An improved version of the DEPTQ experiment yielding the signal and multiplicity information for all carbon types including the signals of quaternary carbons is proposed. It encompasses all the known advantages of the basic DEPT experiment. In comparison to the original version, signals of the sensitivity-limiting quaternary carbons are markedly increased: the initial 13C pulse may be adjusted to the Ernst angle, the NOE build-up period is prolonged by the split relaxation delay and a partial recovery of signal losses due to instrumental imperfections is achieved by the incorporation of composite adiabatic 13C refocussing pulses. Furthermore, pure absorption lineshapes for all carbon types are obtained with only one single scan. These attributes make this experiment attractive for 13C analysis of small molecules (including spectral editing), particularly in high-throughput analysis laboratories.  相似文献

The composition of fluorescent polymer nanoparticles, commonly referred to as carbon dots, synthesized by microwave-assisted reaction of citric acid and ethylenediamine was investigated by 13C, 13C{1H}, 1H─13C, 13C{14N}, and 15N solid-state nuclear magnetic resonance (NMR) experiments. 13C NMR with spectral editing provided no evidence for significant condensed aromatic or diamondoid carbon phases. 15N NMR showed that the nanoparticle matrix has been polymerized by amide and some imide formation. Five small, resolved 13C NMR peaks, including an unusual ═CH signal at 84 ppm (1H chemical shift of 5.8 ppm) and ═CN2 at 155 ppm, and two distinctive 15N NMR resonances near 80 and 160 ppm proved the presence of 5-oxo-1,2,3,5-tetrahydroimidazo[1,2-a]pyridine-7-carboxylic acid (IPCA) or its derivatives. This molecular fluorophore with conjugated double bonds, formed by a double cyclization reaction of citric acid and ethylenediamine as first shown by Y. Song, B. Yang, and coworkers in 2015, accounts for the fluorescence of the carbon dots. Cross-peaks in a 1H─13C HETCOR spectrum with brief 1H spin diffusion proved that IPCA is finely dispersed in the polyamide matrix. From quantitative 13C and 15N NMR spectra, a high concentration (18 ± 2 wt%) of IPCA in the carbon dots was determined. A pronounced gradient in 13C chemical-shift perturbations and peak widths, with the broadest lines near the COO group of IPCA, indicated at least partial transformation of the carboxylic acid of IPCA by amide or ester formation.  相似文献

In the chemistry literature it is common to provide NMR data on both proton and carbon spectra based on one‐dimensional experiments, but often only proton spectra are assigned. The absence of a complete attribution of the carbons is in good part due to the difficulty in reaching the necessary resolution in the carbon dimension of two‐dimensional experiments. It has already been shown that high‐resolution heteronuclear spectra can be acquired within nearly the same acquisition time using a violation of the Nyquist condition. For a spectral width reduction by a given factor k, the resolution increases by the same factor as long as it is not limited by relaxation. The price to pay for such an improvement is a k‐fold ambiguity in the chemical shift of the signal along the folded or aliased dimension. The computer algorithm presented in this paper takes advantage of the peak list stemming from one‐dimensional spectra in order to calculate spectral widths for which the ambiguities in the aliased dimension of heteronuclear experiments are eliminated or at least minimized. The resolution improvement factor is only limited by the natural lineshape and reaches a typical value higher than 100. The program may be set to run automatically on spectrometers equipped with automatic sample changers. Applications to short‐range HSQC experiments and long‐range HMBC spectra of steroids, carbohydrates, a peptide and a mixture of isomers are shown as examples. A new version of the two-dimensional INADEQUATE experiment was designed for detection of small couplings between equivalent carbon atoms separated in the molecule by several bonds, where other techniques fail due to rich line splitting and mutual peak cancellation in many molecules. As the proposed method is suitable for detection of couplings in strongly coupled systems in general, we propose the name STRONG INADEQUATE in the paper. Similar to other methods for detection of couplings between equivalent carbons, the STRONG INADEQUATE experiment utilizes one-bond carbon–proton coupling for creation of the effective chemical shift differences. The STRONG INADEQUATE experiment works superbly for nJCC, where n ≥ 3. Then the F1 pattern is reduced to a simple antiphase doublet with nJCC separation, and this pattern is also preserved when a symmetrical HC···C′H′ system is coupled to other protons. Even in the measurement of 2JCC couplings, the STRONG INADEQUATE experiment generates a much simpler pattern than the original pulse sequences for measurement of couplings between equivalent carbons.  相似文献

