A convergent formal total synthesis of OF 4949 III is described. Arene-ruthenium chemistry was used in the construction of the diaryl ether linkage in high yield, and cycloamidation under high dilution conditions (0.005 M) was achieved using DPPA as coupling reagent. SmI(2) was used to reductively remove the 2-iodoethyl ester protecting group in the presence of DMPU or HMPA.  相似文献

Hapalosin was initially synthesized by macrolactonization, and a second synthesis was achieved by cycloamidation. In both syntheses, three of the five stereocenters in hapalosin were established by two Brown allylboration reactions. The synthesis of the non-N-Me analog of hapalosin involved chelation-controlled reduction of a gamma-amino-beta-keto ester and cycloamidation. In CDCl(3) at 25 degrees C, synthetic hapalosin exists as a 2.3:1 mixture of conformers, while its non-N-Me analog exists only as a single conformer. (1)H,(1)H-NOESY and computation reveal that the configuration of the amide bond is responsible for the conformations of the two compounds. The major conformer of hapalosin is found to be an s-cis amide, the minor conformer an s-trans amide, and the non-N-Me analog an s-trans amide. Applying distance constraints to protons that exhibit NOESY correlations, computation shows that the major conformer of hapalosin and the non-N-Me analog have very different conformations. By contrast, the minor conformer of hapalosin and the non-N-Me analog have very similar conformations.  相似文献

Synthesis of an amino acid analog. N-α-Fmoc-N-ε-tetrabutyl ester EDTA-L-Lysine, suitable for incorporating a strong metal binding site at any internal sequence position in a peptide is described. To overcome the solubility and purification problems during the synthesis, we prepared esters of N-α-Fmoc-N-ε-Boc-L-Lysine. Since the t- butyl group on the amino acid is acid labile and Fmoc group at α-amino group is base sensitive, protection of N-α-Fmoc-N-ε-Boc-L-Lysine requires an ester that can be prepared and removed under neutral conditions. A scheme for selective protection and deprotection of lysine is reported.  相似文献

In an attempt to resolve a racemic mixture of a trishomocubane hydantoin, the synthesis of a pair of novel diastereomers was obtained by protecting the racemic hydantoin with chlorocarbonic acid-(–)(R)-sec-butyl ester. An achiral i-propyl ester was first used to establish the procedure. The NMR elucidation of both the chiral and achiral N-protected hydantoins is described. Some proton and carbon NMR shifts on the cage are reversed when relative small changes on the protection group are introduced. The chiral centre on the protective group induced splitting of some carbon signals in the 13C spectrum on the cage skeleton, but effective separation of the diastereomers could not be obtained. In a further attempt to demonstrate the potential use of the trishomocubane amino acid in peptide synthesis, the ethyl ester of the cage amino acid was synthesised. The structures of the amino acid derivatives were elucidated with 2D NMR techniques and the assignment of the NMR data is presented.  相似文献

An asymmetric synthesis of fused bicyclic amino acids having a hexahydro-cyclopenta[c]pyridine skeleton and carrying besides an enone structural element a substituent at the beta-position is described. The key steps of the synthesis are a highly selective allylation of N-tert-butylsulfonyl imino ester with bis(allylsulfoximine)titanium complexes and a highly diastereoselective Pauson-Khand cycloaddition of sulfonimidoyl-substituted gamma,delta-unsaturated alpha-amino acid esters carrying a substituent at the beta-position and a propargyl group at the N-atom. The cyclization is accompanied by a reductive cleavage of the sulfoximine group of the primary cyclization product. Surprisingly, the removal of the sulfoximine group proceeds with inversion of the configuration at the S-atom and gives N-methyl-phenylsulfinamide with >/=98% ee. Deprotection of the bicyclic N-tert-butylsulfonyl-protected amino acid ester was accomplished through treatment with CF(3)SO(3)H under anhydrous conditions. The enantio- and diastereomerically pure sulfoximine-substituted gamma,delta-unsaturated alpha-amino acid esters used as starting material were obtained through a highly regio- and diastereoselective allylation of N-tert-butylsulfonyl imino ester with acyclic bis(allylsulfoximine)titanium complexes, described previously.  相似文献

