对黑蚂蚁中脂肪酸的组成和含量进行测定。采用索氏提取法提取其脂肪酸,再进行甲酯化处理,以气相色谱-质谱联用仪进行了分离和鉴定。由黑蚂蚁中分离鉴定出11种脂肪酸,占脂肪酸总量的99.78%,不饱和脂肪酸占脂肪酸总量的80.65%,饱和脂肪酸占脂肪酸总量的19.13。通过流动注射化学发光法分析了黑蚂蚁的抗氧化性能,结果表明,黑蚂蚁具有较强的抗氧化性能。  相似文献

首次对小鼠眼球中的脂肪酸组成进行了气相色谱分析。小鼠眼球中的脂肪酸经氯仿-甲醇-盐酸混合液萃取,用甲醇-氢氧化钾法甲酯化,在25m×0.25mmPEG-20M熔融石英毛细管柱内进行分离,眼球中12种主要脂肪酸得到了定性和定量。测试结果提示:正常小鼠眼球中饱和脂肪酸含量较高,占全部脂肪酸含量的52.8%;不饱和脂肪酸含量较少,但以多不饱和脂肪酸居多,占全部脂肪酸含量的18.2%,不饱和脂肪酸与饱和脂肪酸的比值为0.6。可为有关眼疾病的治疗、预防及生化方面的深入研究提供有意义的信息。  相似文献

A high-performance liquid chromatographic method for the analysis of the fatty acid composition of human serum lipids with fluorescence detection was examined. Both free and total fatty acids extracted from serum were derivatized with 9-anthryldiazomethane and were analysed using methanol-water (94.7:5.3) as mobile phase. Twelve kinds of fatty acid were detected, both in the free and total fatty acids, and were well separated. Concentrations of individual fatty acids of serum lipids were estimated from an internal standard, heptadecanoic acid. The results correlated well with those from two other quantitative analyses. These results indicate that the high-performance liquid chromatographic analysis of fatty acids is a reliable method for determining individual fatty acids of human serum lipids. The compositions of free fatty acids and total fatty acids of serum lipids were analysed and compared in 27 normal subjects, 27 diabetics, and 20 angina pectoris patients by this method.  相似文献

Although unsaturated fatty acids have long been known to accompany saturated fatty acids in most lipids, qualitative and quantitative determination of fatty acid patterns only became possible with the advent of modern analytical methods. Present day knowledge of the chemical structure, physical properties, and metabolism of unsaturated fatty acids provides the basis for the development of new concepts of their function. Thus unsaturated fatty acids crucially determine the properties of biological membranes. Moreover, essential fatty acids are precursors of prostaglandins.  相似文献

建立了孔石莼脂肪酸的气相色谱/质谱(GC/MS)测定方法。使用Folch法提取了孔石莼中的总脂,经过2 mol/L HCl-甲醇溶液的甲酯化处理后,采用GC/MS法对其脂肪酸组成进行了分离分析,同时结合有机质谱学规律,分别对饱和脂肪酸甲酯、单不饱和脂肪酸甲酯和多不饱和脂肪酸甲酯的裂解规律和质谱特征进行了分析归纳。通过质谱数据库检索和标准品对照,鉴定出孔石莼中的24种脂肪酸,其中9,12,15-十八碳三烯酸、4,7,10,13-十六碳四烯酸和6,9,2,15-十八碳四烯酸3种主要多不饱和脂肪酸占总脂肪酸含量的45.14%。通过对孔石莼中脂肪酸的分析,表明特征离子在脂肪酸甲酯尤其是多不饱和脂肪酸甲酯的定性方面具有很好的应用价值。  相似文献

 用双 (2 乙基己基 )酚酞酸酯 (DEHP)诱导大鼠肝过氧化物酶体增殖 ,再用蔗糖密度梯度离心法分离大鼠肝细胞过氧化物酶体 ,并用十七烷酸作内标 ,以毛细管气相色谱法在非极性SPB 1石英毛细管柱上对其中的 11种脂肪酸进行分离测定。正常组和诱导组的大鼠肝过氧化物酶体中的不饱和脂肪酸和长链脂肪酸所占总脂肪酸的比例及总脂肪酸的统计结果是 :诱导组的不饱和脂肪酸的含量高于正常组的 (P <0 0 5 ) ,而两个组的脂肪酸总量及长链脂肪酸的含量无明显差别。结果提示 :诱导组的大鼠肝过氧化物酶体的脂肪酸成分发生了变化 ,其膜结构与正常组的不相同。  相似文献

Summary This report describes a modified method for the separation and analysis of polyunsaturated fatty acids such as 20:4, 20:5, and 22:6, using HPLC. The results show that these fatty acids are well separated from the saturated acids. Since the unsaturated fatty acids elute earlier than saturated acids, and this method does not require the fractionation of free fatty acids using thin layer chromatography, a necessary step for the gas chromatographic analysis, the recoveries of polyunsaturated fatty acids were significantly higher as compared to those from gas chromatography. Furthermore, HPLC and gas chromatographic methods gave identical results for the acyl chain composition of phosphatidylserine. The advantages of using HPLC over gas chromatography in determining the acyl chain composition of free fatty acids and phospholipids are also discussed.  相似文献

