In order to increase productivity of drug analysis in the pharmaceutical industry, an efficient and sensitive generic static headspace gas chromatography (HSGC) method was successfully developed and validated for the determination of 44 classes 2 and 3 solvents of International Conference of Harmonization (ICH) guideline Q3C, as residual solvents in drug substance. In order to increase the method sensitivity and efficiency in sample equilibration, dimethylsulfoxide (DMSO) was selected as the sample diluent based on its high capacity of dissolving drug substance, stability and high boiling point. The HS sample equilibration temperature and equilibration time are assessed in ranges of 125–150 °C and 8–15 min, respectively. The results indicate that the residual solvents in 200 mg of drug substance can be equilibrated efficiently in HS sampler at 140 °C for 10 min. The GC parameters, e.g. sample split ratio, carrier flow rate and oven temperature gradient are manipulated to enhance the method sensitivity and separation efficiency. The two-stage gradient GC run from 35 to 240 °C, using an Agilent DB-624 capillary column (30 m long, 0.32 mm I.D., 1.8 μm film thickness), is suitable to determine 44 ICH classes 2 and 3 solvents in 30 min. The method validation results indicate that the method is accurate, precise, linear and sensitive for solvents assessed. The recoveries of most of these solvents from four drug substances are greater than 80% within the method determination ranges. However, this method is not suitable for the 10 remaining ICH classes 2 and 3 solvents, because they are too polar (e.g. formic acid and acidic acid), or have boiling points higher than 150 °C, (e.g. anisol and cumene). In comparison with the previous published methods, this method has a much shorter sample equilibration time, a better separation for many solvents, a higher sensitivity and a broader concentration range.  相似文献

At the present study, a new and rapid headspace solvent microextraction (HSME), for the extraction and pre-concentration of the volatile components of plant sample into a microdrop was applied. The extraction occurred by suspending a microliter drop of the solvent from the tip of a microsyringe to the headspace of a ripen and powdered dry fruit sample (Iranian Pimpinella anisum seed) in a sealed vial for a preset extraction time, then the microdrop was retracted back into the microsyringe and injected directly into a GC injection port. The chemical composition of the HSME extracts were confirmed according to their retention indexes and mass spectra (EI, 70 eV); and quantitative analysis was performed by GC-FID.Parameters such as the nature of the extracting solvent, particle size of the sample, temperatures of the microdrop and sample, volume of sample and the extraction time were studied and optimized, and the method's performance was evaluated. The optimized conditions were: sample particle size, 1 mm; sample volume, 5 ml (in a 15 ml vial); sample temperature, 60 °C; microsyringe needle temperature, 0 °C; and extraction time, 10 min. Finally, accordingly, the percentage of trans-anethole (the major compound of P. anisum) and the relative standard deviation for extraction and determination of trans-anethole (seven-replicated analysis) were determined to be 90% and 3.9%, respectively.  相似文献

Perfluoro-2-methylene-1,3-dioxolane (III) was synthesized and polymerized with an initiator, perfluoro dibenzoyl peroxide, and a white solid product III-P was quantitatively isolated. The polymer was insoluble in organic solvents including fluorinated solvents such as Fluorinert FC 75 and hexafluorobenzene, but dissolved in hexafluorobenzene by heating at around 140 °C in a sealed ampoule. The X-ray measurement showed that III-P was semi-crystalline and melted at 230 °C. The IR spectrum of III-P indicated that the polymer obtained did not show carbonyl peak and it was the vinyl addition product. When the solid product was heated above the melting temperature and pressed under 100-200 kg/cm2, we obtained an amorphous and flexible film, which is transparent from the UV region to the near IR region. The glass transition temperature was 110 °C and refractive indexes were 1.3443, 1.3434 and 1.3373 at 633, 839 and 1544 nm, respectively. The film did not degrade in concentrated sulfuric acid and aqueous sodium hydroxide solutions even heated at 80-90 °C for 2 days. The film was thermally stable and began to decompose at 300 °C under air atmosphere.  相似文献

