Three new homoisoflavanones, 1 – 3 , together with a known one, 4 , were obtained from the AcOEt extract of the tuberous roots of Ophiopogon japonicus (Liliaceae). They were identified as (3R)‐2,3‐dihydro‐7‐hydroxy‐5‐methoxy‐3‐(4‐methoxybenzyl)‐6,8‐dimethyl‐4H‐chromen‐4‐one ( 1 ), (3R)‐3‐(1,3‐benzodioxol‐5‐ylmethyl)‐2,3‐dihydro‐7‐hydroxy‐5‐methoxy‐6,8‐dimethyl‐4H‐chromen‐4‐one ( 2 ), (3R)‐3‐(1,3‐benzodioxol‐5‐ylmethyl)‐2,3‐dihydro‐7‐hydroxy‐5‐methoxy‐6‐methyl‐4H‐chromen‐4‐one ( 3 ), and ophiopogonanone A ( 4 ). Their structures were determined on the basis of extensive NMR‐spectroscopic and mass‐spectrometric analyses. The three new compounds are rare homoisoflavanones which contain a MeO group at C(5). Compounds 1 and 2 showed weak cytotoxicity against the HepG2 (human hepatoma G2), KB (human oral epidermoid carcinoma), and MCF‐7 (human breast adenocarcinoma) cell lines in an MTT assay. Compound 3 exhibited weak cytotoxicity against HepG2 and MCF‐7, and moderate cytotoxicity against KB cell lines. Compound 4 showed moderate cytotoxicity against HepG2, KB, and MCF‐7 cell lines.  相似文献

Compounds 4a-4d, 4f and 10 were prepared and their ring-opening reactions with N-bromosuccinimide (NBS) investigated. Compounds 4a and 4b gave a mixture of products which did not contain any significant quantity of the corresponding aldehydes 5a and 5b whereas compounds 4c, 4d and 4f gave exclusively the aldehydes 5c, 5d and 5f respectively. Compound 10 similarly gave the aldehyde 11 when treated with NBS.  相似文献

Several quinoxaline 1,4-di-N-oxides have been shown to be efficient and selective cytotoxins for hypoxic cells. We present now a series of 4-cyano-2-oxo-1,2,4-oxadiazolo[2,3-a]quinoxaline 5-N-oxides 2a-2k . They were prepared starting from 3-amino-2-quinoxalinecarbonitrile 1,4-di-N-oxides 1a-1k and 2-chloroethyl isocyanate in dry dioxane at 100–110°. A reaction mechanism is proposed. The treatment of 1a with phenyl isocyanate afforded 2a . Reaction of 2c with silica gel yielded 1c . Compounds 2a-2g were heated in the presence of ethanol and 2-propanol giving the corresponding carbamates 3a-3g and 4a-4g . Compound 2d was already obtained by heating a mixture of 1d and ethyl chloroformiate. Compound 2b was prepared when the carbamate 3b was heated at 150°. Quinoxalines were tested as cytotoxic agents both in oxic and hypoxic cells. The most interesting compounds were 3g and 4g .  相似文献

Antioxidant hispidin derivatives from medicinal mushroom Inonotus hispidus   总被引:1,自引:0,他引:1  
Two natural antioxidants, named inonotusin A (1) and B (2), were isolated from the methanolic extract of the fruit bodies of Inonotus hispidus, together with (E)-4-(3,4-dihydroxyphenyl)but-3-en-2-one (3), hispidin (4) and 3,4-dihydroxybenzaldehyde (5). Their structures were identified by means of extensive NMR and MS data analysis. Compounds 1, 2 and 4 exhibited significant scavenging activity against the 2,20-azinobis(3-ethylbenzhiazoline-6-sulfonate) (ABTS) radical cation. Compound 1 also showed moderate cytotoxicity against a human breast carcinoma cell line (MCF-7) with IC(50) values of 19.6 μM.  相似文献

A new cardenolide diglycoside (1) was isolated from Nerium oleander together with ten known cardenolide diglycosides 2-11. The structure of compound 1 was established on the basis of their spectroscopic data. The in vitro anti-inflammatory activity of compounds 1-11 was examined on the basis of inhibitory activity against the induction of the intercellular adhesion molecule-1 (ICAM-1). Compounds 2-5 were active at an IC(50) value of less than 0.8 μM. The cytotoxicity of compounds 1-11 was evaluated against three human cell lines normal human fibroblast cells (WI-38), malignant tumor cells induced from WI-38 (VA-13), and human liver tumor cells (HepG2). Compound 3 was active toward VA-13 cells, and compounds 2-5 were active toward HepG2 cells at IC(50) values of less than 1.3 μM. The multidrug resistance (MDR)-reversal activity of compounds 1-11 was evaluated on the basis of the amount of calcein in MDR human ovarian cancer 2780AD cells in the presence of each compound. Compounds 1 and 8 showed moderate effects on calcein accumulation.  相似文献

