Synthesis and self‐assembly behavior of a novel amphiphilic brush‐coil block copolymer bearing hydrophilic poly(ethylene glycol) segment and hydrophobic polypeptide brush segment were presented in this work. The poly(γ‐benzyl‐L ‐glutamate) (PBLG) brush is synthesized through “grafting from” strategy by ring‐opening polymerization of γ‐benzyl‐L ‐glutamate‐N‐carboxyanhydride (BLG‐NCA) initiated by the flanking terminal primary amino group of macroinitiator. The copolymers were characterized by 1H NMR, gel permeation chromatography, Fourier transform infrared, circular dichroism spectrum, and differential scanning calorimetry. The self‐assembly behavior of the brush‐coil block copolymers in aqueous solution was investigated by means of transmission electron microscopy, scanning electron microscopy, atomic force microscopy, and laser light scattering. Spherical micelles were observed when the length of PBLG brush is shorter. The aggregate morphology transforms to spindle‐like micelles and then to rod‐like micelles, as the length of polypeptide brush increases. © 2009 Wiley Periodicals, Inc. Constructing polymeric toroids with a uniform, tunable size is challenging. Reported herein is the formation of uniform toroids from poly(γ‐benzyl‐l ‐glutamate)‐graft‐poly(ethylene glycol) (PBLG‐g‐PEG) graft copolymers by a two‐step self‐assembly process. In the first step, uniform rodlike micelles are prepared by dialyzing the polymer dissolved in tetrahydrofuran (THF)/N,N′‐dimethylformamide (DMF) against water. With the addition of THF in the second step, the rodlike micelles curve and then close end‐to‐end to form uniform toroids, which resemble a cyclization reaction.  相似文献

Dendron‐like poly(γ‐benzyl‐L ‐glutamate)/linear poly(ε‐caprolactone)/dendron‐like poly(γ‐benzyl‐L ‐glutamate) triblock copolymers having 2m + 1 PBLG branches (denoted as PBLG‐Dm‐PCL‐Dm‐PBLG, m = 0, 1, 2, and 3) were for the first time synthesized by utilizing ring‐opening polymerization (ROP) and click chemistry. The bifunctional azide‐terminated PCL (N3‐PCL‐N3) was click conjugated with propargyl focal point PAMAM‐typed dendrons Dm to generate Dm‐PCL‐Dm, which was then used as macroinitiator for the ROP of BLG‐NCA monomer to produce the targeted PBLG‐Dm‐PCL‐Dm‐PBLG triblock copolymers. Their molecular structures and physical properties were characterized in detail by FTIR, NMR, gel permeation chromatography, differential scanning calorimetry, and wide angle X‐ray diffraction (WAXD). The crystallinity of the central PCL segment within these copolymers is increasingly suppressed by the flanking PBLG wedges, whereas the PBLG segments gradually changed from a β‐sheet conformation to an α‐helix conformation with the increasing PBLG branches. These triblock copolymers formed thermoreversible organogels in toluene, and the dendritic topology of PBLG wedges controlled their critical gelation concentrations. The self‐assembled structure of organogels was further characterized by means of transmission electron microscopy, WAXD, and small‐angle X‐ray scattering. The fibers with flat ribbon morphology were clearly shown, and the gelation occurred through a self‐assembled nanoribbon mechanism. © 2009 Wiley Periodicals, Inc. Well‐defined azobenzene‐containing side‐chain liquid crystalline diblock copolymers composed of poly[6‐(4‐methoxy‐azobenzene‐4′‐oxy) hexyl methacrylate] (PMMAZO) and poly(γ‐benzyl‐L ‐glutamate) (PBLG) were synthesized by click reaction from alkyne‐ and azide‐functionalized homopolymers. The alkyne‐terminated PMMAZO homopolymers were synthesized by copper‐mediated atom transfer radical polymerization with a bromine‐containing alkyne bifunctional initiator, and the azido‐terminated PBLG homopolymers were synthesized by ring‐opening polymerization of γ‐benzyl‐L ‐glutamate‐N‐carboxyanhydride in DMF at room temperature using an amine‐containing azide initiator. The thermotropic phase behavior of PMMAZO‐b‐PBLG diblock copolymers in bulk were investigated using differential scanning calorimetry and polarized light microscopy. The PMMAZO‐b‐PBLG diblock copolymers exhibited a smectic phase and a nematic phase when the weight fraction of PMMAZO block was more than 50%. Photoisomerization behavior of PMMAZO‐b‐PBLG diblock copolymers and the corresponding PMMAZO homopolymers in solid film and in solution were investigated using UV–vis. In solution, trans–cis isomerization of diblock copolymers was