A new type of chiral surfactant, sodium maleopimaric acid (SMA), was synthesized, and employed for the enantioselective micellar electrokinetic chromatographic (MEKC) separation of amino acid enantiomers derivatized with naphthalene-2,3-dicarboxaldehyde (NDA-D/L-AAs). The effect of the surfactant concentration, type and concentration of the BGE, and buffer pH on the resolution was studied, and optimized conditions were used to evaluate the ability of this new surfactant to perform chiral separations toward NDA-D/L-AAs by MEKC. Enantiomeric separations of NDA-D/L-AAs were achieved with a running buffer consisting of 100 mM borate (pH 9.5) and 20 mM SMA in a 58.5 cm length x 50 microm id capillary. Under the conditions selected, two pairs of tested amino acid enantiomers including NDA-D/L-trptophan (Trp) and NDA-D/L-kynurenine (Kyn) were resolved.  相似文献

报道了一种利用具有乙酰鸟氨酸脱乙酰酶活性的固定化细胞拆分D,L-缬氨酸的新方法. 该酶促反应最适条件: pH=6, 反应温度50 ℃, 底物N-乙酰-D,L-缬氨酸浓度200 mmol/L, 固定化细胞用量0.2 g/mL(或100 U/mL). 0.1 mmol/L CoCl2条件对该酶促反应有显著的激活作用. 在以上条件下反应2~3 h, 测得产物L-缬氨酸浓度95 mmol/L. 该固定化细胞连续10次使用, 平均转化率为90.8%(以N-乙酰-L-缬氨酸计), 显示出了良好的工业化应用前景.  相似文献

The formation of D-amino acids (D-aa's) in many fermented foods depends, among other factors, on the particular fermentation conditions, the action and autolysis of the microorganisms involved. In this sense, the analysis of chiral amino acids is an interesting analytical strategy for food scientists, since these compounds can be used as bacterial markers and can help, e.g., to detect adulterations, microbiological contaminations, etc. In this work, a fast and sensitive method based on MEKC-LIF has been developed to analyze and quantitate L-amino acid (L-aa) and D-aa in vinegars. The chiral MEKC-LIF procedure uses 100 mM sodium tetraborate, 30 mM SDS, and 20 mM beta-CD at pH 9.7 as running buffer, obtaining a good separation of the main vinegar L-/D-aa previously derivatized with fluorescein isothiocianate. Namely, L/D proline, alanine, arginine, glutamic, and aspartic acid, plus the nonchiral amino acid gamma-aminobutyric acid are separated in less than 20 min with high efficiency (up to 720,000 plates/m) and good sensitivity (LODs lower than 16.6 nM were achieved). Several D-aa's were detected and quantified in balsamic, sherry, white wine, and cider vinegars using this MEKC-LIF procedure, observing interesting differences in their L-aa and D-aa profiles and contents.  相似文献

An optically active and electroconductive polymeric adsorbent has been developed for the use in chromatographic resolution of nonderivatized amino acids. The chiral selectivity of the adsorbent is based upon ligand exchange of coordinated copper(II) complexes of D or L-amino acids and a molecular imprinting technique by modifying the resin surface with polypyrrole coating. Applying a potential difference of +/-1.5 V to the chiral and conductive column, racemic amino acids are separated according to their charge characteristics, and simultaneously resolved with respect to their optical isomerisms. A pH-controlled mixture of D,L-lysine and D,L-aspartic acid is resolved displaying enantioselectivity values of 1.19 and 2.08, respectively, and a baseline separation of the two amino acids is accomplished by alternating the polarity of the electric field. The synthesized adsorbent also exhibits size exclusion factor discriminating amino acids with larger side chains.  相似文献

A new chiral derivatizing reagent, dehydroabietylisothiocyante (DHAIC), was synthesized and used for the enantiomeric separation of chiral compounds in capillary electrophoresis (CE). The synthetic route to obtain DHAIC is described. The separation conditions for the chiral separation of several chiral compounds, such as protein amino acids and chiral drug DOPA were optimized. Best results for the chiral separation of DHAIC derivatized amino acids and DOPA were obtained in a running buffer consisted of 50 mM borate (pH 9.5), 5 mM sodium dodecyl sulphate (SDS) and 20% acetonitrile for amino acids and 60 mM Na2HPO4 (pH 8.0), 17 mM SDS and 25% acetonitrile for DOPA. Under the conditions studied, chiral separation of five amino acids including Ser, Val, Ala, Thr, Cys and a chiral drug DOPA as their diastereomeric DHAIC derivatives has been achieved by micellar electrokinetic chromatography (MEKC).  相似文献

