A Pseudomonas sp. was cultured which was associated with the Japanese seaweed Diginea sp. Crude extracts prepared from this bacterial culture were found to inhibit the growth of other marine bacterial strains. From this bacterial culture, two new peptides cyclo-[phenylalanyl-prolyl-leucyl-prolyl] (3) and cyclo-[isoleucyl-prolyl-leucyl-alanyl] (4) have been isolated together with two known peptides (1) and (2). The crude extract from a culture of Pseudoalteromonas sp. associated with the Thai sponge Halisarca ectofibrosa was found to inhibit the growth of Bacillus subtilis and Vibrio anguillarum. Isolation studies yielded a fraction containing two peptides that were identified as cyclo-[phenylalanyl-leucyl]2 (5) and cyclo-[leucyl-isoleucyl]2 (6) by means of LC-MS and 2D NMR data. Absolute stereochemistry was confirmed by the synthesis of cyclo-[l-phenylalanyl-l-leucyl]2. Peptides (1)-(3) were also isolated from this bacterial strain. None of the individual peptides isolated in this study showed antibiotic activity.  相似文献

An NMR study of 10 l-alanine- and l-valine-containing peptides was carried out in the native [C2MIM][Cl], [C4MIM][Cl], [C6MIM][Cl], [C4MIM][BF4], [C4MIM][PF6], and [C4Py][BF4] ionic liquid media. A unique high sensitivity of the ionic liquid system to the nature of peptide and ability to tune solvent–solute interactions were observed in contrast to regular organic solvents. The l-valine peptides can be selectively dissolved in [C4MIM][Cl] and [C6MIM][Cl], whereas their solubility in [C2MIM][Cl] and other ionic liquids was dramatically lower. In spite of structural similarity between the amino acids, a distinct behavior was observed for the l-alanine peptides. Solvent–solute interactions with an ionic liquid impose significant changes, and NMR spectroscopy is a useful probe for the molecular-level and nanoscale organization of the studied systems. An even/odd effect of the number of amino acids in the peptide on molecular interactions in ionic liquids was observed. Enhancement of chemical properties of peptides in ionic liquids and application of ionic liquids in the separation of peptides are the areas of practical interest in the studied systems.  相似文献

The synthesis of the Boc-protected 1-(2-aminoethyl)-3-methylimidazolium salts [BocNHCH2CH2ImMe]X [2]X (X = I, PF6) and their straightforward transformation into [NH2CH2CH2ImMe]X [3]X is reported. The reaction between [2]X and Ag2O leads to the formation in the solid state of three different bonding motifs: a biscarbene salt [(NHC-NHBoc)2Ag]PF6 ([4]PF6, NHC-NHBoc = 1-(2-BocNH-ethyl)-3-methyl-imidazolin-2-ylidene), a tetranuclear complex [Ag(NHC-NHBoc)2]2[Ag2I4], (5), and a polymeric silver “staircase” [(NHC-NHBoc)2-Ag4-I4]n, (6) composed of Ag4I4 clusters. The same reaction carried out with [3]I showed that a primary silver mono-NHC-NH2 carbene complex of the type [(NHC-NH2)AgI] (7) is likely to form but it is unstable in solution. The solid state molecular structures of [4]PF6, 5 and 6 were determined by X-ray diffraction analysis, whereas PGSE NMR experiments were employed to investigate the hydrodynamic dimension of the imidazolium salts and silver complexes and, consequently, to gain information on the level of aggregation in solution. PGSE NMR studies were complemented by NOE NMR investigations in order to obtain information on anion-cation relative orientation within aggregates.  相似文献

