Compound‐specific isotope analysis (CSIA) by liquid chromatography coupled to isotope ratio mass spectrometry (LC/IRMS) has until now been based on ion‐exchange separation. In this work, high‐temperature reversed‐phase liquid chromatography was coupled to, and for the first time carefully evaluated for, isotope ratio mass spectrometry (HT‐LC/IRMS) with four different stationary phases. Under isothermal and temperature gradient conditions, the column bleed of XBridge C18 (up to 180 °C), Acquity C18 (up to 200 °C), Triart C18 (up to 150 °C), and Zirchrom PBD (up to 150 °C) had no influence on the precision and accuracy of δ13C measurements, demonstrating the suitability of these columns for HT‐LC/IRMS analysis. Increasing the temperature during the LC/IRMS analysis of caffeine on two C18 columns was observed to result in shortened analysis time. The detection limit of HT‐RPLC/IRMS obtained for caffeine was 30 mg L–1 (corresponding to 12.4 nmol carbon on‐column). Temperature‐programmed LC/IRMS (i) accomplished complete separation of a mixture of caffeine derivatives and a mixture of phenols and (ii) did not affect the precision and accuracy of δ13C measurements compared with flow injection analysis without a column. With temperature‐programmed LC/IRMS, some compounds that coelute at room temperature could be baseline resolved and analyzed for their individual δ13C values, leading to an important extension of the application range of CSIA. Cortex Moutan is a well-known traditional Chinese medicine derived from Paeonia suffruticosa ANDREWS. However, root cortices of P. delavayi and P. decomposita also are used under the name of this drug in some regions such as Yunnan and Sichuan Provinces, respectively. In order to make a comparison of their chemical constituents, the compounds of the three Paeonia species were analyzed by high-performance liquid chromatography-diode array detection/electrospray ionization and quadrupole-time-of-flight tandem mass spectrometry (HPLC-DAD/ESI-MS2). A total of 50 compounds were observed in the 50% (v/v) methanolic extracts, including 17 monoterpenes, 14 galloyl glucoses, 10 acetophenones, 5 phenolic acids, 3 flavonoids and 1 triterpene. These chemical constituents were separated on a C18 column and identified or tentatively characterized based on UV spectra and MS fragmentation behavior. The chemical compositions of the three Paeonia species were found to have many differences. Paeonol was the predominant constituent of P. suffruticosa and P. decomposita, while P. delavayi contained albiflorin and more galloyl glucoses than the other two Paeonia species. Most of these identified compounds have been reported from P. delavayi and P. decomposita for the first time. The ESI-MS fragmentation behavior of monoterpene glycosides, acetophenones and galloyl glucoses was also investigated successively, and appropriate characteristic pathways were proposed. The large differences in chemical compounds among the three Paeonia species strongly encouraged further comparison of the bioactivities of these three species.  相似文献

Changes in sphingolipids have been associated with profound effects in cell fate and development in both plants and animals. Sphingolipids as a group consist of a large number of different compound classes of which numerous individual species may vary in response to environmental stimuli to affect cellular responses. The ability to measure all sphingolipids simultaneously is, therefore, essential to an understanding of the biochemical regulation of sphingolipid metabolism and signaling molecules derived from it. In the model plant Arabidopsis thaliana, the major sphingolipid classes are glycosylinositolphosphoceramides, glucosylceramides, hydroxyceramides and ceramides. Other minor but potentially important sphingolipids are free long-chain bases and their phosphorylated derivates. By using a single solvent system with reversed-phase high-performance liquid chromatography coupled to electrospray ionization tandem mass spectrometry detection we have been able to separate and measure 168 sphingolipids from a crude sample. This greatly speeds up and simplifies the analysis of plant sphingolipids and should pave the way for a better understanding of their role in plant performance.  相似文献