Summary A wide series of L-phenylalanine tetraamide selectors (Phe-n-O-TA) have been synthesized and used as stationary phases for chiral resolution of D,L-amino acids by capillary gas chromatography. The influence of length and polarity of the oxaalkanoyl bridge spacing the diamide chains of the selectors and the effects of steric hindrance on separating power were investigated. The characteristics of the chiral columns and the separation factors of D,L-n-butyl-N-trifluoroacetyl amino esters are reported.  相似文献

Summary 3-[(3-cholamidopropyl)-dimethylammoniol-1-propane sulfonate (CHAPS) can be used as an effective chiral selector for the separation of dansyl-amino acids by capillary electrophoresis (CE). While CHAPS can serve as an chiral selector, better enantiomeric separation can be performed by using CHAPS not as the sole chiral selector but as one of a [CHAPS-SDS-cyclodextrin] three-component system. In this CHAPS-SDS-CD system, enantiomeric separations of the amino acids can be readily accomplished by judiciously adjusting the pH of the solution, concentrations of CHAPS and SDS, and the concentration and type of CD. All amino acids can be baseline resolved in less than 15 minutes with resolution as high as 2.01 at pH 6.5 with 50 mM of CHAPS and 75 mM of SDS. The resolution is also dependent on the size of the CD. Substantial increase in the resolution can be readily achieved by replacing β-CD with γ-CD. For example, theR s for Leu was increased by four-folds (from 1.65 to 6.29) while the elution time still remains as short as 20 min when β-CD was replaced by γ-CD.  相似文献

Simultaneous chiral separations of underivatized amino acids have been performed using a teicoplanin-based chiral stationary phase and ionspray tandem mass spectrometry for their ionisation and detection. Different amino acid enantiomer pairs were separated simultaneously, including those of positional isomeric amino acids (e.g., L,D-Leu/Ile, or L,D-Val/Iva). Due to the specificity of tandem mass spectrometry, co-eluting enantiomers of different amino acids could also be determined. Fifteen chiral underivatized proteinogenic and non-proteinogenic amino acids were analysed simultaneously under isocratic conditions (acetonitrile-water, 75:25) in less than 25 min. For maximum sensitivity, post-column addition of 500 mM aqueous HCOOH was necessary. Detection limits varied from 2.5 to 50 microg l(-1) depending on the amino acid. The signal vs. concentration relationship was linear for all D- and L-amino acids (0.9995 < or = r2 < or = 1) for three orders of magnitude.  相似文献

3-(4-Methylbenzylidene)-camphor (MBC) is a chiral sunscreen agent used in cosmetic products. In this work, the enantioseparation of MBC has been performed by EKC and applied to the analysis of the MBC enantiomers in cosmetic creams. Different experimental conditions (type and concentration of the chiral selector, temperature, and sample solvent) have been optimized. Due to the neutral nature of this compound, anionic CD derivatives were investigated as chiral selectors. Carboxymethylated-beta-CD (CM-beta-CD) showed the highest chiral separation power, observing that a 15 mM concentration of this CD at a working temperature of 15 degrees C enabled to obtain the highest enantioresolution. However, under these conditions, tailing of peaks obtained for the enantiomers was observed. The addition of increasing concentrations of the neutral alpha-CD to CM-beta-CD at a 15 mM concentration in a 100 mM borate buffer at pH 9.0 improved the enantiomeric separation and decreased peak tailing. The use of DMF for the total dissolution of the cosmetic creams, and methanol:water (1:1 v/v) for appropriate dilution enabled to observe good shape and size for the peaks of the MBC enantiomers. After optimizing a method for the preconditioning of the capillary, the analytical characteristics of the chiral separation method for the analysis of MBC were investigated. Linearity, LODs and LOQs, precision (instrumental repeatability, method repeatability, intermediate precision), accuracy, and selectivity were evaluated. The method was applied to analyze MBC enantiomers contained in two commercial cosmetic creams containing racemic MBC and to study the skin absorption of this compound with time.  相似文献