Three different kinds of ring carbo-mers of [5]cyclitol ethers were targeted as challenging examples of functional [5]pericyclynes. Three tertiary pentaaryl-carbo-[5]cyclitol methyl ethers were synthesized through a [11+4] ring-closing double addition of triphenyl- and tri-p-anisyl-undecatetrayn-diides to dibenzoylacetylene. These compounds, obtained as oily mixtures of stereoisomers, are stable and can behave as acetylenic ligands of one or two Co2(CO)6 units. NMR analysis reveals that the broad diasteroisomeric dispersity of a triether, is consistently reduced in the symmetrized pentaether. Three bis-secondary triaryl-carbo-[5]cyclitol methyl ethers with adjacent CH(OR) vertices were synthesized through a similar [11+4] ring-closing process, where the same tetrayn-diides add to both the carbaldehyde ends of the (η2-OCH-CC-CHO)Co2(CO)6 complex. Despite the possibility of tautomeric isomerization, the occurrence of two adjacent bis-propargylic carbinol vertices does not diminish the stability of the [5]pericyclyne framework. Finally, two bis-secondary carbo-[5]cyclitol methyl ethers with non-adjacent CH(OH) vertices were synthesized through an alternative [10+5] ring-closing process. The bis-secondary carbo-[5]cyclitols are regarded as isohypsic equivalents of the challenging [C,C]5carbo-cyclopentadienyl cation. A diphenyl-hexaoxy-[5]pericyclyne with two non-adjacent secondary carbinol vertices was also prepared through a [10+5] ring-closing strategy: this molecule is an isohypsic equivalent of the previously calculated zwitterionic carbo-cyclopentadienone, which could be observed as a DCI/NH3-MS fragment after treatment with SnCl2/HCl. Analytical HPLC showed that the C11 triphenyl-undecatetrayne precursor of the [11+4] strategy was obtained as a statistical 1:2:1 mixture of the three possible diastereoisomers. Semi-preparative HPLC allowed for the resolution of this mixture. The pure major diastereoisomer was employed to prepare a partly resolved sample of pentamethoxy-pentaphenyl-[5]pericyclyne. Analytical HPLC showed that the latter corresponds to the statistical distribution of the expected three residual diastereoisomers. Semi-preparative HPLC finally afforded samples of diastereoisomerically pure pentamethoxy-[5]pericyclyne as crystalline solids.  相似文献

A series of novel N-benzylcarboxamide derivatives of bicyclic compounds, 3,4-dihydropyrido[3,2-f][1,4]oxazepin-5(2H)-one and 2,3,4,5-tetrahydro-6H-pyrido[2,3-b][1,5]oxazocin-6-one, were synthesized by cyclization of N-benzyl-2-chloro-N-(2-hydroxyethyl)- [and -(3-hydroxypropyl)-] nicotinamides, respectively. Atropisomerism was observed in 5-[3,5-bis(trifluoromethyl)benzyl]-7-phenyl-2,3,4,5-tetrahydro-6H-pyrido[2,3-b][1,5]oxazocin-6-ones due to steric hindrance of the carboxamide moiety and restriction of its rotation. Cyclization of N-[3,5-bis(trifluoromethyl)benzyl]-2-chloro-N-[(2S)-3-hydroxy-2-methylpropyl]-5-methyl-4-phenylnicotinamide gave (3S)-5-[3,5-bis(trifluoromethyl)benzyl]-3,8-dimethyl-7-phenyl-2,3,4,5-tetrahydro-6H-pyrido[2,3b][1,5]oxazocin-6-one, which exists predominantly in the thermodynamically stable aR-conformer in CDCl3. This compound showed excellent NK1-antagonistic activity with IC50 value (in vitro inhibition of [125I]-Bolton-Hunter-substance P binding in human IM-9 cells) of 0.47 nM, which is ca. 200-fold more potent than that of its enantiomer, indicating that the atropisomer chirality affects NK1-receptor recognition.  相似文献