Phenolic compounds are the major bioactive constituents of the Chinese herbal drug Tu-Si-Zi, which is prepared from the seeds of Cuscuta chinensis. However, seeds of C. australis also are offered under the name of this drug in the herb market. In order to make a comparison of their chemical constituents, the phenolic compounds of these two Cuscuta species were analyzed by high-performance liquid chromatography/diode-array detection/electrospray ion trap tandem mass spectrometry (HPLC/DAD/ESI-MS(n)). A total of 50 compounds were observed in the methanol extracts, including 23 flavonoids, 20 lignans and 7 quinic acid derivatives. These compounds were separated on a C18 column and identified or tentatively characterized based on UV spectra and MS fragmentation behavior. In contrast to previous reports, the phenolic patterns of these two Cuscuta species were found to be very different. Kaempferol and astragalin were the predominant constituents of C. australis, while hyperoside was the major compound in C. chinensis. Most of the identified compounds, especially the acylated flavonoid glycosides, have not previously been reported from Cuscuta species. In addition, a 30 Da neutral loss observed for flavonols was investigated and could be used to differentiate flavonoid isomers such as kaempferol and luteolin. The ESI-MS fragmentation behavior of furofuran lignans was also investigated, and a characteristic pathway is proposed. The large differences observed between the phenolic constituents of C. chinensis and C. australis strongly encouraged further comparison of the bioactivities of these two species.  相似文献

A liquid chromatographic (LC) method has been developed for the separation of 11 monohydroxybenzo[a]pyrenes (OH BaPs) positional isomers, and for their detection using electrospray ionization mass spectrometry (ESI-MS). All OH BaPs isomers were separated on an octadecylsilyl (C(18))-bonded amorphous organosilica column utilizing gradient elution with acetonitrile-water and triethylamine (TEA) at pH 11.0 and determined by MS, except 2- and 8-OH BaPs which were coeluted. The lower detection limits were in the range from 1.6 micro g/L for 12-OH BaP to 12 micro g/L for 5-OH BaP without any sample enrichment. The relative standard deviations of area response were in the range from 1.8% (9-OH BaP) to 4.9% (12-OH BaP) except for 9.4% (7-OH BaP). The developed method was successfully applied to incubation mixtures of BaP and CYP1A1/epoxide hydrolase. This method identified 1-, 3- and 9-OH BaPs as the major metabolites, and 2- (and/or 8-) and 12-OH BaPs as the minor metabolites in the incubation mixture.  相似文献

Comprehensive analytical methods based on pressurized liquid extraction followed by normal-phase liquid chromatography (NPLC) with ultraviolet detection and reversed-phase liquid chromatography (RPLC)/electrospray mass spectrometry (MS) have been developed for determination of alkylphenol ethoxylates (APEOs) in textile samples. NPLC with an aminosilica column allowed for the chromatographic separation of APEOs according to the increasing number of ethylene units and revealed the exact distribution of individual oligomers. RPLC coupled with electrospray MS was highly sensitive and enabled the complete qualitative and quantitative determination of individual APEOs in textile samples. The 2 analytical methods based on different chromatographic separation mechanisms, i.e., NPLC and RPLC, may provide complementary information of APEOs in textile materials. The 2 detection methods were successfully applied to the investigation of various textile samples, and the data of our research suggested actual pollution in real textile products.  相似文献

Flavonoids are a class of secondary plant metabolites existing in great variety in nature. Due to this variety, identification can be difficult, especially as overlapping compounds in both chromatographic separations and mass spectrometric detection are common. Methods for distinguishing isobaric flavonoids using MS2 and MS3 have been developed. Chromatographic separation of various plant extracts was done with RP‐HPLC and detected with positive ESI‐MS operated in information‐dependent acquisition (IDA) mode. Two methods for the determination of flavonoid identity and substitution pattern, both featuring IDA criteria, were used together with the HPLC equipment. A third method where the collision energy was ramped utilized direct infusion. With the developed strategies, it is possible to differentiate between many isobaric flavonoids. Various classes of flavonoids were found in all of the plant extracts, in the red onion extract 45 components were detected and for 29 of them the aglycone was characterized, while the substituents were tentatively identified for 31 of them. For the strawberry extract, those numbers were 66, 30 and 60, and for the cherry extract 99, 56 and 71. The great variety of flavonoids, several of them isobaric, found in each of the extracts highlights the need for reliable methods for flavonoid characterization. Using a parallel microcolumn switching liquid chromatography set-up coupled to a quadrupole time-of-flight mass spectrometer, a rapid liquid chromatography/mass spectrometric (LC/MS) protein identification method is presented. Without prior sample clean-up up to 300 protein digest samples a day can be processed. Using data-directed acquisition, up to 10 fragmentation analyses for each protein sample can be acquired in the same chromatographic run that can be used for database searching. Using internal peptide sequence information, protein databases and the various nucleic acid databases can both be queried for cross-species identification of the protein sample. The method was evaluated and put into force to generate data for a tobacco cell culture protein database.  相似文献

