Fluorescent probes in the second near‐infrared window (NIR‐II) allow high‐resolution bioimaging with deep‐tissue penetration. However, existing NIR‐II materials often have poor signal‐to‐background ratios because of the lack of target specificity. Herein, an activatable NIR‐II nanoprobe for visualizing colorectal cancers was devised. This designed probe displays H2S‐activated ratiometric fluorescence and light‐up NIR‐II emission at 900–1300 nm. By using this activatable and target specific probe for deep‐tissue imaging of H2S‐rich colon cancer cells, accurate identification of colorectal tumors in animal models were performed. It is anticipated that the development of activatable NIR‐II probes will find widespread applications in biological and clinical systems.  相似文献

Fluorescence imaging is one of the most powerful techniques for monitoring biomolecules in living systems. Fluorescent sensors with absorption and emission in the near-infrared (NIR) region are favorable for biological imaging applications in living animals, as NIR light leads to minimum photodamage, deep tissue penetration, and minimum background autofluorescence interference. Herein, we have introduced a new strategy to design NIR functional dyes with the carboxylic-acid-controlled fluorescence on-off switching mechanism by the spirocyclization. Based on the design strategy, we have developed a series of Changsha (CS1-6) NIR fluorophores, a unique new class of NIR functional fluorescent dyes, bearing excellent photophysical properties including large absorption extinction coefficients, high fluorescence quantum yields, high brightness, good photostability, and sufficient chemical stability. Significantly, the new CS1-6 NIR dyes are superior to the traditional rhodamine dyes with both absorption and emission in the NIR region while retaining the rhodamine-like fluorescence ON-OFF switching mechanism. In addition, we have performed quantum chemical calculations with the B3LYP exchange functional employing 6-31G* basis sets to shed light on the structure-optical properties of the new CS1-6 NIR dyes. Furthermore, using CS2 as a platform, we further constructed the novel NIR fluorescent TURN-ON sensor 7, which is capable of imaging endogenously produced HClO in the living animals, demonstrating the value of our new CS NIR functional fluorescent dyes. We expect that the design strategy may be extended for development of a wide variety of NIR functional dyes with a suitable fluorescence-controlled mechanism for many useful applications in biological studies.  相似文献

Fluorescence imaging in the near‐infrared (NIR) region (650–900 nm) is useful for bioimaging because background autofluorescence is low and tissue penetration is high in this range. In addition, NIR fluorescence is useful as a complementary color window to green and red for multicolor imaging. Here, we compared the photoinduced electron transfer (PeT)‐mediated fluorescence quenching of silicon‐ and phosphorus‐substituted rhodamines (SiRs and PRs) in order to guide the development of improved far‐red to NIR fluorescent dyes. The results of density functional theory calculations and photophysical evaluation of a series of newly synthesized PRs confirmed that the fluorescence of PRs was more susceptible than that of SiRs to quenching via PeT. Based on this, we designed and synthesized a NIR fluorescence probe for Ca2+, CaPR‐1 , and its membrane‐permeable acetoxymethyl derivative, CaPR‐1 AM , which is distributed to the cytosol, in marked contrast to our previously reported Ca2+ far‐red to NIR fluorescence probe based on the SiR scaffold, CaSiR‐1 AM , which is mainly localized in lysosomes as well as cytosol in living cells. CaPR‐1 showed longer‐wavelength absorption and emission (up to 712 nm) than CaSiR‐1 . The new probe was able to image Ca2+ at dendrites and spines in brain slices, and should be a useful tool in neuroscience research.  相似文献

