合成了2-乙基-3-(3-硝基苯基)喹唑啉-4-酮(ENPQO),用紫外光谱法、荧光光谱法、荧光偏振法、离子强度法及分子模拟法研究了ENPQO与小牛胸腺DNA(ct DNA)的相互作用。紫外光谱法显示ct DNA与ENPQO作用后,引起ENPQO紫外吸收光谱的增色效应;在以吖啶橙(AO)为荧光探针的实验中,随着ENPQO浓度的增加,ct DNA-AO体系的荧光被猝灭,其猝灭过程主要是静态猝灭,ENPQO对ct DNA-AO体系的荧光偏振基本无影响;不同浓度的Na Cl溶液未削弱ENPQO对ct DNA-AO的猝灭程度。以上结果均表明ENPQO与ct DNA之间的作用方式主要为沟槽结合。利用分子模拟对接技术预测了ENPQO与ct DNA的结合最优构象,与光谱实验结果一致。 相似文献

双链DNA(dsDNA)中单碱基凸出结构(bulge structure)具有重要生物学意义,这种结构也是DNA靶向药物的目标部位之一.荧光小分子2-氨基-5,6,7-三甲基-1,8-萘啶(ATMND)能够通过氢键识别胞嘧啶(cytosine),因而对dsDNA中凸出的胞嘧啶表现出明显的特异性结合.与其余三种凸出的碱基相比,ATMND与凸出部位胞嘧啶的结合伴随着ATMND荧光的明显猝灭,因而可以用于胞嘧啶凸出结构的识别.利用解旋温度测量、荧光、圆二色光谱对ATMND和存在胞嘧啶凸出结构的dsDNA相互作用进行了研究.荧光滴定结果表明ATMND和dsDNA中凸出部位未配对的胞嘧啶的结合常数K11=4.8×105M?1.通过对含胞嘧啶凸出结构的dsDNA与ATMND结合前后的解旋温度曲线进行解析,发现胞嘧啶凸出结构相邻碱基对凸出的胞嘧啶与ATMND的结合有较大的影响.荧光测量结果也表明ATMND荧光的猝灭效率与凸出结构相邻碱基的类型有关,当相邻碱基为鸟嘌呤(guanine,G)时,荧光猝灭效率最高.基于dsDNA中凸出的碱基对ATMND荧光猝灭效率存在明显差异这一现象,设计了探针DNA实现了乳腺癌相关基因(PGR gene rs3740753)中单核苷酸多态性(G/C变异)的荧光分型. 相似文献

在生理酸度(pH 7.4)下,采用荧光光谱、紫外光谱法并结合溴化乙锭(EB)荧光探针、I-效应、离子强度及DNA热变性效应等实验手段研究了自制的β-二酮Ti(Ⅳ)新型抗肿瘤前药与DNA之间的相互作用。前药能极大地猝灭溴化乙锭(EB)-DNA体系的荧光,其电子吸收光谱在280 nm处的最大吸收峰在加入DNA后产生明显红移和减色效应。实验还发现KI对前药-DNA体系的荧光猝灭效率明显小于自由形式存在的前药的荧光猝灭效率;这些实验结果说明前药以嵌插方式作用于DNA的亲核位点。 相似文献

本文建立了利用Al(Ⅲ)-8-羟基喹啉-5-磺酸络合体系测定痕量F-的荧光猝灭分析方法。在pH=4.90的HAc-NaAc缓冲体系中,Al(Ⅲ)和8-羟基喹啉-5-磺酸能够形成稳定的有较强荧光的络合物,激发和发射波长分别为365nm和494.4nm。该体系中加入F-后络合物荧光不同程度猝灭,当F-浓度为0.2~3.5μg·mL-1时,其荧光猝灭程度与F-的浓度呈线性关系,方法加标回收率为96.2%~104.0%,相对标准偏差(RSD)小于3.5%,检测限为10ng·mL-1。用抗坏血酸-邻菲罗啉掩蔽可消除Fe(Ⅲ)、Cu(Ⅱ)和Ni(Ⅱ)的干扰。该方法简便快速、灵敏度高、选择性好,可用于水样和牙膏中氟化物的测定。 相似文献

研究了酸度、温度对不对称草酰胺:N-邻羧基苯基-N-(2-氨己基)草酰胺(OBZE)荧光特性的影响。测定了该试剂的荧光量子产率。在pH8.0~9.5的H3BO3-NaOH缓冲介质中,NO3^-能使OBZE荧光强度定量猝灭,据此建立了荧光猝灭法测定NO3^-的新体系。OBZE的激发波长为210nm,发射波长为395nm。荧光猝灭值与NO3^-在0.004~0.68mg/L呈范围内线性关系,方法的检出限为0.004mg/L。实验了多种干扰离子的影响,已用于水中硝酸根离子的测定。 相似文献