In this paper we report a study on the mechanism of the enantiomeric separation of unmodified D,L-amino acids in RP-HPLC by copper(II) complexes of two tetradentate diaminodiamido ligands, (S,S)-N,N'-bis(phenylalanyl)ethanediamine (PheNN-2) and (S,S)-N,N'-bis(methylphenylalanyl)ethanediamine (Me2PheNN-2), added to the eluent. The aim is to investigate whether and how a copper(II) complex with no free equatorial positions can perform chiral discrimination of bidentate analytes such as unmodified amino acids. The problem is approached in a systematic way by: (a) varying the different chromatographic parameters (pH, selector concentration, eluent polarity); (b) performing chiral separation with the selector adsorbed on the stationary phase; (c) studying the ternary complex formation of these ligands with D- and L-amino acids in solution by glass electrode potentiometry and electrospray ionization MS. All the experimental data are consistent with a mechanism of chiral recognition, based on ligand exchange, which involves as selectors the species [Cu2L2H(-2)]2+ and [CuLH(-2)] and proceeds by displacement of two binding sites from the equatorial positions, giving rise to the ternary species [CuLA]+ and [CuLH(-1) A]. The most important factor responsible for chiral discrimination seems to be the affinity of the diastereomeric ternary complexes for the stationary phase since no enantioselectivity is observed in solution.  相似文献

Five new chiral macrocycles, 3a-e, have been prepared by the acylation cyclization of chiral diamine dihydrobromide intermediates 2a-c with 2,6-pyridinedicarbonyl dichloride in highly diluted solution at room temperature. The chiral diesters 1a-c needed for the preparation of the macrocycles were obtained from condensation of corresponding N-(Z)-L-amino acids and 2,6-bishydroxymethyl pyridine in the presence of DCC and DMAP. The enantiomeric recognition of chiral macrocycles 3a-e for D- and L-amino acid methyl ester hydrochlorides has been characterized by fluorescence spectra, which indicate that some of them exhibited significant chiral recognition for the enantiomers of D- and L-amino acid methyl ester hydrochlorides. The stoichiometry and binding constants of 3a-L-Am(2) and 3c-L-Am(2) complexes have been determined. An X-ray analysis of the chiral macrocycle 3b show that the chiral ligand is rather rigid and strained.  相似文献

The aim of this work was to assay seasoning D- or L-aromatic amino acids (AAs) in rice-brewed suspensions, Laozao in Chinese, by chiral ligand-exchange CE with UV detection and Zn(II) complex as a chiral selecting system. Resolution and peak retention were found to be parallel to the basicity of the AA chiral ligands, and basic L-Arg was known to work the best at pH 8.20 compared with L-Lys and other AA ligands. Baseline separation of DL-aromatic AAs and partially separation of some FMOC-labeled nonaromatic AAs have been achieved using a running buffer of 5 mM ammonium acetate, 100 mM boric acid, 3 mM ZnSO(4), and 6 mM L-Arg at pH 8.20. The aromatic amino acids in four brands of Laozao were measured in a range of 0.25-20 microg/mL for Typ, 1.00-120 microg/mL for Phe, and 2.50-200 microg/mL for Tyr, with linear regression coefficient all over 0.999. The LOD (S/N=3) was 0.15 microg/mL for Typ, 0.50 microg/mL for Phe, and 1.25 microg/mL for Tyr. The recovery of the method determined by spiking with the supernates of Laozao as background was 94.0-112.9%. The RSDs of migration time and peak area measured from six injections of tyrosine were 0.2 and 2.7%, respectively, for run-to-run, and 1.6 and 3.2%, respectively for day-to-day. Interestingly, there were only L-Trp, D-Tyr, and L-Tyr found in the assayed four brands of Laozao. They may serve as an index to recognize the brand of Laozao.  相似文献

A facile chiral ligand-exchange capillary electrophoretic method has been explored for the enantioseparation and UV detection of dansyl-amino acids with Zn(II) L-arginine complex as a chiral selecting system. Successful enantioseparation of 17 pairs of amino acid enantiomers has been achieved with a buffer of 100 mM boric acid, 5 mM ammonium acetate, 3 mM ZnSO4 and 6 mM L-Arg at pH 8.0, of which 10 pairs were fully resolved with resolution in between 1.59 and 4.21. This new method was shown to be applicable to the separation of some mixed pairs of amino acids and to the quantitative analysis of some real samples such as rice vinegars, with a linear range between 0.8 and 150 microg/mL, correlation coefficient above 0.99 and recovery in between 90.1 and 112.4%. It was found that amino acids with low resistance side chain(s), low tendency to form intramolecular hydrogen bond or high tendency to form intermolecular hydrogen bonds are more easily enantioseparated than those with extra carboxyl and/or phenyl groups. By the use of the suggested buffer, the running pH should be selected at 7.4-9.0 to compromise the resolution and elution speed.  相似文献