[CuII(Ma)(Mb)]?2+ complexes, where Ma and Mb are dipeptides or tripeptides each containing either a tryptophan (W) or tyrosine (Y) residue, have been examined by means of electrospray tandem mass spectrometry. Collision‐induced dissociations (CIDs) of complexes containing identical peptides having a tryptophan residue generated abundant radical cations of the peptides; by contrast, for complexes containing peptides having a tyrosine residue, the main fragmentation channel is dissociative proton transfer to give [Ma + H]+ and [CuII(Mb – H)]?+. When there are two different peptides in the complex, each containing a tryptophan residue, radical cations are again the major products, with their relative abundances depending on the locations of the tryptophan residue in the peptides. In the CIDs of mixed complexes, where one peptide contains a tryptophan residue and the other a tyrosine residue, the main fragmentation channel is formation of the radical cation of the tryptophan‐containing peptide and not proton transfer from the tyrosine‐containing peptide to give a protonated peptide. The absolute configurations of the two algal pheromones multifidene 1 and viridiene 2 were determined as (+)-(3S, 4S)-3-[(Z)-1-butenyl]-4-vinylcyclopentene and (+)-(3R, 4S)-3-[(1Z)-1, 3-butadienyl]-4-vinylcyclopentene, respectively. The strategy involves enzyme-initiated asymmetric synthesis of the ring-saturated pheromone analogues (+)- 8a and (?)- 8b and their subsequent catalytic hydrogenation to the chiral cycloalkanes 9a and 9b , only the letter of which is also obtained from the two natural messengers (+)- 1 or (+)- 2 . Biological activity assays proved these enantiomers of 1 or 2 to be the characteristic pheromones for male gametes of the seaweeds Syringoderma, Cutleria multifida and Chorda tomentosa.  相似文献

The possible inclusion complexes of Cp2NbCl2 into calixarenes hosts have been investigated. The existence of a true inclusion complex in the solid state was confirmed by a combination of NMR, ab-initio calculations, thermogravimetric analysis, FTIR, Raman and PXRD. Ab-initio calculations, 1H NMR solution and solid state 13C CP-MAS NMR results demonstrated that p-sulfonic calix[6]arene does form an inclusion complex with Cp2NbCl2. Raman spectroscopy showed, for the inclusion compound of p-sulfonic calix[6]arene-Cp2NbCl2, a band between 500 and 850 cm−1 characteristic of Nb-O vibration. This result suggests that Nb(V) may engage in coordination with the oxygen of the sulfonate group, as part of the host-guest interaction. However, it is important to mention that the niobocene dichloride (Cp2NbCl2) dissolves in water and undergoes oxidation and hydrolysis processes to yield Cp2NbCl2(OH) species. For that reason this band does not exclude that the Nb-O band belongs to Cp2NbCl2(OH). Solid State 13C CP-MAS NMR and solution 1H NMR spectroscopies together with ab-initio results showed that Cp2NbCl2 is included in the p-sulfonic calix[6]arene cavity, with both Cp rings inside the cavity. In contrast, the solution 1H NMR results demonstrated that calix[6]arene does not form inclusion complex with Cp2NbCl2 in CDCl3 solution. Cp2NbCl2 is not included in the calix[6]arene cavity, possibly due to the lack of sulfonate heads which promote Nb-O interactions and assist the inclusion of Cp2NbCl2 into the cavity.  相似文献

Solid-phase microextraction (SPME) samplings from live insects or natural secretion allowed one to identify the aggregation pheromones of the pest beetles Scapanes australis and Strategus aloeus by efficient and rapid isolation of their highly volatile (72 < M(r) < 116) components. S. australis male pheromone was identified as a 84:12:4 (w/w) mixture of 2-butanol [67:33 (R)-(-):(S)-(+) ratio], 3-hydroxy-2-butanone and 2,3-butanediol [43:17:40 (R,R)-(-):(S,S)-(+):meso ratio], and S. aloeus pheromone as a 95.5:4.0:0.5 (w/w) mixture of 2-butanone, 3-pentanone and sec.-butyl acetate by GC-MS using conventional and chiral capillary columns. This is the first report of Scarabaeidae pheromones based on such small and common molecules.  相似文献

The reaction of [2.2]paracyclophane with ferrocene in the presence of AlCl3 and Al/powder gives the new compounds [1-6-η-[2.2]paracyclophane- (η5-cyclopentadienyl)iron]+[PF6] and [1-6-η; 9-14-η-[2.2]paracyclophane- [(η5-cyclopentadienyl)iron]2]2+[PF6]2 in high yields. The species have been characterized by elemental analysis, and by 1H and 13C NMR spectroscopy.  相似文献