Two new eremophilenolides, 6beta,8beta,10beta-trihydroxyeremophil-7(11)-en-12,8-olide (1) and 3beta-acetoxy-8alpha-hydroxy-6beta-methoxyeremophila-7(11),9-dien-12,8-olide (2) along with twenty-nine other compounds were obtained from the methanolic extracts of the aerial parts and rhizomes of Farfugium japonicum (L.) Kitam. var. formosanum (Hayata) Kitam. (Compositae = Asteraceae). The structures of the isolated compounds were characterized and identified by spectral techniques. Compounds 5, 6, 10, 12-24, 29, and 30, were reported for the first time from this genus. Cytotoxicity and anti-inflammatory activity of the isolated compounds were evaluated. Compounds 3 and 16 possessed moderate cytotoxicity against human breast cancer cell line (MCF 7). Compounds 3, 16, 25 and 26 exhibited moderate cytotoxicity against hepatoma cells (Hep G2 and Hep 3B). With respect to the anti-inflammatory activity, compounds 15 and 16 (each 10 microg/mL) inhibited superoxide anion generation by human neutrophils in response to fMLP/CB by 92.0% and 87.3%, respectively.  相似文献

Two series of territrem B analogues (6, 11) were designed and synthesized from jujubogenin 2. Compound llc inhibited AChE with the ratio of 70% at 10^-4 mol/L. Compounds5a, 5b, 6a and lib showed moderate cytotoxicity on cultured KB cells at 10^-6 mol]L.Compounds 5c and 6b alleviated injury arising from oxygen-glucose deprivation (OGD).  相似文献

The synthesis of a series of 1,5-dichloro-9(10H)-anthracenones bearing O-linked and N-linked substituents in the 10-position are described. Previous studies have shown that 9-acyloxy 1,5-dichloroanthracenes and 9-acyloxy 1,8-dichloroanthracenes displayed a potential cytotoxic effect. These results have encouraged us in further investigation of potential anthracenone derivatives. Therefore, a series of 10-substituted 1,8-dichloro-9(10H)-anthracenone derivatives were synthesized. These compounds were evaluated for their ability to inhibit the growth of human oral epidermoid carcinoma cells (KB cell line), human cervical carcinoma cells of ME 180 (GBM 8401) and Chinese hamster ovary (CHO) cells, respectively. Compounds 3c and 4c of this series compare favorably in the KB cellular assay with mitoxantrone. Compound 4c showed combined inhibitory action against KB, GBM and CHO cell growth, respectively. In addition, redox property of the compounds for the inhibition of lipid peroxidation in model membranes was determined. Compounds 4b and 4d exhibited stronger antioxidant activity than ascorbic acid, (+)-alpha-tocopherol and mitoxantrone, respectively.  相似文献

A new series of novel (2S)-2-({2-[1,4-dihydro-3,5-bis(methoxycarbonyl)-2,6-dimethyl-4-(2-nitrophenyl)pyridin-1-yl]-2-oxoethyl}amino)-3-(4-hydroxyphenyl) propanoic acid (3a) and its analogues 3b-j has been synthesized. These compounds were evaluated for their in vitro antioxidant activity, anti-inflammatory activity and antiulcer activity. Compounds 3b and f exhibited significant antioxidant action comparable with that of standard. Efficacy against inflammation and ulceration was also found to be significant. The chemical structures of these compounds were confirmed on the basis of spectral data.  相似文献

A novel series of diaryl biuret derivatives containing a tetrazole moiety was designed and synthesized.All the target compounds were evaluated for their in vitro antitumor activity against HT-29,HepG2,MCF-7 and A549 cells by MTT assay.Most of them exhibited obvious antitumor activity,and four of them(4a,4c,4h and 7a)were superior to sorafenib in general.Among them,Compound 4h displayed more potent activity than sorafenib in all tested cancer cells.Compound 4c exhibited the most outstanding activity in inhibition of growth of HepG2 cells(IC50=0.55 μmol/L).Further,they both revealed favorable metabolic stability in in vitro assay.Compounds 4c and 4h are promising candidates for further development.  相似文献

To improve the biological anticancer activity of 20(S)-camptothecin, a novel class of 20(S)-20-O-camptothecin β-aminopropionates were designed and synthesized with camptothecins as the starting materials, through acylation with acryloyl chloride followed by Michael′s addition. Twelve esters, 1a-1d, 4a-4d and 5a-5d, were synthesized and evaluated by using MTT assay method. The results demonstrate that all these compounds show a potential cytotoxicity on KB, HT-29, HCT-8, and Bel7402 tumor cell lines. Some compounds, 1d, 4c, 5b, 5c and 5d, show a higher cytoto-xicity on KB and HCT-8 compared with camptothecin.  相似文献

