A novel optical coherence tomography (OCT) system is presented that combines Fourier-domain OCT with incoherent nonlinear time gating. By processing backscattered light in the optical domain, the user can select a restricted depth field of view for improved contrast and acquisition speed. This technique has the additional advantage that imaging is done in the infrared (approximately 1280 nm) but is detected in the visible(approximately 504 nm).  相似文献

We have developed a new algorithm and configuration for self-eliminating the autocorrelation of the object wave in Fourier-domain optical coherence tomography. A self-interferogram of the object wave is acquired synchronously with the standard interferogram of the recombined object and reference waves. The former is then subtracted from the latter after Fourier transformation. The algorithm is validated by numerical simulation and by experimental measurement of a U.S. Air Force target and a feline eye.  相似文献

The possibility of measuring a full Doppler flow depth profile in parallel by use of frequency-domain optical coherence tomography is demonstrated. The method is based on a local phase analysis of the backscattered signal and allows for imaging of bidirectional Doppler flow. The Doppler frequency limit is 5 kHz for the presented measurements and is set by half of the frame rate of the CCD detector array. We measured the flow of 0.3-microm microspheres suspended in distilled water at controlled flow rates and in vitro human blood flow through a 200-microm capillary with a real-time color-encoded Doppler tomogram rate of 2-3/s.  相似文献

A method, novel to our knowledge, for effective mirror image suppression in Fourier-domain optical coherence tomography based on a phase shift between neighboring A-mode scans is demonstrated. By realizing that the phase shifts of the real and mirror images are mutually reversed and assuming that the real image intensities of the two successive A-mode scans are the same, we can solve a set of two coupled equations to obtain the real image signals. The images based on the scanning of a high-resolution spectral-domain optical coherence tomography system are processed to show effective mirror image suppression results. Compared with a similar method of broad application, our approach has the advantages of shorter process time and higher flexibility in selecting the concerned image portions for processing.  相似文献

<正>We demonstrate the subsurface imaging of an articular cartilage using Fourier-domain common-path optical coherence tomography.The bare fiber probe integrated with a hypodermic needle provides the rigidness required to perform lateral scanning with less microscale bending.By submerging both the probe and the specimen into saline solution,we not only reduce the beam divergence,but also increase the signal-to-noise ratio compared with the measurement in free space.Our system can differentiate the characteristic cartilage zones and identity various micro-structured defects in an ex vivo chicken knee cartilage,thus demonstrating that it could be used to conduct early arthritis diagnosis and intraoperative endo-microscopy.  相似文献

We describe an artifact removal setup swept-source optical coherence tomography (OCT) system that enables high-speed full-range imaging. We implement a piezoelectric fiber stretcher to generate a periodic phase shift between successive A-scans, thus introducing a transverse modulation. The depth ambiguity is then resolved by performing a Fourier filtering in the transverse direction before processing the data in the axial direction. The dc artifact is also removed. The key factor is that the piezoelectric fiber stretcher can be used to generate discrete phase shifts with a high repetition rate. The proposed experimental setup is a much improved version of the previously reported B-M mode scanning for spectral-domain OCT in that it does not generate additional artifacts. It is a simple and low-cost solution for artifact removal that can easily be applied.  相似文献

We demonstrate en face swept source optical coherence tomography (ss-OCT) without requiring a Fourier transformation step. The electronic optical coherence tomography (OCT) interference signal from a k-space linear Fourier domain mode-locked laser is mixed with an adjustable local oscillator, yielding the analytic reflectance signal from one image depth for each frequency sweep of the laser. Furthermore, a method for arbitrarily shaping the spectral intensity profile of the laser is presented, without requiring the step of numerical apodization. In combination, these two techniques enable sampling of the in-phase and quadrature signal with a slow analog-to-digital converter and allow for real-time display of en face projections even for highest axial scan rates. Image data generated with this technique is compared to en face images extracted from a three-dimensional OCT data set. This technique can allow for real-time visualization of arbitrarily oriented en face planes for the purpose of alignment, registration, or operator-guided survey scans while simultaneously maintaining the full capability of high-speed volumetric ss-OCT functionality.  相似文献

While human sweat secretion is accepted as a mechanism by which the body cools off, excessive sweating (hyperhidrosis) is now appreciated as a medical condition and the primary site for diagnosis is the palm of the hand. We propose sweat film layer thickness as a potential clinical diagnostic parameter when screening for excessive sweating. In this preliminary study we demonstrate the usefulness of Fourier-domain optical coherence tomography (FD-OCT) for measurement of sweat film thickness in vivo with micron-scale resolution on the hand of a human volunteer. FD-OCT has a superior image acquisition time and identification of active sweat glands, ducts and pores is also possible.  相似文献

We report a new yet simple method to achieve full-range complex Fourier-domain optical coherence tomography (OCT) for in vivo imaging. The method utilizes a scanner that is dedicated for lateral scanning in the system to introduce a constant carrier frequency into the OCT spectral interferograms during the scanning. This is achieved by simply offsetting the sampling beam spot away from the pivot point of the scanning mirror. We demonstrate the method experimentally for in vivo full-range imaging of the anterior segment of a human eye. The method is free from complex conjugate mirror image and self-cross-correlation image artifacts.  相似文献

