Our studies of Levantine viper venom have demonstrated that the venom is a rich source of biomedically important proteins and peptides. The venom contains metalloproteases: thrombolytic, fibrin-degrading lebetase, an endothelial cell apoptosis inducing metalloprotease (VLAIP), factor X activator (VLFXA); serine proteases: factor V activator, bradykinin-releasing serine proteases, β-fibrinogenase, α-fibrinogenase and chymotrypsin-like protease and different other enzymes such as phosphodiesterase, 5`-nucleotidase, ribonuclease, phospholipase A2s and L-amino acid oxidase. Among nonenzymatic components venom contains: nerve growth factor, vascular endothelial growth factor, disintegrins, C-type lectins.Here we report the isolation and characterization of proteins and peptides from Vipera lebetina venom using size exclusion, ion exchange, hydrophobic interaction and affinity chromatography, HPLC, UPLC and MALDI-TOF MS methods. N-terminal sequences and internal sequences of tryptic peptides of different proteins were determined using Edman sequencing and LC-ESI-MS/MS techniques. On the basis of fragmental sequences of proteins the oligonucleotides were designed and used as primers for cDNA cloning. Using cDNA library of the venom gland of a single snake the cDNAs coding proteins were cloned and sequenced. Protein sequences were deduced from cDNA sequences.The substrate specificity of venom proteases against insulin B-chain, bradykinin, substance P, and 6-10 amino acid residues containing peptides synthesized according to potential cleavage regions of fibrinogen, factor X, factor IX, factor V, α2-macroglobulin bait region and pregnancy zone protein were studied using MALDI-TOF mass spectrometry technique.  相似文献

Snake venom hyaluronidases known as “spreading factor” are not extensively studied. Recently, it is argued that beyond its role as a spreading factor, venom hyaluronidase (HYL) deserves to be explored as a possible therapeutic target for inhibiting the systemic distribution of venom/toxins and also for minimizing local tissue destruction. In this context, in the present study, a low-molecular weight HYL has been isolated from Bungarus caeruleus (Indian krait) venom by single step chromatography on HPLC system. The apparent molecular weight determined by SDS-PAGE is 14 ± 2 kDa, as confirmed by zymogen study and LC–MS as well. The enzyme had optimal pH 6 and temperature 37°C. The Michaelis–Menten constant (Km) was found to be 8.48 µg/mL at 37°C. The activity of purified enzyme was completely inhibited by Ba2+ metal ion and N-acetyl imidazole group-specific agents. This work yielded a highly active HYL from B. caeruleus the first one to be isolated. Further studies on its pharmacological actions will be interesting to develop lead molecules for better management of snakebite.  相似文献

Plant lectins are widely used in medical glycosciences and glycotechnology. Many lectin-based techniques have been applied for the detection of disease-associated glycans and glycoconjugates. In this study, Butea monosperma agglutinin (BMA), a lectin purified from seeds of the medicinal plant Butea monosperma, was used for the detection of cholangiocarcinoma (CCA)-associated glycans. Expression of BMA-binding N-acetyl galactosamine/galactose (GalNAc/Gal)-associated glycan (BMAG) in CCA tissues was determined using BMA lectin histochemistry; the results showed that BMAG was undetectable in normal bile ducts and drastically increased in preneoplastic bile ducts and CCA. The study in hamsters showed that an increase of BMAG was associated with carcinogenesis of CCA. Using an in-house double BMA sandwich enzyme-linked lectin assay, BMAG was highly detected in the sera of CCA patients. The level of serum BMAG in CCA patients (N = 83) was significantly higher than non-CCA controls (N = 287) and it was applicable for diagnosis of CCA with 55.4% sensitivity, 81.9% specificity, and 76.0% accuracy. A high level of serum BMAG (≥82.5 AU/mL) was associated with unfavorable survival of CCA patients; this information suggested the potential of serum BMAG as a poor prognostic indicator of CCA. In summary, BMAG was aberrantly expressed in preneoplastic bile ducts and CCA, it was also highly detected in patient serum which potentially used as a marker for diagnosis and prognostic prediction of CCA.  相似文献