We report the preparation of 1-tetrahydro-pyranyloxy dodec 11-yne(1), from castor oil, with an overall yield of 9.6%, and illustrate its use with the novel and practical synthesis of 1-oxo octadec (z) 11-ene(2), the insect sex pheromone of Achroia grisella, a species of wax moth, which are specific enemies of bees1. Parenthetically, it has been found that 1 is a good precursor to the surprisingly large number of pheromones that have the structural unit, [sbnd]CH[dbnd]CH[sbnd](CH2)9CHXY2.  相似文献

A variety of backbone-modified peptides were desorbed by fast atom bombardment and collisionally activated. These peptide modifications involve the replacement of a normal [CONH] peptide linkage with such groups as thiomethylene ether (CH2S), thioamide (CSNH), methyleneamine (CH2NH), and thiomethylene sulfoxide (CH2SO) moieties. Modified linear peptides decompose to give fragmentations characteristic of the modifications as well as typical peptide bond fragments. The presence of a replacement group in cyclic peptides can induce new fragmentations. The presence of other functional groups, such as an exocyclic N-terminal residue, however, can dominate the observed fragmentations. Upon collisional activation, unmodified linear peptides fragment to give N-terminal ions as the most abundant daughter ions. In comparison, ψ[CH2NH] and ψ[CH2S ] modified linear peptides decompose to give prominent C-terminal sequence ions. The ψ[CH2SO] modified linear peptides, however, fragment into both N- and C-terminal ions of high relative abundance. Depending on the modification, daughter ions or internal fragment ions are observed that are characteristic of the amide bond replacement. Useful structural information can therefore be obtained.  相似文献

Both enantiomers of (3Z,9Z)-cis-6,7-epoxy-3,9-octadecadiene, one of which is the major component of the sex pheromone of Ectropis oblique Prout, were synthesized in 23% overall yield for the (?)-(6S,7R)-enantiomer and 18% yield for the (+)-(6R,7S)-isomer. This protocol uses a sequential regioselective ring-opening strategy and provides a convenient and reliable access to other structurally related insect sex pheromones. Preliminary biological studies revealed that (?)-(6S,7R)-2a was roughly as active as the natural pheromone, while racemic (±)-2 was less bioactive and (+)-2b was much less bioactive.  相似文献

New 2′-deoxyadenosine and adenosoine modifications: 8-[(2-dimethylaminoethyl)amino]-2′-deoxyadenosine and 8-[(2-dimethylaminoethyl)amino]adenosine were prepared and their reactivity towards cyclic oxonium adducts of closo-dodecaborate and cobalt-bis-dicarbollide was studied. The cleavage reactions of clusters oxonium rings by N,N-dimethylanio group of modified nucleosides led to the first [B12H12]2− and new [Co(C2B9H11)2] conjugates with adenosine and 2′-deoxyadenosine respectively. The proposed methodology provides a convenient route for the synthesis of libraries of boron cluster modified adenosine and 2′-deoxyadenosine derivatives for biological screening.  相似文献

The identification of the sex pheromone of the lesser date moth, Batrachedra amydraula (Meyrick) was based on GC-MS analysis of volatiles released by virgin females using sequential SPME auto-sampling of headspace and by synthesis of the key component, (4Z,7Z)-4,7-decadien-1-yl acetate. Substantial capture of males in a date palm plantation using a bait consisting of the key component and 5Z-decen-1-yl acetate in a ratio of 1:2 indicated that these are the essential components of the sex pheromone. Addition of 4Z-decen-1-yl acetate and decan-1-yl acetate, which were also identified, did not affect trap-capture. The lesser date moth is the first member of the Batrachedridae whose sex pheromone has been identified. (4Z,7Z)-4,7-Decadien-1-yl acetate is a novel compound among moth sex pheromones.  相似文献