Owing to their applications in biodetection and molecular bioimaging, near‐infrared (NIR) fluorescent dyes are being extensively investigated. Most of the existing NIR dyes exhibit poor quantum yield, which hinders their translation to preclinical and clinical settings. Plasmonic nanostructures are known to act as tiny antennae for efficiently focusing the electromagnetic field into nanoscale volumes. The fluorescence emission from NIR dyes can be enhanced by more than thousand times by precisely placing them in proximity to gold nanorods. We have employed polyelectrolyte multilayers fabricated using layer‐by‐layer assembly as dielectric spacers for precisely tuning the distance between gold nanorods and NIR dyes. The aspect ratio of the gold nanorods was tuned to match the longitudinal localized surface plasmon resonance wavelength with the absorption maximum of the NIR dye to maximize the plasmonically enhanced fluorescence. The design criteria derived from this study lays the groundwork for ultrabright fluorescence bullets for in vitro and in vivo molecular bioimaging.  相似文献

Near-infrared (NIR) fluorescent sensors have emerged as promising molecular tools for imaging biomolecules in living systems. However, NIR fluorescent sensors are very challenging to be developed. Herein, we describe the discovery of a new class of NIR fluorescent dyes represented by 1a/1c/1e, which are superior to the traditional 7-hydroxycoumarin and fluorescein with both absorption and emission in the NIR region while retaining an optically tunable hydroxyl group. Quantum chemical calculations with the B3LYP exchange functional employing 6-31G(d) basis sets provide insights into the optical property distinctions between 1a/1c/1e and their alkoxy derivatives. The unique optical properties of the new type of fluorescent dyes can be exploited as a useful strategy for development of NIR fluorescent sensors. Employing this strategy, two different types of NIR fluorescent sensors, NIR-H(2)O(2) and NIR-thiol, for H(2)O(2) and thiols, respectively, were constructed. These novel sensors respond to H(2)O(2) or thiols with a large turn-on NIR fluorescence signal upon excitation in the NIR region. Furthermore, NIR-H(2)O(2) and NIR-thiol are capable of imaging endogenously produced H(2)O(2) and thiols, respectively, not only in living cells but also in living mice, demonstrating the value of the new NIR fluorescent sensor design strategy. The new type of NIR dyes presented herein may open up new opportunities for the development of NIR fluorescent sensors based on the hydroxyl functionalized reactive sites for biological imaging applications in living animals.  相似文献

Two‐photon microscopy is a powerful tool for studying biological systems. In search of novel two‐photon absorbing dyes for bioimaging, we synthesized a new anthracene‐based dipolar dye (anthradan) and evaluated its two‐photon absorbing and imaging properties. The new anthradan, 9,10‐bis(o‐dimethoxy‐phenyl)‐anthradan, absorbs and emits at longer wavelengths than acedan, a well‐known two‐photon absorbing dye. It is also stable under two‐photon excitation conditions and biocompatible, and thus used for two‐photon imaging of mouse organ tissues to show bright, near‐red fluorescence along with negligible autofluorescence. Such an anthradan thus holds promise as a new class of two‐photon absorbing dyes for the development of fluorescent probes and tags for biological systems.  相似文献

Two-photon fluorescence microscopy (2PFM) emerged as a powerful alternative to conventional one-photon microscopy. 2PFM typically uses two near-infrared (NIR) photons to excite fluorescent dyes, which minimizes light scattering in biological samples. Multiphoton absorption also suppresses background signal and autofluorescence from tissues and allows to achieve higher 3D resolution images with low photodamage and photobleaching. Fluorene dyes possess distinct properties that meet the strict criteria of probes used for 2PFM such as enhanced solubility, photostability, and two-photon absorption cross-section. The fluorene molecule also includes many active positions that allow versatile synthesis, selective functionalization, bioconjugation, and tuning solubility. These properties have led to reporting several fluorene probes including monomers, polymers, and dendrimers with important uses in understanding molecular dynamics and bioimaging. The current review presents a compact summary of fluorene-based fluorophores for 2PFM bioimaging applications, shedding light on structure-photophysical property relationships in fluorenes and polyaromatic probe designs.  相似文献