The semi preparative HPLC method with PDA Detector was proposed for the determination of one phenolic acid, three flavonoids and one polyalcohol from Butea monosperma and Hedychium coronarium in gradient elution system. The influence of composition of the mobile phase concentration of the mix modifier and temperature on the separation of gallic acid, quercetin, iso-butrin, butrin and eugenol for 90 min was studied. Two different gradient programmes were used to separate these components. The lower limit of quantification of phenolic acid, flavonoids and eugenol is 0.050–0.150 μg/mL and was determined by the least square method and a good correlation was obtained for all separated components.  相似文献

This work is intended to examine the microbially influenced corrosion on galvanized steel (GS) caused by sulfate-reducing bacteria (SRB). The efficacy of Butea monosperma (palash) leaf extract to mitigate the corrosion caused by Desulfovibrio desulfuricans was investigated in modified Barr's medium. Weight loss and electrochemical analysis were performed to check the corrosion rate at regular time intervals. SEM images were performed to understand the level of deterioration of the metal surfaces. Image analysis of the unprotected sample showed the presence of pits. From the gravimetric study, the maximum inhibition efficiency (IE) of 98% was obtained with 500 ppm of Palash leaf extract for the fourth-week sample. With the addition of 500 ppm of palash extract, the sulfide concentration decreases to 0 ppm from 123 ppm. Outcomes of potentiodynamic polarization (PP) studies showed that the extract disturbs the cathodic reaction significantly and moves the corrosion potential to a more negative value and IE was about 71% from PP studies. FTIR and GC-MS analysis was performed to recognize the plausible chemical compounds present in Palash leaf powder. EIS results confirmed that the resistance to corrosion increases substantially with the addition of inhibitor. The mechanism for corrosion inhibition has been proposed based on the results obtained.  相似文献

Fractionation of the methanol extract of the stem bark of Turraeanthus africanus led to the isolation of two new alkaloids designated turraeanthin A and B, together with two known alkaloids. The structures of the new alkaloids were elucidated by means of spectroscopic analysis and characterized as 10-O-demethyl-17-O-methyl isoarnottianamide and 11-demethoxyl-12-methoxyl oxynitidine respectively.  相似文献

Two new sphingolipids mucusamide (1) and mucusoside (2) have been isolated from methanol soluble part of the stem bark of Ficus mucuso WELW., together with fifteen known secondary metabolites including cellobiosylsterol (3), β-sitosterol (4), stigmasterol (5), β-sitosterol 3-O-β-D-glucopyranoside (6), lupeol acetate (7), ursolic acid (8), procatechuic acid (9), 2-methyl-5,7-dihydroxychromone 8-C-β-D-glucoside (10), apigenin (11), (-)-epicatechin (12), (+)-catechin (13), N-benzoyl-L-phenylalanilol (14), α-acetylamino-phenylpropyl α-benzoylamino-phenylpropionate (15), asperphenamate (16) and bejaminamide (17). Summary A cation-exchange procedure has been developed for fractionation of venom from Bulgarian snakes (Vipera ammodytes), using a 75×7.5 mm TSK SP-5 PW column for gradient elution of venom components with NH4OAc and UV-detection at 280 nm. The procedure developed was applied for venom component separation and especially to assay the vipoxin content—a previously studied and described toxic protein complex—in the venom of various subspecies of Bulgarianvipera ammodytes. Chromatographic profiles obtained show that venom fromVipera ammodytes ssp.montandoni Boulenger, 1904 andVipera ammodytes ssp.meridionalis Boulenger, 1903 contain vipoxin (content higher in venom ofVipera ammodytes ssp.meridionalis), while in the venom ofVipera ammodytes ssp.ammodytes Linnaeus, 1758 vipoxin is absent. All data obtained show that the procedure developed can be successfully used for reliable subspecies affiliation by venom analysis and assay of vipoxin (in analytical mode) and its purification (in semipreparative mode) also.  相似文献