An efficient method was developed for toxicological drug screening in urine by liquid chromatography coupled with electrospray ionization time-of-flight mass spectrometry. The method relies on a large target database of exact monoisotopic masses representing the elemental formulae of reference drugs and their metabolites. Mass spectral identification is based on matching measured accurate mass and isotopic pattern (SigmaFit) of a sample component with those in the database. Data post-processing software was developed for automated reporting of findings in an easily interpretable form. The mean and median of SigmaFit for true-positive findings were 0.0066 and 0.0051, respectively. The mean and median of mass error absolute values for true-positive findings were 2.51 and 2.17 ppm, respectively, corresponding to 0.65 and 0.60 mTh. For routine screening practice, a SigmaFit tolerance of 0.03 and a mass tolerance of 10 ppm were chosen. Ion abundance differences from urine extracts did not affect the accuracy of the automatically acquired SigmaFit or mass values. The results show that isotopic pattern matching by SigmaFit is a powerful means of identification in addition to accurate mass measurement.  相似文献

The aim of this work was to develop a method for the characterization and determination of diuretics in human urine samples by liquid chromatography (LC) coupled to pneumatically assisted electrospray ionization (ES) mass spectrometry (MS). The diuretics studied were substances forbidden by the IOC such as trichlormethiazide, furosemide, canrenoic acid, benzthiazide, bendroflumethiazide, bumetanide, etacrynic acid and spironolactone. For this purpose, the operational parameters of electrospray, such as counter electrode voltage, capillary voltage, sample cone voltage and source temperature, were optimized in order to obtain the best signal stability and the highest sensitivity for the greatest number of diuretic agents. The optimized separation method was successfully coupled with the MS system to analyze the above-mentioned diuretics extracted from spiked urine samples by a liquid extraction and clean-up procedure at basic pH, using ethyl acetate as solvent and the salting-out effect (NaCl). The mass spectra obtained provide adequate information for identification purposes. Positive urine samples obtained from athletes were also analyzed. The presence of these substances in human urine was confirmed by this method, making LC/ES-MS an analytical tool to be considered in the area of antidoping control.  相似文献

A simple, sensitive and rapid high-performance liquid chromatography/electrospray ionization tandem mass spectrometry method was developed and validated for the assay of granisetron in human plasma. Following liquid-liquid extraction, the analytes were separated using an isocratic mobile phase on a reversed-phase C18 column and analyzed by MS in the multiple reaction monitoring mode using the respective [M+H]+ ions, m/z 313/138 for granisetron and m/z 409/228 for the IS. The assay exhibited a linear dynamic range of 0.1-20 ng/mL for granisetron in human plasma. The lower limit of quantification was 100 pg/mL with a relative standard deviation of less than 5%. Acceptable precision and accuracy were obtained for concentrations over the standard curve range. A run time of 2.0 min for each sample made it possible to analyze more than 400 human plasma samples per day. The validated method has been successfully used to analyze human plasma samples for application in pharmacokinetic, bioavailability or bioequivalence studies.  相似文献

An analytical method based on on-line solid-phase extraction-liquid chromatography coupled to electrospray tandem mass spectrometry (SPE-LC-ESI-MS/MS) for the determination of glyphosate and aminomethylphosphonic acid (AMPA) residues has been applied to the analysis of water samples within a Round Robin Study. The method had been previously validated in a variety of water samples and it fulfilled all the parameters of precision, accuracy, sensitivity and unequivocal confirmation. The results within the study that we participated were highly satisfactory in all cases with the only exception of glyphosate in groundwater samples, where surprisingly recoveries around 15% were obtained despite the use of isotope-labeled glyphosate as internal standard (I.S.). A slight modification has been introduced in the method, simply consisting of the acidification of sample with hydrochloric acid (HCl) to pH 1. Then, the sample is neutralized and immediately derivatized with 9-fluorenylmethylchloroformate (FMOC) before LC-MS/MS determination. Round Robin Study samples were reanalyzed using this approach, and the recoveries increased up to 98%. A possible explanation might be the slow kinetic interaction between glyphosate and some components of the matrix. These components might act as chelating agents, making glyphosate unavailable for derivatization and therefore for analysis. Several water samples collected at the Mediterranean area of Spain, and previously analyzed and being found to contain glyphosate, were also reanalyzed using this approach, obtaining higher concentrations (between 2 and 14 times) in most of cases.  相似文献

