Phlorotannins of brown algae are multifunctional compounds with putative roles in herbivore deterrence, antioxidation and as primary cell wall components. Due to their peripheral localization and absorption at short wavelengths, a photoprotective role is suggested. We examined the induction of phlorotannins by artificial UV radiation in the intertidal kelp Lessonia nigrescens and whether they attenuate the inhibition of photosynthesis and DNA damage, two major detrimental effects of UV. The soluble and cell wall-bound fractions of phlorotannins were quantified in blades collected in summer and winter. Major findings were that (1) the synthesis of phlorotannins (both forms) was induced by UV only in summer; (2) the induction was fast (within 3 days); and (3) there was a positive relationship between of the contents of insoluble phlorotannins and the suppression of photoinhibition and DNA damage, measured as formation of cyclobutane pyrimidine dimers and 6-4 photoproducts. Overall, the photoprotective role of phlorotannins appears to respond to an interplay between the external UV stimulus, seasonal acclimation and intrinsic morpho-functional processes. In summer, when algae are naturally exposed to high UV irradiances, soluble phlorotannins are induced, while their transition to insoluble phlorotannins could be related with the growth requirements, as active blade elongation occurs during this season.  相似文献

The endemic Antarctic brown macroalga Desmarestia anceps is strongly shade‐adapted, but shows also a high capacity to cope with different environmental stressors, e.g. UV radiation and temperature. Therefore, this species colonizes wide depth gradients, which are characterized by changing environmental conditions. In this study, we examine whether the different physiological abilities allowing D. anceps to grow across a wide depth range is determined by high levels of phlorotannins. Photosynthesis, measured by PAM‐fluorometry, the contents of soluble phlorotannins, antioxidant capacities of field grown were analyzed in response to different conditions of radiation (PAR and PAR + UV) and temperature (2, 7 and 12°C). The results show that maximal quantum of fluorescence (Fv/Fm) decreased with increasing doses of UV radiation, but remained unaffected by temperature. High levels of soluble phlorotannins were detected and confirmed by microscopic observation revealing the abundance of large physodes. Exposure to UV radiation and elevated temperature showed that phlorotannins were not inducible by UV but increased at 12°C. ROS scavenging capacity was positively correlated with the contents of phlorotannins. In general, highest contents of phlorotannins were correlated with the lowest inhibition of Fv/Fm in all experimental treatments, highlighting the UV‐protective role of these compounds in D. anceps.  相似文献

Phlorotannins have often been considered to act as UV-protective compounds in zoospores of brown algae. However, only the absorption characteristics of zoospores under UV exposure have been determined and no data are available on the actual content of phlorotannins or on temperature–UV interactions. Therefore, we determined the absorbance spectra and the phlorotannin contents in zoospore suspensions of three Arctic species ( Saccharina latissima , Laminaria digitata , Alaria esculenta ), and in the media surrounding zoospores after exposure to different radiation (400–700, 320–700, 295–700 nm) and temperature (2–18°C) conditions for 8 h. Absorption typical of phlorotannins with a maximum at 276 nm was monitored in zoospore suspensions as well as in the media surrounding zoospores, but the results depended strongly on radiation treatments and on zoospore densities. Surprisingly, the content of UV-absorbing phlorotannins subsequent to different exposures did not change in any of the three species. The observed exceptional absorption properties could, therefore, not be related to phlorotannin contents. These findings are discussed in light of a strong phlorotannin investment from sporophytes during spore release and a minor UV-protective role of phlorotannins for zoospores of Arctic kelp species.  相似文献

Radiation damage can inter alia result in lipid peroxidation of macroalgal cell membranes. To prevent photo-oxidation within the cells, photoprotective substances such as phlorotannins are synthesized. In the present study, changes in total fatty acids (FA), FA composition and intra/extracellular phlorotannin contents were determined by gas chromatography and the Folin-Ciocalteu method to investigate the photoprotective potential of phlorotannins to prevent lipid peroxidation. Alaria esculenta juveniles (Phaeophyceae) were exposed over 20 days to high/low photosynthetically active radiation (PAR) in combination with UV radiation (UVR) in the treatments: PAB (low/high PAR + UV-B + UV-A), PA (low/high PAR + UV-A) or low/high PAR only. While extracellular phlorotannins increased after 10 days, intracellular phlorotannins increased with exposure time and PA and decreased under PAB. Interactive effects of time:radiation wavebands, time:PAR dose as well as radiation wavebands:PAR dose were observed. Low FA contents were detected in the PA and PAB treatments; interactive effects were observed between time:high PAR and PAB:high PAR. Total FA contents were correlated to extra/intracellular phlorotannin contents. Our results suggest that phlorotannins might play a role in intra/extracellular protection by absorption and oxidation processes. Changes in FA content/composition upon UVR and high PAR might be considered as an adaptive mechanism of the A. esculenta juveniles subjected to variations in solar irradiance.  相似文献

