A series of heterocyclic analogues of glutamic acid diethyl ester (GDEE), an antagonist at central excitatory amino acid receptors, have been synthesized and tested biologically. (RS)-Ethyl alpha-amino-alpha-(3-ethoxyisoxazol-5-yl)acetate (7), (RS)-ethyl 2-amino-3-(3-ethoxy-5-methylisoxazol-4-yl)propionate (16) and closely related analogues were synthesized. Compound 7, a diethyl derivative of the naturally occurring excitatory amino acid ibotenic acid (IBO), was synthesized from 3-hydroxy-5-methylisoxazole (1) via 3-ethoxyisoxazol-5-ylacetic acid (5) and its ethyl ester. Nitrosation of this ester followed by catalytic reduction gave 7. The ethyl ester of IBO, 9, was synthesized in a similar manner from 3-benzyloxyisoxazol-5-ylacetic acid (8). Ethyl derivatives of the synthetic excitatory amino acid 2-amino-3-(3-hydroxy-5-methylisoxazol-4-yl)propionic acid (AMPA) were synthesized from 3-hydroxy-4,5-dimethylisoxazole (10) through a diethyl acetylaminomalonate derivative, which upon deprotection gave the 3-ethoxy derivative of AMPA (15). Esterification of 15 gave the diethyl derivative 16 and the ethyl ester of AMPA (18) as well as N-ethylated derivatives of AMPA, 21 and 22 were synthesized. The final products were tested microelectrophoretically. The derivatives 7, 9, 15, 16 and 18 were weak and non-selective excitatory amino acid antagonists, whereas 21 and 22 were found to be inactive.  相似文献

以胆甾醇为原料,经氯铬酸吡啶氧化生成胆甾-4-烯-3,6-二酮(2),2与盐酸羟胺反应合成了(3E)-胆甾-4-烯-3,6-二酮-3-肟(3),总收率66%。利用合成3的反应条件合成了两个3的类似物——(3E)豆甾-4-烯-3,6-二酮-3-肟(6)和(3E)-谷甾-4-烯-3,6-二酮-3-肟(7)。2,3,6和7的结构经NMR和IR表征。  相似文献

By interaction of N‐methyl(ethyl)‐dithiocarbamate sodium salt with 3‐chloro‐pentane‐2,4‐dion the 1‐(3‐alkyl‐4‐methyl‐2‐thioxo‐2,3‐dihydro‐thiazol‐5‐yl)‐ethanones 1 , 2 and corresponding oximes 7 , 8 were synthesized. On the basis of the mentioned compounds hydrazono ( 3 , 4 ), ureayl and thioureayl ( 5 , 6 ) derivatives, substituted oximes ( 9 , 10 ) and azinyl oximes ( 11 , 12 ) were obtained. The structures of synthesized compounds were confirmed by proton nuclear magnetic resonance spectroscopy and elemental analysis. The pesticidal activities of synthesized compounds were studied. Some of the synthesized compounds simultaneously have shown growth stimulant and fungicidal activity.  相似文献

6,8-Dimethoxy-3-[2-(4-methoxyphenyl)ethyl]isocoumarin was synthesized by condensation of 5,7-dimethoxyhompophthalic acid with 3-(4-methoxyphenyl)propanoyl chloride. The structure of the synthesized compound was confirmed by its mass spectrometric studies. The synthesized compound serves as a model for synthesis of DL-agrimonolide.  相似文献

Several 3-alkyl-6-(2,4-dichlorophenoxymethyl)-s-triazolo[3,4-b]-1,3,4-thiadiazoles 3a-3e and 6,6′-bis(3-aryl-s-triazolo[3,4-b]-1,3,4-thiadiazoles) 3f-3j were synthesized. The structures of all the compounds synthesized were elucidated by elemental analyses and spectral data. The representative compounds 3a and 3b exhibited moderate biological activities.  相似文献

赛吲哚霉素的双乙酰衍生物已由吲哚曼 -吲哚法合成。吲哚-3-乙酸经催化氢化, 甲酯化和氨解反应得到吲哚曼-3-乙酰胺(9); 将4-去氧-D-核-吡喃己糖以三苯甲基保护后乙酰化。消除保护基再经氧化和甲酯化等反应, 得到1,2,3-O-三乙酰基-4-去氧-β-D-核-吡喃己糖醛酸甲酯(6), 将化合物6和9先缩合后脱氢即得到赛吲哚霉素的双乙酰衍生物2。赛吲哚霉素的类似物1-(3-氨羰甲基吲哚基)-2,3,4,6-O-四乙酰基-β-D-吡喃葡萄糖苷(13)也由相似的方法合成。  相似文献

A series of α,β‐unsaturated ketones containing quinolone moieties 2 , 3 , 4 , 5 , 6 were synthesized by condensation of 7‐methoxyquinoline‐2,4(1H,3H)‐dione ( 1 ) with different aryl aldehydes. Pyrazole derivatives 8 , 9 , 10 , 11 were also synthesized via refluxing of α,β‐unsaturated ketones 2 , 3 , 4 , 5 , 6 with hydrazine derivatives. Newly synthesized compounds were characterized by elemental analyses, spectral data, and screened for their antioxidant and antitumor activities. Geometrical optimizations of the molecular structures for different synthesized compounds were studied.  相似文献

4(3H)-Quinazolinones have been synthesized from poly(ethylene glycol) (PEG) supported aza-Wittig reaction. 2-Dialkylamino- 4(3H)-quinazolinones 6 were synthesized efficiently by reaction of secondary amine with PEG-supported carbodiimides 4, which were obtained from aza-Wittig reaction of PEG-supported iminophosphoranes 3 with isocyanates.  相似文献