A simple and efficient liquid-phase microextraction (LPME) in conjunction with gas chromatography-electron capture detector (GC-ECD) has been developed for extraction and determination of 11 organochlorine pesticides (OCPs) from water samples. In this technique a microdrop of 1-dodecanol containing pentachloronitrobenzene (internal standard) is delivered to the surface of an aqueous sample while being agitated by a stirring bar in the bulk of solution. Following completion of extraction, the sample vial was cooled by putting it into an ice bath for 5 min. Finally 2 μL of the drop was injected into the GC for analysis. Factors relevant to the extraction efficiency were studied and optimized. Under the optimized extraction conditions (extraction solvent: 1-dodecanol; extraction temperature: 65 °C; sodium chloride concentration: 0.25 M; microdrop and sample volumes: 8 μL and 20 mL respectively; the stirring rate: 750 rpm and the extraction time: 30 min), figures of merit of the proposed method were evaluated. The detection limits of the method were in the range of 7-19 ng L−1 and the RSD% for analysis of 2 μg L−1 of OCPs was below 7.2% (n = 5). A good linearity (r2 ≥ 0.993) and a relatively broad dynamic linear range (25-2000 ng L−1) were obtained. After 30 min of extraction, preconcentration factors were in the range of 708-1337 for different organochlorine pesticides and the relative errors ranged from −10.1 to 10.9%. Finally the proposed method was successfully utilized for preconcentration and determination of OCPs in different real samples.  相似文献

A method based on single-drop microextraction (SDME) combined with electrothermal vaporization (ETV)-ICP-MS was proposed for the determination of trace Cd and Pb. 8-Hydroxyquinoline (8-HQ) was employed as extractant dissolved in several microliters of chloroform and then an organic microdrop was formed at the tip of the microsyringe needle to extract the interest analytes. The vaporization behavior of the metal-8-HQ chelates in graphite furnace was investigated, and the ETV temperature program was optimized. The factors that influenced the extraction efficiency of target analytes (including pH value, flow rate of sample, extraction time and organic microdrop volume) were studied. Under the optimum conditions, the detection limits of the Cd and Pb were 4.6 and 2.9 pg mL−1 with the enrichment factor of 140-fold for Cd and 190-fold for Pb, respectively. The proposed method was applied successfully to the determination of trace Cd and Pb in environmental and biological samples. In order to validate the developed method, a certified reference material of GBW 08501 peach leaves was analyzed and the determined values obtained were in a good agreement with the certified values.  相似文献

A fast and reliable analytical method using microwave assisted extraction has been developed. Several extraction solvents (methanol (MeOH) and ethanol (EtOH), 30-70% in water and water), temperatures (50-150 °C), extraction solvent volume, as well as the sample size (1.0-0.1 g) and extraction time (5-30 min) were studied for the optimization of the extraction protocol. The optimized extraction conditions for quantitative recoveries were: 0.5 g of sample, 50 °C, 20 min and 50% ethanol as extracting solvent. No degradation of the isoflavones was observed using the developed extraction protocol and a high reproducibility was achieved (>95%).  相似文献

The use of natural and synthetic anabolic steroids in animal fattening has been prohibited in Taiwan and many countries because of their potential toxic effect on public health. This paper describes a newly developed gas chromatography-ion trap-mass spectrometry (GC-IT-MS) method for the quantitative determination of various residual anabolic steroids in meat. Anabolic steroid was derivatized with N-methyl-N-trimethylsilytrifluoroacetamide prior to GC-IT-MS analysis. MS2 was employed for quantitative measurement. In addition, 2d-estradiol was used as an internal standard. Quantitative determination was based on the ratio of peak area of steroid derivative to peak area of internal standard derivative. Good linearity of each compound, 0.03-1.0 μg/ml, was determined. Solvent extraction was used to extract residual anabolic compounds in meat samples and a solid phase extraction (SPE) procedure was utilized for sample cleanup and pre-concentration. The limits of detection of anabolic compounds approximately ranged from 0.1 to 0.4 μg/kg. The detection limit was comparable with or better than reported methods and was below the minimum required performance limits (MRPLs) established by the European Community (EC). The application of this newly developed method was demonstrated by analyzing various beef, pork, chicken and several animal internal organ samples from local markets.  相似文献

A miniaturized methodology for the determination of phosphate in waters has been developed by combining directly suspended droplet microextraction (DSDME) with microvolume spectrophotometry. The method is based on the extraction of the ion pair formed between 12-molybdophosphate and malachite green onto a microdrop of methyl isobutyl ketone and subsequent spectrophotometric determination with no dilution. An enrichment factor of 325 was obtained after 7.5 min of microextraction. The detection limit was 6.1 nM phosphate and the repeatability, expressed as relative standard deviation, was 2.7% (n = 6). The method was successfully applied to the determination of dissolved reactive phosphorus in different freshwater samples.  相似文献