A high-speed (47,000 A-scans/s), ultrahigh axial resolution Fourier domain optical coherence tomography (OCT) system for retinal imaging at approximately 1060 nm, based on a 1024 pixel linear array, 47 kHz readout rate InGaAs camera is presented. When interfaced with a custom superluminescent diode (lambda(c) = 1020 nm, Deltalambda = 108 nm, Pout = 9 mW), the system provides 3.3 microm axial OCT resolution at the surface of biological tissue, approximately 4.5 microm in vivo in rat retina, approximately 5.7 microm in vivo in human retina, and 110 dB sensitivity for 870 microW incident power and 21 mus integration time. Retinal tomograms acquired in vivo from a human volunteer and a rat animal model show clear visualization of all intraretinal layer and increased penetration into the choroid.  相似文献

We present the results of studies of the basic principles and describe the design of a low-coherence two-wavelength interferometer based on polarization-maintaining fiber. The interferometer was developed for optical coherence tomography (OCT) imaging of the internal structure of living biological tissue simultaneously at two wavelengths, 830 and 1300 nm. Images of several sites of living biological tissue are presented and analyzed.  相似文献

An experimental tracking optical coherence tomography (OCT) system has been clinically tested. The prototype instrument uses a secondary sensing beam and steering mirrors to compensate for eye motion with a closed-loop bandwidth of 1 kHz and tracking accuracy, to within less than the OCT beam diameter. The retinal tracker improved image registration accuracy to <1 transverse pixel (<60 microm). Composite OCT images averaged over multiple scans and visits show a sharp fine structure limited only by transverse pixel size. As the resolution of clinical OCT systems improves, the capability to reproducibly map complex structures in the living eye at high resolution will lead to improved understanding of disease processes and improved sensitivity and specificity of diagnostic procedures.  相似文献

Scanning optical coherence tomography (OCT) is limited in sensitivity and resolution by the restricted focal depth of the confocal detection scheme. Holoscopy, a combination of holography and Fourier-domain full-field OCT, is proposed as a way to detect photons from all depths of a sample volume simultaneously with uniform sensitivity and lateral resolution, even at high NAs. By using the scalar diffraction theory, as frequently applied in digital holographic imaging, we fully reconstruct the object field with depth-invariant imaging quality. In vivo imaging of human skin is demonstrated with an image quality comparable to conventionally scanned OCT.  相似文献

Spectroscopic optical coherence tomography (OCT), an extension of conventional OCT, is demonstrated for performing cross-sectional tomographic and spectroscopic imaging. Information on the spectral content of backscattered light is obtained by detection and processing of the interferometric OCT signal. This method allows the spectrum of backscattered light to be measured over the entire available optical bandwidth simultaneously in a single measurement. Specific spectral features can be extracted by use of digital signal processing without changing the measurement apparatus. An ultrabroadband femtosecond Ti:Al(2)O(3) laser was used to achieve spectroscopic imaging over the wavelength range from 650 to 1000 nm in a simple model as well as in vivo in the Xenopus laevis (African frog) tadpole. Multidimensional spectroscopic data are displayed by use of a novel hue-saturation false-color mapping.  相似文献

Second-harmonic optical coherence tomography, which uses coherence gating of second-order nonlinear optical responses of biological tissues for imaging, is described and demonstrated. Femtosecond laser pulses were used to excite second-harmonic waves from collagen harvested from rat tail tendon and a reference non-linear crystal. Second-harmonic interference fringe signals were detected and used for image construction. Because of the strong dependence of second-harmonic generation on molecular and tissue structures, this technique imparts contrast and resolution enhancement to conventional optical coherence tomography.  相似文献

A fiber-optic-bundle-based optical coherence tomography (OCT) probe method is presented. The experimental results demonstrate this multimode optical fiber-bundle-based OCT system can achieve a lateral resolution of 12 microm and an axial resolution of 10 microm with a superluminescent diode source. This novel OCT imaging approach eliminates any moving parts in the probe and has a primary advantage for use in extremely compact and safe OCT endoscopes for imaging internal organs and great potential to be combined with confocal endoscopic microscopy.  相似文献

We developed a structured interference optical coherence tomography (SIOCT) to enhance the lateral resolution beyond the diffraction limit. A sinusoidal pattern is created on the interferometric beam with the reference intensity temporally modulated. In the Fourier domain, the high spatial frequencies are shifted into the detectable range, which enhances the lateral resolution beyond the diffraction limit by a factor of 2. The lateral resolution of SIOCT was characterized in our study as ~5.5 μm, surpassing the diffraction limit ~9.6 μm as in conventional Fourier-domain optical coherence tomography. SIOCT was demonstrated on phantoms and ex vivo adipose tissues.  相似文献

Doppler optical coherence tomography (OCT) can image tissue structure and blood flow at micrometer-scale resolution but has limited imaging depth. We report a novel, linear-scanning, needle-based Doppler OCT system using angle-polished gradient-index or ball-lensed fibers. A prototype system with a 19-guage (diameter of approximately 0.9 mm) echogenic needle is constructed and demonstrates in vivo imaging of bidirectional blood flow in rat leg and abdominal cavity. To our knowledge, this is the first demonstration of Doppler OCT through a needle probe in interstitial applications to visualize deeply situated microcirculation.  相似文献
