Nine bisabolangelone reduction derivatives were synthesized and separated as potential anti‐ulcer agent. Their structures were characterized by 2D NMR, IR, ESI‐MS, elemental analysis and single‐crystal X‐ray diffraction analysis. Preliminarily H+/K+‐ATPase activity evaluation indicated that all the target compounds had a certain inhibitory effect, and compounds II and IV exhibited the better inhibitory activity against H+/K+‐ATPase than bisabolangelone and the commercial omeprazole with the IC50 of 23.21 and 65.32 μmol/L, respectively. The initial structure‐activity analysis suggested that the presence of carbonyl group in six‐membered ring and double bond in side‐chain seemed to be necessary to the activity. 相似文献

A novel series of substituted benzoylguanidine derivatives were designed and synthesized in order to evaluate their NHE1 inhibitory activity. Most of them were found to inhibit NHE1‐mediated platelet swelling in a concentration‐dependent manner, and eight compounds showed more potent NHE1 inhibitory activity than Cariporide. Compound 6f with an IC50 value of 1.08×10?10 mol·L?1, was 39 times more potent than lead compound CPU‐X‐050420 in vitro tests. 相似文献

为了寻找对心肌缺血再灌注损伤具有保护作用的药物, 以苯甲酰胍为母核, 在其苯环的4位引入4-(2,3,4-三甲氧基苄基)哌嗪-1-甲基, 3位引入不同的取代苯甲酰胺基, 设计并合成了12个未见文献报道的目标化合物. 其结构经MS, IR, 1H NMR和元素分析确证. 体外血小板肿胀模型(PSA)试验结果表明, 大部分目标化合物显示出较好的Na+/H+交换器1 (NHE1)抑制作用, 其中化合物7g和7l抑制NHE1的IC50值分别为3.72和3.53 nmol8226;L-1, 是卡立泊来德(IC50=12.1 nmol8226;L-1)的3.2和3.4倍. 相似文献

Abstract 6-(Pyrazol-1-yl)pyrazolo[3,4-b]pyridin-3-ol (3a) was synthesized by cyclization reaction from 2,6-dichloro-4-methylnicotinonitrile. The related sulfonyl and acylated product (4–7) were obtained from the compound 3a and different sulfonyl and acyl chloride compounds. The sulfonylation of the compound 3a afforded the O1-sulfonylation, only, to give 3-substituted products 4–6. The acylation of the compound 3a gives 1,3-diacylated products 7 via N2, O1-diacylation. All compounds were characterized by spectroscopic methods and the molecular structures of 3a and 4–7 were determined by X-ray diffraction analysis. The molecular structure of 5 and 7 were studied by density functional theory (DFT) calculations, reproduced the experimental geometry. The energies of the three molecules of the tautomeric forms of the compound 3a were calculated by DFT calculations. It was shown that the 3a predominated in the gas phase. Besides, molecular electrostatic potential (MESP) and frontier molecular orbital (FMO) analysis of the compound 3a were investigated by theoretical calculations. 合成出了一系列新型基于咔唑的单-/双-碳酰腙衍生物3和4.利用1H NMR、13C NMR、IR和元素分析对其进行了结构表征.评价了目标化合物对蛋白酪氨酸磷酸酶1B(PTP1B)的抑制活性,讨论了结构与活性的关系.实验结果显示,大部分化合物对PTP1B具有良好的抑制活性,其中1,5-双[(9-丁基-3-咔唑基)亚甲基]碳酰腙(4c)的抑制活性最高,IC50=(4.81±0.41)mmol/L,且活性高于对照药物齐墩果酸.对目标化合物1-[(9-庚基-3-咔唑基)亚甲基]碳酰腙(3f)和4c进行分子对接研究和密度泛函理论(DFT)计算.分子对接结果表明,化合物3f和4c结合到PTP1B酶由螺旋α3和α6形成的活性位点,与PTP1B酶通过氢键、极性、疏水和p-p等相互作用形成了稳定的复合物. 相似文献

