A systematic study of the reactions of dimethyl acetylenedicarboxylate (DMAD) and methyl propynoate with 5-mercaptoazoles and pyridine-2-thiones has been carried out and as a result, a number of novel imidazo[1,5-b]thiazin-4-ones 6a,b, pyrazolo[1,5-b] thiazin-4-ones 15a-f, imidazo[1,5-b]thiazol-4-ones 7a,b and thiazolo[3,2-a]pyridines 21a-c have been prepared. The influence of the size of the ring of the starting "cyclic" thioamides on the size of the fused ring in the reaction products has been established. The preferred formation of a six-membered thiazine ring took place in the reactions of 5-mercaptoazoles. In contrast, the five membered thiazolidine ring is formed in reactions of pyridine-2-thiones. In both cases the product is a five-membered ring fused to a six-membered heterocycle.  相似文献

Diazo-azoles react as 1,7- or 1,11-dipoles with isocyanates to form the hitherto unknown pyrazolo[5,1-d] [1,2,3,5]tetrazine-4-ones, [1,2,3,5]tetrazino[5,4-b]indazole-4-ones and [1,2,3]triazolo[5,1-d][1,2,3,5]tetrazine-4-ones in a [7+2]- or [11+2]-cycloaddition reaction.  相似文献

Regioselective palladium-catalyzed amination of 2-chloro-3-iodopyridine followed by a subsequent palladium-catalyzed amination leads to 2,3-diaminopyridines. Treatment with triphosgene affords highly functionalized unsymmetrical imidazo[4,5-b]pyridin-2-ones in just three synthetic steps. A two-step synthesis of pseudosymmetrically disubstituted imidazo[4,5-b]pyridin-2-ones, 1,4-disubstituted pyrido[2,3-b]pyrazinediones, and 1,3-disubstituted thiadiazolo[3,4-b]pyridin-2-ones is also described.  相似文献

A solid-phase synthesis of substituted cyclic urea derivatives as potential heterocyclic library scaffolds is described. 2-Amino-3-nitropyridine is attached to Wang resin via a carbamate linkage. Reduction of the nitro group was achieved with SnCl(2).2H(2)O. Reductive alkylation with a range of substituted benzaldehydes followed by cyclative cleavage afforded a small library of 3-substituted imidazo[4,5-b]pyridine-2-ones in 33-45% yield and 59-88% purity. Subsequently, this methodology was applied to the synthesis of 3-substituted imidazo[4,5-f]quinolin-2-ones.  相似文献

The simple MO LCAO method was used for calculations on 4H-imidazo[5,1-b]benzimidazole, imidazo[5,1-b]benzoxazole, imidazo[5,1-b]benzothiazole, and a number of their derivatives. Inclusion of the unshared pairs of the hetero atoms at positions 4 and 9 in the -electron system has a considerable effect on the stability of compounds of this type. The 1H NMR spectra were recorded for 4-methyl-imidazo[5,1-b]benzimidazole and imidazo[5,1-b]benzoxazole derivatives, and a similarity was found between the chemical shifts of the imidazole ring protons and the -electron densities at the adjacent carbon atoms. Calculation of a series of models for the cations and measurements of the basicity constants show protonation of 4-methylimidazo[5,1-b]benzimidazole takes place at the nitrogen atom in position 2. The calculated values for the electronic structure and reactivity indices in the investigated series of tricyclic systems were compared with their chemical properties.  相似文献

Azo compounds obtained by the coupling of 5-diazoimidazoles with diethyl esters of nitro-, chloro-, bromo-, and acetylaminomalonic acids under conditions of base catalysis are cyclized to give 1,4-dihydroimidazo[5,1-c]-1,2,4-triazin-4-ones or imidazo[5,1-c]-1,2,4-triazoles. The chloro, bromo, and nitro groups in the bicyclic products are readily replaced by action of nucleophiles. Abstract

A novel series of bis(tetrahydro[1,2,4]triazolo[5,1-b]quinazolin-8-ones) and bis(tetrahydrobenzo[4,5]imidazo[2,1-b]quinazolinones) containing amide linkages were regionselectively prepared via a three-component reaction of bis(aldehydes) with dimedone and 3-amino-1,2,4-triazole (or 2-aminobenzimidazole) under conventional heating as well as under microwave irradiation.  相似文献

The reaction of 5-benzenesulfonyl-3,4-dihydro-1 H-pyridin-2-one derivatives with azides or isocyanides provided two new classes of compounds, triazolo[4,5-b]pyridin-5-ones 3 or pyrrolo[3,4-b]pyridin-2-ones 4, respectively, in good yields and regioselectivity. Three-component condensation of 5(3)-amino-3(5)-methylpyrazole with aromatic aldehydes and 1,3-cyclohexanedione afforded mixtures of 3-methyl-4-aryl-2,4,6,7,8,9-hexa hydro-5H-pyrazolo[3,4-b]quinolin-5-ones and 2-methyl-9-aryl-5,6,7,9-tetrahydro pyrazolo[5,1-b]quinazolin-8(4H)-ones. The reaction of 3-aminopyrazolo-4-carbonitrile and ethyl 3-aminopyrazolo-4-carboxylate with aldehydes and cyclo hexanedione or dimedome is regioselective and leads to the formation of partially hydrogenated pyrazolo [5,1-b]quinazolin-8-one systems. In all compounds the dihydroazine ring exists in the enamine tautomeric form.  相似文献

