The solubility of nimodipine was measured in aqueous solutions of the following cyclodextrins: -cyclodextrin (-CD), hydroxypropyl--CD (HP--CD), -cyclodextrin (-CD), random substituted methyl--CD (M--CD), three hydroxypropyl--CDs (HP--CD) with mutually different average degree of substitution, and hydroxypropyl--cyclodextrin (HP--CD). From the determined linear solubility diagrams the values of the binding constant K11 of the inclusion complexes of nimodipine with the respective CDs were evaluated. The -CDs efficiently solubilized sparingly soluble nimodipine, the highest value of K11 was found for M--CD (1680 M-1), followed by -CD (550 M-1) and HP--CDs, where the higher degree of substitution lowered K11. Only slight solubilization of nimodipine was observed in the solutions of the -CDs and HP--CD.  相似文献

Aim of the present work was to investigate the effect of somecyclodextrins (CDs) on the solubility and ocular bioavailability of rufloxacin base (RUF), with theultimate goal of developing an ophthalmic formulation. Phase solubility studies of RUF inpH 7.4 buffer were carried out in the presence of -cyclodextrin (-CD),hydroxypropyl--cyclodextrin (HP--CD) and -cyclodextrin(-CD). The effect of hydroxypropyl methylcellulose (HPMC) on RUF solubility was evaluated after heating the solutionscontaining HP--CD at 120 °C.A significant enhancement of RUF solubility was achieved by associatingthe drug with CDs, particularly HP--CD. This CD formed with RUF a less stablecomplex than that formed by -CD, but did not suffer the solubility limitations ofthe parent CD, and showed a higher solubilizing capacity than -CD. Addition of 0.25%(w/v) HPMC to solutions containing HP--CD increased the solubilizing effect of this CD,thus allowing reduction of the amount necessary for solubilization of 0.3% (w/v) RUF.Preliminary pharmacokinetic data in rabbits indicated that theocular bioavailability of 0.3% (w/v) RUF solubilized by HP--CD was higher when compared witha 0.3% (w/v) RUF suspension used as reference.  相似文献

The effect of -cyclodextrin (-CD) and hydroxypropyl--cyclodextrin (HP--CD) upon the photochemically-induced fluorescence (PIF) properties of four phenylurea herbicides, including linuron, diuron, isoproturon and neburon has been studied. Photochemical conversion of these nonfluorescent herbicides into strongly fluorescent photoproducts was shown to occur in -CD and HP--CD aqueous media. The influence of pH, UV irradiation time and photoproduct stability on the fluorescence intensity was also investigated. In addition, the stoichiometry and formation constants of the complexes formed between herbicides and -cyclodextrin (-CD) or 2-hydroxypropyl--cyclodextrin (HP--CD) were determined. The formation constant values, ranging from 184 ± 40 to 1498 ± 245 M-1, were calculated by applying the iterative nonlinear regression (NLR) approach to the PIF data. Linear calibrations graphs were established in the interval 1–12 g/mL, for diuron, linuron and neburon. The IUPAC limits of detection ranged between 580 and 700 ng/mL, according to the compound. Application to the analysis of phenylurea herbicides in spiked river water was also described.  相似文献

The influence of cyclodextrins (CDs) on the chemical stability of the contraceptive steroid progestin, ST1435, in aqueous solution has been studied using reversed phase high performance liquid chromatography. The effects of CD structure, temperature, and CD concentration on the rate of degradation were investigated. It was found that the drug degraded to different extents following a pseudo-first order reaction mechanism. The presence of the host molecules affected the degradation rate as a result of complexation which might result in protection of the labile moiety of the drug molecule against degradation. Hydroxypropyl--cyclodextrin (HP--CD) and hydroxyethyl--cyclodextrin (HE--CD) retarded the degradation in contrast to -cyclodextrin (-CD) which accelerated the steroid degradation. The stabilizing action of HP--CD is larger than that of HE--CD. The degradation rate increased upon increasing temperature and the Arrhenius equation is valid. Lineweaver-Burk equation analysis indicated that the steroid included inside the CD cavity degraded three times more slowly than did the free ST1435 in solution. This equation further supported the formation of a 1 : 1 inclusion complex between ST1435 and HP--CD with a stability constant of 934.5 M-1 at 65°C.  相似文献