Studies were carried out on the effects induced by the main components of airborne particulate matter (APM) as soot, inorganic and cyclohexane soluble substances and by glass fiber filters on photodegradation of pyrene, benz(a)anthracene and benzo(a)pyrene exposed to UV and solar radiation. In both experimental models tested hydrocarbons showed a higher photochemical stability when absorbed on APM. Inorganic components of APM slightly enhance UV photodegradation. In real condition (outdoor exposure to solar radiation) PAH half lives generally showed a good linear correlation with mean solar radiation intensity; only degradation rate of benzo(a)pyrene on APM, exposed to sunlight was practically constant. Pyrene, in particular, showed a higher degradation rate when high ozone concentrations (0.2 ppm) occurred.  相似文献

The photochemical reaction of poly(ether ether ketone) (PEEK) sheets under tensile loads has been investigated. Two types of UV irradiation tests were carried out in a vacuum environment: with and without a cooling apparatus. Chemical structures, thermal properties, and mechanical properties were measured to clarify photo-deterioration. Chemical analysis based on Fourier Transform Infrared Spectrometry (FT-IR) and X-ray Photoelectron Spectroscopy (XPS) showed photochemical scission caused by UV exposure. Thermal properties, measured by Differential Scanning Calorimetry (DSC), indicated that a crosslinking reaction occurred during the radiation tests. Tensile properties of PEEK sheets after UV radiation clearly showed a tendency to embrittlement affected not only by crosslinking but also by the orientation of molecular chains resulting from the temperature rise of the specimens. Furthermore, applied tensile stress during exposure accelerated molecular scission and disturbed the crosslinking effects of the tensile properties.  相似文献

Diatoms have relatively high biomass in mid- to high-latitude oceans, which is also the most sensitive region to climate change. Photoautotrophs are thus predicted to become exposed to both higher temperatures and increased solar irradiance. In this study, we examined the consequences of such changes for the growth and photo-physiology of two diatoms by mimicking the scenarios that correspond to present day and that predicted for the end of this century. Elevated light induced higher rates of damage to photosystem II (PSII) that significantly reduced photochemical yields of both diatoms. Treatments including UV radiation induced ~ 50% inhibition of PSII under present PAR levels. Generally, warming alleviated UVR inhibition, resulting in higher photochemical yields, and faster recovery during dim light exposure. Therefore, concurrent increase of irradiance and temperature mitigated UV inhibition of PSII by 8–15%. The growth was stimulated by warming under PAR treatment, while less stimulation, or even decreased growth rates were found under the PAR + UVR treatment. Results suggest that ocean warming could fully offset the inhibition of high light on PSII. However, under the latter higher UVR stress scenario, the energetic expenditure required by the diatoms to repair damage could lead to their lower overall growth in future oceans.  相似文献

To assess the effects of UV radiation (280–400 nm) on development, oxidative damage and antioxidant defence in larvae of the tropical sea urchin Tripneustes gratilla, a field experiment was conducted at two depths in Aitutaki, Cook Islands (18.85°S, 159.75°E) in May 2008. Compared with field controls (larvae shielded from UV-R but exposed to VIS-radiation), UV-B exposure resulted in developmental abnormality and increases in oxidative damage to proteins (but not lipids) in embryos of T. gratilla held at 1 m depth. Results also indicated that larvae had the capacity to increase the activities of protective antioxidant enzymes when exposed to UV-B. The same trends in oxidative damage and antioxidant defence were observed for embryos held at 4 m, although the differences were smaller and more variable. In contrast to UV-B exposure, larvae exposed to UV-A only showed no significant increases in abnormality or oxidative damage to lipids and proteins compared with field controls. This was true at both experimental depths. Furthermore, exposure to UV-A did not cause a significant increase in the activities of antioxidants. This study indicates that oxidative stress is an important response of tropical sea urchin larvae to exposure to UV radiation.  相似文献

Measurements were conducted at San Ya, China (18.4°N, 109.7°E, altitude 18 m) to investigate the diurnal variation of ocular exposure to ultraviolet (UV) radiation. The experimental apparatus was composed of a manikin and a dual-detector spectrometer to simultaneously measure ocular and ambient UV data. The experimental apparatus was rotated clockwise to simulate three different types of exposure. When the manikin was facing into the sun, the ocular exposure to UV radiation on a summer day was bimodally distributed. The maximum ocular UV irradiance occurred at solar elevations of around 40° and 50° for UVA and UVB respectively. The spectral irradiances were measured at specific wavelength to obtain the ocular biologically effective UV (UV(BE) ) irradiances for photokeratitis, photoconjunctivitis and cataract, and the UV index (UVI) was calculated at the same time point for comparison. When the manikin faced the sun, the maximal ocular UV(BE) irradiance values were obtained at the solar elevation where the UVI value was 8. The results of this study showed that protection against ocular overexposure during outdoor activities should be taken not only at noon but also at other times.  相似文献