slower than that of the corresponding PMMAZO homopolymers. We report the design and synthesis of new fully biodegradable thermoresponsive amphiphilic poly(γ‐benzyl L ‐glutamate)/poly(ethyl ethylene phosphate) (PBLG‐b‐PEEP) block copolymers by ring‐opening polymerization of N‐carboxy‐γ‐benzyl L ‐glutamate anhydride (BLG? NCA) with amine‐terminated poly(ethyl ethylene phosphate) (H2N? PEEP) as a macroinitiator. The fluorescence technique demonstrated that the block copolymers could form micelles composed of a hydrophobic core and a hydrophilic shell in aqueous solution. The morphology of the micelles as determined by transmission electron microscopy (TEM) was spherical. The size and critical micelle concentration (CMC) values of the micelles showed a decreasing trend as the PBLG segment increased. However, UV/Vis measurements showed that these block copolymers exhibited a reproducible temperature‐responsive behavior with a lower critical solution temperature (LCST) that could be tuned by the block composition and the concentration.  相似文献

Tuning the secondary structure of polypeptide is an effective strategy to modulate the assembly behaviors of polypeptide‐based copolymers. In this study, ring‐opening polymerization of l ‐alanine (Ala) and γ‐benzyl‐l ‐glutamate (BLG) N‐carboxyanhydrides was adopted using mPEG‐NH2 as the initiator to prepare mPEG‐poly(l ‐alanine‐co‐γ‐benzyl‐l ‐glutamate) (PEAB) copolymers with various Ala to BLG ratios. 1H NMR spectra and GPC test confirmed their well‐defined chemical structures. FT‐IR spectra indicated that at the powder state, all copolymers adopted both β‐sheet and αhelical conformations. With the content of PBLG increased, the crystallization temperature and melting points of PEAB copolymers first rose then fell indicated by DSC curves. The self‐assembly of PEAB copolymers in dilute aqueous solution studied by DLS, TEM and circular dichroism spectra showed that PEAB copolymers self‐assembled into nanostructures with diverse morphologies and sizes due to distinct polypeptide conformations. Rheological analysis indicated that the alteration of the polypeptide composition can effectively modulate the modulus of PEAB assemblies in concentrated solutions. In all, copolymerization of two hydrophobic amino acid N‐carboxyanhydrides into the polypeptide block maybe an effective approach for modulating the assembly properties of PEGylated polypeptide. Besides, nanosilver‐encapsulated PEA or PEAB hydrogel showed promising antibacterial effect against Staphylococcus aureus and Bacillus subtillis. © 2017 Wiley Periodicals, Inc. J. Polym. Three amphiphilic rod‐coil diblock copolymers, poly(2‐ethyl‐2‐oxazoline‐b‐γ‐benzyl‐L ‐glutamate) (PEOz‐b‐PBLG), incorporating the same‐length PEOz block length and various lengths of their PBLG blocks, were synthesized through a combining of living cationic and N‐carboxyanhydride (NCA) ring‐opening polymerizations. In the bulk, these block copolymers display thermotropic liquid crystalline behavior. The self‐assembled aggregates that formed from these diblock copolymers in aqueous solution exhibited morphologies that differed from those obtained in α‐helicogenic solvents, that is, solvents in which the PBLG blocks adopt rigid α‐helix conformations. In aqueous solution, the block copolymers self‐assembled into spherical micelles and vesicular aggregates because of their amphiphilic structures. In helicogenic solvents (in this case, toluene and benzyl alcohol), the PEOz‐b‐PBLG copolymers exhibited rod‐coil chain properties, which result in a diverse array of aggregate morphologies (spheres, vesicles, ribbons, and tube nanostructures) and thermoreversible gelation behavior. © 2008 Wiley Periodicals, Inc. We have investigated the morphology and packing manner of graft copolymers consisting of rigid‐rod‐like poly(γ‐benzyl L ‐glutamate) (PBLG) main chains and grafted diblock copolymers of amorphous poly(propylene glycol) (PPG) and crystalline poly(ethylene glycol) (PEG). The results of differential scanning calorimetry and wide‐angle X‐ray scattering measurements for graft copolymers with higher side‐chain volume fractions suggest that the rodlike main chains and crystallized PEG chains exist in segregated domains. Small‐angle X‐ray scattering profiles for these samples show diffraction intensity maxima accompanied by higher order peaks, the positions of which suggest the formation of an ordered layered