It is critical to develop a cost-effective quantitative/semiquantitative assay for rapid diagnosis and on-site detection of toxic or harmful substances. Here, a naked-eye based semiquantitative immunochromatographic strip (NSI-strip) was developed, on which three test lines (TLs, TL-I, TL-II and TL-III) were dispensed on a nitrocellulose membrane to form the test zone. Similar as the traditional strip assay for small molecule, the NSI-strip assay was also based on the competitive theory, difference was that the analyte competed three times with the capture reagent for the limited number of antibody binding sites. After the assay, the number of TLs developed in the test zone was inversely proportional to the analyte concentration, thus analyte content levels could be determined by observing the appeared number of TLs. Taking aflatoxin B1 as the model analyte, visual detection limit of the NSI-strip was 0.06 ng mL−1 and threshold concentrations for TL-I–III were 0.125, 0.5, and 2.0 ng mL−1, respectively. Therefore, according to the appeared number of TLs, the following concentration ranges would be detectable by visual examination: 0–0.06 ng mL−1 (negative samples), and 0.06–0.125 ng mL−1, 0.125–0.5 ng mL−1, 0.5–2.0 ng mL−1 and >2.0 ng mL−1 (positive samples). That was to say, compared to traditional strips the NSI-strip could offer more parameter information of the target analyte content. In this way, the NSI-strip improved the qualitative presence/absence detection of traditional strips by measuring the content (range) of target analytes semiquantitatively.  相似文献

A rapid sandwich immunoassay (IA) with enhanced signal response for human fetuin A (HFA) was developed by modifying the surface of a KOH-treated polystyrene microtiter plate (MTP) with agarose and 3-aminopropyltriethoxysilane (APTES). The agarose-APTES complex binds covalently to the hydroxyl moiety of the MTP plate to serve as a binding platform for bioconjugation of EDC-activated anti-HFA antibody (Ab) via carbodiimide coupling. The one-step kinetics-based sandwich enzyme-linked immunosorbent assay (ELISA) enabled the detection of HFA in 30 min with a limit of detection (LOD) and a linear range of 0.02 ng mL−1 and 1–243 ng mL−1, respectively. It detected HFA spiked in diluted human whole blood and serum, and HFA in ethylenediaminetetraacetic acid (EDTA)-plasma of patients with high precision similar to that of conventional ELISA. The anti-HFA Ab-bound agarose-functionalized MTPs retained their functional activity after 6 weeks of storage in 0.1 M PBS, pH 7.4 at 4 °C.  相似文献

A simple dispersive solid-phase micro-extraction method based on CoFe2O4 nano-particles (NPs) functionalized with 8-hydroxyquinoline (8-HQ) with the aid of sodium dodecyl sulfate (SDS) was developed for separation of Al(III) ions from aqueous solutions. Al(III) ions are separated at pH 7 via complex formation with 8-HQ using the functionalized CoFe2O4 nano-particles sol solution as a dispersed solid-phase extractor. The separated analyte is directly quantified by a spectrofluorometric method at 370 nm excitation and 506 nm emission wavelengths. A comparison of the fluorescence of Al(III)–8-HQ complex in bulk solution and that of Al(III) ion interacted with 8-HQ/SDS/CoFe2O4 NPs revealed a nearly 5-fold improvement in intensity. The experimental factors influencing the separation and in situ monitoring of the analyte were optimized. Under these conditions, the calibration graph was linear in the range of 0.1–300 ng mL−1 with a correlation coefficient of 0.9986. The limit of detection and limit of quantification were 0.03 ng mL−1 and 0.10 ng mL−1, respectively. The inter-day and intra-day relative standard deviations for six replicate determinations of 150 ng mL−1 Al(III) ion were 2.8% and 1.7%, respectively. The method was successfully applied to direct determine Al(III) ion in various human serum and water samples.  相似文献

