建立了一种离子色谱-串联质谱(IC-MS/MS)测定白酒、黄酒、干红葡萄酒3种酒类样品中10种有机酸含量的方法。白酒样品氮吹后,经去离子水稀释,用IC-MS/MS分析检测;干红葡萄酒样品和黄酒样品,对比不同固相萃取小柱净化能力,最终选择石墨化炭黑固相萃取小柱进行净化,经去离子水稀释,用IC-MS/MS分析检测。选用高容量、强亲水性的Dionex IonPacTM AS11-HC型阴离子分析柱进行分离,以淋洗液自动发生器在线产生的KOH水溶液为淋洗液,进行梯度淋洗。淋洗液经抑制器抑制后直接进入电喷雾电离串联质谱(ESI-MS/MS),采用负离子模式电离,多反应监测(MRM)模式检测,外标法定量。在该实验条件下:草酸、富马酸、马来酸、苹果酸、酒石酸、柠檬酸、奎尼酸和乌头酸在0.05~2 mg/L范围内线性关系良好;琥珀酸在0.05~5 mg/L范围内线性关系良好;乳酸在0.05~10 mg/L范围内线性关系良好(相关系数r2>0.99)。10种有机酸的检出限(S/N=3)在1.0~8.0 μg/L范围内,定量限(S/N=10)在3.5~26.5 μg/L范围内,在3个不同浓度的添加水平下,平均回收率在83.0%~112.1%之间,相对标准偏差≤9.1%,满足检测要求。该方法前处理简单,不使用有机溶剂,不需进行衍生化处理,测定快速、准确,灵敏度高,适用于3种酒类样品中10种有机酸的定量分析,为酒类食品的风味及品质测定提供方法支持。  相似文献

Near infrared (NIR) spectroscopy was used to simultaneously predict the concentrations of malvidin-3-glucoside (M3G), pigmented polymers (PP) and tannins (T) in red wine. A total of 495 samples from 32 commercial scale red wine fermentations over two vintages using two grape varieties (Cabernet Sauvignon and Shiraz), and also including as additional variables two types of fermenters, two different yeasts, and three fermentation temperatures were used. Samples were scanned in transmission mode (400-2500 nm) using a monochromator instrument (NIRSystems6500). Calibration equations were developed from high performance liquid chromatography (HPLC) and NIR data using partial least squares (PLS) regression with internal cross validation. Using PLS regression, very good calibration statistics (Rcal2>0.80) were obtained for the prediction of M3G, PP and T with standard deviation (S.D.)/standard error in cross validation (SECV) ratio (residual predictive deviation, RPD)) ranging from 1.8 to 5.8. It was concluded that near infrared spectroscopy could be used as rapid alternative method for the prediction of the concentration of phenolic compounds in red wine fermentations.  相似文献

建立了基于脂肪酸的漂浮液滴固化分散液液微萃取(FA-DLLME-SFO)方法,与高效液相色谱联用检测了酒类样品中的4-乙基苯酚和4-乙基愈创木酚。该方法萃取时间仅需4 min,仅需脂肪酸、氨水和硫酸3种对环境友好的试剂。对影响方法萃取效率的因素(包括样品的体积、萃取剂的种类和用量、氨水和硫酸的体积以及盐的加入量)进行了详细的考察。在最佳萃取条件下(10 mL样品,100 μL辛酸萃取剂,110 μL 25%~28%(质量分数)的氨水,0.8 mL 98%(质量分数)的浓硫酸,3.0 g NaCl),4-乙基苯酚和4-乙基愈创木酚在0.02~1.0 mg/L范围内呈良好的线性关系,相关系数分别为0.9997和0.9999,相对标准偏差(n=3)分别为6.2%和3.5%,检出限分别为6.33和5.81 μg/L,富集倍数分别为79和86。在啤酒和白葡萄酒样品中,加标回收率为81.4%~108.7%,相对标准偏差(n=3)小于8.9%。该方法简单、对环境友好,可用于酒类样品中4-乙基苯酚和4-乙基愈创木酚的检测。  相似文献

Abstract

A method using high performance liquid chromatography (HPLC) for the analysis of primary amino acids in human placenta is described. This method involves separation of primary amino acids by high performance ion-exchange chromatography followed by post column derivatization using O-phlthalaldehyde (OPA) and 2-mercaptoethanol and fluorescence (excitation 340 nm and emission 410 nm) detection of derivatives. Waters 840 HPLC Amino Acid System was used for this purpose.