A method is described for the determination of fatty acids in dried sweat spot and plasma samples using gas chromatography with flame ionization detection. Plasma and dried sweat spot samples were obtained from a group of blood donors. The sweat was collected from each volunteer during exercise. Sweat was spotted onto collection paper containing butylated hydroxytoluene. Fatty acids were derivatized with acetyl chloride in methanol to form methyl esters of fatty acids. The fatty acids in dried sweat spot samples treated with butylated hydroxytoluene and stored at –20°C were stable for 3 months. Our results indicate that sweat contains, among fatty acids with short chain, also fatty acids with long chain and unsaturated fatty acids. Linear relationships between percentage content of selected fatty acids in dried sweat spot and plasma were observed.  相似文献

A new approach to the visualization of fatty acids in mouse liver and retinal samples has been developed using silver nanoparticles (AgNPs) in nanoparticle-assisted laser desorption/ ionization imaging mass spectrometry (nano-PALDI-IMS) in negative ion mode. So far, IMS analysis has concentrated on main cell components, such as cell membrane phospholipids and cytoskeletal peptides. AgNPs modified with alkylcarboxylate and alkylamine were used for nano-PALDI-IMS to identify fatty acids, such as stearic, oleic, linoleic, arachidonic, and eicosapentaenoic acids, as well as palmitic acid, in mouse liver sections; these fatty acids are not detected using 2,5-dihydroxybenzoic acid (DHB) as a matrix. The limit of detection for the determination of palmitic acid was 50 pmol using nano-PALDI-IMS. The nano-PALDI-IMS method is successfully applied to the reconstruction of the ion images of fatty acids in mouse liver sections. We verified the detection of fatty acids in liver tissue sections of mice by analyzing standard lipid samples, which showed that fatty acids were from free fatty acids and dissociated fatty acids from lipids when irradiated with a laser. Additionally, we applied the proposed method to the identification of fatty acids in mouse retinal tissue sections, which enabled us to learn the six-zonal distribution of fatty acids in different layers of the retina. We believe that the current approach using AgNPs in nano-PALDI-IMS could lead to a new strategy to analyze basic biological mechanisms and several diseases through the distribution of fatty acids.  相似文献

The biosynthesis of iso-fatty acids was investigated in the myxobacteria Myxococcus xanthus and Stigmatella aurantiaca by feeding labeled precursors of these fatty acids and by conducting detailed analysis of the fatty acid profiles and fatty acid MS and NMR spectra. These experiments revealed that in S. aurantiaca all iso-even fatty acids are formed via alpha-oxidation of iso-odd fatty acids and not by using valine-derived isobutyryl-CoA.  相似文献

用色谱-质谱(GC-MS)联用技术分析了银环蛇蛇蜕的脂肪酸成分,鉴定出36种脂肪酸(饱和酸28种不饱和酸8种),在蛇蜕中发现16种饱和二元酸,并发现了自然界中少见的奇数碳脂肪酸14种及支链酸2种,用氨基酸自动分析仪测定了蛇蜕水提取液中的13种游离氨基酸,酸性水解液中17种氨基酸,在蛇蜕中发现存在牛 磺酸。  相似文献

采用2-(11H-苯[a]咔唑)乙基对甲苯磺酸酯(BCETS)为柱前荧光衍生试剂,通过梯度洗脱使得18种脂肪酸在BDS-C8柱上得到良好的分离.方法应用于大果白刺不同部位中游离脂肪酸的分析,结果表明大果白刺的果皮果肉和叶子中均含有大量的不饱和脂肪酸,其总不饱和脂肪酸含量分别为70.74%和73.47%.大果白刺种子中不饱和脂肪酸的含量相对较少,仅占总脂肪酸含量的57.21%,其不饱和脂肪酸组成主要是C18∶1(油酸)和C18∶2(亚油酸).其中,大果白刺的果皮果肉中,不饱和脂肪酸主要是C18∶1、C18∶2和C18∶3(亚麻酸).其叶子中的不饱和脂肪酸主要是C18∶3,所占总脂肪酸比例为48.34%.首次对大果白刺中的脂肪酸进行了分析,可以为大果白刺在食品、药品中的进一步开发应用和质量控制提供一定的数据支持.  相似文献

Lipids in foods contain a wide variety of fatty acids differing in chain length, degree of unsaturation, position and configuration of double bonds and the presence of special functional groups. Modern capillary gas chromatography offers excellent separation of fatty acids. Fused-silica capillary columns with stationary phases of medium polarity and non-polar methylsilicone stationary phases successfully separate most of the natural fatty acids. Special applications, such as the separation of complex cis-trans fatty acid mixtures and cyclic fatty acids, required particular chromatographic conditions, including the use of very long capillary columns or more polar stationary phases. The derivatization methods for the preparation of fatty acid esters also need to be optimized to obtain accurate quantitative results. This paper reviews the derivatization techniques, capillary columns and stationary phases commonly used in the gas chromatography of fatty acids in food.  相似文献