A rapid, simple, and sensitive headspace solid phase microextraction coupled to ion mobility spectrometry (HS-SPME-IMS) method is presented for analysis of the highly specific angiotensin-converting enzyme (ACE) inhibitor, captopril (CAP). Positive ion mobility spectra of CAP were acquired with an ion mobility spectrometer equipped with a corona discharge ionization source. Mass-to-mobility correlation equation was used to identify product ions. A dodecylsulfate-doped polypyrrole (PPy-DS) coating was used as a fiber for SPME. The results showed that PPy-DS based SPME fiber was suitable for successfully extracting CAP from human blood plasma and pharmaceutical samples. The HS-SPME-IMS method provided good repeatability (R.S.D.s < 4%) for aqueous and spiked plasma samples. The calibration graphs were linear in the range of 10-300 ng mL−1 (R2 > 0.99) and detection limits were 7.5 ng mL−1 for aqueous and 6.3 ng mL−1 for plasma blank samples. Finally, a standard addition calibration method was applied to HS-SPME-IMS technique for the analysis of blood plasma samples and tablets. Purpose method seemed to be suitable for the analysis of CAP in plasma samples as it is not time consuming (state total time from sample preparation to analysis), it required only small quantities of the sample, and no derivatization was required.  相似文献

A novel fluorescence turn-on detection method of human serum albumin (HSA) and bovine serum albumin (BSA) in aqueous solution is investigated using 2,4-dihydroxyl-3-iodo salicylaldehyde azine (DISA). Upon the addition of DISA to HSA/BSA solution, a fluorescence turn-on effect at 529 nm can be observed with a large stokes shift of ∼129 nm based on hydrophobic binding-mode between protein and dye. Under the optimal condition, the linear ranges of fluorescence intensity for HSA and BSA are 0.1-30 μg mL−1 with the relative correlation coefficient of R2 = 0.991 (n = 10) and 0.3-50 μg mL−1 with R2 = 0.997 (n = 10); and the detection limits for HSA and BSA based on IUPAC (CDL = 3Sb/m) are 20 ng mL−1 and 50 ng mL−1, respectively.  相似文献

Hemimicelles of tetradecanoate chemisorbed onto magnetic nanoparticles (MNPs) are here proposed as a sorbent for the single-step extraction and cleanup of bisphenol A (BPA) in soft drinks. The purpose of this work was to develop a simple, rapid and low-cost sample treatment suitable to assess the human exposure to BPA from this type of high consumption food. The nanoparticles were easily coated by mixing commercially available magnetite of 20–30 nm mean particle diameter with tetradecanoate at 85 °C for 30 min. The extraction/cleanup procedure involved stirring the samples (3 mL) with 200 mg of tetradecanoate-coated MNPs for 20 min, isolating the sorbent with a Nd–Fe–B magnet and eluting BPA with methanol. The extraction efficiency was not influenced by salt concentrations up to 1 M and pH values over the range 4–9. No cleanup of the extracts was needed, and the method proved matrix-independent. The extracts were analyzed by liquid chromatography, electrospray ionization tandem mass spectrometry. Quantitation was performed by internal standard calibration using BPA-13C12. The limit of quantitation obtained for the method, 0.03 ng mL−1, was below the usual range of concentrations reported for BPA in soft drinks (0.1–3.4 ng mL−1). The proposed method was successfully applied to the determination of BPA in different samples acquired from various supermarkets in southern Spain; the concentrations found ranged from 0.066 to 1.08 ng mL−1. Recoveries from samples spiked with 0.33 ng mL−1 of BPA ranged from 91% to 105% with relative standard deviations from 3% to 8%.  相似文献

An amplified electrochemical impedimetric aptasensor for ochratoxin A (OTA) was developed with picomolar sensitivity. A facile route to fabricate gold nanoparticles covalently bound reduced graphene oxide (AuNPs–rGO) resulted in a large number of well-dispersed AuNPs on graphene sheets with tremendous binding sites for DNA, since the single rGO sheet and each AuNP can be loaded with hundreds of DNA strands. An aptasensor with sandwich model was fabricated which involved thiolated capture DNA immobilized on a gold electrode to capture the aptamer, then the sensing interface was incubated with OTA at a desired concentration, followed by AuNPs–rGO functionalized reporter DNA hybridized with the residual aptamers. By exploiting the AuNPs–rGO as an excellent signal amplified platform, a single hybridization event between aptamer and reporter DNA was translated into more than 107 redox events, leading to a substantial increase in charge-transfer resistance (Rct) by 7∼ orders of magnitude compared with that of the free aptamer modified electrode. Such designed aptasensor showed a decreased response of Rct to the increase of OTA concentrations over a wide range of 1 pg mL−1–50 ng mL−1 and could detect extremely low OTA concentration, namely, 0.3 pg mL−1 or 0.74 pM, which was much lower than that of most other existed impedimetric aptasensors. The signal amplification platform presented here would provide a promising model for the aptamer-based detection with a direct impedimetric method.  相似文献