This study is aimed to isolate the phytoactives from the stem bark of Artocarpus hirsutus and evaluate their in vitro anti-acne activity. The ethanolic stem bark extract of A. hirsutus provided two major phytoactive constituents: (i) pyranocycloartobiloxanthone A, (1) and (ii) Artonine E, (2) whose structures were determined by NMR and MS spectroscopic analysis. The present study is the first to report compound 1 as a mixture of two anomers (α and β), approximately 70:30 ratio. Both compounds 1 and 2 were isolated for the first time from this plant. In vitro anti-acne activity of compounds 1 and 2 were evaluated by agar well diffusion method and the minimum inhibition was determined by broth micro dilution method. The result of anti-microbial activity (MIC = 2.0 μg/mL each) is comparable to antibiotic, Clindamycin (MIC = 0.03 μg/mL) and clearly demonstrate their potential as anti-acne agents.  相似文献

The chemical investigation of the ethyl acetate extract of the stem bark of Staudtia kamerunensis and sap led to the isolation of six compounds which included three isoflavonoids: biochanin A (1), formononetin (2) and 3-(1,3-benzodioxol-5-yl)-5,6,7-trimethoxy-4H-1-benzopyran-4-one (3), one flavonoid: (-) epicatechin (4) and two pentacyclic triterpenoids (oleanan-12-ene-2α,3β -diol (5) and 2α,3β-dihydroxylup-20-ene (6). They were characterized by HREIMS (High Resolution Electron Ionisation Mass Spectrometry), NMR spectroscopy (1D and 2D) and comparison with existing data in literature. The crude extract and isolates were tested against twelve bacterial strains namely; Bacillus subtilis, Staphylococcus epidermidis, Enterococcus faecalis, Mycobacterium smegmatis, Staphylococcus aureus, Enterobacter cloacae, Klebsiella oxytoca, Pseudomonas aeruginosa, Proteus vulgaris, Escherichia coli, Proteus mirabilis and Klebsiella pneumonia. Streptomycin, nalidixic acid and ampicillin were used as standard antibacterial drugs. The results revealed significant antibacterial activity for both the ethyl acetate partition and for the tested compounds, with the lowest MIC value being 15.625 μg/mL. A synergistic activity of the isolated triterpenoids was evaluated with interesting results. On a general note, the antibacterial activity of compound 5 was doubled specifically against Gram-negative bacterial strains. This could be a therapeutic antimicrobial pathway in face of the rising bacterial resistance. To the best of our knowledge, it is the first time that flavonoids and triterpenoids are isolated from this genus and species. It is also the first report of antibacterial studies on this species.  相似文献

The phytochemical study of stem bark of Allanblackia gabonensis has resulted in the isolation and characterisation of one new xanthone derivative, named allanxanthone D, together with 10 known compounds, including 6 xanthones derivatives, allanxanthone A, 1,5-dihydroxyxanthone, 1,7-dihydroxyxanthone and 1,3,6,7-tetrahydroxy-2-(3-methylbut-2-enyl)xanthone, forbexanthone, 6-deoxyisojacareubin, one polyisoprenylated benzophenone, guttiferone F, one flavanol, epicathechin, two phytosterols, beta-sitosterol and campesterol. The structures of these compounds were established on the basis of one- and two-dimensional NMR homo- and hetero-nuclear evidence. These compounds were evaluated for their activity against Leishmania amazonensis in vitro and antimicrobial activities against a range of Gram negative and Gram positive bacteria.  相似文献

