A procedure based on QuEChERS extraction and a simultaneous liquid–liquid partition clean-up was developed. The procedure involved extraction of hydrated soil samples using acetonitrile and clean-up by liquid–liquid partition into n-hexane. The hexane extracts produced were clean and suitable for determination using gas chromatography–tandem mass spectrometry (GC–MS/MS). The method was validated by analysis of soil samples, spiked at five levels between 1 and 200 μg kg−1. The recovery values were generally between 70 and 100% and the relative standard deviation values (%RSDs) were at or below 20%. The procedure was validated for determination of 19 organochlorine (OC) pesticides. These were hexachlorobenzene (HCB), α-HCH, β-HCH, γ-HCH, heptachlor, heptachlor epoxide (trans), aldrin, dieldrin, chlordane (trans), chlordane (cis), oxychlordane, α-endosulfan, β-endosulfan, endosulfan sulfate, endrin, p,p′-DDT, o,p′-DDT, p,p′-DDD and p,p′-DDE. The method achieved low limits of detection (LOD; typically 0.3 μg kg−1) and low limits of quantification (LOQ; typically 1.0 μg kg−1). The method performance was also assessed using five fortified soil samples with different physico-chemical properties and the method performance was consistent for the different types of soil samples. The proposed method was compared with an established procedure based on Soxtec extraction. This comparison was carried out using six soil samples collected from regions of Pakistan with a history of intensive pesticide use. The results of this comparison showed that the two procedures produced results with good agreement. The proposed method produced cleaner extracts and therefore led to lower limits of quantification. The proposed method was less time consuming and safer to use. The six samples tested during this comparison showed that soils from cotton growing regions contained a number of persistent OC residues at relatively low levels (<10 μg kg−1). These residues were α-HCH, γ-HCH, heptachlor, chlordane (trans), p,p′-DDT, o,p′-DDT, p,p′-DDD, p,p′-DDE, β-endosulfan and endosulfan sulfate.  相似文献

The structural, elastic, mechanical, electronic, optical properties and effective masses of CuMIIIBO2 (MIIIB = Sc, Y, La) compounds have been investigated by the plane-wave ultrasoft pseudopotential technique based on the first-principles density-functional theory under local density approximation. The equilibrium structural parameters are in good agreement with previous experimental and theoretical data. To our knowledge, there are no available data of elastic constants for comparison. The bulk, shear and Young's modulus, ratio of B/G, Poisson's ratio and Lamé's constants of CuMIIIBO2 have been studied. The electronic structures of CuMIIIBO2 are consistent with other calculations. The population analysis, charge densities and effective masses have been shown and analyzed. The imaginary and real parts of the dielectric function, refractive index and extinction coefficient of CuMIIIBO2 are calculated. The interband transitions to absorption of CuMIIIBO2 have been analyzed.  相似文献

The present study reports the development and validation of a high-throughput high-performance liquid chromatographic (HPLC) assay for the determination of caffeine in food samples. The analyte was separated rapidly from sample matrix using a short monolithic column (50 mm × 4.6 mm i.d.). The flow rate was 3.0 mL min−1, while the mobile phase consisted of ACN/water (10:90, v/v). Caffeine was detected directly at 274 nm. Under the optimal HPLC conditions, the sampling rate was 60 h−1. The assay was validated for linearity, LOD and LOQ, precision, selectivity and ruggedness. The case of external calibration versus standard addition for the analysis of real samples was also examined. The proposed assay was applied to the analysis of beverages and coffee samples.  相似文献