A new triflate-type fluorescence chiral derivatizing reagent, (S)-(+)-1-methyl-2-(6,7-dimethoxy-2,3-naphthalimido)ethyl trifluoromethanesulfonate, [S-(+)-MDNE-OTf], has been developed for the determination of the enantiomers of carboxylic acids. By introducing the two methoxy groups on the naphthalimido ring moiety, the red shift in the fluorescence spectrum and a high resolution in reversed-mode separation of the diastereomers of chiral carboxylic acids have been achieved. The detection limits (S/N=3) with ultraviolet and fluorescence detection are 8 fmol (λmax=283 nm) and 4 fmol (λex=283 nm, λem=467 nm), respectively.  相似文献

(+)-18-crown-6 tetracarboxylic acid (18C6H4) has been used as a chiral selector for D/L-amino acids in HPLC, where L-isomer is usually eluted prior to D-isomer, except for the case of serine. To clarify why serine exhibits the reverse order for the elusion, the chiral interactions of D- and L-serines with (+)-18C6H4 were investigated by the X-ray single crystal analyses, together with the case of D- and L-glutamic acids, which exhibit the usual elution order in HPLC. The backbone structures (amino, Calpha-H and carboxyl groups) of these four amino acids showed the nearly same interaction with (+)-18C6H4 despite their different chirality. In contrast, the hydroxyl group of L-serine side chain formed a hydrogen bond with the carboxyl group of (+)-18C6H4, whereas such a interaction was not formed for the side chain of D-serine and D- and L-glutamic acids. Thus, it was shown that the exception of D/L-serine from the first elution rule of L-isomer in HPLC is due to the presence and absence of a hydrogen bond formation of its side chain OH group.  相似文献

An effective micellar electrokinetic capillary chromatography with laser-induced fluorescence detection (MEKC-LIF) method has been proposed for the separation and the determination of 16 amino acids and two catecholamines using a new fluorogenic reagent, 3-(4-chlorobenzoyl)-2-quinolinecarboxaldehyde (Cl-BQCA), as the derivatizing reagent. The highest derivatization efficiency was achieved in pH 8.0 borate buffer at 50 °C for 50 min. The optimal separation of Cl-BQCA-labeled amines was obtained with a running buffer (pH 9.15) containing 120 mM boric acid, 38.5 mM sodium dodecyl sulfate, and 17% acetonitrile. The detection limit (S/N = 3) was found to be as low as 1.4 nM. The present method has been successfully used to detect amino acids and catecholamines in HEK293 and PC12 cell samples. This study explores the potential of MEKC-LIF with Cl-BQCA labeling as a tool for monitoring amino acids and catecholamines during the complex physiological and behavioral processes in various matrices.  相似文献

In this work, chiral and nonchiral MEKC methods have been combined with LIF detection (MEKC-LIF) to identify and quantify a group of D- and L-amino acids (D/L-aa) in different microalgae samples. The combination of the nonchiral and chiral-MEKC-LIF methods made the identification of the microalgae amino acids easier, previously derivatized with FITC, providing a double proof on the correct detection of these analytes. Three microalgae species, Spirulina platensis, Dunaliella salina, and Tetraselmis suecica, were compared in terms of their content in D-Arg, L-Arg, D-Lys, L-Lys, D-Ala, L-Ala, D-Glu, L-Glu, D-Asp, and L-Asp. Also, a comparison between two Spirulina platensis samples dried under different conditions (i.e., hot air or lyophilized) was carried out in order to investigate the effect of the thermal processing on the amino acid content. Moreover, two procedures for the extraction of amino acids from microalgae (i.e., a classical procedure and pressurized liquid extraction (PLE)) together with different conditions for amino acid derivatization were studied in order to increase the sensitivity of the whole analytical method. By using the selected chiral-MEKC-LIF conditions (100 mM sodium tetraborate, 30 mM SDS, and 20 mM beta-CD at pH 9.7) the main microalgae D/L-aa are separated in less than 25 min with efficiencies up to 840 000 plates/m and good sensitivity (i.e., 330 ng of D-Arg per gram of microalga could be detected by this procedure for an S/N of 3). Several D-aa were detected in all the microalgae, observing interesting differences in their D/L-aa profiles, what corroborates the usefulness of the chiral-MEKC-LIF approach to characterize different microalgae species as well as different microalgae drying processes. Moreover, the use of PLE can selectively extract different free amino acids from microalgae.  相似文献