The chemical composition of the volatile extract (yield ≈ 0.13%, v/w) from the bark of Quercus leucotrichophora (Fagaceae) was analysed for the first time by GC-MS. Twenty-three constituents, amounting to 93.0% of the total detected contents of the volatile extract, were identified. The volatile extract contained approximately 86.36% monoterpenoids, 6.53% sesquiterpenoids and 0.11% aliphatic aldehydes. 1,8-Cineol (40.359%) followed by γ-terpinene (16.369%) were the major monoterpene constituents of the volatile extract. The residue of volatile extract (0.00025-250 μg mL(-1)) exhibited a potent antimicrobial activity against Streptococcus pyogenes ATCC 19615. This study concludes that residues of the volatile extract of Q. leucotrichophora could serve as an important bioresource for the extraction and isolation of monoterpenoids exhibiting antimicrobial activity, and thus has good potential for use in the pharmaceutical industry.  相似文献

Two new lignans, 4-methoxymagnaldehyde B (1) and coumanolignan (2), were isolated from the stem bark of Magnolia obovata, together with 11 known compounds (3-13). The structures of compounds 1 and 2 were determined to be 5'-allyl-2'-hydroxyphenyl-4-methoxy-3-cinnamic aldehyde (1) and 6-allyl-8-(5'-allyl-2'-hydroxyphenyl)coumarin (2) on the basis of spectroscopic and physicochemical analyses including 2D NMR and high-resolution EI-MS. Compounds 1-8, 11, 12, and 13 were tested in vitro for their cytotoxic activities against the HeLa, A549, and HCT116 cancer cell lines. Among the compounds tested, compound 1 showed the strongest cytotoxic activity against the HCT116 cancer cell line, with an IC(50) value of 1.3 microg/ml.  相似文献

Agastache rugosa Kuntze (Korean mint) is used as a spice and in folk medicine in East Asia. The present study identified a total of 18 polyphenols from the flower, leaf, stem and roots of this plant using high‐performance liquid chromatography–tandem mass spectrometry. Fourteen of these compounds had not previously been identified in these plant tissues. Each polyphenol was validated in comparison with external calibration curves constructed using structurally related compounds, with determination coefficients >0.9993. The limits of detection and quantification were 0.092–0.650 and 0.307–2.167 mg/L, respectively. Recoveries of 61.92–116.44% were observed at two spiking levels, with 0.91–11% precision, expressed as relative standard deviation (except anthraquinone spiked at 10 mg/L). Hydroxycinnamic acid was the most abundant compound in the root, while the flowers showed the highest total flavonoid level. Antioxidant activities, determined in terms of reducing power, Fe2+ chelating activity and the radical scavenging activities using α,α‐diphenyl‐β‐picrylhydrazyl and 2‐2?‐azino‐bis‐3‐ethylbenzothiazoline‐6‐sulfonic acid, increased in a concentration‐dependent manner; the highest activity was identified in the stems, followed by leaves > flowers > roots. These findings indicate that A. rugosa is a good source of bioactive compounds and can be used as a functional food. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

A 1:1 thioredoxin-Pt(bpy) complex was prepared by adding [Pt(bpy)(en)]Cl(2)(bpy = 2,2'-bipyridine, en = ethylenediamine) to Thermus thermophilus HB8 thioredoxin in pH 8 phosphate buffer. Matrix-assisted laser desorption-ionization time of flight mass spectrometry (MALDI-TOF MS) and UV spectra of indicate the formation of Pt(bpy)(cys-Ala-Pro-cys-containing peptide fragment). These findings suggest that the Pt(bpy)(2+) unit binds to the active site of thioredoxin. The thioredoxin-platinum complex has no catalytic activity for the reduction of glutathione disulfide in the presence of NADPH and thioredoxin reductase, so that the platinum complex functions as an inhibitor.  相似文献

