The intramolecular cyclization of -ketocarboxylic acids and their esters gives 5-alkylsubstituted 3H-thiolen-2-ones and 3H-furan-2-ones. We have studied thionylation of 5-R-3H-furan-2-ones and 4-oxoalkanoates using phosphorus pentasulfide and the Lawesson reagent. It was found that the structure of the reaction products dependent on the thionylating agent. Preparative methods have been developed for the synthesis of 5-R-5H- and 5-R-3H-thiophen-2-ones and 5-R-3H-furan-2-thiones.N. G. 2-Alkoxy-5H-furan-4-ones (7,8) and 4-alkoxy-5H-furan-2-ones (4,5) were prepared regiospecifically and in high yields from tetronic acids (4-hydroxy-5H-furan-2-ones) (2) in the first case by acetylating the 4-OH group and then reacting with trialkyloxonium tetrafluoroborate, and in the second case by alkylating tetrabutylammonium tetronates with dialkyl sulfate, respectively. Direct alkylation of tetronic acids with trialkyloxonium tetrafluoroborate gave in four cases regiospecific 2-O-alkylation, in one case 4-O-alkylation and in two other cases mixtures of 2- and 4-alkoxy derivatives.  相似文献

Regioselective alkylation of 2-alkyl-5,6,7,8-tetrahydro-3H-cycloheptimidazol-4-one (1) and 2-alkyl-3H-cycloheptimidazol-4-one (2) was investigated. 3-[2'-(1-tert-Butyl-1H-tetrazol-5-yl)biphenyl-4-ylmethyl]-2-propyl-5,6,7,8-tetrahydro-1H-cycloheptimidazol-4-one (6) was preferentially obtained under the conditions by using NaH in DMF or THF. On the other hand, 3-[2'-(1-tert-butyl-1H-tetrazol-5-yl)biphenyl-4-ylmethyl]-2-propyl-5,6,7,8-tetrahydro-3H-cycloheptimidazol-4-one (5), the synthetic intermediate compound of Pratosartan, was obtained selectively in the presence of n-Bu(4)NBr in toluene by using aqueous sodium hydroxide as a base. In this reaction, it was found that the concentration of the alkaline solution influences its regioselectivity. This selectivity was observed even for aldehyde and ester derivatives.  相似文献

The Michael condensation in the series of 5-aryl-3-arylidene-3H-pyrrol-2-ones and 5-aryl-3-arylidene-3H-furan-2-ones, containing an activated C=C bond, with cyclohexanone was investigated. It was shown that the condensation products were 1,5-dicarbonyl compounds containing a heterocyclic fragment. Several 1-alkyl-2-methylazetidin-3-ones were prepared in good yield by the hydride-induced cyclization of the corresponding β-bromo-α,α-dimethoxyketimines, the resulting 3,3-dimethoxyazetidines being hydrolyzed by acid. Imination of these 1,2-disubstituted azetidin-3-ones, followed by alkylation under kinetic control conditions resulted in regioisomeric mixtures of 2,4- and 2,2-dialkylated compounds. Analytical samples of the major 2,4-disubstituted derivatives were obtained after extensive chromatographic separation. The cis stereochemistry of the major 2,4-dialkylated isomer was demonstrated on the basis of NMR data.  相似文献

A preparative method for the synthesis of 2-alkyl-1,2,4-benzotriazin-3(2H)-ones 3 was developed. The method involves treatment with sodium cyanate of 2-alkylbenzotetrazinium tetrafluoroborates, which exist in solutions in equilibrium with o-(alkylazo)aryldiazonium tetrafluoroborates. Reduction of compounds 3 with zinc in acetic acid afforded 2-alkyl-1,4-dihydro-1,2,4-benzotriazin-3(2H)-ones. Published in Russian in Izvestiya Akademii Nauk.