In the present work, a novel heterocyclic hybrid of a spirooxindole system was synthesized via the attachment of ferrocene and triazole motifs into an azomethine ylide by [3 + 2] cycloaddition reaction protocol. The X-ray structure of the heterocyclic hybrid (1″R,2″S,3R)-2″-(1-(3-chloro-4-fluorophenyl)-5-methyl-1H-1,2,3-triazole-4-carbonyl)-5-methyl-1″-(ferrocin-2-yl)-1″,2″,5″,6″,7″,7a″-hexahydrospiro[indoline-3,3″-pyrrolizin]-2-one revealed very well the expected structure, by using different analytical tools (FTIR and NMR spectroscopy). It crystallized in the triclinic-crystal system and the P-1-space group. The unit cell parameters are a = 9.1442(2) Å, b = 12.0872(3) Å, c = 14.1223(4) Å, α = 102.1700(10)°, β = 97.4190(10)°, γ = 99.1600(10)°, and V = 1484.81(7) Å3. There are two molecules per unit cell and one formula unit per asymmetric unit. Hirshfeld analysis was used to study the molecular packing of the heterocyclic hybrid. H···H (50.8%), H···C (14.2%), Cl···H (8.9%), O···H (7.3%), and N···H (5.1%) are the most dominant intermolecular contacts in the crystal structure. O···H, N···H, H···C, F···H, F···C, and O···O are the only contacts that have the characteristic features of short and significant interactions. AIM study indicated predominant covalent characters for the Fe–C interactions. Also, the electron density (ρ(r)) at the bond critical point correlated inversely with the Fe–C distances. 相似文献

Efficient Synthesis,Crystal Structure and Antibacterial Activity of Two Novel 1,3-Oxazin Derivatives
Two new 1,3-oxazin derivatives, C22H24N2O5(3I) and C19H16N2O5(3II), have been synthesized via an unusual cascade reaction. The attractive aspect of this cascade reaction is that the novel construction of 1,3-oxazine and the direct C-N bond formation from C-C bond can be easily achieved via pyridine-mediated acylation in a one-pot operation. Both compounds have been synthesized and characterized by elemental analysis, IR, NMR spectra and X-ray single-crystal diffraction. Compound 3I crystallizes in monoclinic, space group P21/n with a = 16.282(4), b = 7.4117(18), c = 17.256(5) , β = 103.193(9)°, V = 2027.4(9) 3, Mr = 396.43, Z = 4, Dc = 1.299 g/cm3, F(000) = 840, MoKa radiation(λ = 0.71073 ), the final R = 0.0771 and wR = 0.1582 for 3662 were observed reflections with I 2σ(I). Compound 3II crystallizes in triclinic, space group P1 with a = 7.1265(9), b = 10.1071(13), c = 23.529(3), α = 97.463(9), β = 96.981(9), γ = 94.345(9)°, V = 1600.5(4) 3, Z = 4, Dc = 1.409 g/cm3, F(000) = 736, CuKa radiation(λ = 1.54186 ), the final R = 0.0515 and wR = 0.1241 for 4920 observed reflections with I 2σ(I). The preliminary antibacterial activities of 2 and 3 against E. coli and S. aureus were investigated. The results showed that the inhibiting effect of 3 was higher than that of 2. 相似文献

A facile synthesis of heavy alkali metal octahydrotriborates (MB3H8; M=K, Rb, and Cs) has been developed. It is simply based on reactions of the pure alkali metals with THF?BH3, does not require the use of electron carriers or the addition of other reaction media such as mercury, silica gel, or inert salts as for previous procedures, and delivers the desired products at room temperature in very high yields. However, no reactions were observed when pure Li or Na was used. The reaction mechanisms for the heavy alkali metals were investigated both experimentally and computationally. The low sublimation energies of K, Rb, and Cs were found to be key for initiation of the reactions. The syntheses can be carried out at room temperature because all of the elementary reaction steps have low energy barriers, whereas reactions of LiBH4/NaBH4 with THF?BH3 have to be carried out under reflux. The high stability and solubility of KB3H8 were examined, and a crystal structure thereof was obtained for the first time. 相似文献

