The synthesis of the novel oxazole building block, 4-bromomethyl-2-chlorooxazole, and its palladium-catalysed cross-coupling reactions to make a range of 2,4-disubstituted oxazoles, is described. Selectivity for the 4-bromomethyl position is observed, with Stille coupling effected in good to excellent yields, or Suzuki coupling in moderate yields, to provide a range of 4-substituted-2-chlorooxazoles. Subsequent coupling at the 2-chloro-position can be achieved through either Stille or Suzuki reactions in excellent yields.  相似文献

A soluble perylene monoanhydride diester has been designed and synthesized. It was successfully utilized as an efficient intermediate for the synthesis of the first unsymmetric perylene tetracarboxylic tetraester and a series of unsymmetric perylene diester monoimides in good to excellent yields. Its application in synthesis of unsymmetric perylene diimides was also demonstrated. Availability of such a versatile intermediate would enable the convenient integration of perylene tetracarboxylic tetraesters and perylene biester monoimides into intricate functional molecular systems.  相似文献

A scheme combining the preparation of building blocks in solution followed by solid-phase combinatorial chemistry has been developed to side-chain diversify 5-(hydroxymethyl)oxazole scaffold (1) into aryl ethers, thioethers, sulfones, sulfonamides, and carboxamides. Protected heterocyclic scaffolds 2 were linked to the solid phase and N-terminal derivatized using active ester chemistry, providing chemset 4?1-4,1-4?. The free side-chain hydroxyl of 4 was smoothly converted to aryl ethers 6 under Mitsunobu conditions, with a broad range of substituted phenols. Alternatively, quantitative conversion of hydroxyl to bromide followed by displacement with alkyl and aryl thiols gave thioethers 8. Thioethers were optionally oxidized to sulfones 9. Bromide displacement by azide, followed by reduction to amine and acylation with a range of carboxylic acids and sulfonyl chlorides gave carboxamides 11 and sulfonamides 13, respectively. Crude purity at typically >90% was observed for each of the five modifications detailed. A series of 20 compounds, exemplifying each modification, was reprepared, purified, and fully characterized.  相似文献

A practical synthesis of 7-mono-substituted quinolines has been achieved. Selective reduction of the inexpensive commercial reagent 4,7-dichloroquinoline affords 7-chloroquinoline, which has been converted into more complex 7-mono-substituted quinolines through a series of Pd-catalyzed cross coupling reactions. These studies include the first examples of Suzuki reactions for the preparation of 7-mono-substituted quinolines as well as the first application of the Sonagashira reaction for the synthesis of 7-substituted quinolines. This strategy has been extended to the preparation of 2,7-di-substituted quinolines.  相似文献

A novel general synthetic route to tetraaryltetrabenzoporphyrins (Ar(4)TBP) with various peripheral functional groups is developed. The procedure includes (i) Barton-Zard condensation of 1-nitro- or 1-phenylsulfonylcyclohexenes with isocyanoacetic acid esters, (ii) condensation of the resulting 4,5,6,7-tetrahydroisoindoles with aromatic aldehydes to give fused tetraaryltetracylohexenoporphyrins (Ar(4)TCHP), and (iii) aromatization of the metal complexes of Ar(4)TCHP's into the corresponding Ar(4)TBP's. Cu and Zn complexes of Ar(4)TBP's are further demetalated to give the corresponding Ar(4)TBP free bases. The overall yields for the sequence range from 15% to 40%, making the method suitable for the preparation of gram quantities of Ar(4)TBP's in a single run. The scope of the method, the selection of the peripheral substituents, the choice of the metal ions, and their influence on the yields of aromatization are discussed. The basic spectroscopic properties of newly synthesized Ar(4)TBP's and Ar(4)TCHP's are reported together with the first X-ray crystallographic structure of the NiAr(4)TBP complex.  相似文献

Convenient access to 1,3-diacetyl-2-methylcyclopentene 2 is reported by three routes: acetylation of methylcyclopentene ( or cyclohexane), condensation of acetylacetone with 1,2-dibromoethane, and the isomerisation of O-acrtylated dienolate of 1-actyl-2-methylcyclopentene. A straightforward route for preparing derivatives of ethyl 5-trichloromethyl-1,2,4-oxadiazole-3-carboxylate, in which the 5-trichloromethyl and/or the ester functionalities are displaced by various nucleophiles, has been designed. Mild reaction conditions were chosen to enable the formation of products, which constitute a new series of bifunctional 3,5-disubstituted 1,2,4-oxadiazole building blocks capable of being integrated into biologically relevant molecules.  相似文献

The key steps involve cyclization of a suitably functionalized glutamic acid derivative and concomitant alkylation of the resulting beta,beta-diketoester system, C-acylation of the cyclic alpha-amidoketone 9, and decarboxylbenzylation of 12. Phenacyl bromides are used as a building blocks in synthetic organic chemistry, which serves as a key model for the development of various biologically important heterocyclic compounds and other industrially important scaffolds. This review covers recent synthetic advances in the preparation of heterocyclic compounds utilizing various phenacyl bromides. A wide range of five- and six-membered heterocyclic compounds as well as fused heterocyclic compounds were synthesized efficiently from the phenacyl bromide precursors through one-pot multicomponent reactions.  相似文献

An efficient and versatile method has been developed for the synthesis of 1-aminohydantoin derivatives which can be substituted at all the possible positions of the hydantoin ring. The starting materials are aldehydes, ketones, carboxylic acids and hydrazides as well as isocyanates readily available from commercial sources. The semicarbazide-type reaction product of an N-acyl-N′-(1-cyanoalkyl)hydrazine, obtained from the above materials by methods known from the literature, and an isocyanate is cyclized in the presence of a basic catalyst to yield 1-acylamino-4-imino-2-oxoimidazolidine derivatives whose acid catalyzed hydrolysis leads to 1-aminohydantoin derivatives in good to excellent yields. The last two steps are carried out in a single reaction medium.  相似文献

The synthesis of racemic nitrile (+/-)-9 was accomplished in four steps and 58% overall yield from the known pyrrolidinone 5. Nitrile (+/-)-9 was resolved via preparative chiral HPLC to afford optically pure nitriles (+)-9 and (-)-9, from which 3,3-dimethylprolines (+)-1 and (-)-1 and 3,3-dimethylprolinamides (+)-2 and (-)-2 could be accessed in nearly quantitative yield, without loss of optical purity. The absolute configurations of the resolved prolines and prolinamides were determined by correlation with an intermediate of known absolute stereochemistry.  相似文献