Gamma radiation alters the molecular structure of biomolecules and is able to mitigate the action of snake venoms and their isolated toxins. The effect of γ-radiation on the folding of Bothrops lecurus venom lectin was measured by a hemagglutinating assay, intrinsic and bis-ANS fluorescence. Intrinsic and bis-ANS fluorescence analyses indicated that irradiation caused unfolding followed by aggregation of the lectin. Our results suggest that irradiation can lead to significant changes in the protein structure, which may promote the loss of its binding property and toxic action.  相似文献

Helicobacter pylori is a major risk factor for gastritis, ulcers and gastric cancer. This study was aimed to determine the antimicrobial activity of the stem bark of Bridelia. micrantha on H. pylori isolated in South Africa. Extracts and clarithromycin were tested against 31 clinical strains, including a standard strain (NCTC 11638) of H. pylori, by measuring the diameters of the corresponding inhibition zones, followed by determination of the Minimum Inhibitory Concentration (MIC) (using metronidazole, and amoxicillin as control antibiotics) and the rate of kill. Preliminary phytochemical screening was also done. Inhibition zone diameters which ranged from 0-23 mm were observed for all five of the extracts and 0-35 mm for clarithromycin. Marked susceptibility of strains (100%) was noted for the acetone extract (P < 0.05), followed by ethyl acetate extract (93.5%). The MIC?? values ranged from 0.0048 to 0.156 mg/mL for the ethyl acetate extract and 0.0048 to 0.313 mg/mL for the acetone extract. The MIC?? values ranged from 0.0048 to 2.5 mg/mL for the ethyl acetate extract and 0.078 to > 0.625 mg/mL for the acetone extract, respectively. Insignificant statistical difference in potency was observed when comparing the crude ethyl acetate extract to metronidazole and amoxicillin (P > 0.05). Complete killing of strain PE430C by the ethyl acetate extract was observed at 0.1 mg/mL (2 × MIC) and 0.2 mg/mL (4 × MIC) at 66 and 72 h. For strain PE369C, 100% killing was observed at 0.1 mg/mL (2 × MIC) in 66 and 72 h. The ethyl acetate extract could thus be a potential source of lead molecules for the design of new anti-Helicobacter pylori therapies as this study further confirmed the presence of phytochemicals including alkaloids, flavonoids, steroids, tannins and saponins.  相似文献

Plant-derived compounds are emerging as an alternative choice to synthetic fungicides. Chloroform–methanol extract, obtained from the bark of Zanthoxylum rhoifolium, a member of Rutaceae, showed a fungistatic effect on Botrytis cinerea, Sclerotinia sclerotiorum, Alternaria alternata, Colletotrichum gloeosporioides and Clonostachys rosea, when added to the growth medium at different concentrations. A fraction obtained by gel separation and containing the alkaloid O-Methylcapaurine showed significant fungistatic effect against B. cinerea and S. sclerotiorum, two of the most destructive phytopathogenic fungi. The underlying mechanism of such an inhibition was further investigated in B. cinerea, a fungus highly prone to develop fungicide resistance, by analysing the expression levels of a set of genes (BcatrB, P450, CYP51 and TOR). O-Methylcapaurine inhibited the expression of all the analysed genes. In particular, the expression of BcatrB gene, encoding a membrane drug transporter involved in the resistance to a wide range of xenobiotic compounds, was strongly inhibited (91%).  相似文献

The CH2Cl2/MeOH extract of the stem bark of Oriciopsis glaberrima ENGL. afforded four new acridone alkaloids namely oriciacridone C, D, E and F along with six known compounds: atalaphyllidine, oleanolic acid, butulinic acid, beta-sitosterol, stigmasterol, glucoside of stigmasterol and one synthetically known acridone: 1,3,5-trihydroxy-4-prenylacridone. The structures were established on the basis of MS, 1D and 2D NMR experiments. The acridones 1, 4 and 5 showed potent activity against alpha-glucosidase, while the acridones 1-5 showed moderate free radical scavenging activity against 1,1-diphenyl-2-picrylhydrazyl (DPPH).  相似文献