Intramolecular base catalyzed C-C bond formation led to exclusive stereoselective syntheses of trans-6,7-dihydro-5H-[1,2,4]triazolo[3,4-b][1,3,4]thiadiazines, trans-7,8-dihydro-6H-[1,2,4]triazino[3,4-b][1,3,4]thiadiazin-4-ones, and trans-3,4-dihydro-2H-[1,3,4]thiadiazino[2,3-b]quinazolin-10-one. trans-6,7-Dihydro-5H-[1,2,4]triazolo[3,4-b][1,3,4]thiadiazines isomerize slowly in CDCl3 and more rapidly in DMSO-d6 into the corresponding cis-stereoisomers. The other trans-6,7-dihydro-[1,3,4]thiadiazines isomerize also in DMSO-d6 into the corresponding cis-stereoisomers. Pyrolytic conversion of these heterocyclic condensed dihydrothiadiazines into their corresponding pyrazolo[5,1-b][1,2,4]triazoles, pyrazolo[5,1-c][1,2,4]triazin-4-ones and pyrazolo[4,3-b]quinazolin-9-ones via desulfurization ring contraction is described.  相似文献

Syntheses, Single-Crystal X-Ray Analyses and Solid-State 29Si NMR Studies of a Zwitterionic λ5-Spirosilicate and a Cage-like Octa(silasesquioxane) The zwitterionic λ5-spirosilicate bis[2,3-naphthalenediolato(2 ?)][2-(dimethylammonio)phenyl]silicate ( 1 ; isolated as 1 · 1/2 CH3CN) was synthesized by reaction of the [2-(dimethylamino)phenyl]dimethoxyorganosilanes 5, 6 and 7 [2-(Me2N)C6H4Si(OMe)2R: R = Ph ( 5 ), cyclo? C6H11 ( 6 ), Me ( 7 )] with 2,3-dihydroxynaphthalene in acetonitrile at room temperature. Reaction of 1 · 1/2 CH3CN or [2-(dimethylamino)phenyl]trimethoxysilane ( 3 ) with water in acetonitrile yielded the cage-like octa{[2-(dimethylamino)phenyl]silasesquioxane} ( 2 ). The crystal structures of 1 · 1/2 CH3CN and 2 were studied by X-ray diffraction. In addition, 1 · 1/2 CH3CN and 2 were characterized by solid-state (29Si CP/MAS) and solution NMR studies (1H, 13C, 29Si).  相似文献

Starting from tryptophan methyl ester, a three-step synthesis of fused tetrahydro-β-carbolinequinoxalinones in two new ionic liquids, [bdmim][Tf2N] and [bdmim][PFBuSO3], was described. Both ionic liquids can be readily prepared from commercially available starting materials in high yields. Unlike the commonly used [PF6]-based ionic liquids that evidently undergo slow hydrolysis of the PF6 anion with the concomitant release of HF, ionic liquids of [bdmim][Tf2N] and [bdmim][PFBuSO3] are not only chemically stable but also apparently inert to hydrolysis and therefore organic reactions carried out in both ionic liquids proceed smoothly with good yields. The overall isolated yields for this three-step synthesis of tetrahydro-β-carbolinequinoxalinones were 34-55%. To the best of our knowledge, the preparation of fused tetrahydro-β-carbolinequinoxalinones was unprecedented.  相似文献

An investigation of a skeletal Wagner-Meerwein rearrangement of variously substituted or quinoline-annulated 3a,6;4,5-diepoxyisoindol-1-ones is reported. Optimum reaction conditions (Ac2O, BF3·OEt2, rt) were discovered for the formation of the target 4,6-epoxycyclopenta[c]pyridines in 40-80% yields. It was shown that the direction of the sigmatropic rearrangement of 3a,6;4,5-diepoxyisoindol-1-ones depended dramatically on the carboxyl group position (exo-/endo-) in the oxabicyclo[2.2.1]heptane moiety. The spatial structure of previously unknown 7,9-epoxycyclopenta[4,5]pyrido[1,2-a]quinolines derived from Wagner-Meerwein rearrangement of 2,11b-epoxyoxireno[6,7]isoindolo[2,1-a]quinolines was established based on the X-ray analysis data. The skeletal rearrangement proceeded regio- and stereospecifically in all the cases examined due to the absence of the epimerization of the carbon atoms adjacent to the carbocation centres.  相似文献