The cyclization of imidazolylhydrazines, synthesized from 5-diazoimidazoles and cyanoacetic acid derivatives, to 4-aminoimidazo[5,1-c][1,2,4]triazines and imidazo[5,1-c][1,2,4]triazin-4-ones has been studied. It was established that electron-withdrawing substituents at position 4 of the imidazole ring had a weak effect on the cyclization process. 2,6-Di-tert-butyl-4-{[2-(2-aryl-2-oxoethylthio)-1H-imidazol-1-yl]imino}cyclohexa-2,5-dien-1-ones under the action of bases give products of the 2,3-dihydroimidazo[2,1-b]thiazol- 3-ol series by subsequent recyclization reaction of the intermediate imidazo[2,1-b][1,3,4]thiadiazoles. The structure of imidazo[2,1-b]thiazol-3-ol is supported by the X-ray diffraction. The features of the cyclization processes of quinone imine derivatives were stimulated by DFT calculations using the wB97XD/6-311++G** method.  相似文献

The reaction of 2,6-diphenyl and 2,6-di-p-tolyltetrahydro-4H-thiopyran-4-ones with benzaldehyde afforded 2,6-diphenyl and 2,6-di-p-tolyl-3,5-dibenzy-lidenetetrahydro-4H-thiopyran-4-ones, which, on treatment with hydroxylamine hydrochloride, hydrazine hydrate and thiourea, gave thiopyrano[4,3-c]isoxazole, thiopyrano[4,3-c]pyrazole and thiopyrano[4,3-d]pyrimidine derivatives, respectively. Also, the reaction of dibenzylidenetetrahydrothiopyran-4-ones with malononitrile in piperidine and malononitrile in ammonim acetate afforded thiopyrano[4,3-b]pyran and thiopyrano[4,3-b]pyridine derivatives, respectively, while treatment with ethyl acetoacetate gave acetyl thiopyrano[4,3-b]pyran derivatives. On the other hand, treatment of 2,6-diphenyl and 2,6-di-p-tolyltetrahydro-4H-thiopyran-4-ones with elemental sulfur and malononitrile in the presence of diethylamine gave thieno[2,3-c]thiopyran derivatives. A new procedure for preparation of 2-R-5-aryl-5,6-dihydro-7H-[1,2,4]triazolo[5,1-b][1,3]thiazin-7-ones by condensation of 5-R-1,2,4-triazole-3-thiones with 3-arylacryloyl chlorides was developed. The thiazine ring of the [1,2,4]triazolo[5,1-b][1,3]thiazin-7-ones is easily cleaved by treating with ammonia and hydrazine affording amides and hydrazides of 3-aryl-3-(1H-1,2,4-triazol-5-ylsulfanyl)propanoic acids. The latter react with isothiocyanates furnishing carbamoyl thiohydrazides of 3-aryl-3-(1H-1,2,4-triazol-5-ylsulfanyl)propanoic acids that in alkaline media undergo cyclization into 4-aryl-5-[2-(4H-1,2,4-triazol-5-ylsulfanyl)-2-phenylethyl]-2,4-dihydro-3H-1,2,4-triazole-5-thiones.  相似文献

The reactions of 4-amino derivatives of imidazo[4.5-c]pyridin-2-ones with -bromomethylketones have been studied. Depending on the nature of the reagents and the reaction conditions, either 5-acylmethyl salts of the starting amine or 2-substituted imidazo[4.5-c]imidazo[1,2-a]pyridin-8-ones can be obtained. A series of novel 3-substituted imidazo[4,5-c]quinolin-4(5H)-ones (2a-w) was prepared by the reaction of imidazo[4,5-c]quinolin-4(5H)-ones (6) with several electrophiles under basic conditions. The bronchodilatory activity of these compounds was evaluated on the basis of their protective effects against antigen-induced contraction (the Schultz-Dale reaction) of guinea-pig trachea (in vitro) and antigen inhalation-induced bronchospasm in passively sensitized guinea-pigs (in vivo). Although correlations between in vitro and in vivo activities were not clear, short alkyl chains such as the methyl and ethyl groups at the 3-position were important for potent activity, especially in vivo. Substituents at the 5-position were more tolerant of the activity than those at the 3-position. 5-Ethyl-3-methyl-3H-imidazo[4,5-c]quinolin-4(5H)-one (21) exhibits the most potent bronchodilatory activity among our tested compounds and is at least 5-fold more active than theophylline in vivo.  相似文献

[reaction: see text] N-Methyl-4-alkoxy-3-alkynylpyridin-2(1H)-ones readily undergo iodine-promoted 5-endo-heteroannulation under mild conditions to 3-iodofuropyridinium triiodide salts in moderate to good yields. The latter may be dealkylated in situ upon exposure to an iodide anion to provide the corresponding 3-iodofuro[2,3-b]pyridin-4(1H)-ones. The reaction of 3-haloanthranilic acids with o-bromomethylphenylacetonitrile gave 2-(2-carboxy-6-halophenyl)-1,4-dihydro-3(2H)-isoquinolinium bromides. 2-Chlorophenylisoquinolinium bromides are readily converted into 4-R-6,11-dihydro-13H-isoquino[3,2-b]quinazolin-13-ones by heating >145°C, but 2,4-dibromophenylisoquinolinium bromide only on fusing with anthranilic acid. The effect of the nature and position of substituents in the quinazoline fragment of 7,12-dihydro-5H-isoquino[2,3-a]quinazolin-5-ones on the rate of the rearrangement into 6,11-dihydro-13H-isoquino[3,2-b]quinazol-13-ones has been studied. Cyclization of azo compounds, synthesized from 5-diazoimidazoles and 2-naphthol or p-substituted phenols, into naphth[2,1-e]imidazo[5,1-c]-1,2,4-triazines and benz[e]imidazo[5,1-c]-1,2,4-triazines occurs only in the presence of p-toluenesulfonic acid.