Certain phytochemicals, such as the stilbene, resveratrol (RES, found in red grapes and berries), and the triterpenoid, ursolic acid (UA, found in waxy berries and herbs such as rosemary and oregano), have antioxidant, anti‐inflammatory and antiproliferative effects. Two human‐derived cell lines, hTERT‐RPE with a nonmalignant phenotype derived from retinal pigment epithelium, and ATCC CRL‐11147 derived from a malignant skin melanoma, were used as in vitro models of photooxidative stress produced by exposure to the broadband output of a 150 W Hg vapor arc lamp at an irradiance of 19–26 mW cm?2. In untreated cells, UV–VIS broadband light exposure produced a loss of proliferative ability, an activation of NF‐κB and an increase in protein carbonyl adducts at 24 h postexposure. Pretreatment of the cells with RES or UA at 1–2 μm significantly reduced the amount of phosphorylated NF ‐κB at 24 h postexposure. RES pretreatment reduced the burden of light‐induced protein carbonyl adducts by up to 25% in exposed cells. UA treatment markedly increased the sensitivity of melanoma cells to UV radiation, while conferring some photoprotection to RPE cells. These observations indicate that phytochemicals modulate the cellular response to photochemical stress by interacting with specific cell‐signaling pathways.  相似文献

We examined the effects of ultraviolet (UV) radiation in combination with high levels of infrared (IR) radiation on the spectral transmittance of plastic filters. The biological action spectrum for damage to the human eye and skin changes dramatically in the 300-400 nm wavelength range. Cut-off filters used in this region to shape the spectrum of exposure sources are thus critical to the design of experiments which use broadband light sources. The changes in transmittance of three types of plastic filters were observed over an exposure period of 1000 h. One set of three filters was exposed mainly to UV radiation, while the other set was exposed to UV radiation plus IR radiation. Filters exposed to both UV and IR radiation showed spectral changes in their transmittance, while the filters exposed to UV only showed no measurable changes.  相似文献

Coral bleaching is the manifestation of the dysfunction of the symbiosis between scleractinian corals and dinoflagellates of the diverse genus Symbiodinium and is induced by elevated temperatures and high irradiance. We investigated the photophysiological response of two genetically distinct Symbiodinium subtypes within clade A upon exposure to elevated temperatures at two light intensities for 3 weeks. While both subtypes displayed a characteristic photoacclimation to high light (HL) (decrease in light-harvesting pigments, lower photochemical efficiency of photosystem II, increased xanthophyll pool sizes), the tolerance toward thermal stress clearly differed between the two subtypes. Symbiodinium Ax was highly susceptible to chronic photoinhibition at temperatures ≥30°C, which was exacerbated under HL conditions. A1 showed a capacity for photoacclimation and high thermal tolerance, which might be related to higher cellular concentrations of photoprotective xanthophylls and the low-molecular antioxidant glutathione (GSx) along with the dynamic regulation of these photoprotective pathways. Whereas HL conditions induced both accumulation of diatoxanthin and GSx, thermal stress further stimulated xanthophyll cycling, which might compensate for diminished amounts of GSx at elevated temperatures. Our results show that the two clade A subtypes clearly differ in their strategies to cope with thermal stress in combination with high irradiance.  相似文献

UV solar spectra have been measured, using a double-grating spectroradiometer, during population studies carried out across Europe for the EC Framework 7 funded ICEPURE project on the impact of climatic and environmental factors on personal UV radiation exposure and human health. Spectral field measurements have been conducted at ambient temperatures which varied between 11.5 and 33.5 °C. This temperature variation might affect instrument performance. The effect of ambient temperature was quantified and verified, and a model for temperature correction of spectral data is presented.  相似文献