A rapid and sensitive method has been developed for the simultaneous detection of cyromazine and melamine in chicken eggs using the quick, easy, cheap, effective, rugged and safe (QuEChERS) method coupled with liquid chromatography–tandem mass spectrometry (LC–MS/MS). The optimal extraction solvent for the liquid–liquid extraction was 5 mL of acetonitrile with a 0.1 M hydrochloric acid aqueous solution (99.5:0.5, v/v). The extract was cleaned with 0.5 g of anhydrous magnesium sulfate and 10 mg of graphitized carbon black. The analysis of cyromazine and melamine was accomplished by combining the use of an anion exchange LC column with tandem mass spectrometry in the positive electrospray ionization mode with selected reaction monitoring mode (SRM). The detection limits were 1.6 ng g−1 for cyromazine and 8 ng g−1 for melamine, and the quantitation limits were 5.5 ng g−1 for cyromazine and 25 ng g−1 for melamine. The recoveries of cyromazine and melamine in the spiked egg samples were 83.2% and 104.6%, respectively, with an relative standard deviation (RSD) of less than 18.1%. The intra-day and inter-day precisions, represented by the RSD, ranged from 1.5% to 8.8% and 6.8% to 14.3%, respectively. The proposed method was tested by analyzing chicken eggs from the markets and from the veterinary medicine laboratory. The concentrations of cyromazine and melamine detected in these samples were in the range of 20–94 ng g−1. The results demonstrated that the QuEChERS method combined with LC–MS/MS is a simple, rapid and inexpensive method for the analysis of cyromazine and melamine in eggs.  相似文献

Ferruginol, a diterpene phenol, has recently received attention for its extensive pharmacological properties, including anti‐tumor, antibacterial, cardio‐protective and gastroprotective effects. In the present study, a high‐performance liquid chromatographic (HPLC) method was developed for determination of ferruginol in rat plasma and applied for the pharmacokinetics study. The HPLC assay was performed with a VP ODS‐C18 column. The mobile phaseconsisted of methanol and 1% acetic acid solution (90:10, v/v). The flow rate was 1.0 mL/min, and the wavelength was set at 270 nm. This method was linear over the studied range of 0.1–10.0 µg/mL for ferruginol. The correlation coefficient was 0.9998. The intra‐day and inter‐day precisions were better than 4 and 5%, respectively. The extraction recovery and accuracy were greater than 97 and 96%, respectively. The detection limit was 30 ng/mL. The mean maximum concentration of ferruginol in rat plasma was 3.14 µg/mL at 40 min after oral administration at a dose of 20 mg/kg. Ferruginol was absorbed quickly p.o. with t1/2ka = 14.86 min and had a high rate of elimination with t1/2 = 41.73 min. Co3xNi3−3x(PO4)2·8H2O (x = 1, 0.8, 0.6, 0.4, 0.2, and 0) were synthesized via simple wet chemical reaction and energy saving route method. The final decomposition products of hydrates are corresponding anhydrous tri(cobalt nickel) diphosphates. The metal and water contents of the synthesized hydrates were confirmed by AAS and TG/DTG/DTA techniques, respectively. The observed metal and water contents agree well with the formula of the title compounds. The crystal structures and lattice parameters as well as crystallite sizes of the studied compounds were determined using XRD data. The results from XRD and TG/DTG/DTA techniques confirmed that Co3xNi3−3x(PO4)2·8H2O at all ratios were the single phase. The FTIR spectra of studied compounds were recorded and assigned. The thermal behaviours of single and binary tri(cobalt nickel) diphosphate octahydrates were studied for the first time. The morphologies of the studied compounds were investigated by using the SEM technique. The micrographs of all studied compounds exhibited the thin plated morphology. The surface area and the pore size data of anhydrous forms were measured by N2 adsorption at −190 °C according to the BET method. The anhydrous forms of binary metal phosphate at x = 0.8, Co2.4Ni0.6(PO4)2, exhibits the highest surface area and expects to improve the catalytic activity.  相似文献

A modified sol-gel method was used for synthesis of zinc doped black TiO2 nanoparticles. The modified sol-gel synthesised catalyst was utilised for degradation of 2, 4, 6 tri-chloro-phenol under visible light irradiation. The catalyst was characterized using XRD, SEM, TEM, BET and DRS analysis. The nanoparticles were crystalline in nature and in anatase phase. The size of zinc doped black TiO2 nanoparticles was 5 nm. The synthesised nanoparticles were mesoporous in nature and the specific surface area was found to be 34.15 m2/g. The band gap energy of zinc black TiO2 nanoparticles was found to be 2.73 eV. The point of zero charge of zinc doped black TiO2 nanoparticles was 6.7. The maximum degradation of 2, 4, 6 tri-chloro-phenol using 2 mol% zinc doped black TiO2 was found to be 95%.  相似文献

