A novel method for the preparation of amides and phthalimides has been developed. The process involves a palladium catalyzed aminocarbonylation of an aryl halide, using a carbodiimide and formic acid as the carbonyl source. Experimental data suggest that the mechanistic pathway for this process involves in‐situ generation of carbon monoxide from the reaction of formic acid with a carbodiimide in the presence of a palladium catalyst. The method can be used to produce a variety of amides and N‐substituted phthalimides efficiently.  相似文献

Transition‐metal‐catalyzed hydroamination reactions are sustainable and atom‐economical C? N bond‐forming processes. Although remarkable progress has been made in the inter‐ and intramolecular amination of olefins and 1,3‐dienes, related intermolecular reactions of amides are still much less known. Control of the regioselectivity without analogous telomerization is the particular challenge in the catalytic hydroamidation of alkenes and 1,3‐dienes. Herein, we report a general protocol for the hydroamidation of electron‐deficient N‐heterocyclic amides and sulfonamides with 1,3‐dienes and vinyl pyridines in the presence of a catalyst derived from [{Pd(π‐cinnamyl)Cl}2] and ligand L7 or L10 . The reactions proceeded in good to excellent yield with high regioselectivity. The practical utility of our method is demonstrated by the hydroamidation of functionalized biologically active substrates. The high regioselectivity for linear amide products makes the procedure useful for the synthesis of a variety of allylic amides.  相似文献

A new type of chiral sulfonium salts that are characterized by a bicyclic system has been designed and synthesized from α‐amino acids. Their corresponding ylides, which were prepared by basic treatment of the sulfonium salts, reacted smoothly with a broad array of simple and chiral aldehydes to provide trans‐epoxy amides in reasonable to very good yields and excellent stereoselectivities (>98 %). The obtained epoxy amides were found to be useful as synthetic building blocks. Thus, they were reduced into their corresponding epoxy alcohols and subjected to oxirane‐ring‐opening reactions with different types of nucleophiles.  相似文献

Palladium complexes incorporating chiral N‐heterocyclic carbene (NHC) ligands catalyze the asymmetric intramolecular α‐arylation of amides producing 3,3‐disubstituted oxindoles. Comprehensive DFT studies have been performed to gain insight into the mechanism of this transformation. Oxidative addition is shown to be rate‐determining and reductive elimination to be enantioselectivity‐determining. The synthesis of seven new NHC ligands is detailed and their performance is compared. One of them, L8 , containing a tBu and a 1‐naphthyl group at the stereogenic centre, proved superior and was very efficient in the asymmetric synthesis of fifteen new spiro‐oxindoles and three azaspiro‐oxindoles often in high yields (up to 99 %) and enantioselectivities (up to 97 % ee; ee=enantiomeric excess). Three palladacycle intermediates resulting from the oxidative addition of [Pd(NHC)] into the aryl halide bond were isolated and structurally characterized (X‐ray). Using these intermediates as catalysts showed alkene additives to play an important role in increasing turnover number and frequency.  相似文献

A polystyrene‐cross‐linking tricyclohexylphosphine (PS‐TCP) was synthesized through radical emulsion polymerization of 4‐tert‐butylstyrene as a monomer and tris(trans‐4‐styrylcyclohexyl)phosphine as a threefold cross‐linker. The PS‐TCP showed enhanced ligand performance compared to the corresponding polystyrene‐triphenylphosphine hybrid PS‐TPP and tricyclohexylphosphine in Pd‐catalyzed Suzuki–Miyaura and Buchwald–Hartwig reactions of aryl chlorides.  相似文献

Chagas disease, a chronic and silent disease caused by Trypanosoma cruzi, is currently a global public health problem. The treatment of this neglected disease relies on benznidazole and nifurtimox, two nitroheterocyclic drugs that show limited efficacy and severe side effects. The failure of potential drug candidates in Chagas disease clinical trials highlighted the urgent need to identify new effective chemical entities and more predictive tools to improve translational success in the drug development pipeline. In this study, we designed a small library of pyrazole derivatives (44 analogs) based on a hit compound, previously identified as a T. cruzi cysteine protease inhibitor. The in vitro phenotypic screening revealed compounds 3g, 3j, and 3m as promising candidates, with IC50 values of 6.09 ± 0.52, 2.75 ± 0.62, and 3.58 ± 0.25 µM, respectively, against intracellular amastigotes. All pyrazole derivatives have good oral bioavailability prediction. The structure–activity relationship (SAR) analysis revealed increased potency of 1-aryl-1H-pyrazole-imidazoline derivatives with the Br, Cl, and methyl substituents in the para-position. The 3m compound stands out for its trypanocidal efficacy in 3D microtissue, which mimics tissue microarchitecture and physiology, and abolishment of parasite recrudescence in vitro. Our findings encourage the progression of the promising candidate for preclinical in vivo studies.  相似文献

Novel and highly soluble hybrid conjugated organic oligomers consisting of oligodiacetylene and thiophene units have been synthesized in high purity through iterative and divergent approaches based on a sequence of Sonogashira reactions. The series of thiophene‐containing oligodiacetylenes (ThODAs) and homocoupled ThODAs (HThODAs) show—both in solution and in the solid state—a strong optical absorption, which is progressively red shifted with increasing chain length. The linear correlation of the absorption maximum (λAmax) with the inverse of conjugation length (CL=number of double and triple bonds) shows that the effective conjugation length of this system is extended up to at least CL=20. Furthermore, absorption measurements of dropcast thin films display not only a bathochromic shift of the absorption maxima but also a higher wavelength absorption, which is attributed to increased π–π interactions. The wavelength of the maximum fluorescence emission (λEmax) also increases with CL, and emission is maximal for oligomers with CL=7–12 (fluorescence quantum yield ΦF=~0.2). Both longer and shorter oligomers display marginal emission. The calculated Stokes shifts of these planar materials are relatively large (0.4 eV) for all oligomers, and likely due to excitation to the S2 state, thus suggesting that the presence of enyne moieties dominates the ordering of the lowest excited states. The fluorescence lifetimes (τF) are short (τF,max=?1 ns) and closely follow the tendency obtained for the fluorescence quantum yield. The anisotropy lifetimes show a near‐linear increase with CL, in line with highly rigid oligomers.  相似文献