Isoflavone derivatives from freeze-dried soybeans were extracted by pressurized liquid extraction (PLE) and determined by reverse-phase high performance liquid chromatography (HPLC) with both photo diode array and mass spectrometry (MS) detection. Both real and spiked samples were used in the development of the method.Several extraction solvents (methanol (MeOH) and ethanol (EtOH), 30-80% in water and water), temperatures (60-200 °C), pressures (100-200 atm), as well as the sample size (0.5-0.05 g) and cycle length (5-10 min) were studied for the optimization of the extraction protocol. The optimized extraction conditions for quantitative recoveries were: 0.1 g of sample, 100 °C, three (7 min) static extraction cycles and ethanol 70% as extracting solvent. The stability of the isoflavones during the PLE was also determined. Under PLE conditions, degradation of malonyl glucoside forms of the isoflavones takes place using temperatures higher than 100 °C whereas degradation of glucosides takes place above 150 °C. Using the optimized protocol, isoflavones can be extracted from freeze-dried soybeans without degradation.  相似文献

Headspace solvent microextraction (HSME) was shown to be an efficient preconcentration method for extraction of some polycyclic aromatic hydrocarbons (PAHs) from aqueous sample solutions. A microdrop of 1-butanol (as extracting solvent) containing biphenyl (as internal standard) was used in this investigation. Extraction occurred by suspending a 3 μl drop of 1-butanol from the tip of a microsyringe fixed above the surface of solution in a sealed vial. After extraction for a preset time, the microdrop was retracted back into the syringe and injected directly into a GC injection port. The effects of nature of extracting solvent, microdrop and sample temperatures, stirring rate, microdrop and sample volumes, ionic strength and extraction time on HSME efficiency were investigated and optimized. Finally, the enrichment factor, dynamic linear range (DLR), limit of detection (LOD) and precision of the method were evaluated by water samples spiked with PAHs. The optimized procedure was successfully applied to the extraction and determination of PAHs in different water samples.  相似文献

A method for the determination of 11 UV-filter compounds in sludge has been developed and evaluated. The procedure includes the use of non-porous polymeric membranes in combination with pressurised liquid extraction (PLE). Firstly, the solid sample, wetted with the extraction solvent, was enclosed into tailor-made bags prepared with low density polyethylene. Secondly, these packages were submitted to a conventional PLE (70 °C, 4 cycles of 5 min static time). Finally, the analytes were determined by liquid chromatography–atmospheric pressure photoionisation–tandem mass spectrometry. The main advantage of this procedure is the reduction of time, solvent and labour effort ought to the combination of extraction and clean-up in a single step. Although the extraction is not quantitative (thus, standard addition is recommended for quantification) selectivity is clearly gained using the membrane as a consequence of the differences of permeation and transport through the membrane between the analytes and other sample matrix components. The optimised protocol provides limits of detection ranging from 0.3 ng g−1 (ethylhexyl dimethyl p-aminobenzoate (OD-PABA)) to 25 ng g−1 (ethylhexyl triazone (EHT)) with only 0.5 g of sludge sample. All the studied UV filters were found in the samples at concentration levels between 1.4 and 2479 ng g−1, emphasising the high adsorption potential of this kind of environmental pollutants onto solid samples such as sludge. Also, this method has permitted the determination of seven of the studied UV filters in sludge samples for the first time.  相似文献

A new approach using single-drop microextraction (SDME) and gas chromatography–mass spectrometry for the determination of six haloacetic acids (HAAs) in water samples was presented. n-Octanol was used as extractant and derivatization reagent. HAAs were derivatized both simultaneously during the extraction in the solvent microdrop, and after extraction, inside a glass microvial (1.1 mm I.D.). Trifluoroacetic anhydride (TFAA) was used as the reaction catalyst. The influence of catalyst amount, derivatization time and temperature on the yield of the in-microvial derivatization was investigated. Derivatization reaction was performed using 1.2 μL of TFAA at 100 °C for 20 min. Extraction was performed using 1.8 μL of n-octanol containing TFAA (10%, v/v). Experimental parameters, such as, exposure time, sample pH and extraction temperature were controlled and optimized. Analytical parameters such as linearity, precision and limit of detection were also evaluated. The proposed method was proved to be a suitable analytical procedure for HAAs in water with limits of detection 0.1–1.2 μg/L. The relative recoveries range from 82.5 to 97.6% for all the target analytes. Precision values were from 5.1 to 8.5% (as intra-day relative standard deviation, RSD) and 8.8–12.3% (as inter-day RSD).  相似文献

