stable adsorption has a benzene-ringlike structure with the highest adsorption energy of 5.11 e V per molecule and the minimum distortion of carbon covalent bond angle.  相似文献

We examine the network of forces to be expected in a static assembly of hard, frictionless spherical beads of random sizes, such as a colloidal glass. Such an assembly is minimally connected: the ratio of constraint equations to contact forces approaches unity for a large assembly. However, the bead positions in a finite subregion of the assembly are underdetermined. Thus to maintain equilibrium, half of the exterior contact forces are determined by the other half. We argue that the transmission of force may be regarded as unidirectional, in contrast to the transmission of force in an elastic material. Specializing to sequentially deposited beads, we show that forces on a given buried bead can be uniquely specified in terms of forces involving more recently added beads. We derive equations for the transmission of stress averaged over scales much larger than a single bead. This derivation requires the ansatz that statistical fluctuations of the forces are independent of fluctuations of the contact geometry. Under this ansatz, the d(d+1)/2-component stress field can be expressed in terms of a d-component vector field. The procedure may be generalized to nonsequential packings. In two dimensions, the stress propagates according to a wave equation, as postulated in recent work elsewhere. We demonstrate similar wave-like propagation in higher dimensions, assuming that the packing geometry has uniaxial symmetry. In macroscopic granular materials we argue that our approach may be useful even though grains have friction and are not packed sequentially.  相似文献

Dissimilar materials welded joints provide many advantages in power, automotive, chemical, and spacecraft industries. The weld bead integrity which is determined by process parameters plays a significant role in the welding quality during the fiber laser welding (FLW) of dissimilar materials. In this paper, an optimization method by taking the integrity of the weld bead and weld area into consideration is proposed for FLW of dissimilar materials, the low carbon steel and stainless steel. The relationships between the weld bead integrity and process parameters are developed by the genetic algorithm optimized back propagation neural network (GA-BPNN). The particle swarm optimization (PSO) algorithm is taken for optimizing the predicted outputs from GA-BPNN for the objective. Through the optimization process, the desired weld bead with good integrity and minimum weld area are obtained and the corresponding microstructure and microhardness are excellent. The mechanical properties of the optimized joints are greatly improved compared with that of the un-optimized welded joints. Moreover, the effects of significant factors are analyzed based on the statistical approach and the laser power (LP) is identified as the most significant factor on the weld bead integrity and weld area. The results indicate that the proposed method is effective for improving the reliability and stability of welded joints in the practical production.  相似文献

We performed numerical simulations of one-bead collision on the surface of a static granular medium. The simulations have been done for two- and three-dimensional packings of beads. The effect of the incident bead velocity, the shot angle, the mechanical parameters and the packing structure are analyzed for ordered and disordered 2D packings and only disordered 3D packings. The 2D results are in good agreement with experimental available data. The 3D simulations give good preliminaries results about the shock-wave propagation through the stacking and provides new insights in the ejection process (“splash function”).  相似文献

Raman measurements on solid hydrogen show a maximum in the vibrational frequency of a H2 molecule as a function of pressure, around 350 Kbar. We give an interpretation of this experimental data by considering a model in which a single H2 molecule is immersed in a dielectric medium, and enclosed in a spheroidal box to simulate the effect of pressure. The maximum results from a balance between a decreasing equilibrium bond length of H2 with pressure at low pressures and a term which reflects the pressure dependence of the dielectric constant. Our results are consistent with available calculations which give a much lower vibrational frequency at a much higher pressure, for a H2 molecule embedded in a metallic medium.  相似文献

We examine the sources of error in the histogram reweighting method for Monte Carlo data analysis. We demonstrate that, in addition to the standard statistical error which has been studied elsewhere, there are two other sources of error, one arising through correlations in the reweighted samples, and one arising from the finite range of energies sampled by a simulation of finite length. We demonstrate that while the former correction is usually negligible by comparison with statistical fluctuations, the latter may not be, and give criteria for judging the range of validity of histogram extrapolations based on the size of this latter correction.  相似文献

We present a simple method to calibrate optical tweezers by using only videomicroscopy to measure the position autocorrelation function of the trapped bead in the potential well of the tweezers. To accomplish this task, we use a high-speed camera with ??500 fps (frames per second), which provides a precise measurement of the relaxation time of the bead Brownian fluctuations. We also study the variation of the trap stiffness as a function of some parameters of interest such as the laser power and the distance from the bead center to the microscope coverslip, showing that the presented method returns precise results.  相似文献

We study the jamming of bead assemblies placed in a cylindrical container whose bottom is pierced with a circular hole. Their jamming behavior is quantified here by the median jamming diameter, that is the diameter of the hole for which the jamming probability is 0.5. Median jamming diameters of monodisperse assemblies are obtained numerically using the Distinct Element Method and experimentally with steel beads. We obtain good agreement between numerical and experimental results. The influence of friction is then investigated. In particular, the formation of concentric bead rings is observed for low frictions. We identify this phenomenon as a boundary effect and study its influence on jamming. Relying on measures obtained from simulation runs, the median jamming diameter of bidisperse bead assemblies is finally found to depend only on the volume-average diameter of their constituting beads. We formulate this as a tentative law and validate it using bidisperse assemblies of steel beads.  相似文献

A statistical method for measuring the modal density of elastic waves through direct mode counting in strongly scattering disordered systems is presented. To illustrate this approach, the results of ultrasonic experiments in a highly porous sintered glass bead network are reported. This method is shown to yield a reliable and robust measurement of the density of states, enabling mode-counting techniques to be applied to increasingly complex systems, where modal overlap and sensitivity to experimental conditions have previously hampered definitive results.  相似文献

We develop a statistical model for a confined homopolymeric chain molecule based on a monomer grand ensemble representation. The molecule is subject to a confining external field, a backbone interaction, and an attractive interaction between any pair of monomers. An exact minimum principle for the thermodynamics of the backbone in an external field is obtained, and a controlled mean field approximation results in a modified minimum principle from which relevant physical quantities such as monomer density can be found. We explore the limit in which the chain is subject to tight confinement, and make a preliminary investigation of a prototypical system.  相似文献

This paper presents a study carried out on 3.5 kW cooled slab laser welding of 904 L super austenitic stainless steel. The joints have butts welded with different shielding gases, namely argon, helium and nitrogen, at a constant flow rate. Super austenitic stainless steel (SASS) normally contains high amount of Mo, Cr, Ni, N and Mn. The mechanical properties are controlled to obtain good welded joints. The quality of the joint is evaluated by studying the features of weld bead geometry, such as bead width (BW) and depth of penetration (DOP). In this paper, the tensile strength and bead profiles (BW and DOP) of laser welded butt joints made of AISI 904 L SASS are investigated. The Taguchi approach is used as a statistical design of experiment (DOE) technique for optimizing the selected welding parameters. Grey relational analysis and the desirability approach are applied to optimize the input parameters by considering multiple output variables simultaneously. Confirmation experiments have also been conducted for both of the analyses to validate the optimized parameters.  相似文献