structure. From these observations, the graft copolymers are estimated to form repeated layered structure consisting of segregated PBLG, PPG, and PEG layers. A proposed model for molecular packing of the graft copolymers is consistent with the experimental observation that the repeating distance for the layered structure decreases with an increase in the volume fraction of side chains. © 2002 Wiley Periodicals, Inc. In this study, we used click chemistry to synthesize a new macromolecular self‐assembling building blocks, linear polypeptide‐b‐polyhedral oligomeric silsesquioxane (POSS) copolymers, from a mono‐azido–functionalized POSS (N3‐POSS) and several alkyne‐poly(γ‐benzyl‐L ‐glutamate) (alkyne‐PBLG) systems. The incorporation of the POSS unit at the chain end of the PBLG moiety allowed intramolecular hydrogen bonding to occur between the POSS and PBLG units, thereby enhancing the α‐helical conformation in the solid state, as determined through Fourier transform infrared spectroscopy and wide‐angle X‐ray diffraction analyses. POSS‐b‐PBLG underwent hierarchical self‐assembly, characterized using small‐angle X‐ray scattering, to form a bilayer‐like nanostructure featuring α‐helical or β‐sheet conformations and POSS aggregates. Thermogravimetric analysis indicated that the thermal degradation temperature increased significantly after incorporation of the POSS moiety, which presumably formed an inorganic protection layer on the nanocomposite's surface. © 2011 Wiley Periodicals, Inc. Solution property of poly(γ‐benzyl‐L ‐glutamate)‐b‐polyisoprene‐b‐poly(γ‐benzyl‐L ‐glutamate) (GIG copolymer) was studied by using dynamic light scattering and static light scattering for N,N‐dimethylformamide (DMF) solution and DMF/toluene mixed solutions. GIG copolymer proved to aggregate in DMF and under DMF‐rich condition, that is, high‐polar region. The aggregate decreased in size, and completely disappeared under toluene‐rich condition, that is, low‐polar region. The correlation between solubility parameter and aggregate size of GIG copolymer in the DMF/toluene solution systems quantitatively demonstrated how strongly polarity caused by hydrogen bond made an impact on the aggregation behavior. Because the main driving force to the aggregation under DMF‐rich condition originates with polyisoprene (PIP) blocks, the aggregate in DMF is considered to be a core‐shell micelle consisting of flexible PIP core surrounded by rigid poly(γ‐benzyl‐L ‐glutamate) (PBLG) shell. The values of dimensionless parameter ρ, defined as the ratio of radius of gyration 〈S21/2 to hydrodynamic radius RH, revealed that a single chain of GIG copolymer had the form of rigid rod with flexibility, that is, once‐broken rod, caused by the incorporation of a flexible PIP chain between two rigid PBLG rods in the DMF/toluene solution system. © 2010 Wiley Periodicals, Inc. Hybrid rod‐rod diblock copolymers, poly(γ‐benzyl L‐glutamate)‐poly(4‐cyano‐benzoic acid 2‐isopropyl‐5‐methyl‐cyclohexyl ester) (PBLG‐PPI), with determined chirality are facilely synthesized through sequential copolymerization of γ‐benzyl‐L‐glutamate N‐carboxyanhydride (BLG‐NCA) and phenyl isocyanide monomers bearing chiral menthyl pendants using a Ni(cod)(bpy) complex as the catalyst in one‐pot. Circular dichroism and absorption spectra reveal that each block of the block copolymers possesses a stable helical conformation with controlled helicity in solution due to the induction of chiral pendants. The two diastereomeric polymers self‐assemble into helical nanofibrils with opposite handedness due to the different chiral induction of the L‐ and D‐menthyl pendants, confirmed by transmission electron micro­scopy (TEM). Deprotection of the benzyl groups of the PBLG segment affords biocompatible amphiphilic diblock copolymers, poly(L‐glutamic acid)‐poly(4‐cyano‐benzoic acid 2‐isopropyl‐5‐methyl‐cyclohexyl ester) (PLGA‐PPI), that can self‐assemble into well‐defined micelles by cosolvent induced aggregation. Very interestingly, a chiral rhodamine chromophores RhB(D) can be selectively encapsulated into the chiral polymeric micelles, which is efficiently internalized into living cells when directly monitored with a confocal microscope. This contribution will be useful for developing novel rod‐rod biocompatible hybrid block copolymers with a controlled helicity, and may also provide unique chiral materials for potential bio‐medical applications.