The lactic acid bacterium,Lactobacillus casei, produces an intracellular β-glucosidase when grown on Man-Rogosa-Sharpe (MRS) medium with cellobiose as carbon source. The β-glucosidase activity is produced intracellulary, and no extracellulary activity was detected. The enzyme was purified by ion-exchange chromatography and gel filtration. The molecular mass of the purified intracellular β-glucosidase as estimated by gel filtration was 480 kDa, consisting of six probably identical subunits. The enzyme exhibited optimum activity at 35°C and pH 6.3 with citrate-phosphate buffer. The enzyme was active against soluble glycosides with (1→4)-β configuration and from Lineweaver Burk plots, Km value of 16 mmol/L was found for β-pNPG. The β-glucosidase was competitively inhibited by glucose, and no glycosyl transferase activity was observed in the presence of ethanol.  相似文献

A fluorescence quenching method has been developed to determine cerium with rhodamine 6G. The method is based on the oxidation of rhodamine 6G by cerium(IV) in sulfuric acid solution. The linear calibration range is 4.0 × 10–7 -1.6 × 10–6mol/L. The detection limit is 1.0 × 10–7 mol/L. The relative standard deviation was 1.0% (n = 5). The method has been used to determine cerium in rare earth ores, with satisfactory results. The method offers the advantages of simplisity, sensitivity and selectivity.  相似文献

A fast extraction procedure has been developed for Sb(III) and Sb(V) oxoanions speciation in airborne particulate matter samples. Different extraction media (diammonium tartrate, hidroxilammonium clorhidrate, citric acid + ascorbic acid, phosphoric acid and citrate solutions) were tried, with assistance of an ultrasonic probe. The operation power and time of extraction were also optimized. The higher extraction recoveries were obtained with a 100 mmol L−1 hidroxilammonium clorhidrate aqueous solution assisted by the ultrasound probe operated at 50 W during 3 min. The extracts were analyzed by HPLC-HG-AFS. The chromatographic separation of Sb(III) and Sb(V) was also optimized using diammonium tartrate and phthalic acid as mobile phases. The separation of both Sb species was performed in less than 3 min under isocratic conditions, using a 200 mmol L−1 diammonium tartrate solution. The proposed extraction procedure and the HPLC-HG-AFS instrumental coupling have been successfully applied to airborne particulate matter samples, with high Sb content, collected in heavy traffic streets from Buenos Aires (Argentina). The results showed the presence of both Sb species at similar concentrations in the ng m−3 level. The extraction yield was higher than 90% for all the analyzed samples.  相似文献

 A simple and sensitive spectrophotometric method is described for the determination of some phenolic antibiotics namely: cefadroxil, amoxicillin and vancomycin. The method is based on the measurement of the orange yellow species produced when the drugs are coupled with diazotized benzocaine in triethylamine medium. The method is applicable over the range of 0.8–12 μg/ml for cefadroxil, 2–16 μg/ml for amoxicillin and 2–18 μg/ml for vancomycin. The formed compounds absorb at 455 nm for both cefadroxil and amoxicillin and at 442 nm for vancomycin. The proposed method has detection limits of 0.018 μg for cefadroxil, 0.0034 μg for amoxicillin and 0.0156 μg for vancomycin. The stoichiometric ratio for the studied compounds was found to be 1:1 and a proposal of the reaction pathway was made. The proposed method was applied for the analysis of the cited drugs in their pharmaceutical preparations. The results are in good agreement with those obtained by the official methods. A sensitive and specific method was developed for the determination of sophoridine (SRI), sophocarpine (SC) and matrine (MT) in rabbit plasma by HPLC-MS. After an administration of Kuhuang by injection, blood samples were collected and extracted with methanol. The extract solutions were analysed by HPLC-MS method. The separation was performed on a ZORBAX Extend-C18 column using methanol/water/diethylamine (50:50:0.07, v/v/v) as mobile phase. The quinolizidine alkaloids were detected by using mass spectrometry in the SIM mode. There was a good linear relationship between peak area and concentration of analytes over the concentration range of 13.2–995.0 ng mL–1 for SRI, 7.0–530.0 ng mL–1 for SC and 8.8–655.0 ng mL–1 for MT, respectively. The absolute recovery of this method was more than 57% for SRI, 87% for SC and 91% for MT. The accuracy of assay was more than 90%. The limits of detection (LODs) were 6.8 ng mL–1 for SRI, 3.5 ng mL–1 for SC and 4.2 ng mL–1 for MT, respectively. The limits of quantitation (LOQs) were 13.2 ng mL–1 for SRI, 7.0 ng mL–1 for SC and 8.8 ng mL–1 for MT, respectively. The intra-day and inter-day coefficients of variation (RSDs) were less than 10.1, 6.3 and 5.8% for SRI, SC and MT, respectively. The developed method was applied to determine the concentration–time profiles of SRI, SC and MT in rabbit plasma after injection of Kuhuang.  相似文献