A novel microextraction technique, air-assisted liquid–liquid microextraction (AALLME), which is a new version of dispersive liquid–liquid microextraction (DLLME) method has been developed for extraction and preconcentration of phthalate esters, dimethyl phthalate (DMP), diethyl phthalate (DEP), di-iso-butyl phthalate (DIBP), di-n-butyl phthalate (DNBP), and di-2-ethylhexyl phthalate (DEHP), from aqueous samples prior to gas chromatography–flame ionization detection (GC–FID) analysis. In this method, much less volume of an organic solvent is used as extraction solvent in the absence of a disperser solvent. Fine organic droplets were formed by sucking and injecting of the mixture of aqueous sample solution and extraction solvent with a syringe for several times in a conical test tube. After extraction, phase separation was performed by centrifugation and the enriched analytes in the sedimented phase were determined by GC–FID. Under the optimum extraction conditions, the method showed low limits of detection and quantification between 0.12–1.15 and 0.85–4 ng mL−1, respectively. Enrichment factors (EFs) and extraction recoveries (ERs) were in the ranges of 889–1022 and 89–102%, respectively. The relative standard deviations (RSDs) for the extraction of 100 ng mL−1 and 500 ng mL−1 of each phthalate ester were less than 4% for intra-day (n = 6) and inter-days (n = 4) precision. Finally some aqueous samples were successfully analyzed using the proposed method and three analytes, DIBP, DNBP and DEHP, were determined in them at ng mL−1 level.  相似文献

Polyclonal antibody (PAb) with broad-specificity for O,O-diethyl organophosphorus pesticides (OPs) against a generic hapten, 4-(diethoxyphosphorothioyloxy)benzoic acid, was produced. The obtained PAb showed high sensitivity to seven commonly used O,O-diethyl OPs in a competitive indirect enzyme-linked immunosorbent assay (ciELISA) using a heterologous coating antigen, 4-(3-(diethoxyphosphorothioyloxy)phenylamino)-4-oxobutanoic acid. The 50% inhibition value (IC50) was 348 ng mL−1 for parathion, 13 ng mL−1 for coumaphos, 22 ng mL−1 for quinalphos, 35 ng mL−1 for triazophos, 751 ng mL−1 for phorate, 850 ng mL−1 for dichlofenthion, and 1301 ng mL−1 for phoxim. The limit of detection (LOD) met the ideal detection criteria of all the seven OP residues. A quantitative structure-activity relationship (QSAR) model was constructed to study the mechanism of antibody recognition using multiple linear regression analysis. The results indicated that the frontier-orbital energies (energy of the highest occupied molecular orbital, EHOMO, and energy of the lowest unoccupied molecular orbital, ELUMO) and hydrophobicity (log of the octanol/water partition coefficient, log P) were mainly responsible for the antibody recognition. The linear equation was log(IC50) = −63.274EHOMO + 15.985ELUMO + 0.556 log P − 25.015, with a determination coefficient (r2) of 0.908.  相似文献

Six polyclonal antisera to chloramphenicol (CAP) were successfully raised in camels, donkeys and goats. As a comparison of sensitivity, IC50 values ranged from 0.3 ng mL−1 to 5.5 ng mL−1 by enzyme-linked immunosorbent assay (ELISA) and from 0.7 ng mL−1 to 1.7 ng mL−1 by biosensor assay. The introduction of bovine milk extract improved the sensitivity of four of the antisera by ELISA and two by biosensor assay; a reduction in sensitivity of the remaining antisera ranged by a factor of 1.1-2.6. Porcine kidney extract reduced the sensitivity of all the antisera by a factor ranging from 1.1 to 7 by ELISA and a factor of 1.5 to 4 by biosensor. A low cross-reactivity with thiamphenicol (TAP) and florfenicol (FF) was displayed by antiserum G2 (1.2% and 18%, respectively) when a homologous ELISA assay format was employed. No cross-reactivity was displayed by any of the antisera when a homologous biosensor assay format was employed. Switching to a heterologous ELISA format prompted three of the antisera to display more significant cross-reactivity with TAP and FF (53% and 82%, respectively, using D1). The heterologous biosensor assay also increased the cross-reactivity of D1 for TAP and FF (56% and 129%, respectively) and of one other antiserum (G1) to a lesser degree. However, unlike the ELISA, the heterologous biosensor assay produced a substantial reduction in sensitivity (by a factor of 6 for D1).  相似文献