A liquid chromatography-tandem mass spectrometric (LC/MS/MS) method was developed for the determination of a selective Na(+)/H(+) exchanger inhibitor 4-cyano(benzo[b]thiophene-2-carbonyl)guanidine (KR-33028) in rat plasma. KR-33028 and the internal standard, linezolid, were extracted from rat plasma with ethyl acetate at neutral pH. The analytes were separated on an XBridge C(18) column with a mixture of methanol-0.1% formic acid (35:65, v/v) as mobile phase and detected using an electrospray ionization tandem mass spectrometry in the multiple-reaction-monitoring mode. The standard curve was linear (r = 0.9998) over the concentration range of 2.0-1000 ng/mL. The coefficients of variation of intra- and inter-assay were 1.3-6.8% and the relative error was 0.8-5.0%. The recoveries of KR-33028 and linezolid were 70.5 and 84.6%, respectively. The lower limit of quantification for KR-33028 was 2.0 ng/mL using 50 microL plasma sample. This method was successfully applied to the pharmacokinetic study of KR-33028 in rats. 相似文献

A series of symmetrical dibenzylidene derivatives of cyclobutanone were synthesized with the goal of studying the physicochemical properties of cross-conjugated dienones (ketocyanine dyes). The structures of the products were established and studied by X-ray diffraction and by NMR and electronic spectroscopy. All the products had E,E-geometry. The oxidation and reduction potentials of the dienones were determined by cyclic voltammetry. The potentials were shown to depend on the nature, position, and number of substituents in the benzene rings. A linear correlation was found between the difference of the electrochemical oxidation and reduction potentials and the energy of the long-wavelength absorption maximum. This correlation can be employed to analyze the properties of other compounds of this type. Quantum chemistry was used to explain the observed regularities in the electrochemistry, absorption, and fluorescence of the dyes. The results are in good agreement with the experimental redox potentials and spectroscopy data. 相似文献

In this paper, we report on the design and synthesis of a novel series of quinazoline-2,4(1H,3H)-dione derivatives as fluoroquinolone-like inhibitors of bacterial gyrase and DNA topoisomerase IV to identify and develop antimicrobial agents to prevent bacterial resistance problems. Their structures were confirmed using spectroscopic analyses (IR, NMR, and EI-MS). The novel quinazoline-2,4(1H,3H)-dione derivatives were evaluated for their antimicrobial activities against Gram-positive and Gram-negative bacterial strains using the Agar well diffusion method to study the antimicrobial activities and compared them with the standard drugs. Most compounds displayed moderate activity. Among the tested compounds, the most promising compounds 13 and 15 provided broad bioactive spectrum against Gram-positive and Gram-negative strains compared to the standard drugs. 相似文献

In search of novel anti tubercular agents, a series of twelve 4‐(substituted benzylidene)‐2‐p‐tolyloxazol‐5(4H)‐ones (5a – 5l) has been synthesized, characterised and subjected to evaluate their antitubercular activity for the first time against Mycobacterium tuberculosis H37Rv (ATCC 27294). The out‐put of these studies disclosed that all the synthesized target molecules of the series displayed good to moderate activity with MIC values ranging 2–32 μg/mL in comparison with the standard first line antitubercular drugs Rifampicin and Isoniazid. Compound 5e with three methoxy groups meta to each other, is the most distinctive compound identified amongst the series, because of its remarkable in vitro antitubercular activity and thus may act as a promising lead molecule for further explorations. 相似文献

为了寻找新的高活性农药, 利用2-甲基苯甲腈与不同的醛反应, 再通过2,3-二氯-5,6-二氰基-1,4-苯醌(DDQ)脱氢或NaH和CH3I氮甲基化, 合成了3个系列共18个5-取代氨基-12-取代苯并[c]菲啶衍生物, 其中15个是未见文献报道的化合物. 它们的结构均经1H NMR, 13C NMR, IR和高分辨质谱表征; 用X射线衍射测定了化合物5-氨基-12-(4-二乙氨基苯基)-11,12-2H-苯并[c]菲啶盐酸盐(3c)的晶体结构. 初步的生物活性测试结果表明, 部分化合物具有一定的杀菌活性和促进黄瓜子叶生根活性. 相似文献
