A membrane-based chiral separation system for the separation of racemic tryptophan solutions is developed by the covalently binding beta-cyclodextrin onto the surface of commercial cellulose membranes. The immobilization process is monitored by XPS. AFM demonstrates the evolutionary transition of membrane surface morphology before and after the CD immobilization. Due to their different complexation with immobilized CD, dialysis transport experiments show d-tryptophan preferential permeability through the immobilized CD membranes, and the enantioselectivity is 1.10. A model based on the existence of a thin chiral solution layer of amino acid at the interface between the feed solution and the membrane has been proposed. This chiral separation model has been verified using the chiral separation results of racemic amino acids and binding constants of amino acids with CD. The effect of membrane's pore size on enantioselectivity has also been investigated. The immobilized CD membrane, having MWCO 1000, exhibits the highest enantioselectivity to the racemic tryptophan solution.  相似文献

Twelve basic analytes, including ephedrine and its structurally related compounds, were used to study the influence of capillary temperature on enantioselectivity in CE enantioseparations under reversed polarity mode using sulfated β‐CD (S‐β‐CD) as chiral selectors. All of the effective mobility changes of (+)‐enantiomers between 35 and 20°C were higher than those of (–)‐enantiomers whosoever enantioselectivity increased or decreased with an increase in temperature. However, the unusual temperature effect that enantioselectivity was increased with an increase of temperature was observed for the compounds with hydroxyl substitution on phenyl ring and had relationship with the molecular structures. With performing NMR studies on the selected selector‐analyte complexes, the results led to a deeper understanding of the chiral discrimination process. Inspection of the complexation‐induced chemical shifts (CICS) of the enantiomers showed that the phenyl ring sits deeply and slantways in the cavity of S‐β‐CD and alkyl chain pointed out of the wider rim of S‐β‐CD with ion–ion and hydrogen bond interactions between analytes and S‐β‐CD.  相似文献

In order to obtain a better enantioselectivity of phenylalanine enantiomers and establish the optimal chiral extraction conditions, the distribution behavior was investigated in aqueous two‐phase systems which were composed of polyethylene glycol and ammonium sulfate containing combinatorial chiral selector: β‐cyclodextrin and HP‐β‐cyclodextrin. The influence of the molar concentration ratio of combinatorial chiral selectors, the total molar concentration of combinatorial chiral selectors, pH value, buffer type and its concentration were thoroughly studied, respectively. The results show that the enantioselectivity reaches 1.53 under the optimal chiral extraction conditions. This extraction is a potential economical and effective way for chiral resolution.  相似文献

The enantiomers of fourteen O,O-dialkyl-2-benzyloxycarbonyl-aminoarylmethyl-phosphonates are directly separated on the tris(3,5-dimethylphenylcarbamate) cellulose column (Chiralcel OD-R) under reversed phase mode. The results of the chiral separation are different from the results obtained in the normal phase mode. The mobile phase plays an essential role in chiral discrimination when using Chiralcel OD-R. The influences of the mobile phase composition on the retention and the enantioselectivity are investigated. The influences on chiral separation of the length and steric hindrance of alkoxy groups of the phosphonate ester and of the nature of the substituent on the benzene ring that is attached to the chiral carbon atom are also discussed.  相似文献

Two modified β‐cyclodextrins, H‐2 and H‐3 , having a flexible appended moiety were studied for the chiral discrimination of the enantiomers of various amino acids by means of fluorescence as signaling option. These hosts quenched the fluorescence intensities of amino acids upon binding. The d‐ enantiomers were better recognized by these hosts. The association constants (Ks) and enantioselectivity factors (α) of the host?guest complexes were calculated.  相似文献

