A three step, convergent synthesis of 4-aryl-3-pyrrolin-2-ones from a tetramic acid has been developed. The key transformation utilized a Suzuki-Miyaura cross-coupling reaction between a 4-tosyloxy-3-pyrrolin-2-one and an arylboronic acid. This work also provides access to 4-arylpyrrolidin-2-ones, cyclic analogs of γ-aminobutyric acid (GABA). Hydrogenation of 4-(4′-chlorophenyl)-3-pyrrolin-2-one proceeded smoothly to give baclofen lactam.  相似文献

trans-4-Aryl-3-chloro-1-(2-chloroethyl)azetidin-2-ones, prepared through cyclocondensation of chloroketene and the appropriate imines in a diastereoselective way, were unexpectedly transformed into 3-aryl-2-(ethylamino)propan-1-ols using LiAlH(4) in THF under reflux. A stepwise analysis showed that the initially formed 1-(1-aryl-2-chloro-3-hydroxypropyl)aziridines were converted into trans-2-aryl-3-(hydroxymethyl)aziridines, most probably via N-spiro bis-aziridinium intermediates, which were subsequently prone to undergo ring opening by LiAlH(4) to afford 3-aryl-2-(ethylamino)propan-1-ols.  相似文献

The reactivity of 4-aryl-1-(2-chloroethyl)azetidin-2-ones and 4-aryl-1-(3-bromopropyl)azetidin-2-ones with regard to lithium aluminium hydride has been evaluated for the first time. 4-Aryl-1-(2-chloroethyl)azetidin-2-ones were transformed into novel 1-(1-aryl-3-hydroxypropyl)aziridines through an unprecedented conversion of beta-lactams into 2,3-unsubstituted aziridine derivatives. Unexpectedly, 4-aryl-1-(3-bromopropyl)azetidin-2-ones underwent dehalogenation towards 3-aryl-3-(N-propylamino)propan-1-ols upon treatment with LiAlH(4). 1-(1-Aryl-3-hydroxypropyl)aziridines were further elaborated by means of ring opening reactions using benzyl bromide in acetonitrile towards 3-aryl-3-[N-benzyl-N-(2-bromoethyl)amino]propan-1-ols and using aluminium(iii) chloride in diethyl ether, affording 3-aryl-3-[N-(2-chloroethyl)amino]propan-1-ols.  相似文献

Depending on the reaction condition, 1-(4-aminosulfonylphenyl)-5-aryl-4-aroyl-3-hydroxy-3-pyrrolin-2-ones reacted with aromatic amines to yield 3-arylamino-3-pyrrolin-2-ones or 4-[aryl (arylamino) methylene]tetrahydropyrrole-2,3-diones. Reactions of 1-(4-aminosulfonylphenyl)-5-aryl-4-aroyl-3-hydroxy-3-pyrrolin-2-ones with hydrazine hydrate afforded pyrrolo[3,4-c]pyrazol-6-ones.  相似文献

Aryl hydrazines (1) possessing an electron-withdrawing substituent on the phenyl ring, on treatment with diethyl (ethoxymethylene)malonate in ethanolic sodium ethoxide solution, have directly afforded 2-aryl-4-ethoxy-carbonyl-3-pyrazolin-5-ones (4). The reactions of 4-isopropylidene-1-aryl-3-methyl-2-pyrazolin-5-ones 4a-d were investigated under a variety of conditions. In the presence of thiols or piperidine, 4a-d failed to yield conjugate addition products, presumably due to the steric bulk provided by the two methyl substituents of the isopropylidene side chain. Reaction of 4a-d with hydrazine derivatives gave the 1-aryl-3-methyl-2-pyrazolin-5-ones 3a-d and isopropyl-hydrazones. Treatment of 4a with potassium cyanide yielded a stable conjugate addition product which exists as a mixture of tautomers in different solvents. Also, oxidation of 4a with hydrogen peroxide gave a spiroepoxide 22 , while m-chloroperbenzoic acid oxidation afforded both the spiroepoxide 22 , and a small quantity of a hydroxyspiroepoxide 23.  相似文献

A number of new 3,3-dimethoxy-2-aryl-2,3-dihydro-1-oxacyclopenta[l]phenanthren-2-ols were synthesized by the base-catalyzed reaction between phenanthrenequinone and 4-substituted acetophenones in methanol. These furanols are unstable and are easily converted to stable 3-methoxy-3-aryl-3H-1-oxacyclopenta[l]phenathren-2-ones in excellent yields. The furanols are converted to 2-aryl-1-oxacyclopenta[l]phenanthren-3-ones on treatment with acids. Plausible mechanisms for some of these new transformations are proposed.  相似文献

Palladium-catalyzed Suzuki cross-coupling of 2-aryl-4-chloro-3-iodoquinolines with excess arylboronic acids (2.5 equiv.) in the presence of tricyclohexylphosphine afforded the 2,3,4-triarylquinolines in one-pot operation. The incipient 2,3-diaryl-4-chloroquinolines were also prepared and transformed to the primary 4-amino-2,3-diarylquinolines and 2,3-diarylquinolin-4(1H)-ones.  相似文献