In the present work, a novel method for the determination of trihalomethanes (THMs) such as chloroform, dichlorobromomethane, chlorodibromomethane and bromoform in drinking water has been described. It is based on coupling headspace liquid-phase microextraction (HS-LPME) with gas chromatography-electron capture detector (GC-ECD). A microdrop of organic solvent at the tip of a commercial microsyringe was used to extract analytes from aqueous samples. Three organic solvents—xylene, ethylene glycol and 1-octanol—were compared and 1-octanol was the most sensitive solvent for the analytes. Extraction conditions such as headspace volume, extraction time, stirring rate, content of NaCl and extraction temperature were found to have significant influence on extraction efficiency. The optimized conditions were 15 ml headspace volume in a 40 ml vial, 10 min extraction time and 800 rpm stirring rate at 20 °C with 0.3 g ml−1 NaCl. The linear range was 1-100 μg l−1 for THMs. The limits of detection (LODs) ranged from 0.15 μg l−1 (for dichlorobromomethane and chlorodibromomethane) to 0.4 μg l−1 (for chloroform); and relative standard deviations (RSD) for most of THMs at the 10 μg l−1 level were below 10%. Real samples collected from tap water and well water were successfully analyzed using the proposed method. The recovery of spiked water samples was from 101 to 112%.  相似文献

A cold column trapping-cloud point extraction (CCT-CPE) method coupled to high performance liquid chromatography (HPLC) was developed for preconcentration and determination of curcumin in human urine. A nonionic surfactant, Triton X-100, was used as the extraction medium. In the proposed method, a low surfactant concentration of 0.4% v/v and a short heating time of only 2 min at 70 °C were sufficient for quantitative extraction of the analyte. For the separation of the extraction phase, the resulted cloudy solution was passed through a packed trapping column that was cooled to 0 °C. The temperature of the CCT column was then increased to 25 °C and the surfactant rich phase was desorbed with 400 μL ethanol to be directly injected into HPLC for the analysis. The effects of different variables such as pH, surfactant concentration, cloud point temperature and time were investigated and optimum conditions were established by a central composite design (response surface) method. A limit of detection of 0.066 mg L−1 curcumin and a linear range of 0.22–100 mg L−1 with a determination coefficient of 0.9998 were obtained for the method. The average recovery and relative standard deviation for six replicated analysis were 101.0% and 2.77%, respectively. The CCT-CPE technique was faster than a conventional CPE method requiring a lower concentration of the surfactant and lower temperatures with no need for the centrifugation. The proposed method was successfully applied to the analysis of curcumin in human urine samples.  相似文献

In this article, we report a new method that involves headspace single-drop microextraction and ion chromatography for the preconcentration and determination of fluoride. The method lies in the in situ hydrogen fluoride generation and subsequent sequestration into an alkaline microdrop (15 μL) exposed to the headspace above the stirred aqueous sample. The NaF formed in the drop was then determined by ion chromatography. The influences of some crucial single-drop microextraction parameters such as the extraction temperature, extraction time, sample stirring speed, sulphuric acid concentration and ionic strength of the sample, on extraction efficiency were investigated. In the optimal condition, an enrichment factor of 97 was achieved in 15 min. The calibration working range was from 10 μg L−1 to 2000 μg L−1 (R2 = 0.998), and the limit of detection (signal to noise ratio of 3) was 3.8 μg L−1 of fluoride. Finally, the proposed method was successfully applied to the determination of fluoride in different milk samples. The recoveries of fluoride (at spiked concentrations of 200 μg L−1 and 600 μg L−1 into milk) in real samples ranged from 96.9% to 107.7%. Intra-day precision (N = 3) in terms of peak area, expressed as relative standard deviation, was found to be within the range of 0.24-1.02%.  相似文献

A CPE-HPLC (UV) method was developed for the determination of Sudan (I-IV) dyes, non-ionic surfactant Triton X-100 was used to extract and preconcentrate Sudan dyes from chilli powder prior to their determination by HPLC-UV. The separation and determination of Sudan dyes was then carried out in the HPLC-UV system with isocratic elution, and the detector was set at 500 nm. The parameters and variables that affect the extraction were investigated. Under optimum conditions: 3% of Triton X-100 (W/V), 10% of Na2CO3 (W/V), heat-assisted at 70 °C for 30 min. Recoveries of the Sudan dyes ranged from 80.70% to 85.45% in chilli powder by CPE method, with all the relative standard deviations of less than 3%. Limit of detection (LOD) and limit of quantification (LOQ) were in the range of 2.0-4.0 and 7.0-12.0 μg kg−1 in chilli powder, respectively.  相似文献