We synthesized the six presumed metabolites (2--7) of 4-(2-methyl-1H-imidazol-1-yl)-2,2-diphenylbutanamide [KRP-197/ONO-8025, 1], a urinary incontinence therapeutic agent, in order to confirm the structures of the metabolites. Metabolite (2) was synthesized via glucuronidaion of compound (1) and methyl 2,3,4-tri-O-benzoyl-1-methanesulfonyl-alpha-D-glucopyranuronate. Metabolite (3) was synthesized via 3-(tert-butoxycarbonyl)-2-methyl-1,3-imidazolidine-4,5-dione. Metabolites (4--7) were synthesized via 4-amino-2-diphenylbutanamide, respectively. The structures of the metabolites (2--7) in humans were identified by means of synthesis of the authentic compounds.  相似文献

Chelates of the type M(L)2 {where, M ?= ?Co(II), Ni(II) and Cu(II), and L ?= ?3-{(E)-[(2-hydroxy-3-methoxyphenyl)methylidene]amino}pyridin-4(1H)-one)} were synthesized by using the Schiff base ligand in the stochiometric ratio 2:1 (L:M) and Schiff base ligand (L) was synthesized by simple condensation between 2-hydroxy-3-methoxybenzaldehyde with 3-aminopyridin-4-ol. The structure and formation of synthesized compounds were established by different analytical and spectroscopic methods like, elemental analysis, UV- spectroscopy, FT-IR, Proton and Carbon NMR, mass spectrometry and Powder XRD. Further, the synthesized chelates screened for the DNA binding studies of Calf Thymus (CT)-DNA by exploiting electronic absorption spectra, relative viscosity measurements and thermal denaturation methods. The proposed DNA binding mode supports the enhancement in the binding activity of the complexes in presence of newly synthesized ligand. The cleavage activities of the PUC-18 DNA in the presence and the absence of the complexes were recorded with the help of gel-electrophoresis. The cleavage experiment results reveals that all the synthesized chelates can cleave pUC-18 DNA effectively.  相似文献

The versatile multifunctional unreported pyridine‐2,6‐bis(2‐cyano‐N‐phenyl‐3‐oxopropanethioamide) ( 3 ) was prepared starting from pyridine‐2,6‐bis‐(3‐oxopropanenitrile) ( 1 ). Several new series of polysubstituted thiophenes and 1,3,4‐thiadiazoles incorporating 2,6‐pyridine moiety were efficiently synthesized. The newly synthesized compounds were evaluated for their in vitro anticancer activity against human cancer cell lines: hepatocellular liver carcinoma (HEPG2) and Caucasian breast adenocarcinoma (MCF‐7). Some of the newly synthesized compounds exhibited better activity than doxorubicin as a reference drug.  相似文献

2-amino-5-(3-fluoro-4-methoxyphenyl)thiophene-3-carbonitrile derivatives have been synthesized from 1-(3-fluoro-4-methoxyphenyl)ethanone, malononitrile, mild base, and sulfur powder using the Gewald method through a multistep reaction sequence. The structures of newly synthesized compounds were established on the basis of their elemental analyses, IR, 1H NMR, 13C NMR, and mass spectral data, and then synthesized compounds were screened for their in vitro antimicrobial activity. Among them, derivatives 3b (thiphene), 3f (pyrazole), and 3d (halogen) showed good activity and remaining derivatives exhibited moderate activity.  相似文献

以染料木素为起始原料,经两步反应制得中间体7-O-炔丙基染料木素(3);以CuI为催化剂,3与叠氮乙酰基糖经Click反应合成了7-O-[1-(2,3,4,6-四-O-乙酰基-β-D-糖苷)1H-1,2,3-三氮唑-4-甲基]-染料木素(5a~5c); 5a~5c经去乙酰基制得三种含1,2,3-三氮唑的氮苷染料木素糖缀合物(6a~6c)。3, 5和6均为新化合物,其结构经1H NMR, 13C NMR和HR-MS表征。  相似文献

Several substituted-quinazolin-3(4H)-ones 8-11ad were synthesized by condensation of 2-chloro-N-(4-oxo-substituted-quinazolin-3(4H)-yl)-acetamides with various substituted imidazoles through one pot reaction. Elemental analysis, IR, (1)H-NMR and mass spectral data confirmed the structure of the newly synthesized compounds. Synthesized quinazolin-4-one derivatives were investigated for their antitubercular, antibacterial and antifungal activities. Some of the tested compounds showed good antitubercular activity. 以丙二酸二乙酯为原料,经水解、硝化得到关键中间体偕二硝基乙酸乙酯(2);2与乙醛、叠氮化钠缩合环化得到4-甲基-5-硝基-1,2,3-连三唑(3)。3的结构经NMR,IR和元素分析表征。初步探讨了硝化反应的反应历程,并对制备2的合成进行了工艺改进,发现以NaNO2-98%HNO3为硝化试剂的新方法,2的收率达62.1%,纯度93.1%。  相似文献

The new monomer, 5'-O-methacryloyl-3'-azido-3'-deoxy-thymidine (MAZT), was synthesized from methacryloyl chloride (MAC) and 3'-azido-3'-deoxythymidine (AZT). Poly(MAZT), poly(MAZT-co-AA) and terpoly(MAZT-FUR-MAH) were synthesized by radical polymerizations. The average molecular weights of synthesized polymers were in the range of 8,800 ∼23,000 depending on polymers. The in vivo antitumor activities of polymers at 800mg/kg were increased in the following order: terpoly(MAZT-FUR-MAH) >x poly(MAZT-co-AA) > poly(MAZT) > AZT > 5-FU. The in vitro anti-HIV activities of synthesized polymers were less effective than those of AZT and D4T. But the cytotoxicities of the polymers on the MT-4 cell line were found to be much less toxic than AZT.  相似文献