Two multi‐walled carbon nanotube (MWCNT)‐based nanohybrids, MWCNT–ZnTPP and MWCNT–TPP (TPP=5‐[4‐{2‐(4‐formylphenoxy)‐ ethyloxy}phenyl]‐10,15,20‐triphenylporphyrin, ZnTPP=5‐[4‐{(4‐formylphenyl)ethynyl}phenyl]‐10,15,20‐triphenylporphinatozinc(II)), were prepared directly from pristine MWCNTs through 1,3‐dipolar cycloaddition reactions. Covalent attachment of the porphyrins to the surfaces of the MWCNTs was confirmed by Fourier transform infrared spectroscopy, ultraviolet/visible absorption, fluorescence, Raman, and X‐ray photoelectron spectroscopy, elemental analysis, transmission electron microscopy, and thermogravimetric analysis. Attachment of the porphyrin moieties to the surface of the MWCNTs significantly improves the solubility and ease of processing of these MWCNT–porphyrin composite materials. Z‐scan studies reveal that these MWCNT–porphyrin nanohybrids exhibit enhanced nonlinear optical properties under both nanosecond and picosecond laser pulses at λ=532 nm in comparison with free MWCNTs and the free porphyrin chromophores, whereas superior optical limiting performance was displayed by MWCNT–porphyrin composite materials rather than MWCNTs/ZnTPP and MWCNTs/TPP blends, which is consistent with a remarkable accumulation effect as a result of the covalent linkage between the porphyrin and the MWCNTs.  相似文献

Covalent triazine frameworks (CTFs) with polypyridyl ligands are very promising supports to anchor photocatalytic complexes. Herein, we investigate the photophysical properties of a series of ligands which vary by the extent of the aromatic system, the nitrogen content and their topologies to aid in selecting interesting building blocks for CTFs. Interestingly, some linkers have a rotational degree of freedom, allowing both a trans and cis structure, where only the latter allows anchoring. Therefore, the influence of the dihedral angle on the UV-Vis spectrum is studied. The photophysical properties are investigated by a combined computational and experimental study. Theoretically, both static and molecular dynamics simulations are performed to deduce ground- and excited state properties based on density functional theory (DFT) and time-dependent DFT. The position of the main absorption peak shifts towards higher wavelengths for an increased size of the π-system and a higher π-electron deficiency. We found that the position of the main absorption peak among the different ligands studied in this work can amount to 271 nm; which has a significant impact on the photophysical properties of the ligands. This broad range of shifts allows modulation of the electronic structure by varying the ligands and may help in a rational design of efficient photocatalysts.  相似文献

Difunctional magnesium, zinc, and tin reagents M?C6H4?O?C6H4?M (M = MgBr, ZnCl, SnBu3) in the presence of palladium or nickel catalysts undergo cross-coupling polymerizations with aromatic, heteroaromatic, benzylic, and allylic dihalides to give oligomeric and polymeric materials. Tin reagents lead to products of higher molecular weight than Mg and Zn reagents. The reaction is sensitive to the solvent and enhanced by magnesium halides. Increased reaction temperatures lead only to moderate increases in the degree of polymerization and are limited by catalyst decomposition above 200°C. The new poly(ether ketone) and poly(ether sulfone) type polymers prepared show high thermal stability. In contrast to conventional poly(ether sulfones)s, the biphenyl-based sulfone polymers reported here are crystalline. © 1992 John Wiley & Sons, Inc.  相似文献

Using a flow microreactor system, carbamoyllithium compounds were successfully generated and used for reactions with electrophiles to give various amides, including α‐ketoamides. The present method could be applied to the three‐component synthesis of functionalized α‐ketoamides using a carbamoyllithium compound, methyl chloroformate, and a functionalized organolithium reagent.  相似文献

The linear optical (LO) and nonlinear optical (NLO) properties of a series of isocyanurates functionalized by donor arms at the periphery are reported herein. These octupolar derivatives were obtained in a straightforward way from commercial isocyanate derivatives and were fully characterized. Although several of these compounds are known, those that exhibited the largest NLO activities are all new compounds. In terms of second-order activity, several of these derivatives exhibit remarkable activity/transparency tradeoffs. In terms of third-order activity, the longer derivatives with the stronger donor groups (X=NH(2) , NMe(2) , or NPh(2) ) were shown to possess significant two-photon absorption cross sections. These strongly luminescent derivatives exhibit two-photon absorption cross sections up to 410?GM. DFT computations were also conducted to unravel their electronic structures and to rationalize their NLO properties. To our knowledge, the present study is the first concerned with the nonlinear optical properties of these original cyclotrimers.  相似文献

The reaction of allyl ethyl carbonates with isocyanides in the presence of a catalytic amount of Pd(OAc)2 provided ketenimines through β‐hydride elimination of the allyl imidoylpalladium intermediates. The insertion of the isocyanide into the π‐allyl Pd complex proceeded via an unusual η1‐allyl Pd species. The resulting ketenimines were hydrolyzed to β,γ‐unsaturated carboxamides during purification by flash column chromatography on silica gel or converted in situ into 1,5‐disubstituted tetrazoles by [3+2] cycloaddition with hydrazoic acid or trimethylsilyl azide.  相似文献