利用萘酰亚胺衍生物N-氨基-4-(N-甲基哌嗪)-1,8-萘酰亚胺(AMN)为荧光探针,建立了一种荧光猝灭法分析测定蛋白质的新方法。在pH=3.5的磷酸盐缓冲体系中,AMN的激发和发射波长分别为387nm和533nm。牛血清蛋白(BSA)的加入能够使其荧光发生不同程度的猝灭,猝灭程度与0.1~12.0μg·mL-1浓度范围的BSA呈线性关系。该方法简便快速,选择性好且灵敏度高,对BSA的检测限为1.05×10-8 g·mL-1;用于血清中总蛋白质含量的测定,加标回收率为98.7%~103.2%,相对标准偏差(RSD)在3.63%以内。 相似文献

用溶胶-凝胶法以磷钼酸(MPA)的镍盐溶液水解钛酸四丁酯制备了NiPMo/TiO2催化剂.使用ICP、 XRD、 TG-DTA、 IR、 TPD-MS和微反应技术研究了催化剂的化学组成、热稳定性、化学吸附性质和催化反应性能.杂多钼酸盐与TiO2通过O2-在TiO2表面发生了键合.在623 K下,杂多阴离子仍保持原有的Keggin结构.CO2在Lewis酸位Ni(Ⅱ)和Lewis碱位Ni-O-Mo的桥氧协同作用下生成CO2卧式吸附态Ni(Ⅱ)←O-(CO)←(O--Ni).丙烯有多种吸附态在催化剂上吸附.在563 K、 1 MPa和空速1500 h-1的反应条件下,丙烯的摩尔转化率为3.2%,产物MAA选择性为95%. 相似文献

The Langevin paramagnetic theory can’t describe the relation between magnetization of ferrofluids and applied magnetic field. The structuralization of ferrofluids, which is considered the main influence factor of the magnetization, is regarded. The part of magnetization works is deposited when the structure is forming. This action influences the magnetization of ferrofluids directly or indirectly. On the base of the “compressing” model, the Langevin function that usually describes the magnetization of ferrofluid is modified, and a well-fitted curve is obtained. An equation of the relation between the equivalent volume fraction after being “compressed” and the intensity of magnetic field is discovered, which approximately describes the process of magnetization. The relation between the approximate initial susceptibility and the volume fraction can be obtained from modified formula. 相似文献

The highly regioselective Buchwald–Hartwig amination at C-2 of the cheap and readily accessible reagent, 2,4-dichloropyridine with a range of anilines and heterocyclic amines is described. This new methodology is robust and provides a facile access to 4-chloro-N-phenylpyridin-2-amines on 0.25 mol scale. These intermediates undergo a further Buchwald–Hartwig amination at higher temperature to enable rapid exploration of the chemical space at C-4 and to provide a library of 2,4-bisaminopyridines. 相似文献

KMnO4-mediated oxidative CN bond cleavage of tertiary amines producing secondary amine was introduced, which was trapped by electrophiles (acyl chloride and sulfonyl chloride) to form amides and sulfonamides. A general synthesis of previously unknown semicarbazone-based α-amidoalkylating reagents, 4-(tosylmethyl)semicarbazones, has been developed. The synthesis involved three-component condensation of semicarbazones of aliphatic or aromatic aldehydes with the same or other aldehydes and p-toluenesulfinic acid. The scope and limitations of this reaction were investigated. The compounds obtained were demonstrated to be an efficient α-(4-semicarbazono)alkylating agents. They were reacted with H- (sodium borohydride), O- (sodium methylate), S- (sodium phenylthiolate), N- (pyrrolidine, sodium succinimide), P- (trialkyl phosphites), and C-nucleophiles (sodium diethyl malonate) to give the corresponding products of the tosyl group substitution, 4-substituted semicarbazones, including analogues of nitrofurazone. Among the prepared compounds tested in vitro for antibacterial and antifungal activity, three nitrofuryl-containing semicarbazones exhibited high biological activities with minimum inhibitory concentration (MIC) values of 8–32 μg/mL. 相似文献

Regiospecific and direct imidation of the methyl C(sp3)–H bond of thioanisoles is realized under mild and metal-free conditions with N-fluorobis(benzenesulfonyl)imide as an oxidant and nitrogen source. Proposed mechanism suggests that thionium ion intermediates and a Pummerer-type reaction are involved. The imidation has advantages such as high step-economy, excellent functionality tolerance, and regiospecificity, giving structurally diverse imidation products. 相似文献

A small library of new chiral bidentate hydroxyalkyl-imidazolium salts 1 is conveniently synthesized on multi-gram scale from inexpensive and commercially available chiral pool amino acids. The corresponding carbenes, generated by deprotonation of imidazolium salts 1, in combination with palladium(II) chloride were tested in the Mizoroki–Heck coupling reaction. The most significant results in terms of yields and reactivities were achieved with low catalyst loading. The catalytic activities of these imidazolium salts were also investigated in the asymmetric addition of diethylzinc to benzaldehyde. The use of MgO nanoparticles as an additive in conjunction with these ligands played a crucial role in increasing the efficiency of these reactions. 相似文献