Significant amounts of fatty acids and alcohols, in addition to straight-chain hydrocarbons, were observed in the vacuum pyrolysate of a young kerogen isolated from a fresh-water sediment. The amounts and the distribution patterns of fatty acids and alcohols were compared with those indigenous to the original kerogen, as determined by solvent extraction followed by saponification extraction. Straight-chain fatty acids in the pyrolysate were 12.7 mg/g kerogen, while those extracted from the original kerogen were 10.5 mg/g. The amounts of fatty alcohols in the pyrolysate and in the extracts were 1.31 and 1.10 mg/g. respectively. The excess amounts in the pyrolysate were explained by the release of a strictly bound form of fatty acids and alcohols, which were left in the kerogen matrix even after the saponification extraction. The results indicated that most fatty acids and alcohols were not decomposed as a result of the vacuum pyrolysis and that neither fatty acids nor fatty alcohols should be viewed as the prime precursors of the straigth-chain hydrocarbons (4.6 mg/g) simultaneously generated during vacuum pyrolysis.  相似文献

A high-performance liquid chromatographic (HPLC) method for the separation and quantitation of retinal fatty acids containing long-chain polyunsaturated fatty acids is described. Fatty acids from frog retinal lipids were converted to the corresponding phenacyl derivatives which were separated on a C18 reversed-phase column and detected at 242 nm. Molar absorptivities (peak area units/nmol) of up to seventeen fatty acid phenacyl derivatives were determined and used for quantitation of fatty acids separated by HPLC. Compared with gas chromatography, the HPLC method gave a similar molar percent distribution of the fatty acids and was twenty to fifty times more sensitive. This HPLC method provides a useful means for the study of chemistry and metabolism of long-chain polyunsaturated fatty acids in retina and other tissues where amounts of material may be limited or recovery of individual components desirable.  相似文献

合成了氨基化氧化石墨烯(NH_(2)-PEG-GO),利用界面张力仪研究了脂肪酸质量浓度、NH_(2)-PEG-GO质量浓度以及脂肪酸链长度对于NH_(2)-PEG-GO与脂肪酸在水-油界面共吸附行为的影响。结果表明,当NH_(2)-PEG-GO与脂肪酸共同存在于液相且质量浓度较低时,此时的水-油界面张力值要明显低于体系中只添加NH_(2)-PEG-GO或脂肪酸时的张力值。脂肪酸的链长越长,NH_(2)-PEG-GO与脂肪酸共吸附降低界面张力能力越强。由于相对于NH_(2)-PEG-GO,脂肪酸尺寸较小,所以初期的界面张力降低由脂肪酸吸附引起,而且,脂肪酸在水-油界面的吸附能够吸引NH_(2)-PEG-GO向界面扩散,表现出吸附的协同效应。而在吸附后期,界面张力降低现象由NH_(2)-PEG-GO主导,NH_(2)-PEG-GO与脂肪酸之间存在竞争吸附。  相似文献

首次研究了维药细穗柽柳(Tamarix leptostachys Bunge)实中挥发油和脂肪酸的化学成分。 分别采用药典中的挥发油提取法和索式取提法提取柽柳实中的挥发油和脂肪酸,使用气质联用技术获取总离子流图,各色谱峰相应的质谱图经过NIST2011标准谱库检索定性,并采用峰面积归一化法进行定量分析,计算各成分的相对百分含量。 两种方法分别鉴定出48种挥发油和19种脂肪酸。 挥发油主要成分为芳香类化合物(43.71%)、芳香性醛酮类(20.58%)、脂肪酸类(13.03%)、酯类(17.36%)和醇类(4.19%)等。 脂肪酸主要成分为棕榈酸(35.61%)、亚油酸(27.26%)和油酸(11.33%)等,其中不饱和脂肪酸含量占总脂肪酸含量的38.65%。 维药细穗柽柳实中富含丰富的挥发油和不饱和脂肪酸,具有很好的开发利用价值。  相似文献

采用超声提取-毛细管气相色谱法分析测定了海水养殖和淡水养殖的南美白对虾的肌肉和肝胰腺(中肠腺)中脂肪酸的组成与含量,以用于对虾繁殖与营养的研究。采用Folch 液超声提取对虾组织中的脂质,脂质经氢氧化钾-三氟化硼法甲酯化后上机分析。采用BPX70(60 m×0.25 mm i.d.)石英毛细管柱分离,火焰离子化检测器检测。以线性程序升温等当碳长度(ECLT)值结合标准品对照的方法对17种脂肪酸定性,用面积归一化法定量。结果表明,两种对虾的脂肪酸组成均以不饱和脂肪酸为主,而且不饱和多烯酸的含量明显高于不饱和单烯酸。两种对虾的肌肉脂肪酸中高度不饱和脂肪酸,如二十碳五烯酸(EPA)与二十二碳六烯酸(DHA)的总含量均较高,达20%以上;肝胰腺中EPA和DHA的总含量较低,为11%~17%。淡水虾中n-6族不饱和脂肪酸含量比海水虾高,海水虾中n-3族与n-6族脂肪酸含量之比值比淡水虾高。  相似文献