In an effort to modify the solubility and dissolution rate of the contraceptive steroid, 19-norprogesterone in order to improve its bioavailability, the cyclodextrin complexation approach was chosen. In solution, the complex formation with -cyclodextrin (-CD), hydroxyethyl -cyclodextrin (HE--CD) and hydroxypropyl -cyclodextrin (HP--CD) was confirmed by using solubility, UV, IR and 1H-NMR spectrophotometric techniques. The phase solubility diagrams were categorized as AL-type. The complexing affinity of the CDs investigated were ranked as follows: -CD > HP--CD > HE--CD. The complexation thermodynamic parameters were obtained from the temperature dependence of the dissociation constants. In the solid state, differential scanning calorimetery (DSC) and optical microscopy methods were utilized to characterize the complexes. Dissolution studies showed that such molecularly encapsulated forms offered a marked improvement in the dissolution rate compared to the parent drug.  相似文献

Determinations of drug fluxes through semi-permeable cellophanemembranes are used to evaluate cyclodextrin complexes and cyclodextrin containingdrug formulations. In the present study we investigated how the cyclodextrin concentration,the membrane thickness and the molecular weight cut off (MWCO) of the membraneinfluence drug fluxes. The cyclodextrin used was 2-hydroxypropyl--cyclodextrin(HP--CD) and the sample drug was hydrocortisone. The MWCO ofthe membranes ranged from 500 to 14,000 and the HP--CD concentrationranged from 0 to 25% (w/v). The hydrocortisone flux from saturated solutions through the MWCO 500 membrane was unaffected by the cyclodextrin concentration. When MWCO of the membrane was greater than the molecular weight of the complex the flux fromsolutions saturated with hydrocortisone increased with increasing HP--CD concentration.This increase showed negative deviation from linearity. When the flux was correctedfor the viscosity increase with increasing HP--CD concentration then the fluxpattern could be described on the basis of Fick's first law and Stokes–Einsteinequation. However the flux did not correlate with the viscosity when it was increasedby adding polymer to the saturated drug solutions. It was shown that the observed fluxpattern was consistent with self-association of cyclodextrin complexes in the aqueousdonor phase.  相似文献

FT-Raman spectra of 2-, 3-, and 4-chlorostyrene included in-cyclodextrin (CD),glycerol ether -CD, -CD,sulfated -CD, andglycerol ether -CD were recorded.In the inclusion complexes, the area of the vinyl(C=C) band decreased remarkably, whereasthe area of the phenyl (C=C) band increasedcompared to those of liquid 2-, 3-, and 4-chlorostyrene,respectively. The effect of 2-hydroxypropyl--cyclodextrin (HP--CyD) on the release of a water-soluble 1-selective adrenoreceptor antagonist, metoprolol (Met), from ternary Met/HP--CyD/ethylcellulose (EC) tablets was investigated. The release rate of Met from the ternary tablets was dependent on amounts of HP--CyD in the tablets, i.e., the rate decreased when small amounts of HP--CyD were added, while large amounts of HP--CyD accelerated the rate. The slowest rate was observed for the tablet consisted of a 30/10/60 weight ratio of Met/HP--CyD/EC. The analyses of the release rates by the Korsmeyer equation and their temperature dependence suggested that Met is released from the EC matrix containing HP--CyD according to the diffusion-controlled mechanism. The water penetration studies and the micro- and macroscopic observations suggested that the retarding effect of HP--CyD is attributable to a viscous gel formation in small pores on the surface of the tablets, where HP--CyD gels may work as a barrier for the water penetration into the tablets and the release of the drug from the tablets. The in-vitro release property of the ternary tablets was reflected in the in-vivo absorption profile in dogs. The results indicated that a combination of HP--CyD and EC is useful for the release control of water-soluble drugs such as Met.  相似文献

Inclusion complexation between -cyclodextrin (-CD),hydroxypropyl--cyclodextrin (HP--CD), water-solublepolymers (PVP and HPMC) and vinpocetine was studied in aqueous solution and in the solid state.Phase solubility studies were used to evaluate the complexation in aqueous solution at roomtemperature. Stability constants (Kc) of binary and ternary complexes were determined spectrophotometrically. Differential scanning calorimetry (DSC)was used to characterize kneaded, co-evaporated and lyophilised binary and ternary systems.The Kc values obtained were 70.14 M-1 and 35.01 M-1 for vinpocetine--CD and vinpocetine-HP--CD and increased in a range of 17% to 94%by addition of water-soluble polymers. Some preliminary evidences ofinclusion complexation were obtained from DSC suggesting that co-evaporated and lyophilised binary andternary systems were truly inclusion complexes.  相似文献