An immunoaffinity (IA) sorbent with antibody fragments was prepared for the analysis of opioid peptides by on-line immunoaffinity solid-phase extraction capillary electrophoresis–mass spectrometry (IA-SPE-CE–MS). The antibody fragmentation was evaluated by MALDI-TOF-MS. Fab′ fragments obtained from a polyclonal IgG antibody against Endomorphins 1 and 2 (End1 and End2) were covalently attached to succinimidyl silica particles to prepare the IA sorbent. An IA-SPE-CE–MS methodology was established analyzing standard solutions of End1 and End2 and acceptable repeatability, linearity ranges and LODs (0.5 and 5 ng mL−1, respectively) were obtained. The LOD of End1 was slightly better than that previously obtained using an IA sorbent with intact antibodies (1 ng mL−1). In human plasma samples, End1 and End2 could be detected at 1 and 50 ng mL−1, respectively, which meant an improvement of 100 and 2-fold with regard to the LODs using an IA sorbent with intact antibodies (100 ng mL−1).  相似文献

An orthogonal array design (OAD) was applied to optimize microwave-assisted derivatization (MAD) for analysis of trace amphetamine (AM) and methamphetamine (MA) by negative chemical ionization gas chromatography–mass spectrometry (NCI GC–MS). The 2,3,4,5,6-pentafluorobenzoyl chloride (PFBC) was used as a derivatization reagent. Experimental factors including solvent, microwave power, and irradiation time at four-levels were studied in 16 trials by OAD16 (44). The significance of these factors was investigated using analysis of variance (ANOVA) and percent contribution (PC). Solvent is statistically demonstrated a chief factor; microwave power and irradiation time are secondary factors. Under the optimum condition, calibration curve of AM is linear over a range from 0.01 to 100 ng mL−1 with correlation coefficient 0.9988, and MA from 0.1 to 1000 ng mL−1 with correlation coefficient 0.9951. The limit of detection (LOD) is 1.20 pg mL−1 for AM and 13.04 pg mL−1 for MA. An applicability of the method was tested by analyzing urine samples from amphetamine-type stimulants (ATS)-abusing suspects. Consequently, the OAD method not only optimizes the MAD condition for determination of trace AM and MA, but identifies the effects of factor solvent, microwave power and irradiation time on the MAD performance.  相似文献

A detailed study of iron (III)–citrate speciation in aqueous solution (θ = 25 °C, Ic = 0.7 mol L−1) was carried out by voltammetric and UV–vis spectrophotometric measurements and the obtained data were used for reconciled characterization of iron (III)–citrate complexes. Four different redox processes were registered in the voltammograms: at 0.1 V (pH = 5.5) which corresponded to the reduction of iron(III)–monocitrate species (Fe:cit = 1:1), at about −0.1 V (pH = 5.5) that was related to the reduction of FeL25−, FeL2H4− and FeL2H23− complexes, at −0.28 V (pH = 5.5) which corresponded to the reduction of polynuclear iron(III)–citrate complex(es), and at −0.4 V (pH = 7.5) which was probably a consequence of Fe(cit)2(OH)x species reduction. Reversible redox process at −0.1 V allowed for the determination of iron(III)–citrate species and their stability constants by analyzing Ep vs. pH and Ep vs. [L4−] dependence. The UV–vis spectra recorded at varied pH revealed four different spectrally active species: FeLH (log β = 25.69), FeL2H23− (log β = 48.06), FeL2H4− (log β = 44.60), and FeL25− (log β = 38.85). The stability constants obtained by spectrophotometry were in agreement with those determined electrochemically. The UV–vis spectra recorded at various citrate concentrations (pH = 2.0) supported the results of spectrophotometric–potentiometric titration.  相似文献