A set of DNA tetranucleotides, which are 3'- or 5'-end extended versions of GCA, was used as chiral selectors for the discrimination of enantiomers of alpha-amino acids. The [X+Y-2H](2-) ions of the 1:1 complexes were generated by electrospraying a mixture of tetranucleotide (X) and amino acid (Y) solution. Chiral discrimination was achieved by studying the collision-induced dissociation spectra of the [X+Y-2H](2-) ion and the ratio of relative abundance of precursor ion to that of the product ion was used to measure the extent of discrimination. Among the tetranucleotides used, GCAA and GGCA exhibited better discrimination, in which GCAA showed D-selectivity and GGCA showed L-selectivity for the studied amino acids. Selective recognition of enantiomers of amino acids valine, methionine, phenylalanine, serine, and tyrosine by a binuclear copper complex with an azomethine obtained from 2,6-diformyl-4-methylphenol and L-valine was studied by means of spectrophotometry. The binding constants of individual enentiomers were estimated for valine, phenylalanine, methionine, serine, and tyrosine, and the enantioselectivity factors were evaluated. The isomers of serine and tyrosine (with respect to the first stage) were recognized with a considerable enantioselectivity (1.34 and 5.46 respectively), whereas the binding constants of valine and phenylalanine enantiomers are virtually indistinguishable.  相似文献

A novel member of a new class of chiral uranyl-salophen complexes has been synthesised. The chiral recognition ability of this receptor toward the enantiomers of two primary amines, a sulfoxide, and a quaternary ammonium chloride has been evaluated for the first time. The enantioselectivities obtained are encouraging. The NMR method developed for this purpose allows a fast, quantitative determination of the enantioselectivity of the host directly from its racemic mixture and could find application as a preliminary screening tool in the search for new receptors using combinatorial methods. The experiments carried out in this context demonstrated also that the activation barrier for the racemisation of such chiral uranyl-salophen receptors is much higher than the lower limit of 21 kcal mol(-1) previously reported.  相似文献

The chiral recognition mechanism of a cinchona alkaloid based chiral selector for N-protected peptide enantiomers was investigated. A chiral stationary phase derived from this selector was employed for liquid chromatographic enantiomer separations. It showed exceptionally high enantiomer discrimination for the (all-R)- and (all-S)-enantiomers of dialanine (alpha = 20), while a pronounced loss of chiral recognition occurred upon the insertion of an additional alanine residue into the peptide backbone. This reduction of enantioselectivity was investigated in great detail by NMR spectroscopy of complexes of the chiral selector and the analyte enantiomers accompanied by molecular modeling studies. Investigation of intramolecular NOEs provided the conformational states of the free and complexed forms of the selector. The analysis of complexation-induced shifts yielded information on intermolecular interactions and allowed us to propose binding models, which were further supported by the observation of intermolecular NOEs, indicating the relative arrangements of selector and analytes. Stochastic molecular dynamics simulations were able to reproduce the chromatographic retention orders and energy differences, as well as the intermolecular NOEs. The computational data were used to evaluate the intermolecular forces responsible for analyte binding. In addition, the relative contributions of the fragments of the chiral selector to the enantioselective binding event were assessed. A spatial arrangement of the chiral selector and the analyte allowing the primary ionic interaction as well as hydrogen bonding and pi-pi-stacking to take place simultaneously was found to be essential to obtain very high enantioselectivities.  相似文献

The performance of four commercially available cellulose tris(3,5-dimethylphenylcarbamate) based chiral stationary phases (CSPs) was evaluated with parallel high performance liquid chromatography (HPLC) and super critical fluid chromatography (SFC). Retention, enantioselectivity, resolution and efficiency were compared for a set of neutral, basic and acidic compounds having different physico-chemical properties by using different mobile phase conditions. Although the chiral selector is the same in all the four CSPs, a large difference in the ability to retain and resolve enantiomers was observed under the same chromatographic conditions. We believe that this is mainly due to differences in the silica matrix and immobilization techniques used by the different vendors. An extended study of metoprolol and structure analogues gave a deeper understanding of the accessibility of the chiral discriminating interactions and its impact on the resolution of the racemic compounds on the four CSPs studied. Also, a clear difference in enantioselectivity is observed between SFC and LC mode, hydrogen bonding was found to play an important role in the differential binding of the enantiomers to the CSPs.  相似文献