Photoacclimation properties were investigated in two marine microalgae exposed to four ambient irradiance conditions: static photosynthetically active radiation (PAR: 400–700 nm), static PAR + UVR (280–700 nm), dynamic PAR and dynamic PAR + UVR. High light acclimated cultures of Thalassiosira weissflogii and Dunaliella tertiolecta were exposed outdoors for a maximum of 7 days. Dynamic irradiance was established by computer controlled vertical movement of 2 L bottles in a water filled basin. Immediate (<24 h), short-term (1–3 days) and long-term (4–7 days) photoacclimation was followed for antioxidants (superoxide dismutase, ascorbate peroxidase and glutathione cycling), growth and pigment pools. Changes in UVR sensitivity during photoacclimation were monitored by measuring UVR-induced inhibition of carbon assimilation under standardized UV conditions using an indoor solar simulator. Both species showed immediate antioxidant responses due to their transfer to the outdoor conditions. Furthermore, upon outdoor exposure, carbon assimilation and growth rates were reduced in both species compared with initial conditions; however, these effects were most pronounced in D. tertiolecta . Outdoor UV exposure did not alter antioxidant levels when compared with PAR-only controls in both species. In contrast, growth was significantly affected in the static UVR cultures, concurrent with significantly enhanced UVR resistance. We conclude that antioxidants play a minor role in the reinforcement of natural UVR resistance in T. weissflogii and D. tertiolecta .  相似文献

In vivo progressive effects of UV irradiation on the lens epithelium were studied using various histomorphological and biochemical parameters. Fifteen day old rat pups were exposed to 600 mW/m2 of radiation, including UV-A and UV-B, 12 h daily for 90, 120, 150 and 180 days. Biochemical parameters such as protein-bound and non-protein-bound sulfhydryl groups in both soluble and insoluble fractions and enzymes, which play an important role in combating the oxidative stress, were studied. Decreased cell density of lens epithelial cells (LEC) was observed in all three zones along with the decrease in the levels of soluble sulfhydryls (S-SH), glutathione reductase (GR), superoxide dismutase (SOD), glutathione peroxidase (GPx) and catalase (CAT). On the other hand, an increase in insoluble sulfhydryls was observed. Because of the decrease in S-SH and GR activities, the LEC became vulnerable to oxidative stress. Decreased activities of SOD, GPx and CAT suggest elevated oxidative stress. This effect of UV radiation may lead to cell death that may be responsible for the observed decrease in the cell density in all three zones of the lens epithelium.  相似文献

Chlorogenic acid is a natural potent antioxidant. It can be used in cosmetics formulations, but for this purpose its photochemical stability should be determined to ensure that the compound will not be degraded after UV radiation exposure. To evaluate this possibility, the concentration of a chlorogenic acid solution was determined by HPLC before and after UVA and UVB irradiation. The results indicate that chlorogenic acid is not degraded under UVA or UVB irradiation.  相似文献

UV sensitivity of the vegetative and reproductive tissues of two Antarctic brown macroalgae was compared. Photosynthesis as well as the content and localization of phenolic substances were determined. Responses to UV radiation were quantified as chlorophyll fluorescence (Fv/Fm). Ascoseira mirabilis showed high UV tolerance, while in Cystosphaera jacquinotii Fv/Fm decreased by 15–21%, the receptacles being more tolerant than the vegetative blades. The phlorotannin contents showed an opposite pattern: the soluble fraction dominated in C. jacquinotii while in A. mirabilis the insoluble fraction was more abundant. Soluble phlorotannins were higher in the reproductive than in vegetative tissues in both species. Images of tissue cross‐sections under violet‐blue light excitation confirmed a high allocation of phenolic compounds (as blue autofluorescence) in C. jacquinotii, both in reproductive and vegetative blades. The allocation and proportions of the soluble and insoluble phlorotannins could be related with the observed UV tolerance of the vegetative and reproductive tissues.  相似文献

Solar ultraviolet (UV) radiation follows people during their whole life. Exposure to UV radiation is vital but holds serious risks, too. The quantification of human UV exposure is a complex issue. UV exposure is directly related to incoming UV radiation as well as to a variety of factors such as the orientation of the exposed anatomical site with respect to the sun and the duration of exposure. The use of badge‐sensors allows assessing the UV exposure of differently oriented body sites. Such UV devices have been available for over 40 years, and a variety of measuring campaigns have been undertaken since then. This study provides an overview of those studies which reported measurements of the personal UV exposure (PE) during outdoor activities of people not related to their occupation. This overview is given chronologically to show the progress of knowledge in this research and is given with respect to different activities. Special focus is put on the ratio of personal exposure to ambient UV radiation. This ratio, when given as a function of solar elevation, allows estimating PE at any other location or date if ambient UV radiation is known.  相似文献

The aim of the present study is to evaluate the occurrence of oxidative stress in the cladoceran Daphnia longispina exposed to UV-A and UV-B radiation. The activity of antioxidant enzymes and lipid peroxidation markers is investigated and the protective action of ascorbic acid determined. Results show differences in the lethality radioinduced by UV-A and UV-B. Both UV-A and UV-B exposure cause an important increase in malonaldehyde (MDA) concentration and catalase activity. Ascorbic acid addition reduces the MDA concentration, indicating that the oxidative stress caused by either UV-A or UV-B radiation can be controlled by antioxidants. The increase of the antioxidant enzymes may be a response mechanism to oxidative stress.  相似文献