The solubilizing potential and complexing tendencies of six cyclodextrins (CyD) with nifedipine in aqueous solution were evaluated using phase solubility methods. Solubility curves of nifedipine with -CyD, 2-hydroxypropyl--CyD (2HP--CyD) and 2-hydroxypropyl--cyclodextrin (2HP--CyD) were classified as type AL, while for heptakis (2,6-dimethyl)--CyD (DIMEB), randomly methylated--CyD (RAMEB) and -CyD, Ap type phase behaviour was observed. Stability constants, calculated from phase solubility diagrams, decreased in the order: DIMEB > RAMEB > -CyD > 21HP--CyD > -CyD > 2HP--CyD.  相似文献

Stereoselective hydrogenation of thymol over Rh/alumina in the presence of various equivalents of -cyclodextrin (-CD) and itsderivatives in the solid state was studied. Hydrogenation of thymol in the absence of -CD gave 76.8% epimeric alcohols with a menthol/neomenthol (M/N) ratio of 5.6 and an alcohol/ketone ratio of 4.5, whereas the presence of 0.1 equivalent (to thymol) of -CD gave rise to 94.6% of epimeric alcohols with a M/N ratio of 6.3 and an alcohol/ketone ratio of 45.4. The effect of -cyclodextrin and its derivatives on the modification of the yield and the proportion of epimeric alcohols formed were found to be the salient features of this investigation. Inclusion complexation of thymol by -CD studied by UV-Visible spectroscopyindicated a 2:1 stoichiometry ofthymol: -CD complex with a binding constant value of 480 ±40 M-2.  相似文献

Summary The effect of tri-O-methyl--cyclodextrin(methyl--CD) on the partition coefficients of drugs, such as p-nitrophenol, salicylic acid, benzoic acid, and aspirin, was studied at 25°C. The partition coefficients of these drugs were increased linearly with methyl--CD concentration. The increase of partition coefficients was interpreted by the 11 complex formation between methyl--CD and the drug in CHCl3 phase.The interaction between p-nitrophenol and methyl--CD in solution was studied by UV and PMR spectroscopies. It was concluded that p-nitrophenol is included in the cavity of methyl--CD in both aqueous solution and CHCl3 solution.Inclusion compounds of these drugs with methyl--CD in the solid state were studied by X-ray diffractometry, IR spectroscopy, and DSC measurements. 11 crystalline inclusion compounds were obtained from hot water. It is also suggested that amorphous inclusion compound was obtained by the grinding of drug with methyl--CD.The dissolution rate and the bioavailability of ketoprofen were significantly increased in the presence of methyl--CD. The bioavailability of ketoprofen after oral administration with methyl--CD to rats was 3.7 times that of ketoprofen alone.  相似文献

The interactions of nabumetone (NAB) with -cyclodextrin (-CD) and-cyclodextrin (-CD) were studied in aqueous solution by meansof phase-solubility analysis. Solid dispersions of NAB with -cyclodextrin(-CD), -cyclodextrin (-CD), methyl- (M-CD),hydroxypropyl-cyclodextrin (HP-CD) were prepared by coevaporationand kneading and also by coprecipitation in the case of -CD. X-ray diffractometry, thermal analysis and infrared spectroscopy (FTIR) were used to study the possibility of complexation of the drug with the different cyclodextrins. Solid dispersions of nabumetone with -CD showed a remarkable improvement in the dissolution rate of nabumetone.  相似文献

The fluorescence quenching of naphthalene, 1-methylnaphthalene, and acenaphthene by trimethylamine (TMA) was studied in aqueous -cyclodextrin (-CD) solutions to know the structural requirements for guest molecules to form three-component complexes. The apparent rates for the fluorescence quenching of the naphthalene derivatives by TMA markedly increased in the presence of -CD. The fluorescence quenching of 1-methylnaphthalene and acenaphthene by TMA was accelerated by -CD more efficently than that of naphthalene. These results suggest the structures of the three-component complexes as the arene-capped -CD including TMA in its cavity.  相似文献