We investigated the enantioseparations of racemic hydrobenzoin, together with benzoin and benzoin methyl ether, in capillary electrophoresis (CE) using the single-isomer heptakis(2,3-dihydroxy-6-O-sulfo)-beta-cyclodextrin (SI-S-beta-CD) as a chiral selector in the presence and absence of borate complexation and enantiomer migration reversal of hydrobenzoin with a dual CD system consisting of SI-S-beta-CD and beta-CD in the presence of borate complexation at pH 9.0 in a borate buffer. The enantioselectivity of hydrobenzoin increased remarkably with increasing SI-S-beta-CD concentration and the enantioseparation depended on CD complexation between hydrobenzoin-borate and SI-S-beta-CD. The (S,S)-enantiomer of hydrobenzoin-borate complexes interacted more strongly than the (R,R)-enantiomer with SI-S-beta-CD. The enantiomers of hydrobenzoin could be baseline-resolved in the presence of SI-S-beta-CD at a concentration as low as 0.1% w/v, whereas the three test analytes were simultaneously enantioseparated with addition of 0.3% w/v SI-S-beta-CD or at concentrations >2.0% w/v in a borate buffer and 0.5% w/v in a phosphate background electrolyte at pH 9.0. Compared with the results obtained previously using randomly sulfated beta-CD (MI-S-beta-CD) in a borate buffer, enantioseparation of these three benzoin compounds is more advantageously aided by SI-S-beta-CD as the chiral selector. The enantioselectivity of hydrobenzoin depended greatly on the degree of substitution of sulfated beta-CD. Moreover, binding constants of the enantiomers of benzoin compounds to SI-S-beta-CD and those of hydrobenzoin-borate complexes to SI-S-beta-CD were evaluated for a better understanding of the role of CD complexation in the enantioseparation and chiral recognition. Enantiomer migration reversal of hydrobenzoin could be observed by varying the concentration of beta-CD, while keeping SI-S-beta-CD at a relatively low concentration. SI-S-beta-CD and beta-CD showed the same chiral recognition pattern but they exhibited opposite effects on the mobility of the enantiomers.  相似文献

The direct chiral separations of pesticide enantiomers by high-performance liquid chromatography by applying self-prepared cellulose triphenylcarbamate chiral stationary phase are performed. The mobile phase is n-hexane modified by isopropanol as a polar modifier. Nine chiral pesticides (benalaxyl, vinclozolin, diclofop-methyl, tebuconazole, quizalofop-ethyl, hexaconazole, lactofen, isocarbophos, and paclobutrazol) show enantioselectivity on the chiral stationary phase. An online circular dichrorism detector is used for identifying the pesticide enantiomers. The influences of the volume content of isopropanol and column temperature on the separations are investigated. The thermodynamic parameters related to the chiral distinguish mechanisms are also calculated.  相似文献

A chiral covalent organic framework was synthesized, characterized, and incorporated into organic polymer monolithic capillary columns to provide chiral stationary phases for enantioseparations. The prepared monolithic capillary columns were characterized by scanning electron microscopy and elemental analysis. To obtain better enantioseparations, the columns’ preparation conditions, and enantioseparation conditions were optimized. Baseline resolutions of several chiral compounds were obtained with good reproducibility and stability. Furthermore, the mechanism of chiral recognition was investigated using molecular docking with AutoDock. Docking results showed that the enantioselectivity factor rather than resolution is correlated with the binding free energy difference between enantiomers with the chiral covalent organic framework. And abundant acetoxy and nitrile groups as well as benzene rings in the chiral covalent organic framework are responsible for the enantioseparation ability of the chiral monolithic capillary columns.  相似文献