Erigeron breviscapus is an important herbal drug for the treatment of cardiovascular and cerebral vessel diseases. In this study, phenolic compounds were extracted from the whole plant of E. breviscapus and analyzed by high-performance liquid chromatography/electrospray ionization mass spectrometry. A total of 53 compounds were identified or tentatively characterized based on their UV and mass spectra. These compounds included caffeoylquinic acids (CQAs), CQA glucosides, malonyl-CQAs, acetyl-CQAs, caffeoyl-2,7-anhydro-3-deoxy-2-octulopyranosonic acids (CDOAs), caffeoyl-2,7-anhydro-2-octulopyranosonic acids (COAs), flavones, flavonols and flavonones. Most of them were reported for the first time from E. breviscapus and nineteen of them belonged to new compounds. The MS(n) spectra of CQA glucosides were similar to CQAs and they were discriminated by their retention times. No caffeic acid related ions (X(0) (-), Y(0) (-) and Z(0) (-)) were observed in MS(n) spectra of acyl-CQAs compared to those of CQAs. Fragment ions ((2,5)O(-), (3,6)O(-) and (4,6)O(-)) corresponding to ring cleavage were shown in MS(n) spectra of CDOAs and COAs, characteristic of this class of compounds. The 5,6,7-trihydroxyl-substituted flavones were dominant in E. breviscapus. Their [A--H](-) ions underwent the loss of a molecule of H(2)O, followed by the loss of CO, which was used to discriminate from other hydroxyl-substituted flavones. Our results are the first comprehensive analysis of E. breviscapus constituents and will be helpful for the quality control of the herb of E. breviscapus and its related preparations.  相似文献

A method based on the on-line coupling of reversed phase liquid chromatography with gas chromatography/mass spectrometry (RPLC-GC-MS) for the chiral evaluation of characteristic constituents of fruit beverage aroma was investigated. The consideration of a variety of parameters involved in the transfer step allowed to achieve relative standard deviations ranging from 0.4 to 10% in most cases and detection limits from 0.2 to 2.5 mg/l. By applying the developed method to fruit beverages, racemic mixtures of ethyl 2-methylbutanoate and gamma-nonalactone were found. This fact suggests the eventual addition of artificial aromas. The method proposed in the present work can be useful to assess reliably the authenticity of aqueous samples, such as fruit beverages.  相似文献

A rapid and sensitive method for the determination of the phthalate monoesters monoethyl phthalate (MEP), monobutyl phthalate (MBP), monobenzyl phthalate (MBzP) and monoethylhexyl phthalate (MEHP), in human urine, using packed capillary column liquid chromatography coupled to electrospray quadrupole-ion trap mass spectrometry (ESI-QITMSn) has been developed. Sample volumes of 200 L of deconjugated and diluted urine were loaded onto a precolumn of 30 mm×0.32 mm I.D. packed with Hypercarb 5 m particles, using a sample carrier consisting of acetonitrile/water (15/85, v/v, adjusted to pH 2 using HCl) with a flow rate of 20 L/min. Backflushed elution onto a 100 mm×0.32 mm I.D. analytical column packed with 5 m Hypercarb particles was conducted using a tetrahydrofuran/water gradient where both solvents contained 10 mM ammonium acetate, at a flow rate of 4 L/min. Determination of the monophthalates was achieved within 8 min. Ionization was performed in the negative mode and the analytes were observed as [M-H] at m/z=193.1, 221.1, 255.1 and 277.0 for MEP, MBP, MBzP and MEHP, respectively. Quantification was performed in the multiple reaction monitoring (MRM) mode monitoring the fragments at m/z=121.1, 177.0, 183.0 and 233.0 for MEP, MBP, MBzP and MEHP, respectively. The method was validated over the concentration range 2.5–125 ng/mL in pretreated urine samples, corresponding to 25–1250 ng/mL untreated urine, yielding correlation coefficients in the range 0.996–0.999. The within-assay (n=6) and between-assay (n=6) repeatabilities were in the range 4.0–18% and 4.8–15% RSD, respectively. The mass limits of detection were in the range 32–70 pg, corresponding to concentration limits of detection of 1.6–3.5 ng/mL of untreated urine.  相似文献