Menthone on reduction with sodium dithionite,showed a good amount of mentholformation in the water/DMF system, withincreasing -cyclodextrin (-CD)concentration from 47.0% for 0.1 equivalent of-CD to 93.5% for 1 equivalentof -CD. Increasing hydroxypropyl--cyclodextrin(HP-CD) gave higher menthol/neomenthol (M/N) ratiosfrom 2.8 to 3.5. In the case of pulegone,increasing -CD showed an increase in the formationof menthols in thewater/DMF system from 13.3% for 0.1 equivalent of-CD to 78.1% for 1equivalent of -CD with greater proportions ofneomenthol and neoisomenthol.However, HP-CD which showed only marginalenhancement in the formationof menthol from menthone (32.1–41%), exhibiteda greater proportion of mentholformation in the case of pulegone (55.1%).However, the phase-transfer capabilityof HP-CD was not found to be significant.  相似文献

Heat capacities of anhydrous -and -cyclodextrin were measured by adiabaticcalorimetry between 10 K and 300 K. The thermalbehaviour of the two compounds exhibits significantdifferences. -Cyclodextrin shows an anomalousexcess heat capacity in the entire region between 70 Kand 210 K. In the case of -cyclodextrin, anendothermic effect is observed at 240K. This effect isanalysed through the study of the correspondingentropy change and discussed in terms ofintramolecular organization.Using the known heat capacity values of anhydrous-CD, a comparative analysis has been developed.For each cyclodextrin, the average behaviour of abound -D-glucopyranose has been calculated andcompared. The complexation process between gentian violet (CV+) and four different cyclodextrins (-, -, -, and HP--CDs) has been investigated under different reaction conditions (pH, solvent, and temperature) by electronic absorption and 1H NMR (NOE and NOESY) spectroscopies. All the binding constants were determined by the direct spectroscopic method. The H and S complexation values have been evaluated and discussed according to the diverse factors that affect the chemical interactions in these systems. A simple association takes place between the secondary hydroxyl or the hydroxypropyl groups of and HP--cycloamyloses, respectively, with the amine group of the gentian violet, while the binding between CV+ and - or -CDs corresponds to a real inclusion. Also, a CV22+ dimmeric species within the -CD cavity was detected in aqueous solution, while two molecules of -CD react with one molecule of gentian violet in DMSO at 294 K. In all the reaction media the -CD forms 1 : 1 complexes, but in the buffered aqueous solution at pH 7.5 the inclusion is deeper than in the other solvents. It is important to point out that the solvophobic effect is the most important binding factor in the complexation of the CV+ with the - and HP--CDs, while the complexes with -, and -cyclodextrins are mainly stabilized by van der Waals interactions between the guest and the host cavity. In all cases, the inclusion orientation is probably determined by the ion-dipole interactions between gentian violet and the solvent.  相似文献

Sterically controlled styrene-acrylonitrile (SAN)copolymers were prepared by inclusion polymerizationusing molar equivalents of -cyclodextrin(-CD) and its derivative. Comonomercompositions, reactivity ratios, monomer sequence andaverage styrene and acrylonitrile sequence lengthswere determined using 13C NMR spectroscopy. Themolecular weight of the polymers prepared increasedwith decreasing -CD concentration, but wasfound to be less than the control. With increase inthe styrene: -CD ratios the averageacrylonitrile sequence length and run number of thepolymers increased. The glass transition temperature(g) also increased along with the meltingtemperature (m) of the polymers at higherstyrene:-CD ratios.  相似文献

Effects of Cyclodextrins on Deodoration of ``Aging Odor'   总被引:2,自引:0,他引:2  
Hexenal, octenal and nonenal are known causes of unpleasant body odor and are present at markedly increased levels in the middle-aged and elderly. The odor of such unsaturated aldehydes is therefore called ``aging odor'. The present study investigated the effects of cyclodextrins (CDs) on deodoration of crotonaldehyde, pentenal, hexenal, heptenal, octenal, nonenal, decenal, undecenal and dodecenal. -, - and -CD formed inclusion complexes with the majority of unsaturated aldehydes in aqueous solution. The -CD inclusion complex contained the highest amount of guest molecule. One molecule of -CD was estimated to include 1 molecule of short chain aldehyde and 2 molecules of long chain aldehyde. Deodorant testing was conducted by headspace gas analysis using sealed vials. All CDs decreased the concentrations of unsaturated aldehyde. With nonenal, the deodorant power of parent CDs was -CD > -CD > -CD, and that of chemically modified CD was Me-CD > HP-CD > G2-CD > MCT-CD. CDs were demonstrated to reduce ``aging odor'. Me-CD was the most effective type of CD for the deodoration of ``aging odor'.  相似文献