A highly stereoselective and efficient total synthesis of trans-dihydronarciclasine from a readily available chiral starting material was developed. The synthesis defines two of the five stereogenic centers of the natural product by an amino acid ester-enolate Claisen rearrangement. The other three stereogenic centers are created in a highly stereocontrolled fashion via a six-ring vinylogous ester intermediate, which is generated from the γ,δ-unsaturated ester functional group of the Claisen rearrangement product in an efficient three-step sequence. This concise total synthesis exemplifies the use of a highly regioselective Friedel-Crafts-type cyclization to form the B ring via an isocyanate intermediate derived from an N-Boc group, which is superior to the conventional method using an imino triflate intermediate. This same N-Boc group is employed to give high selectivity in the Claisen rearrangement earlier in the sequence.  相似文献

Elucidation of the mechanism for peptide bond formation observed when an amino acid ester is treated with anhyd CuCl2 in an anhyd alcoholic solvent, was attempted using results of IR and visible spectra measurements of the amino acid ester-CuCl2 complex and accumulated experimental data.This novel reaction proceeds through a mechanism in which the amino anion produced by elimination of the proton from the Cu(II)-coordinating amino group, attacks the non-activated ester CO group of the amino acid ester which shares a common Cu(II) ion. It differs completely from the peptide formation reaction featuring a Co(III)-amino acid ester complex.  相似文献

A seven-step solid-phase synthesis of spirohydantoins and an eight-step solid-phase synthesis of spiro-2,5-diketopiperazines is reported. Key intermediate in the synthesis of both compound libraries is the resin-bound cyclic alpha,alpha-disubstituted alpha-amino ester, which can be obtained after selective homogeneous reduction of the aliphatic nitro ester using tin(II) chloride dihydrate. Nitro ester, in turn, is synthesized by a high-pressure-assisted [4 + 2] cycloaddition of resin-bound nitro alkene and butadiene, whereas nitro alkene is obtained by a Knoevenagel condensation of resin-bound nitro acetate with an imine. Novel spirohydantoins are obtained by isocyanate coupling with the resin-bound amino ester 5, followed by cyclization cleavage using a base. Novel spiro-2,5-diketopiperazines are obtained by PyBOP coupling of a Fmoc-protected amino acid with resin-bound amino ester, followed by Fmoc deprotection and an acid-assisted cyclization cleavage. After preparation of seven different resin-bound alpha,alpha-disubstituted alpha-amino esters, a 7 x 8 compound library of spirohydantoins was synthesized using eight different isocyanates, and a 7 x 8 compound library of spiro-2,5-diketopiperazines was synthesized using eight different Fmoc amino acids.  相似文献

The synthesis of the novel amino acid 6-(hydroxymethyl)-2-piperidinecarboxylic acid ( 1a ) and its ethyl ester 1b is reported. In the hydrogenation of 6-(hydroxymethyl)pyridine-2-carboxylates, hydrogenolysis of the alcohol group appeared as an unusual side reaction. Optimization of the reaction conditions allowed us to minimize hydrogenolysis and afforded pure 1 .  相似文献

[see structure]. A fullerene derivative containing a free amino group has been condensed with N-Fmoc-L-glutamic acid alpha-tert-butyl ester to give a C60-functionalized amino acid. The carboxylic end of this amino acid has been deprotected in acidic conditions, and the resulting acid has been used for solid-phase peptide synthesis. The final peptide, cleaved from the resin, was very soluble in water solutions and showed antimicrobial activity against two representative bacteria.  相似文献