Summary The influence of column temperature (0–28 °C) and solute molecular size on the retention and enantioselectivity of a series of D, L dansyl amino acids with a non-polar side chain (valine, leucine, phenylalanine and tryptophan) were investigated using a vancomycin-based chiral stationary phase (CSP). The enthalpic and entropic terms for the solute-CSP association were determined from the linear vant Hoff plots. Two solute groups were distinguished in relation to these thermodynamic quantities: the solute group I (dansyl valine, dansyl leucine, dansyl phenylalanine) for which large negative values of enthalpic terms were obtained and the solute group II (dansyl tryptophan) for which H value was much less negative. The enthalpy-entropy compensation study revealed that the interaction mechanism was identical for the group I solute enantiomers but changed for D, L dansyl tryptophan. This was further exemplified as the group I compound enantiomers were resolved over the temperature range while the enantiomers of dansyl tryptophan were not separated in the operating conditions. Relationships between both the solute retention factors and apparent enantioselectivity, and the accessible surface area of the amino acid side chain indicated that when the solute molecular size increased (i) the retention was enhanced by the hydrophobic effect and (ii) the chiral discrimination decreased dependent, at least in part, on a steric hindrance phenomenon at the vancomycin aglycone pocket.  相似文献

[reaction: see text] 1,2-Bis(N-benzenesulfonyl-N-methylamino)benzene (2), which has no fixed asymmetric element, was crystallized from ethyl acetate as chiral crystals belonging to space group P4(1)2(1)2 (No. 92) or P4(3)2(1)2 (No. 96). The array of molecules built by the CH-pi interaction along the c-axis forms an enantiomeric helical superstructure in each individual crystal. The absolute configurations of the chiral crystals of 2 were determined by X-ray crystal structure analysis using the Flack parameter method. The solid-state CD spectra of the chiral crystals in KBr were mirror images. The equilibrium between the two enantiomers in solution is fast during crystallization at ambient temperature, and the energy barrier (DeltaG()) is estimated to be 11.7 +/- 0.3 kcal/mol (233 K).  相似文献

The two enantiomers of metoprolol and the four enantiomeric forms of alpha-hydroxymetoprolol were separated by liquid chromatography on a Chiralcel OD column containing a cellulose tris(3,5-dimethyl-phenylcarbamate) chiral stationary phase. The column efficiency was strongly dependent on the flow-rate and the enantioselectivity was influenced by temperature. Of utmost importance for the chiral separation was the water content of the mobile organic phase. The separation system was used for the separation and determination of the enantiomers in plasma and urine samples. The metoprolol enantiomers could be determined by fluorescence down to 10 nmol/l of each in plasma with a relative standard deviation of less than 15%.  相似文献

A series of six benzoxazolinone aminoketones and height aminoalcohols has been synthetized as agonist and antagonist ligands for adrenergic receptors. For those benzoxazolinone derivatives which contain one or two chiral carbons, a stereoselective liquid chromatographic method, using silica-based cellulose tris-3,5-dimethylphenylcarbamate (Chiralcel OD-H) or tris-4-methylbenzoate (Chiralcel OJ) as chiral stationary phase, has been developed. A better separation was achieved on cellulose carbamate phase compared to the cellulose ester phase. The effects of concentration of various aliphatic alcohols in the mobile phase were studied. In this work, chiral separation of enantiomers of three amino acids was achieved using capillary electrophoresis technique with α-cyclodextrin (αCD) as a running buffer additive. Only tryptophan has exhibited baseline separation in the presence of αCD, while the enantiomers of the other two amino acids, phenylalanine and tyrosine, were only partially separated. The addition of 18-crown-6 (18C6) as a second additive imparted only slight improvement to the separation of all enantiomers. On the other hand, all three racemic amino acid mixtures demonstrated no indication of separation when the larger cavity cyclodextrin members, β- and γCD, are used as running buffer chiral additives. However, remarkable improvements in the separation of the enantiomers of phenylalanine and tyrosine were obtained when 18C6 is used together with βCD as a running buffer additive. Surprisingly, tryptophan enantiomers were not separated by the dual additive system of cyclodextrin and crown ether. Using electrospray ionization mass spectrometry (ESI-MS), all amino acids were found to form stable binary complexes with individual hosts as well as ternary compounds involving the crown ether and the cyclodextrin. Furthermore, we used molecular dynamics (MD) simulations to build a clear picture about the interaction between the guest and the hosts. Most of these complexes remained stable throughout the simulation times, and the molecular dynamics study allowed better understanding of these supramolecular assemblies.  相似文献
