Auflichtphotometrie von Mikroautoradiogrammen für quantitative Einbaustudien an Einzelzellen

By means of micro-autoradiography incorporation studies of labelled compounds can be performed at the cellular level. The procedure described here aims at determining the amount of substances in terms of moles, incorporated into single cells. ¹⁴C is used as a label, since in contrast to tritium, the influence of β-self-absorption of this isotope is negligible. Every autoradiograph is exposed together with a standard consisting of ¹⁴C-methacrylate. Hereby the relation of grain density to radioactivity in an autoradiograph is given. Grain counts are performed in an incident light microphotometer which makes use of the linear relationship between silver grain number and amount of light reflected by the grains. Therefore, measurements are performed in an integrating manner. The usual transmitted light is available for selecting the labelled cells for measurement.     

Synthese und Oxidation von14C-kernmarkierten Bikreosolen

Bicreosol, labelled with¹⁴C in ring position 1.1-, 4.4-, 5.5-, in the methyl- or in the methoxy substituent was oxygenated in 0.25m-NaOH. Isolation of low molecular fragments and determination of their specific activities indicate that at least two fragmentation pathways take place simultaneously: one with ring opening between the oxygen substituted carbon atoms, and one in which the bond between these atoms is not cleaved.

Zusammenfassung Zur Untersuchung der oxydativen Ringspaltung von Bikreosol wurde, ausgehend von spezifisch ring-¹⁴C-markierten Guajacolen, Vanillin und daraus Bikreosol (2,2-Dihydroxy-3,3-dimethoxy-5,5-dimethylbiphenyl-1,1) synthetisiert. Die Einführung der Aldehydgruppe in den aromatischen Ring mittels Methyldichlormethylsulfid wurde näher untersucht.

Specifically ring-¹⁴C-labelled phenol derivatives (II)¹. synthesis of [¹⁴C]vanillin and [¹⁴C]bicreosol

For oxidative ring-fragmentation-studies of bicreosol, vanillin and bicreosol were synthesized, starting from differently specifically ring-labelled guaiacols. Introduction of the CHO-group into the aromatic ring with methyldichlormethylsulfide was investigated.

10-Methoxy-des-bases of the codeinone series

Action of sodium methoxide solution upon 7-bromoneopinonedimethylacetal-methoperchlorat^* (1) yields twodes-bases, (10S)-methoxycodeinonedimethylaeetal-⁹⁽¹⁴⁾-methine (3 d) and 10-methoxyneopinonedimethylacetal-methine (4 d). The structures of the new compounds3 d and4 d are proved and the mechanism of reaction is elucidated.

Spektroskopische und theoretische Untersuchungen am Borazolmolekül und seinen Bortrihalogenoderivaten

Zusammenfassung Es werden Hückel-MO-Rechnungen am Borazol und den B-Trihalogenoborazolen durchgeführt und die entsprechenden Parameter an den teils bekannten und teils gemessenen UV-Spektren geeicht. Die für diese Verbindungen bestimmten chemischen Verschiebungen der ¹¹B- und ¹⁴N-Kernresonanz lassen sich mit den berechneten -Elektronendichten erklären.

Hückel-MO calculations are performed for Borazine and the B-Trihalogenoborazines; the parameters used are calibrated with the UV-spectra, which had either been measured here or obtained from the literature. The calculated -electron densities compare favorably well with the ¹¹B- and ¹⁴N-nuclear magnetic resonances determined for the above mentioned compounds.

Mikrobestimmung von Bromid (Chlorid) durch radioaktive Extraktion

Zusammenfassung Quecksilber(II)-bromid und -chlorid lassen sich durch Extraktion mit Diäthyläther von Quecksilber(II)-ionen trennen. Die Bromid oder Chlorid enthaltende Lösung wird mit einem Überschuß einer mit²⁰³Hg markierten Quecksilbernitratlösung versetzt, dann die entstandenen Halogenidkomplexe mit Äther extrahiert und die- Aktivität der ätherischen Phase gemessen. Die Methode eignet sich zur Bromidbestimmung erheblich besser als zur Chloridbestimmung.