Syntheses of Phosphonic- and Phosphinic Analogues of Pantothenic Acid Ethyl Ester and of the Phosphonic Analogue of Pantetheine The replacement of amino acids in peptides by phosphono-analogous (aminoalkyl)phosphonic acids 1 , (2-aminoethyl)phosphonic acid ( 2 ) and substituted derivatives has been an important aspect of peptides research in the last years. In pantothenic acid ( 3 ), there is a peptide linkage between (2R)-2,4-dihydroxy-3,3-dimethylbutyric acid and the amino group of β-alanine, and in pantetheine ( 4 ), there is a second peptide linkage between the β-alanine and cysteamine. The synthesis of phosphono and phosphino analogues of pantothenic acid ethyl ester, where the β-alanine is replaced by the diethyl ester of (2-aminoethyl)phosphonic acid and the ethyl ester of (2-aminoethyl)methylphosphonic acid, respectively, and the syntheses of the phosphono analogue of pantetheine, where the β-alanine is replaced by (2-aminoethyl)phosphonic acid, are described.  相似文献

The glycosyl amino acids α-GalNAc-Ser and α-GalNAc-Thr are fundamental building blocks for glycopeptide synthesis, Schmidt’s synthesis method often being chosen for this purpose. Methyl esters used as orthogonal carboxylic acid protecting group in this procedure were found to be an efficient and inexpensive alternative to other groups. The mild selective methyl ester deprotection by LiI improved the efficiency of the synthesis method.  相似文献

The bromacetamido group, allowing the reversible anchoring of the first amino aci by an ester bound deriving from a glycolic amide, fits the conditions required by solid-phase synthesis on polyacrylic resins. Final clivage of the peptide from the support can be realised in smooth, quantitative and non racemizing conditions, allowing alternatively the isolation of a terminal acid, ester or amide function. Synthesis of the 1-14 residue of human angiotensinogen illustrates this new methodology.  相似文献

Benzotriazol-1-yloxytris(dimethylamino)phosphonium hexafluorophosphate reagent (BOP) serves as an efficient and versatile coupling reagent for the coupling of conjugated carboxylic acid with methyl ester amino acids hydrochloride allowing the synthesis of various substituted amino acid derivatives in high chemical yields of up to 90%. Stereoselective synthesis of non-proteinogenic amino acids via Michael addition to 3,4-didehydropyroglutamate derivative in which the carboxyl function is protected as a 2,7,8-trioxabicyclo[3.2.1]octane (ABO ester) group is described. The obtained 3-substituted pyroglutamic ABO ester was directly converted to 3-substituted glutamic acids such as chlorpheg by acidic hydrolysis, whereas 3-substituted proline derivatives were obtained by chemoselective reduction of the lactam carbonyl group. A formal total synthesis of baclofen, a γ-aminobutyric acid (GABA) analogue, is also described.  相似文献

The key strategy involves a stereoselective synthesis of the polyketide unit and synthesis of the D-tyrosine derivative, followed by assembly of the fragments by an esterification and cycloamidation reaction sequence. The synthesis of the polyketide fragment was achieved by an iterative asymmetric synthesis to install stereoselectively both 1,3-dimethyl groups and the 1,3-diol unit by utilizing asymmetric cyclopropanations and Sharpless asymmetric epoxidations as the key steps.  相似文献

A novel, ethylene glycol-based protecting group is designed and synthesized for use in solid phase peptide synthesis. Ether and ester type protected amino acids are prepared. The acid stability of the new protecting group showed complete Fmoc/t-Bu compatibility. The new derivatives are tested in solid phase peptide synthesis, with a ‘difficult’ sequence to examine the disruption of peptide aggregation.  相似文献

A new type of fused oxazepinones 9a and 9b , which are analogues of sclerotigenin and circumdatin F, were obtained in a two step synthesis from 2‐(2‐amino‐benzoylamino)‐benzoic acid or the corresponding methyl ester. Secondly a new synthesis of circumdatin F arose from this work, where 2‐(2‐propionylamino‐benzoylamino)‐benzoic acid methyl ester was used as an intermediate.  相似文献