For analysis, villus tissue was extracted with acetonitrile, and the recovered amino acids were reconstituted in a sodium diluent (pH 2.2). The complete profile of the primary amino acids in the sample could be constructed in about 90 minutes. Up to 44 samples can be analyzed without special attention. Using this method, essential amino acids (threonine, valine, methionine, isoleucine, leucine, tyrosine, phenylalanine, lysine, histidine) and nonessential amino acids (aspartic acid, serine, glutamic acid, glycine, alanine, arginine) were detected and quantified in human placental villus in pmol quantities. Plots of peak heights (or areas) were linear for several amino acids. The same method was also used for (a) the assay of free primary amino acids in umbilical bloods, (b) the efflux of amino acids from isolated human placental villus, and (c) to study the uptake of α-aminoisobutyric acid (AIB), a non-metabolizable amino acid, by the isolated placental villus.  相似文献

To meet the need for a new and validated analytical method for simultaneous determination of inorganic and organic acid anions in beverage drinks, a capillary zone electrophoresis (CZE) procedure had been developed based on a new background electrolyte (BGE) system containing 3 mmol/L 1,3,5-benzenetricarboxylic acid (BTA), 15 mmol/L tris(hydroxymethyl)aminomethane and 1.5 mmol/L tetraethylenepentamine (TEPA) at pH 8.4. Baseline separation of anions commonly found in beverage drinks could be achieved in less than 14 min with indirect UV detection at 240 nm. Comigration problems for hydroxycarboxylic acids could be solved using TEPA as BGE additive. The results indicate excellent repeatability for migration time (RSD, 0.27-0.67%, n = 5) and good precision for both peak height (RSD, 3.2-4.2%, n = 5) and peak area (RSD, 3.1-4.5%, n = 5). Under the optimized conditions and using corrected peak area for quantitation, an excellent linear dynamic range (with correlation coefficient > 0.997 in a concentration range from 0.005 to 2.0 mmol/L) and low detection limit (1-4 micro mol/L) were obtained for all the anions investigated. The applicability and reliability of the CE procedure developed were established by parallel method determination using established ion chromatography procedure for the analysis of inorganic and organic acid anions in orange juice and wine samples. Our CZE procedure provided a sensitive and economic technique for simultaneous determination of inorganic and organic acid anions in orange juice, red and white wine samples.  相似文献

固相萃取-高效液相色谱-荧光检测土壤中喹诺酮类抗生素   总被引:7,自引:0,他引:7  
建立了固相萃取-高效液相色谱-荧光(HPLC-FLD)检测土壤中4种喹诺酮类抗生素的分析方法.样品采用50% Mg(NO_3)_2-10% NH_3 · H_2O(96∶ 4,V/V)超声提取,过HLB柱富集净化,再用乙腈-0.067 mol/L H_3PO_4溶液洗脱.采用高效液相色谱-荧光检测器,以乙腈-0.067 mol/L H_3PO_4(用三乙胺调节至pH 2.5)为流动相,于激发波长280 nm、发射波长450 nm处进行检测.土壤样品中4种喹诺酮类抗生素的加标回收率在60.4%~99.3%之间,检出限为0.58~1.0 μg/kg.对蔬菜基地土壤样品分析结果表明,本方法能够满足实际样品的分析要求,4种喹诺酮类抗生素均被检出,土壤中抗生素污染问题值得关注.  相似文献

This work presents a RP-HPLC method for the simultaneous quantification of free amino acids and biogenic amines in liquid food matrices and the results of the application to honey and wine samples obtained from different production processes and geographic origins. The developed methodology is based on a pre-column derivatization with o-phthaldialdehyde carried out in the sample injection loop. The compounds were separated in a Nova-Pack RP-C(18) column (150 mm x 3.9 mm, 4 microm) at 35 degrees C. The mobile phase used was a mixture of phase A: 10 mM sodium phosphate buffer (pH 7.3), methanol and tetrahydrofuran (91:8:1); and phase B: methanol and phosphate buffer (80:20), with a flow rate of 1.0 ml/min. Fluorescence detection was used at an excitation wavelength of 335 nm and an emission wavelength of 440 nm. The separation and quantification of 19 amino acids and 6 amines was carried out in a single run as their OPA/MCE derivatives elute within 80 min, ensuring a reproducible quantification. The method showed to be adequate for the purpose, with an average RSD of 2% for the different amino acids; detection limits varying between 0.71 mg/l (Asn) and 8.26 mg/l (Lys) and recovery rates between 63.0% (Cad) and 98.0% (Asp). The amino acids present at the highest concentration in honey and wine samples were phenylalanine and arginine, respectively. Only residual levels of biogenic amines were detected in the analysed samples.  相似文献