A novel class of biomimetic glycopolymer–polypeptide triblock copolymers [poly(L ‐glutamate)–poly(2‐acryloyloxyethyllactoside)–poly(L ‐glutamate)] was synthesized by the sequential atom transfer radical polymerization of a protected lactose‐based glycomonomer and the ring‐opening polymerization of β‐benzyl‐L ‐glutamate N‐carboxyanhydride. Gel permeation chromatography and nuclear magnetic resonance analyses demonstrated that triblock copolymers with defined architectures, controlled molecular weights, and low polydispersities were successfully obtained. Fourier transform infrared spectroscopy of the triblock copolymers revealed that the α‐helix/β‐sheet ratio increased with the poly(benzyl‐L ‐glutamate) block length. Furthermore, the water‐soluble triblock copolymers self‐assembled into lactose‐installed polymeric aggregates; this was investigated with the hydrophobic dye solubilization method and ultraviolet–visible analysis. Notably, this kind of aggregate may be useful as an artificial polyvalent ligand in the investigation of carbohydrate–protein recognition and for the design of site‐specific drug‐delivery systems. © 2004 Wiley Periodicals, Inc. A series of pyrenyl‐terminated poly(γ‐benzyl‐l ‐glutamate)s (py‐PBLGs) with controlled polymer molecular weight (MW = 2.3–14.8 kg mol?1) and molecular weight distribution (PDI = 1.17–1.55) have been prepared from 1‐pyrenemethylamine hydrochloride‐mediated ring‐opening polymerization (ROP) of γ‐benzyl‐l ‐glutamic acid based N‐carboxyanhydride (BLG‐NCA). FTIR analysis revealed that the py‐PBLG9 was conformationally heterogeneous with 35.0% α‐helix, 55.6% β‐sheet, and 9.4% random coil conformations in the solid state, whereas the py‐PBLG66 adopts 100% α‐helix conformation. Py‐PBLGs promote the dispersion of SWCNTs in organic solvents and in the PBLG solid through π–π interaction, as evidenced by the Raman spectroscopic studies. WAXD analysis revealed that the SWCNTs significantly affect the ordering of the py‐PBLG self‐assembly: the long range hexagonal packing of py‐PBLG66 rods is notably enhanced by the addition of SWCNTs, whereas the lamellar packing of py‐PGLG9 β‐sheets is weakened. In the hexagonal lattice, the SWCNTs are intercalated parallel to the py‐PBLG66 rods, in contrast to the normal orientation of the SWCNTs with respect to the extended py‐PBLG9 chains in the β‐sheets. The relative packing structure also affects the intermolecular interaction among the PBLGs: SWCNTs promote the interaction among the py‐PBLG9 chains packed in a lamellar structure and weaken the intermolecular interaction among the py‐PBLG66 columnar hexagonal array. © 2013 Wiley Periodicals, Inc. The self‐assembling nature and phase‐transition behavior of a novel class of triarm, star‐shaped polymer–peptide block copolymers synthesized by the combination of atom transfer radical polymerization and living ring‐opening polymerization of α‐amino acid‐N‐carboxyanhydride are demonstrated. The two‐step synthesis strategy adopted here allows incorporating polypeptides into the usual synthetic polymers via an amido–amidate nickelacycle intermediate, which is used as the macroinitiator for the growth of poly(γ‐benzyl‐L ‐glutamate). The characterization data are reported from analyses using gel permeation chromatography and infrared, 1H NMR, and 13C NMR spectroscopy. This synthetic scheme grants a facile way to prepare a wide range of polymer–peptide architectures with perfect microstructure control, preventing the formation of homopolypeptide contaminants. Studies regarding the supramolecular organization and phase‐transition behavior of this class of polymer‐block‐polypeptide copolymers have been accomplished with X‐ray diffraction, infrared spectroscopy, and thermal analyses. In this contribution, the preparation of rod‐coil‐rod triblock copolymers based on polydimethylsiloxane and polypeptide [poly(γ‐benzyl‐L ‐glutamate)] is reported. Firstly, self‐assembly in rod‐like structures was demonstrated via polarized optical microscopy and small‐angle light scattering. Further structuration details were obtained using X‐ray scattering and AFM imaging to establish the formation of a double‐hexagonal structure and to accurately define the morphological dimensions of the rodlike structures. The thermal behavior of these structures was investigated using dynamic mechanical analysis and differential scanning calorimetry. We conclude by addressing an unexpected reversible thermal transition within the 130–150 °C temperature range and the ensuing associated organizational modifications. © 2006 Wiley Periodicals, Inc. Macroinitiator‐amino