A non-aqueous reversed-phase liquid chromatographic method coupled to electrospray ionisation (ESI) tandem mass spectrometry was developed for the analysis of triacylglycerols (TGs). The synthetic TGs studied were separated according to their equivalent carbon number with a gradient of methanol (containing 0.01% (v/v) formate adjusted to pH 5.3 with ammonia) and chloroform. ESI mass spectra of TGs yielded positive ion current signals for [M + NH(4)](+) and [M + NH(4)-RCOONH(4)](+). The mass spectra also showed signals believed to arise from [M + K](+). Collision-induced dissociation (CID) of the [M + NH(4)](+) precursor ion yielded [M + NH(4) - RCOONH(4)](+), [RCO + 74](+) and [RCO](+) product ions as aids for the structural elucidation of the TGs. In addition, [RCO - 18](+) and small amounts of [RCO - 2](+) product ions were also found. The latter ions were observed only for TGs containing unsaturated fatty acids. CID of ammoniated 1-stearoyl-2-oleoyl-3-linoleoyl-glycerol (18:0/18:1/18:2) indicated that neutral loss of the sn-2 fatty acid was energetically less favourable than loss of the fatty acid from the sn-1 or sn-3 position.  相似文献

The simultaneous determination of morphine and the glucuronide metabolites [morphine-3-beta-D-glucuronide (M3G) and morphine-6-beta-D-glucuronide (M6G)] in rat hair and rat plasma was carried out using reversed-phase high-performance liquid chromatography (HPLC) coupled with electrospray ionization mass spectrometry (ESI-MS). The chromatographic separation of the analytes was achieved using a semi-micro-HPLC column (3 microm particle size; 100 x 2.0 mm id) by gradient elution with 50 mM ammonium acetate and acetonitrile as eluents. After separation, morphine and the glucuronides were determined by selected ion monitoring (SIM) of ESI-MS using the quasi-molecular ions [M + H]+ at m/z = 286 and 462, respectively. The calibration curves were linear between the concentration of the analytes and the deuterium-labelled morphine (M-d3) selected as internal standard. The method was applied for the determination of the incorporation of morphine and the glucuronides into the hair shafts and hair roots of Dark Agouti rats after single intraperitoneal administration of morphine hydrochloride. Plasma concentrations of morphine and glucuronides were simultaneously determined after administration. Morphine and M3G were detected in the hair shafts and the hair roots. The concentrations of M3G in the hair root were lower than those of morphine in all sampling periods. In contrast, M3G concentrations in plasma were relatively higher at each sampling time. Small quantities of M6G were also identified in the plasma up to 4 h after administration. The concentration difference between the hair root and plasma seems to be due to the incorporation ratio of morphine and glucuronide into hair. As M3G was also identified in the hair shaft 1 week after administration, the incorporation of glucuronide metabolites into hair is obvious. This is the first report of the identification of morphine glucuronide in hair samples without the use of acid hydrolysis or enzyme digestion.  相似文献

The quantitative determination of nucleotides from DNA modified by styrene oxide is described using a combination of inductively coupled plasma high-resolution mass spectrometry (ICP-HRMS) and electrospray ionization mass spectrometry (ESI-MS), both interfaced to reversed-phase high-performance liquid chromatography (HPLC). LC/ICP-MS (resolution > 1500 to discriminate against 15N16O+ and 14N16OH+) was employed to determine quantitatively the content of modified nucleotides in standard solutions based on the signal of phosphorus; phosphoric acid served as an internal standard. By means of the standard addition technique the sensitivity of the LC/ESI-MS approach was subsequently determined. Since a comparison of UV, ICP and ESI-MS data suggested that in ESI-MS the ionization efficiency of the adducts is identical within the error limits, quantitative determination of all adducts is possible. For LC/ESI-MS with single ion monitoring, the detection limit for styrene oxide adducts of nucleotides was determined to be 20 pg absolute or 14 modified in 10(8) unmodified nucleotides in a 5 micrograms DNA sample, which comes close to the best methods available for the detection of chemical modifications in DNA.  相似文献