A set of modified HSQC experiments designed for the study of 13C‐enriched small molecules is introduced. It includes an improved sensitivity‐enhanced HSQC experiment eliminating signal artifacts because of high‐order 13C magnetization terms generated at high 13C enrichment. A broadband homonuclear 13C decoupling sequence based on Zangger and Sterk's method simplifies the complex 13C–13C multiplet structure in the F1 dimension of HSQC. When recording spectra at high resolution, the combination with a multiple‐site modulation of the selective pulse outperforms the constant‐time HSQC in terms of sensitivity and reliability. Finally, two pulse sequences reintroducing selected JCC couplings with selective pulses facilitate their assignments and measurements either in the splitting of the resulting doublets or by modulation of the signal amplitude. A new experiment for the single‐scan measurement of diffusion coefficients is presented. The principle is to introduce a spatial variation in the parameters of a conventional pulse sequence, so that all of the scans required to determine some physical parameter can be recorded simultaneously from different parts of the sample. The spectrum is acquired in the presence of a weak read gradient so that the resulting lineshapes contain the information required. The pulse sequence is described in detail and demonstrated on a sample containing three components; its advantages and limitations are discussed in relation to those of existing techniques. For uncrowded spectra with high signal‐to‐noise ratio, this experiment provides an order of magnitude reduction in experiment time compared with conventional methods and is likely to be of most benefit where samples are changing rapidly with time or where a long period of polarization, which may be difficult to reproduce accurately, prohibits the use of multiple‐scan techniques. Performance of homonuclear coupling sign edited (HCSE) experiment applied to detection of signed carbon–carbon couplings is discussed using a set of already measured samples of nine monosubstituted benzenes. It is shown that coupling sign detection is insensitive to the settings of carbon–carbon polarization transfer delays. The HCSE spectra of ten from the total of 43 measured carbon–carbon couplings were considerably influenced by relaxations and proton–proton strong couplings. These effects are quantitatively discussed. The results of HCSE and SLAP experiments are compared. N‐acetyl‐4‐nitrotryptophan methyl ester (2), N‐acetyl‐5‐nitrotryptophan methyl ester (3), N‐acetyl‐6‐nitrotryptophan methyl ester (4) and N‐acetyl‐7‐nitrotryptophan methyl ester (5) were synthesized through a modified malonic ester reaction of the appropriate nitrogramine analogs followed by methylation with BF3‐methanol. A one‐step method was reported for the synthesis of 6‐acetamido‐3‐(N‐(2‐(dimethylamino) ethyl) sulfamoyl) naphthalene‐1‐yl 7‐acetamido‐4‐hydroxynaphthalene‐2‐sulfonate by treating 7‐acetamido‐4‐hydroxy‐2‐naphthalenesulfonyl chloride with equal moles of N, N‐dimethylethylenediamine in acetonitrile in the presence of K2CO3. The chemical structure of the obtained compounds was characterized by MS, FTIR, 1H NMR, 13C NMR, gCOSY, TOCSY, gHSQC, and gHMBC. cis‐ and trans‐2‐imino‐1,3‐ and ‐3,1‐perhydrobenzoxazines and the N‐methyl derivatives of the latter were synthesized from the corresponding cyclic 1,3‐amino alcohol with cyanogen bromide. The configurations of the studied compounds were confirmed by 1H and 13C NMR spectra. All trans‐fused compounds exist in biased chair–chair conformations as expected, whereas the cis‐fused 1,3‐benzoxazines attain exclusively the O‐in conformations. The cis‐fused 3,1‐benzoxazines, especially the 1‐methyl‐substituted derivatives, tend to favor the N‐out form, obviously owing to the favorable axial orientation of this N‐methyl. Experiments for 1H‐detected heteronuclear 1H,X correlation spectroscopy with 31P‐relayed coherence transfer are described which allow the indirect detection of δX and nJ(X,P) even in the absence of a direct J(X,H) coupling. Signed values of all intra‐ring 2,3,4J(C,C) couplings in nine monosubstituted benzenes (C6H5‐X where X = F, Cl, Br, CH3, OCH3, Si(CH3)3, C ≡ N, NO, NO2) are experimentally determined as well as nine couplings to substituent carbons. It is confirmed that while all the vicinal intra‐ring 3J(C,C) are positive and all geminal 2J(C2,C4) are negative, both signs are found for geminal 2J(C1,C3) couplings. We report the 1H NMR and 13C NMR chemical shifts and J(H,H), J(H,F) and J(C,F) coupling constants of 13 2,4‐diamino‐10‐methylpyrimido[4,5‐b]‐5‐quinolone derivatives, some of them with moderate activity against Plasmodium falciparum in vitro. A modified pulse field gradient (PFG)‐enhanced inverse (1H)‐detected 2D heteronuclear Overhauser effect spectroscopy (HOESY) pulse sequence is demonstrated for the acquisition of 1H–7Li heteronuclear correlations. In practice, t1 noise artifacts were observed using the original PFG‐enhanced inverse‐detected HOESY pulse sequence, which degraded the ability to detect accurately weak heteronuclear Overhauser signals. Experimentally it is shown that a simple modification of the PFG‐enhanced inverse‐detected HOESY pulse sequence greatly reduces the t1 noise that may result from variations in magnetic susceptibility, and allows improved detection of weak 1H–7Li correlations. A combination of homo‐ and heteronuclear 1D and 2D NMR techniques provided the assignment of the 1H and 13C resonances of the major component of a reaction product consisting of the two possible diastereomers of (5S)‐1‐[2‐(2‐hydroxyethyl)tetrahydropyran‐5‐yl]‐1H‐pyrimidine‐2,4‐dione and showed that the tetrahydropyranyl ring in the major 5S,2S‐isomer adopts the twist conformation.