terminated poly(ethylene glycol) (PEG) (NH2‐PEO‐NH2) was prepared by converting both terminal hydroxyl groups of PEG to more reactive primary amino groups. The synthetic route involved reactions of chloridize, phthalimide and finally hydrazinolysis. Furthermore, poly(γ‐benzyl‐L ‐glutamate)‐poly(ethylene oxide)‐poly(γ‐benzyl‐L ‐glutamate) (PBLG‐PEO‐PBLG) triblock copolymer was synthesized by polymerization of γ‐benzyl‐L ‐glutamate N‐carboxyanhydride (Bz‐L‐GluNCA) using NH2‐PEO‐NH2 as macroinitiator. The resultant NH2‐PEO‐NH2 and triblock copolymer were characterized by FT‐IR, 1H‐NMR and gel permeation chromatography (GPC) techniques. The results demonstrated that the degree of amination of the NH2‐PEO‐NH2 could be up to 1.95. The molecular weight of the PBLG‐PEO‐PBLG triblock copolymer could be adjusted easily by controlling the molar ratio of Bz‐L ‐Glu NCA to the macroinitiator NH2‐PEO‐NH2. Single‐walled carbon nanotubes (SWCNTs) have been functionalized with poly(γ‐benzyl‐L ‐glutamate) (PBLG) by ring‐opening polymerizations of γ‐benzyl‐L ‐glutamic acid‐based N‐carboxylanhydrides (NCA‐BLG) using amino‐functionalized SWCNTs (SWCNT‐NH2) as initiators. The SWCNT functionalization has been verified by FTIR spectroscopy and transmission electron microscopy. The FTIR study reveals that surface‐attached PBLGs adopt random‐coil conformations in contrast to the physically absorbed or bulk PBLGs, which exhibit α‐helical conformations. Raman spectroscopic analysis reveals a significant alteration of the electronic structure of SWCNTs as a result of PBLG functionalization. The PBLG‐functionalized SWCNTs (SWCNT‐PBLG) exhibit enhanced solubility in DMF. Stable DMF solutions of SWCNT‐PBLG/PBLG with a maximum SWCNTs concentration of 259 mg L?1 can be readily obtained. SWCNT‐PBLG/PBLG solid composites have been characterized by differential scanning calorimetry, thermogravimetric analysis, wide/small‐angle X‐ray scattering (W/SAXS), scanning electron microscopy, and polarized optical microscopy for their thermal or morphological properties. Microfibers containing SWCNT‐PBLG and PBLG can also be prepared via electrospinning. WAXS characterization reveals that SWCNTs are evenly distributed among PBLG rods in solution and in the solid state where PBLGs form a short‐range nematic phase interspersed with amorphous domains. © 2010 Wiley Periodicals, Inc. Single‐wall carbon nanotubes (SWCNTs) have been added to high molecular weight poly(γ‐benzyl‐L ‐glutamate), or PBLG, to evaluate their effects on the polymer's shear piezoelectricity. While the addition of SWCNTs increased various PBLG physical properties such as electrical conductivity, dielectric constant, several mechanical properties, and electrostriction coefficient, the shear piezoelectricity remained constant up to a 0.3 wt % SWCNT concentration. The composite crystallinity, orientation, and SWCNT alignment (measured by X‐ray diffraction, birefringence, and polarized Raman spectroscopy, respectively) were found to be constant up to this same concentration, corroborating the shear piezoelectric findings. PBLG composites made with acid‐treated (and therefore less electrically conductive) SWCNTs exhibited similar shear piezoelectric behavior, indicating that neither the SWCNT type, concentration (up to the percolation threshold), nor electrical conductivity influences PBLG shear piezoelectricity. © 2010 Wiley Periodicals, Inc. We have developed a metal free synthetic pathway to homopolypeptide rod‐coil block copolymers. The concept was proven for the synthesis of poly(benzyl‐L ‐glutamate‐b‐styrene). A dual initiator containing a primary amine and a nitroxide group was used in a macroinitiation approach with high initiation efficiency. Good control over the molecular weight in the ring opening polymerization of benzyl‐L ‐glutamate N‐carboxyanhydride was obtained in DMF at 0 °C yielding poly(benzyl‐L ‐glutamates) with low polydispersities around 1.1. The almost quantitative incorporation of the dual initiator was confirmed by MALDI‐ToF analysis. Macroinitiation of styrene by nitroxide‐mediated controlled radical polymerization yielded the block copolymer with high structural control. The diblock structure was confirmed by molecular weight increase upon macroinitiation by size exclusion chromatography and retention time comparison with homopolymers using gradient polymer elution chromatography. Both polymerizations were also successfully conducted in one pot without intermediate isolation owing to the high compatibility of both polymerization techniques. © 2008 Wiley Periodicals, Inc.