4-Acetyl-1-alkoxyaryl-5-aryl-3-hydroxy-2,5-dihydro-1H-pyrrol-2-ones reacted with amines to give 1-alkoxyaryl-5-aryl-4-(1-R-aminoethylidene)pyrrolidine-2,3-diones. Reactions of amines with 4-benzoyl-substituted analogs involve nucleophilic attack on the C3 atom in the heteroring to produce the corresponding 3-R-amino-1-alkoxyaryl-5-aryl-4-benzoyl-2,5-dihydro-1H-pyrrol-2-ones. Reactions of the title compounds with hydrazine hydrate, regardless on the substituent on C4, afforded 4-aryl-3-methyl(phenyl)-5-[2(4)-methoxyphenyl]-4,6-dihydropyrrolo[3,4-c]pyrazol-6-ones.  相似文献

The electroreduction of chiral aromatic alpha-imino esters prepared from (S)-alpha-amino acids, such as (S)-valine, (S)-leucine, and (S)-phenylalanine, in the presence of chlorotrimethylsilane and triethylamine afforded four-membered cyclized products, mixed ketals of cis-2,4-disubstituted azetidine-3-ones, stereospecifically (>99% de, 85-99% ee). The best result of the electroreductive cyclization was obtained using Bu(4)NClO(4) as a supporting electrolyte and a Pt cathode. The absolute stereochemistry of the obtained single stereoisomers was confirmed to be 2R,3R,4S by X-ray crystallography. Calculations for the transition states of the cyclization support the stereospecific formation of the (2R,3R,4S)-isomers.  相似文献

1-Aryl-3-piperidino(morpholino)-2-phenylpropan-1-ols were synthesized by reduction of the corresponding 1-aryl-3-piperidino(morpholino)-2-phenylpropan-1-ones with lithium tetrahydridoaluminate, and the products were separated into erythro and threo isomers by fractional crystallization. Their relative configuration and diastereoisomeric purity was determined by 1H NMR spectroscopy.  相似文献

3-Aroyl-6-bromochromen-2-ones and 2-benzoylbenzo[f]chromen-3-one reacted with zinc enolates derived from 1-aryl-2-bromoalkanones and 1-aryl-2-bromo-2-phenylethanone to give, respectively, 4-(1-alkyl-2-aryl-2-oxoethyl)-3-aroyl-6-bromochroman-2-ones and 1-(2-aroyl-1-methyl-2-oxoethyl)-2-benzoyl-1,2-dihydrobenzo[f]chromen-3-ones as a single stereoisomer. Treatment with acetic anhydride of the intermediate product obtained from 3-benzoyl-6-bromochromen-2-one and [1-(4-chlorophenyl)-2-phenylethen-1-yloxy]-zinc(II) bromide resulted in the formation of 3-(1-acetoxy-1-phenylmethylidene)-6-bromo-4-[2-(4-chlorophenyl)-2-oxo-1-phenylethyl]chroman-2-one.  相似文献

Halocyclocarbamation of benzyl N-(1-phenyl-3-butenyl)carbamates afforded 6-functionalized 4-aryl-1,3-oxazinan-2-ones with moderate to excellent diastereoselectivity depending on the N-substituent. Importantly, in contrast to reported cyclocarbamations of homoallylic carbamates, which are generally trans-diastereoselective, cyclization of N-unsubstituted Cbz-protected homoallylamines led to cis-1,3-oxazinan-2-ones, predominantly. The use of N-benzylated and in situ prepared N-silylated derivatives resulted however in trans-selectivity. Transition states are proposed to explain the stereochemical influence of the N-substituent on the cyclocarbamations. The functionalized 1,3-oxazinan-2-ones could be further elaborated towards biologically or synthetically important 6-arylpiperidine-2,4-diones and 3-aryl-1,3-aminoalcohols.  相似文献

One-pot synthesis of 1-([6-bromo-2-hydroxy-naphthalen-1-yl]-aryl-phenyl)methyl)-3-chloro-4-(aryl-phenyl)azetidin-2-ones has been reported in the present research work via Staudinger [2 + 2] ketene-imine cycloaddition reaction pathway. The reaction of 1-((Benzylideneamino)(aryl)methyl)-6-bromo-naphthalen-2-ols with chloroacetic acid and triethylamine afforded 1-([6-bromo-2-hydroxynaphthalen-1-yl]aryl-phenyl)methyl)-3-chloro-4-(aryl-phenyl)azetidin-2-ones. For the structural elucidation of series of compounds, different analytical and spectroscopic techniques such as elemental analysis, IR spectra, 1H-NMR spectra and mass spectra were used. All the newly synthesized compounds were tested for their anti-bacterial activity studies. It revealed that some of the compounds possesses moderate to good activities as compared to standard drugs. The widest spectrum of anti-bacterial activities against both gram-positive and gram-negative bacterial strains among the examined compounds possessed having more hydroxyl group along with β-lactam ring compared to other substituted azetidinones.  相似文献

Treatment of 1-chloro-3-phenylsulfonyl-2-propanone (2) with arenediazonium chlorides gave 1-arylhydrazono-3-chloro-1-phenylsulfonyl-2-propanones 4 , which were cyclized in the presence of sodium acetate to 1-aryl-3-phenylsulfonylpyrazol-4-ols 5B. Reactions of aryl trifluoromethyl ketone imines with 2-sulfanylacetic and 3-sulfanylpropanoic acids afforded 2-aryl-2-trifluoromethyl-1,3-thiazolidin-4-ones and 2-aryl-2-trifluoromethyltetrahydro-4H-1,3-thiazin-4-ones, respectively. Their subsequent oxidation with hydrogen peroxide gave the corresponding 2-aryl-2-trifluoromethyl-1,3-thiazolidin-4-one 1-oxides and 2-aryl-2-trifluoromethyltetrahydro-4H-1,3-thiazin-4-one 1-oxides and 1,1-dioxides.  相似文献