A novel bare-eye based one-step signal amplified semi-quantitative immunochromatographic assay (SAS-ICA) was developed for detection of the pesticide imidacloprid. This method was based on competitive immunoreactions. Signal amplification was achieved by dual labeling of the test lines (TLs) on the strip using high affinity nanogold-biotinylated anti-imidacloprid mAb (BAb) and nanogold-streptavidin (Sa) probes. The relative color intensities of three TLs (TL-I, TL-II and TL-III) on a nitrocellulose (NC) membrane were used for direct visual analysis of the SAS-ICA strips, and could be used for semi-quantitation of analyte concentrations by observing what TLs disappeared in the amplification zone. Under optimized conditions, the following imidacloprid concentration ranges would be detected by visual examination of the SAS-ICA strip: 0–5 ng mL−1 (negative samples), and 5–25 ng mL−1, 25–250 ng mL−1, 250–1000 ng mL−1 and >1000 ng mL−1 (positive samples). The sensitivity (the visual detection limit (VDL) of TL-III) and semi-quantitative analytical capacity (when TL-III disappeared completely) of the SAS-ICA strip were 10-fold and 160-fold higher than those of traditional ICA, respectively. The developed SAS-ICA strip was applied to the analysis of spiked and authentic contaminated Chinese cabbage samples in the laboratory and under field conditions, and the results were validated by high-performance liquid chromatography (HPLC). This process could be adopted as a potential generous technique for all ICA-based detection methods.  相似文献

Orthogonal array design (OAD) was utilized for the first time to optimize the experimental conditions of ultrasound-assisted emulsification–microextraction (USAEME) for determining chlorinated phenoxyacetic acids (CPAs) in river water samples. The use of ultrasound facilitates the mass transfer of CPAs from an aqueous phase into a water-immiscible organic extraction solvent (dichloromethane, DCM) without adding dispersive solvent to form numerous microdroplets. The water-immiscible extractant was collected by centrifugation, dried under low pressure, reconstituted in methanol–water mixture (1:1), and injected into a HPLC system for the determination of CPAs. The linear range was 2–1000 ng mL−1 (2, 5, 10, 50, 200, 500 and 1000 ng mL−1) for each analyte and the relative standard deviations of CPAs among the seven different concentrations were in the range of 1.5–17.0% (n = 3). The detection limits (signal-to-noise ratio of 3) of CPAs ranged from 0.67 to 1.50 ng mL−1. The ranges of intra-day precision (n = 3) for CPAs at the levels of 5 and 200 ng mL−1 were 3.6–11.9% and 5.3–9.5%, respectively. The range of inter-day precision (n = 3) at 5 and 200 ng mL−1 were 1.4–7.7% and 8.5–12.2%, respectively. The applicability of USAEME for environmental analysis was demonstrated by determining CPAs in river water. The recoveries of CPAs from five-spiked river water samples at 10 and 200 ng mL−1 were 96.3–112.5% and 94.8–109.4%, respectively. The maximum contaminant level (MCL) of 2,4-D in drinking water and the tolerance of residues in food for p-CPA are 70 and 200 μg L−1, respectively, according to the US EPA regulations. These contaminant levels fall in the linear range investigated in this study. In addition, this USAEME method provided detection limits lower than their contaminant levels, which made USAEME an effective sample preparation method for determining organic environmental contaminants, such as CPAs, in river water samples with little consumption of organic solvent.  相似文献

A novel ultrasound-assisted surfactant-enhanced emulsification microextraction (UASEME) coupled with high performance liquid chromatography-diode array detection has been developed for the extraction and determination of six carbamate pesticides (metolcarb, carbofuran, carbaryl, pirimicarb, isoprocarb and diethofencarb) in water samples. In the UASEME technique, Tween 20 was used as emulsifier, and chlorobenzene and chloroform were used as dual extraction solvent without using any organic dispersive solvent that is normally required in the previously described common dispersive liquid–liquid microextraction method. Parameters that affect the extraction efficiency, such as the kind and volume of the extraction solvent, the type and concentration of the surfactant, ultrasound emulsification time and salt addition, were investigated and optimized for the method. Under the optimum conditions, the enrichment factors were in the range between 170 and 246. The limits of detection of the method were 0.1–0.3 ng mL−1 and the limits of quantification were between 0.3 and 0.9 ng mL−1, depending on the compounds. The linearity of the method was obtained in the range of 0.3–200 ng mL−1 for metolcarb, carbaryl, pirimicarb, and diethofencarb, 0.6–200 ng mL−1 for carbofuran, and 0.9–200 ng mL−1 for isoprocarb, with the correlation coefficients (r) ranging from 0.9982 to 0.9998. The relative standard deviations varied from 3.2 to 4.8% (n = 5). The recoveries of the method for the six carbamates from water samples at spiking levels of 1.0, 10.0, 50.0 and 100.0 ng mL−1 were ranged from 81.0 to 97.5%. The proposed UASEME technique has demonstrated to be simple, practical and environmentally friendly for the determination of carbamates residues in river, reservoir and well water samples.  相似文献