Summary Mercury(II) bromide and chloride can be separated from mercury(II) ions by extraction with diethyl ether. The solution containing bromide or chloride is treated with an excess of a mercury nitrate solution labelled with²⁰³Hg; the resulting halogenide complex is extracted into ether and the-activity of the ether phase is measured. The method is much better suited for determining bromide than chloride.

Eine Studie derβ-Rückstreuung zum Zwecke ihrer Anwendung in der analytischen Chemie

Zusammenfassung Die Anwendung der-Rückstreuung zur zerstörungsfreien Werkstoffanalyse an Hüttenprodukten wird diskutiert. Unter Verwendung von 20 mC⁹⁰Sr-⁹⁰Y und einer einfachen Apparatur werden Anwendungen zur Aschebestimmung in Kohlen unter Berücksichtigung der Feuchtigkeit und Korngröße beschrieben. Auf weitere zahlreiche Anwendungsmöglichkeiten wird hingewiesen.

Summary The application of the -scattering for the nondestructive analysis of metallurgical materials is discussed. The applications for the determination of ash in coals, taking account of the moisture content and the grain size, are described in which 20 mC⁹⁰Sr-⁹⁰Y, and simple equipment are employed. Numerous other possibilities of using this method are indicated.

Zusammenfassung Die Extraktiv-Photometrie von Wismut als Tributylammoniumchlorobismutat(III) mit Methylenchlorid erlaubt die Bestimmung von Wismutmengen ab 5 g neben etwa jeweils 10⁴fachen Überschüssen an Al³⁺, Cr³⁺, Fe³⁺, Co²⁺, M²⁺, Mn²⁺. In Kupfer, Stählen oder Leichtmetalllegierungen können Gehalte von 0,01% Bi bestimmt werden.

Summary Extractive photometry of bismuth as tributylammonium chloro bismutate(III) using dichloromethane permits a determination of 5 g of Bi in the presence of a 10⁴-fold excess of Al³⁺, Cr³⁺, Fe³⁺, Co²⁺, Ni²⁺ and Mn²⁺. Determinations may be performed in copper, steels and light alloys with contents of 0.01% of Bi.

Kontinuierliche Messung von 14C-markierten Aminos?uregemischen mit Hilfe eines Scintillatorschlauches nach der Trennung an Ionenaustauschers?ulen

Zusammenfassung Es wird eine Methode beschrieben, die es gestattet, während einer automatischen Aminosäuretrennung an Ionenaustauschersäulen die Radioaktivität der einzelnen ¹⁴C-markierten Aminosäuren zu messen. Die aufgetrennten Fraktionen werden unmittelbar am Säulenausgang durch eine Meßanordnung geschickt, bestehend aus einem Scintillatorschlauch, der sich zwischen zwei in Koinzidenz geschalteten Photomultipliern befindet. Anschließend wird mit Ninhydrin angefärbt. Die Nachweisgrenze beträgt 2 · 10^–3 C. Die Methode ist einfach zu handhaben, die Meßgeräte, einschließlich der Meßzelle, sind im Handel erhältlich.

Summary A method is described for the continuous measurement of ¹⁴C-labelled amino-acids during automatic separation by ion-exchanger columns. Immediately after emerging from the column the single fractions are passed through a measuring device consisting of a plastic scintillator between two photomultipliers in a coincidence circuit. Subsequently the eluate is reacted with ninhydrin. The limit of detection is 2 · 10^–3 C. The procedure is simple and the measuring device is commercially available.

Mikroanalyse mit Hilfe von Ionenaustauschern. XXVI

Zusammenfassung Eine neue Harztüpfelmethode mit Bathocuproindisulfonat zum spezifischen Nachweis von ng Kupfer(II) wird vorgeschlagen. Bei Anwendung eines Kügelchens weißer, weitmaschiger, stark basischer Anionenaustauscher, Diaion PA-Harze, beträgt die Erfassungsgrenze 10,4 ng Kupfer in 10l Probelösung (1:1 · 10⁶).

Summary A new resin spot test with bathocuproine disulfonate for the specific detection of ng amounts of copper(II) is described. Using 1 grain of white strongly basic popcorn-resin, Diaion PA 3-series, the identification limit for copper is 10.4 ng in 10l solution (1:1 · 10⁶).