The redox properties of heterogenized RhI, RhII, and RhIII complexes with different, particularly organophosphorus, ligands were studied by cyclic voltammetry (CVA). The support is a carbon-paste electrode based on a fibrous-carbon material and activated carbon. The electrochemical reduction of RhIII produces Rh metal, which further catalyzes hydrogen evolution. After the reduction of water-soluble binuclear RhII complexes, the CVA curves exhibit peaks of electrocatalytic hydrogen evolution and irreversible RhI→RhII oxidation. The RhII complexes with organophosphorus ligands are characterized only by the peak of RhI→RhII oxidation. After reduction, the RhI complexes behave as a pseudo-reverse Rh0/RhI pair. The electron-donating properties of the ligand determine the reversibility of the system. The degree of structurization of the carbon matrix and the presence of phosphorus(v) atoms in it affect the electrochemical activity of the RhII and RhI complexes. A new procedure for determining nitrogenated bases by forming ion pairs with Fe(SCN)63– is studied. The method consists of extracting the ion pair formed by a nitrogenated base and the inorganic complex Fe(SCN)63– into an organic phase, and measuring the iron in the organic phase by AAS at 248.3 nm. The optimal experimental conditions for determining amylocaine, avacan, bromhexine, diphenhydramine and papaverine are studied. The organic phase used is 1,2-dichloroethane. The standard deviation of the method varies between 10–1 and 10–2. The interferences produced by various substances are studied.  相似文献

The design of an appropriate analytical method for assessing the quality of pharmaceuticals requires a deep understanding of science, and risk evaluation approaches are appreciated. The current study discusses how a related substance method was developed for Nintedanib esylate. The best possible separation between the critical peak pairs was achieved using an X-Select charged surface hybrid Phenyl Hexyl (150 × 4.6) mm, 3.5 μm column. A mixture of water, acetonitrile, and methanol in mobile phase-A (70:20:10) and mobile phase-B (20:70:10), with 0.1% trifluoroacetic acid and 0.05% formic acid in both eluents. The set flow rate, wavelength, and injection volumes were 1.0 ml/min, 285 nm, and 5 μl, respectively, with gradient elution. The method conditions were validated as per regulatory requirements and United States Pharmacopeia general chapter < 1225 >. The correlation coefficient for all impurities from the linearity experiment was found to be > 0.999. The % relative standard deviation from the precision experiments ranged from 0.4 to 3.6. The mean %recovery from the accuracy study ranged from 92.5 to 106.5. Demonstrated the power of the stability-indicating method through degradation studies; the active drug component is more vulnerable to oxidation than other conditions. Final method conditions were further evaluated using a full-factorial design. The robust method conditions were identified using the graphical optimization from the design space.  相似文献

Pure gas sorption and transport properties of polyesters based on bisphenol-A and both pure isophthalic and pure terephthalic acid chloride were obtained for He, N2, O2, CH4, and CO2 at 35°C. The polymers were synthesized in our laboratory and amorphous films were prepared with a specialized solvent casting procedure. The polymer containing m-phenylene groups shows higher permselectivity for most of the gas pairs. The ideal selectivity of O2/N2 was increased by 33% when p-phenylene units were replaced by m-phenylene ones. On the other hand, the polyester containing only p-phenylene groups, shows higher permeability to all the gases studied. The polymer based on pure terephthalic acid chloride has a 75% higher oxygen permeability and a 1.1-fold higher carbon dioxide permeability than the isophthalic acid derivative. The polyester containing meta-phenylene units has lower Tg, higher permselectivity, lower permeability, lower fractional free volume (FFV), and lower d-spacing. The values of FFV, and lower d-spacing. The values of FFV and d-spacing were only slightly different between the two isomers. Moreover, for the sub-Tgγ transition the maximum in tan δ occured at essentially the same temperature (?55°C). The polymer with a higher concentration of p-phenylene units shows somewhat larger area under the γ-peak, indicating slightly more sub-Tg motion. The Distribution of FFV is considered to be the determining factor for the differences in transport properties observed. © 1993 John Wiley & Sons, Inc.  相似文献