CeO2-deposited mesoporous silica nanoparticles were synthesized as a probe to determine carcinoembryonic antigen (CEA) in serum by inductively coupled plasma-mass spectrometry (ICP-MS). The prepared mesoporous nanoparticles were modified and tagged to the target for sandwich-type immunoassay. Fe3O4 magnetic nanoparticles (MNPs) were also synthesized and immobilized with antibody to extract the target biomarker. The calibration curve of the synthesized CeO2-deposited silica nanoparticles, which was plotted by the signal ratio of 140Ce/57Fe measured by ICP-MS vs. the concentration of CEA, showed excellent linearity and sensitivity owing to the signal amplification and low spectral interference. Under optimal conditions, the sandwich-type analytical method was applied to determine CEA in serum spiked in the range of 0.001–5 ng mL−1 and showed a limit of detection of 0.36 ng mL−1. Since the deposited CeO2 in the mesoporous silica layer can be substituted by other metal compounds, various kinds of metal-deposited nanoparticles can be prepared as probe materials for multiplex detection in bioanalysis.  相似文献

A new method was developed for determination of micro amounts of nucleic acids based on near-infrared (near-IR) fluorescence recovery, employing a two-reagent system which is composed of an anionic tetracarboxy aluminum phthalocyanine (AlC4Pc) and a cationic tetra-N-hexadecylpyridiniumyl porphyrin (TC16PyP). The fluorescence of the AlC4Pc, with the maximum emission wavelength at 701 nm, could be quenched by TC16PyP at its proper concentration, but recovered by adding nucleic acids. Under optimal conditions, the recovered fluorescence is proportional to the concentration of nucleic acids. The calibration graphs are linear over the range of 1-200 ng mL−1 for fish sperm DNA (FS DNA) and 2-400 ng mL−1 for calf thymus DNA (CT DNA). The corresponding detection limits are 0.59 ng mL−1 for FS DNA and 0.82 ng mL−1 for CT DNA, respectively. Four synthetic and three real nucleic acid samples were determined with satisfactory results.  相似文献

A highly sensitive method was developed for the identification and quantification of fatty alcohols in biological tissues. In the presence of pyridine-d0 and triflic anhydride (Tf2O), fatty alcohols were converted into permanently charged N-alkylpyridinium ions. Stable isotope-labeled derivatives were generated by pyridine-d5 and added as internal standard (IS). The mixture was analyzed by liquid chromatography coupled to positive electrospray ionization tandem mass spectrometry (LC–ESI-MS/MS). This method was optimized and validated in terms of reaction time, derivatization efficiency, stability, desalting, and ion suppression effect. Besides, fatty alcohols exhibited good linear relationship (r2 > 0.993) over the concentration range of 10 ng mL−1–1 μg mL−1. The limits of detection (LODs) were lowered from previously reported 0.1 ng mL−1 to 0.25 pg mL−1. Precision (RSD% < 15.6%), accuracy (93.0–107.2%), matrix effect, and recovery (in thyroid tissues) were validated as well. A simple extraction method for the analysis of PGE2 and PGF in gonad samples from Atlantic cod and further quantification by using liquid chromatography–tandem mass spectrometry is proposed. The evaluation of the best solvent extraction conditions and the analytical performance parameters are reported. The method was highly selective for both prostaglandins and the calibration curves, based on the internal standard method, were linear between 5 and 1000 ng mL−1 for PGE2 and PGF, with limits of detection of 1 ng mL−1 and 1.5 ng mL−1 and recovery values of 99.999 ± 0.002 and 99.967 ± 0.023 respectively. The homogenization of samples using liquid nitrogen combined with the developed extraction protocol can be implemented in different types of biological tissues.  相似文献