In this study, a simple, fast, accurate and sensitive spectrophotometric method has been developed for the determination of tranexamic acid in bulk and pharmaceutical preparations. The method is based on the reaction of ninhydrin with the primary amino group of tranexamic acid in the basic medium at pH 8.0. The reaction produces a bluish-purple color which absorbs maximally at 565 nm. Beer's law was obeyed in the range of 3-40 microg ml(-1) with molar absorptivity of 5.093 x 10(3) L mol(-1) cm(-1). The effects of various factors such as temperature, heating time, concentration of reagent, color stability and interferences were investigated to optimize the procedure. The results have been validated analytically and statistically. The proposed method has been applied for the determination of tranexamic acid in bulk and pharmaceutical preparations with good results.  相似文献

A new rapid and sensitive high performance liquid chromatography (HPLC) method for determining histamine in red wine samples, based on continuous flow derivatization with 1,2-naphthoquinone-4-sulfonate (NQS), is proposed. In this system, samples are derivatized on-line in a three-channel flow manifold for reagent, buffer and sample. The reaction takes place in a PTFE coil heated at 80 degrees C and with a residence time of 2.9 min. The reaction mixture is injected directly into the chromatographic system, where the histamine derivative is separated from other aminated compounds present in the wine matrix in less than ten minutes. The HPLC procedure involves a C18 column, a binary gradient of 2% acetic acid-methanol as a mobile phase, and UV detection at 305 nm. Analytical parameters of the method are evaluated using red wine samples. The linear range is up to 66.7 mg L(-1) (r = 0.9999), the precision (RSD) is 3%, the detection limit is 0.22 mg L(-1), and the average histamine recovery is 101.5% +/- 6.7%. Commercial red wines from different Spanish regions are analyzed with the proposed method.  相似文献

建立了高效液相色谱-串联质谱法快速测定葡萄酒中白藜芦醇、黄酮类、多酚类功效成分的分析方法。葡萄酒样品直接稀释后进样,用C18柱进行分离,以乙腈-0.1%(体积分数)甲酸水溶液为流动相进行梯度洗脱,通过多反应监测(MRM)模式进行检测。13种功效成分在各自线性范围内呈良好的线性关系,相关系数均大于0.99。除表没食子儿茶素、没食子儿茶素、儿茶素没食子酸酯、花旗松素的检出限为1.0、1.0、3.0、3.0μg/L外,其他9种化合物的检出限均小于1.0μg/L。回收率为80.9%~112.3%,相对标准偏差小于10%。该方法快速、准确、灵敏度高,适用于葡萄酒中功效成分的快速分析。对实际样品的检测表明,所测葡萄酒样品中均含有儿茶素、表儿茶素、表没食子儿茶素、没食子儿茶素、表儿茶素没食子酸酯/儿茶素没食子酸酯、白藜芦醇、大豆黄素等功效成分,不同品种葡萄酒中这些功效成分含量差异显著。  相似文献

建立了黄酒中5-羟甲基糠醛和9种多酚(儿茶素、表儿茶素、绿原酸、芦丁、咖啡酸、原儿茶酸、丁香酸、阿魏酸、p-香豆酸)的高效液相色谱检测方法。采用Diamonsil C18柱(150mm×4.6mm,5μm)分离,柱温为42℃,检测波长为280nm,流动相为乙腈和3%乙酸水溶液,梯度洗脱,20min内10种物质得到了较好的分离。各化合物回归方程的相关系数r为0.9911~0.9995,检出限为0.2~0.5mg/L;相对标准偏差RSD≤2.4%;10种组分的平均回收率为89.4%~98.3%;能够满足定量分析要求。实验结果表明,本方法适用于不同种类和年份黄酒中5-羟甲基糠醛和9种多酚的检测。  相似文献