A new sol–gel hybrid coating, polydimethylsiloxane–2-hydroxymethyl-18-crown-6 (PDMS–2OHMe18C6) was prepared in-house for use in solid phase microextraction (SPME). The three compositions produced were assessed for its extraction efficiency towards three selected organophosphorus pesticides (OPPs) based on peak area extracted obtained from gas chromatography with electron capture detection. All three compositions showed superior extraction efficiencies compared to commercial 100 μm PDMS fiber. The composition showing best extraction performance was used to obtain optimized SPME conditions: 75 °C extraction temperature, 10 min extraction time, 120 rpm stirring rate, desorption time 5 min, desorption temperature 250 °C and 1.5% (w/v) of NaCl salt addition. The method detection limits (S/N = 3) of the OPPs with the new sol–gel hybrid material ranged from 4.5 to 4.8 ng g−1, which is well below the maximum residue limit set by Codex Alimentarius Commission and European Commission. Percentage recovery of OPPs from strawberry, green apple and grape samples with the new hybrid sol–gel SPME material ranged from 65 to 125% with good precision of the method (%RSD) ranging from 0.3 to 7.4%.  相似文献

Liquid-liquid-liquid phase microextraction (LLLME) coupled with high-performance liquid chromatography (HPLC) for the analysis of some aromatic amines is described. These compounds were extracted from 4.0 mL aqueous sample that adjusted to pH 13 with, NaOH-NaCl buffer solution (donor phase, P1) into an organic phase (P2) 150 μl benzyl alcohol and ethyl acetate (2:1) and then back extracted into a microdrop of aqueous acceptor phase (P3), adjusted at pH 2, with Na2HPO4-H3PO4 buffer solution. The extraction time, T1 (from P1 to P2) was 20 min and T2 (from P2 to P3) was 1 min. Different crown ethers as complexing agents for amines were added to the acceptor phase to improve the extraction time. Factors such as organic solvents, extraction times, and addition of crown ethers to acceptor phase and stirring rate were optimised. The method was applied for determination of aromatic amines in wastewater samples. Enrichment factors ranged from 184.5 to 389.7. The linearity range was from 3 to 1000 ng/ml and the detection limits varied from 0.8 to 1.80 ng/ml. Relative standard deviations (%, n = 5) were found (at S/N 3) in the range of 1.9 to 10.1. All experiments were carried out at room temperature, 22 ± 0.5 °C.  相似文献

Bimetallic Ni-Cu particles were synthesized from either suspensions of nickel carbonate and copper carbonate, and solutions of nickel nitrate and copper nitrate in ethylene glycol which acts both as solvent and reducing agent. The nature and composition of the powders depend on both the reaction temperature and time, and the reactants. Using the carbonates, bimetallic Ni-Cu powders composed of a nickel-rich and a copper-rich solid solution were obtained after 39 h at 140°C. Increasing the reaction temperature to 190°C gives a Ni-Cu powder composed of a copper-rich solid solution and nickel. Particles obtained under these conditions, however, are polydisperse. The nitrate solution gave bimetallic Ni-Cu particles with a narrow size distribution of about 140 nm after 4 h of reaction at 196°C. These particles are made of a copper core and a nickel shell. The mechanism of bimetallic particle formation is controlled by the solubility of the reactants, the formation of intermediate metal glycolates and Cu2O, and the required reduction temperature.  相似文献

A simple and stability-indicating high performance liquid chromatographic method was developed and validated for the determination of miconazole nitrate in bulk and cream preparations. The extraction step for cream samples consisted in a warming, cooling and centrifugation procedure that assures the elimination of the lipophilic matrix component, in order to avoid further precipitation in the chromatographic system. Separation was achieved on a ZORBAX Eclipse XDB - C18 (4.6 mm × 150 mm, 5 μm particle size) column, using a mobile phase consisting of water, methanol and acetonitrile, in a flow and solvent gradient elution for 15 min. The column was maintained at 25 °C and 10 μL of solutions were injected. UV detection was performed at 232 nm, although employment of a diode array detector allowed selectivity confirmation by peak purity evaluation. The method was validated reaching satisfactory results for selectivity, precision and accuracy. Degradation products in naturally aged samples could be simultaneously evaluated, without interferences in the quantitative analysis.  相似文献