Hydrogen sulfide (H2S) is an important endogenous signaling molecule with a variety of biological functions. Development of fluorescent probes for highly selective and sensitive detection of H2S is necessary. We show here that dual‐reactable fluorescent H2S probes could react with higher selectivity than single‐reactable probes. One of the dual‐reactable probes gives more than 4000‐fold turn‐on response when reacting with H2S, the largest response among fluorescent H2S probes reported thus far. In addition, the probe could be used for high‐throughput enzymatic assays and for the detection of Cys‐induced H2S in cells and in zebrafish. These dual‐reactable probes hold potential for highly selective and sensitive detection of H2S in biological systems.  相似文献

Near‐infrared (NIR) imaging techniques have attracted significant attention for biological and medicinal applications due to the ability of NIR to penetrate deeply into tissues. However, there are very few stable, activatable molecular probes that can utilize NIR light in the wavelength range beyond 800 nm. Herein, we report a new activatable NIR system for photoacoustic imaging based on tautomeric benziphthalocyanines (BPcs). We found that the existence of a free hydroxyl group is crucial for NIR absorption of BPcs. Synthesized water‐soluble hydroxy BPcs exhibited high photostability and no fluorescence, which are desirable features for photoacoustic imaging. We synthesized BPcs in which the free hydroxyl group was masked by an esterase‐labile or an H2O2‐labile group. The photoacoustic signals of these hydroxy‐masked BPcs were increased upon NIR excitation at 880 nm in the presence of esterase or H2O2, respectively. These are rare examples of activatable probes utilizing NIR light at around 900 nm.  相似文献

A set of new azide‐ and alkyne‐bearing lepidinium‐based fluorophores were synthesized for bioorthogonal labeling schemes. These fluorescent dyes all show large Stokes‐shifts with emission maxima in the near‐infrared (NIR) region of the electromagnetic spectrum. The applicability of these dyes in the construction of energy‐transfer systems was tested using one of these new fluorescent tags and daunomycin (Dau), an anticancer drug with fluorescent features. These daunomycin conjugates are the very first examples of fluorescently modulated constructs of this anticancer agent. The dually labeled architectures proved that the applied fluorescent dye can be utilized as an efficient quencher for daunomycin. Enzymatic cleavage of a dually labeled enzyme substrate resulted in full recovery of the fluorescence of daunomycin. Such fluorescently modulated Dau conjugates can provide useful information for the mechanism of action of Dau‐regulated cell death processes.  相似文献

Fluorescence imaging is a promising tool for the visualization of biomolecules in living systems and there is great demand for new fluorescent dyes that absorb and emit in the near‐infrared (NIR) region. Herein, we constructed three new fluorescent dyes ( NBC dyes) based on keto‐benzo[h]coumarin ( k‐BC ) and benzopyrilium salts. These dyes showed large Stokes shifts (>100 nm) and NIR emission (>800 nm). The relationship between the structures and optical properties of these dyes was further investigated by using density functional theory calculations at the B3LYP/6‐3G level of theory. Fluorescence images indicated that the fabricated dyes exhibited good photostability and low cytotoxicity and, thus, have potential applications as imaging agents in living cells and animals.  相似文献

We report the synthesis, crystallographic, optical, and triplet and singlet oxygen generation properties of a series of BF2‐rigidified partially closed chain bromotetrapyrroles as near infrared emitters and photosensitizers. These novel dyes were efficiently synthesized from a nucleophilic substitution reaction between pyrroles and the 3,5‐bromo‐substituents on the tetra‐ and hexabromoBODIPYs and absorb in the near‐infrared region (681–754 nm) with high molar extinction coefficients. Their fluorescent emission (708–818 nm) and singlet oxygen generation properties are significantly affected by alkyl substitutions on the two uncoordinated pyrrole units of these dyes and the polarity of solvents. Among them, dyes 4 ca and 4 da show good singlet oxygen generation efficiency and good NIR fluorescence emission (fluorescence quantum yields of 0.14–0.43 in different solvents studied).  相似文献