Amperometric biosensors for the determination of l-malic and l-lactic acids were optimised and used to monitor micro-malolactic fermentations (micro-MLFs) in red wine. Platinum-based probes, coupled with appropriate enzymes, were assembled in electrochemical flow-injection analysis systems. A classical lactate oxidase based sensor was used for l-lactic acid, while l-malic acid was detected via a new biosensor based on the malic enzyme immobilised in a reactor using phenazine methosulphate as mediator. After a preliminary optimisation phase, a recovery study to evaluate the effect of the matrix (red wine) on biosensor performance was carried out by the addition of different standard solutions of the two analytes to the samples. Recoveries from 93 to 100% and from 94 to 102% were observed for l-malic acid and l-lactic acid, respectively. These optimised biosensors were finally employed to monitor micro-MLFs induced by inoculation of two different strains of Oenococcus oeni into red wine. During the micro-MLFs, samples of wine were collected and assayed for l-malic, l-lactic, and citric acids by use of both biosensors and spectrophotometric techniques. In parallel the viable bacterial cell count was also evaluated. The kinetics of bacterial growth, degradation of l-malic and citric acids, and production of l-lactic acid was found to be a function of the strains inoculated.  相似文献

采用毛细管电泳-间接紫外检测法同时分离测定蜂蜜中的赖氨酸、色氨酸、谷氨酸等9种氨基酸。考察了磷酸浓度、进样方式和缓冲液pH对分离效率和重现性的影响。在分离电压为-15 kV、检测波长为220 nm条件下,以含有0.5 mmol/L十六烷基三甲基溴化铵、20 mmol/L烟酸、10%甲醇的10 mmol/L磷酸二氢钠缓冲溶液(pH 10.2)为运行缓冲液,9种组分在11 min内达到基线分离;检出限最低可达到0.3 mg/L;线性范围为1.0~1000 mg/L;日间及日内精密度为0.64%~5.83%。实际样品中除甲硫氨酸外的8种氨基酸的加标回收率为60.00%~118.37%。将该方法应用于不同蜜源植物和产地的蜂蜜样品的测定,在市售的5种蜂蜜中均检测到脯氨酸、丝氨酸和天冬氨酸,而只在荔枝蜜中检测到苏氨酸。该方法可以为蜂蜜的蜜源鉴别及质量评估提供借鉴方法。  相似文献

建立了保健酒中16种邻苯二甲酸酯类物质(PAEs)的气相色谱-三重四极杆质谱(GC-QQQ-MS)测定法。样品采用正己烷提取,使用GC-QQQ-MS测定,监测方式为选择离子监测(SIM),利用保留时间和碎片离子丰度比值判断定性结果,利用特征离子质量色谱峰峰面积-浓度标准曲线法定量,并用建立的方法分析了81批实际样品。实验结果表明:目标化合物特征离子质量色谱峰的峰面积与其质量浓度在各自的浓度范围内线性关系良好(r2≥0.9959)。在低、中、高3个浓度的添加水平下样品加标回收率除邻苯二甲酸二甲酯(DMP)为52.3%~58.7%外,其余15种化合物为88.6%~107.3%,相对标准偏差(RSD,n=6)为0.1%~6.8%。检出限为0.002~0.061 mg/L,定量限为0.005~0.202 mg/L。该方法具有灵敏、简单、准确、线性范围宽等优点,可满足保健酒中邻苯二甲酸酯类物质的检测需要。  相似文献