A new anthraquinone, 4‐hydroxy‐1,2,3‐trimethoxy‐6‐methylanthracene‐9,10‐dione ( 1 ), and two novel tetrahydroanthraquinones, prisconnatanones A and B ( 2 and 3 , resp.), together with 15 known anthraquinones, 4 – 18 , were isolated from the roots of Prismatomeris connata. Their structures were elucidated on the basis of spectroscopic data. Compounds 1 – 18 were evaluated for their cytotoxicity against A549 and LAC human cancer cell lines. Convenient synthetic procedures for preparing two kinds of methyl-substituted lariat ethers containing a 13-crown-4 ring, 1 and 2 , are described. Compounds 1a-1c were obtained from the reaction of 2-bromomethyl-2-methyl-13-crown-4 ( 5 ) with the appropriate alkoxide. Compound 5 was prepared without the need for prior protection of the bromomethyl group. For the synthesis of compounds 2a-2c , which possess an electron-donating group on the central carbon of the trimethyl moiety of the 13-crown-4 ring, the substituents were introduced before cyclization.  相似文献

以2-巯基苯并咪唑(1)为原料,经缩合和还原得到2-(4-氨基苯硫基)苯并咪唑(3),再与异硫氰酸苯甲酰酯或异硫氰酸烃基酯反应得到取代硫脲(5和7),最后与卤代烃反应得到20个新的S-烃基-1-烃基-3-[4-(苯并咪唑-2-巯基)苯基]异硫脲化合物(6和8),其结构经IR,1HNMR,MS及元素分析确证.初步的药理试验表明,20个目标化合物均有不同程度的iNOS抑制活性,其中化合物6b,8d和8f的iNOS抑制活性与阳性对照药氨基胍相当.  相似文献

Novel pyridine derivatives 1d and 7 , pyrido[2,3‐d]pyrimidine derivatives 2a , 2b , 2c , 2d , 3a , 3b , 3c , 3d , 4a , 4b , 4c , 4d , 5a , 5b , 5c , 5d , 6a , 6b , 6c , 6d , 8a , 8b , and 9a , 9b were synthesized and screened for their anti‐microbial and cytotoxic activities. Compounds 2b , 2c , 3b , 3c , 5b , 5c , 6b , 6c , and 7 showed excellent anti‐microbial activities against all the tested bacteria and fungi compared to the reference drugs. Furthermore, they exhibited high safety profile in cytotoxicity test except 2c and 3c . The structures of the newly synthesized compounds were confirmed by spectral data and elemental analysis.  相似文献

The neutral pentacoordinate monoamidinatosilicon(IV) complex 1 (SiN(2)Cl(3) skeleton) and the neutral hexacoordinate monoamidinatosilicon(IV) complexes 2-9 (SiN(3)OF(2), SiN(3)OCl(2), SiN(3)OBr(2), SiN(5)O and SiN(3)O(3) skeletons) were synthesised and characterised by elemental analyses, single-crystal X-ray diffraction (except for 1) and NMR spectroscopy in the solid state and in solution. Compounds 2-9 contain one bidentate monoanionic N,N'-diisopropylbenzamidinato ligand, one bidentate monoanionic ligand derived from 8-hydroxyquinoline and (i) two identical monoanionic ligands (F, Cl, Br, N(3), NCO, NCS, OSO(2)CF(3)) or (ii) one bidentate dianionic benzene-1,2-diolato ligand. The dynamic behavior of 2-4 (SiN(3)OX(2) skeleton; X = F, Cl, Br) and 9 (SiN(3)O(3)) in solution was studied by multinuclear variable-temperature NMR experiments. Compound 1 was synthesised by reaction of SiCl(4) with the corresponding lithium amidinate, and compound 2 was obtained by reaction of 1 with 8-hydroxyquinoline and triethylamine. Compound 2 served as the starting material in the syntheses of 3-9, in which the two chloro ligands of 2 were substituted by two identical (pseudo)halogeno ligands, two trifluoromethanesulfonato ligands or one benzene-1,2-diolato ligand. Compounds 3 and 4 contain the novel SiN(3)OBr(2) and SiN(3)OF(2) skeletons, while compounds 5-7 are the first neutral hexacoordinate silicon(IV) complexes with an SiN(5)O skeleton.  相似文献

A series of novel 3-[4-phenyl-1-piperazinyl]-1-[5-methyl-1-(2-pyrimidinyl)-4-pyrazolyl]-1-trans-propenes and related compounds were synthesized and evaluated by their cytotoxic activity against several tumor cell lines in vitro and in vivo antitumor activity against some tumor models when administered both intraperitoneally and orally. Compounds with the 3-chloropyridin-2-yl group (9g) and the 3-fluoro-5-substituted phenylpiperazinyl group (29b, c, and e) showed significantly potent cytotoxicity by in vitro testing. Among them, the 3-cyano-5-fluorophenyl derivative (29b) exhibited potent antitumor activity against several tumor cells including human carcinoma without causing undesirable effects in mice.  相似文献