T he use of nonfluorescent azo dyes as dark quenchers in activatable optical bioprobes based on the Förster resonance energy transfer (FRET) mechanism and designed to target a wide range of enzymes has been established for over two decades. The key value of the azo moiety (−N=N−) to act as an efficient “ON–OFF” switch of fluorescence once introduced within the core structure of conventional organic‐based fluorophores (mainly fluorescent aniline derivatives) has recently been exploited in the development of alternative reaction‐based small‐molecule probes based on the “profluorescence” concept. These unprecedented “azobenzene‐caged” fluorophores are valuable tools for the detection of a wide range of reactive (bio)analytes. This review highlights the most recent and relevant advances made in the design and biosensing/bioimaging applications of azo‐based fluorogenic probes. Emphasis is also placed on relevant achievements in the synthesis of bioconjugatable/biocompatible azo dyes used as starting building blocks in the rational and rapid construction of these fluorescent chemodosimeters. Finally, a brief glimpse of possible future biomedical applications (theranostics) of these “smart” azobenzene‐based molecular systems is presented.  相似文献

Herein, we report the development of two fluorescent probes for the highly selective and sensitive detection of H2S. The probes take advantage of a CuII? cyclen complex, which acts as a reaction center for H2S and as a quencher of BODIPY (boron‐dipyrromethene)‐based fluorophores with emissions at 765 and 680 nm, respectively. These non‐fluorescent probes could only be turned on by the addition of H2S, and not by other potentially interfering biomolecules, including reactive oxygen species, cysteine, and glutathione. In a chemical system, both probes detected H2S with a detection limit of 80 nM . The probes were successfully used for the endogenous detection of H2S in HEK 293 cells, for measuring the H2S‐release activity of dietary organosulfides in MCF‐7 cells, and for the in vivo imaging of H2S in mice.  相似文献

Si‐rhodamine (SiR) is an ideal fluorophore because it possesses bright emission in the NIR region and can be implemented flexibly in living cells. Currently, several promising approaches for synthesizing SiR are being developed. However, challenges remain in the construction of SiR containing functional groups for bioimaging application. Herein, we introduce a general and simple approach by a condensation reaction of diarylsilylether and arylaldehyde in o‐dichlorobenzene to synthesize a series of SiRs bearing various functional substituents. These SiRs have moderate to high quantum efficiency, tolerance to photobleaching, and high water solubility as well as NIR emitting, and their NIR fluorescence properties can be controlled through the photoinduced electron transfer (PET) mechanism. Fluorescence OFF‐ON switching effect is observed for SiR 9 in the presence of acid, which is rationalized by DFT/TDDFT calculations. Moreover, reversible stimuli response toward temperature is achieved. Since positive charge enables mitochondrial targeting ability and chloromethyl unit can covalently immobilize the dyes onto the mitochondrial via click reaction between the benzyl choride and protein sulfhydryls, SiR 8 is identified as a valuable fluorescent marker to visualize the morphology and monitor the temperature change of mitochondria with high photostability.  相似文献

The two signaling molecules H2S and H2O2 play key roles in maintaining intracellular redox homeostasis. The biological relationship between H2O2 and H2S remains largely unknown in redox biology. In this study, we rationally designed and synthesized single‐ and dual‐response fluorescent probes for detecting both H2O2 and H2S in living cells. The dual‐response probe was shown to be capable of mono‐ and dual‐detection of H2O2 and H2S selectively and sensitively. Detailed bioimaging studies based on the probes revealed that both exogenous and endogenous H2O2 could induce H2S biogenesis in living cells. By using gene‐knockdown techniques with bioimaging, the H2S biogenesis was found to be majorly cystathionine β‐synthase (CBS)‐dependent. Our finding shows the first direct evidence on the biological communication between H2O2 (ROS) and H2S (RSS) in vivo.  相似文献