建立了一种专属、灵敏的同时测定黄酒和葡萄酒中安赛蜜、糖精、甜蜜素、阿斯巴甜、苯甲酸、山梨酸、甜菊糖苷、纽甜和脱氢乙酸等9种防腐剂和甜味剂的超快速液相色谱-串联质谱(UFLC-MS/MS)分析方法。不同类型的黄酒和葡萄酒经纯水稀释后,以乙腈和0.01%三氟乙酸-2.5 mmol/L乙酸铵水溶液为流动相,采用梯度洗脱方式在Shim-pack XR-ODSII色谱柱(100 mm×2.0 mm, 2.2 μm)上进行分离,以电喷雾负离子多反应监测(MRM)模式进行质谱分析。实验表明,9种防腐剂和甜味剂在检测范围内均具有良好的线性关系(r2>0.998);方法的检出限(以信噪比大于3计)为0.03~15.0 μg/L,定量限(以信噪比大于10计)为0.1~50.0 μg/L;在黄酒中的回收率为96.2%~100.5%,相对标准偏差(RSDs)为0.6%~5.4%;在葡萄酒中的回收率为96.0%~104.0%, RSDs为0.7%~4.8%。同时研究了这9种防腐剂和甜味剂的二级质谱特征,阐释了其二级质谱裂解途径。本方法灵敏度高、重现性好、分析速度快,可用于黄酒和葡萄酒中防腐剂和甜味剂的快速确证检测。  相似文献

改进的毛细管电泳-间接紫外吸收法采用了自制隔离池,以对氨基苯甲酸(PAB)为背景电解质,对茶叶中的氨基酸进行了测定。PAB能够提高分离效率,降低检出限。隔离池的使用避免了PAB的电极反应,降低了基线噪声,维持了两缓冲液池间的电流导通。研究了背景电解质的浓度、pH值以及电渗流改性剂的种类和浓度对氨基酸分离的影响。在优化的实验条件下,16种氨基酸在14 min内达到了基线分离,峰面积的相对标准偏差小于5%(n=5),检出限为1.7~4.5 μmol/L,回收率为83.0%~106%。该法快速、便捷和灵敏,已成功应用于茶叶中11种游离氨基酸的检测。  相似文献

A fully automated, rapid and sensitive method was developed to analyze fourteen different biogenic amines in food. Using a Technicon C4 ion-exchange resin column (20 m X 0.5 cm), adapted to an automatic Technicon TSM amino acid analyzer, the following amines were separated and quantified: adrenaline, noradrenaline, 1,3-diaminopropane, putrescine, cadaverine, histamine, spermidine, dopamine, spermine, agmatine, tyramine, serotonin, phenethylamine and tryptamine. Five buffers were required to elute the amines using a gradient of pH from 5.6 to 12.7; the column temperature was maintained at 65 degrees C. The method was also assayed on ground beef, cheese and wine samples. Amines from cheese and ground beef samples were extracted with 0.6 M perchloric acid. No extraction of wine samples was necessary.  相似文献

Phanquinone (4,7-phenanthroline-5,6-dione) has been investigated as a pre-column derivatization fluorogenic reagent for liquid chromatographic determination of primary amino acids in biological samples. The derivatization reaction was carried out at 68 degrees C both in the presence of aqueous phosphate buffer (pH 8) for 30 min and without buffer for 60 min to allow the determination of basic amino acids (Orn, Lys, Arg). The resulting derivatives were separated under reversed-phase HPLC and detected at lambda(em) = 460 nm with lambda(ex) = 400 nm. The proposed method was validated and applied to the determination of a variety of amino acids directly in urine and after deproteinization with 5-sulfosalicylic acid in plasma samples. The detection and quantitation limits were found in the range 10-450 and 35-1400 fmol, respectively.  相似文献

建立了离子排斥色谱法(ion-exclusion chromatography,IEC)测定黄酒中有机酸含量的分析方法。使用Waters离子排斥色谱柱(300 mm×7.8 mm,7 μm),流动相为H2SO4溶液(A)与乙腈(B)的混合溶液(体积比为98:2),线性梯度程序:0~40 min,流动相A的浓度由0.01 mol/L 上升到0.02 mol/L;40~50 min,流动相A的浓度为0.01 mol/L ;流速为0.5 mL/min,柱温50 ℃,进样量10 μL,检测波长210 nm。结果表明,该方法可在30 min内实现草酸、马来酸、柠檬酸、酒石酸、苹果酸、抗坏血酸、琥珀酸、乳酸、富马酸、乙酸、丙酸、异丁酸和丁酸的完全分离与定量,13种有机酸在0.001~1.000 g/L范围内线性关系良好,回归方程的线性相关系数在0.9997以上。黄酒中13种有机酸的加标回收率为93.4%~103.8%,相对标准偏差为0.1%~1.5%(n=5)。该方法简单快捷、准确、重复性好,可用于黄酒中有机酸的测定。  相似文献