In this work, the use of cetyltrimethylammonium bromide as surfactant for the preparation of oil-in-water emulsions for the determination of Cu and Cr in gasoline by electrothermal atomic absorption spectrometry (ET AAS) was evaluated. The surfactant amount was tested in the range of 25 to 300 mg, added to 2 ml of gasoline, and completed to 10 mL with 0.1% (v/v) nitric acid solution. 150 mg of surfactant was found optimum, and a sonication time of 10 min sufficient to form an oil-in-water emulsion that was stable for several hours. The ET AAS temperature program was established based on pyrolysis and atomization curves. The pyrolysis temperatures were set at 700 and 1300 °C for Cu and Cr, respectively and the selected atomization temperatures were 2400 and 2500 °C. The time and temperature of the drying stage and the atomization time were experimentally tested to provide optimum conditions. The limits of detection were found to be 5 μg L− 1 and 1.5 μg L− 1 for Cu and Cr, respectively in the original gasoline samples. The relative standard deviation (RSD) ranged from 4 to 9% in oil-in-water emulsions spiked with 5 μg L− 1 and 15 μg L− 1 of each metal, respectively. Recoveries varied from 90 to 98%. The accuracy of the proposed method was tested by an alternate procedure using complete evaporation of the gasoline sample. The method was adequate for the determination of Cu and Cr in gasoline samples collected from different gas stations in Salvador, BA, Brazil.  相似文献

Chitinase was purified from the culture medium of Bacillus licheniformis SK-1 by colloidal chitin affinity adsorption followed by diethylamino ethanol-cellulose column chromatography. The purified enzyme showed a single band on sodium dodecyl sulfate polyacrylamide gel electrophoresis. The molecular size and pI of chitinase 72 (Chi72) were 72 kDa and 4.62 (Chi72) kDa, respectively. The purified chitinase revealed two activity optima at pH 6 and 8 when colloidal chitin was used as substrate. The enzyme exhibited activity in broad temperature range, from 40 to 70°C, with optimum at 55°C. It was stable for 2 h at temperatures below 60°C and stable over a broad pH range of 4.0–9.0 for 24 h. The apparent K m and V max of Chi72 for colloidal chitin were 0.23 mg ml−1 and 7.03 U/mg, respectively. The chitinase activity was high on colloidal chitin, regenerated chitin, partially N-acetylated chitin, and chitosan. N-bromosuccinamide completely inhibited the enzyme activity. This enzyme should be a good candidate for applications in the recycling of chitin waste.  相似文献

A simple isocratic reversed-phase high-performance liquid chromatographic method (RP-HPLC) was developed for the simultaneous determination of buprenorphine hydrochloride, naloxone hydrochloride dihydrate and its major impurity, noroxymorphone, in pharmaceutical tablets. The chromatographic separation was achieved with 10 mmol L−1 potassium phosphate buffer adjusted to pH 6.0 with orthophosphoric acid and acetonitrile (17:83, v/v) as mobile phase, a C-18 column, Perfectsil Target ODS3 (150 mm × 4.6 mm i.d., 5 μm) kept at 35 °C and UV detection at 210 nm. The compounds were eluted isocratically at a flow rate of 1.0 mL min−1. The average retention times for naloxone, noroxymorphone and buprenorphine were 2.4, 3.8 and 8.1 min, respectively. The method was validated according to the ICH guidelines. The validation characteristics included accuracy, precision, linearity, range, specificity, limit of quantitation and robustness. The calibration curves were linear (r > 0.996) over the concentration range 0.22-220 μg mL−1 for buprenorphine hydrochloride and 0.1-100 μg mL−1 for naloxone hydrochloride dihydrate and noroxymorphone. The recoveries for all three compounds were above 96%. No spectral or chromatographic interferences from the tablet excipients were found. This method is rapid and simple, does not require any sample preparation and is suitable for routine quality control analyses.  相似文献
