The model morpholine‐1‐carbothioic acid (2‐phenyl‐3H‐quinazolin‐4‐ylidene) amide (1) reacts with phenacyl bromides to afford N4‐(5‐aryl‐1,3‐oxathiol‐2‐yliden)‐2‐phenylquinazolin‐4‐amines (4) or N4‐(4,5‐diphenyl‐1,3‐oxathiol‐2‐yliden)‐2‐phenyl‐4‐aminoquinazoline ( 5 ) by a thermodynamically controlled reversible reaction favoring the enolate intermediate, while the 4‐[4‐aryl‐5‐(2‐phenylquinazolin‐4‐yl)‐1,3‐thiazol‐2‐yl]morpholine ( 8 ) was produced by a kinetically controlled reaction favoring the C‐anion intermediate. 1H nmr, 13C nmr, ir, mass spectroscopy and x‐ray identified compounds ( 4 ), ( 5 ) and ( 8 ).  相似文献

A series of twelve new 7‐chloro‐5‐[(o‐ and p‐R1)phenyl]‐1‐R2‐3H‐[1,4] benzo‐diazepin‐2‐ones, which have possible pharmacological properties were synthesized. The synthesis of all the final compounds was carried out by four steps. The structure of all final products was corroborated by ir, 1H nmr, 13C nmr and ms, and have been obtained in 35‐94% yield.  相似文献

The 1H and 13C nmr spectra of the rotational isomers 3a and 3b of 6‐N‐methyl‐N‐formylaminomefhyl)‐thioquinanthrene were completely assigned with a combination of 1D and 2D nmr techniques. The key‐parts of this methodology were long‐range proton‐carbon correlations and NOE experiments with N‐methyl‐N‐formylaminomethyl substituent. The X‐ray study of 4‐methyl‐2‐N‐methyl‐N‐formylaminomethyl)quinoline 4a as well as 1H and 13C nmr spectra show that N‐methyl‐N‐formylaminomethyl substituent in 4a and 4b has a different steric arrangement than the same substituent in 3a and 3b .  相似文献

Benzo[h]pyrimido[4,5‐b]quinolines ( 3 ) have been synthesized via a regiospecific cyclocondensation reaction between 6‐aminopyrimidines ( 1 ) and 2‐dimethylaminomethylentetralone hydrochloride ( 2 ). The linear structure of the final compounds were determined by nmr measurements, especially by 1H,1H, 1H,13C COSY and DEPT experiments.  相似文献

The preparation and spectral properties often novel methyl 5‐[(o‐, m‐, and p‐substituted)‐phenylthio]‐2‐benzimidazolecarbamates with possible pharmacological activity as antihelmintics is described; by condensation and cyclization between 5‐methylthioures sulfate chloroformic acid methyl ester and 3,4‐diaminophenyl‐substituted‐phenylthio ether dissolved in ethanol. The ureidation reactions of 2‐ and 4‐picoline N‐oxides with 2‐chloro‐4,5‐dihydroimidazole are described. A mechanism of novel thioureidation reaction of 4‐picoline N‐oxide with 2‐(4,5‐dihydro‐1H‐imidazol‐2‐ylthioxy)‐4,5‐dihydro‐1H‐imidazole is proposed. Structural assignment is confirmed by 1H and 13C nmr as well as by X‐ray crystallography.  相似文献

N‐(Substituted aryl/cyclohexyl)‐N'‐[5‐bromo‐5‐nitro‐2‐oxido‐1,3,2‐dioxaphosphorinane‐2‐yl]ureas RR'P(O)NHC(O)NHR' (5) were synthesized by the reactions of 2‐bromo‐2‐nitro‐1,3‐propanediol (4) with chlorides of aryl/cyclohexyl carbamidophosphoric acids (3) in the presence of triethylamine at room temperature. Several 2‐alkylcarbamato/thiocarbamato/aryloxy/trichloromethyl‐2,3‐dihydro‐5‐propoxy‐1H‐1,3,2‐benzodiazaphosphole 2‐oxides ( 4 and 6 ) were synthesised by reacting 4‐propoxy‐o‐phenylenediamine ( 1 ) with various N‐dichlorophosphinyl carbamates ( 3 ), aryl phosphorodichloridates ( 5a‐f ) and trichloromethyl phosphonic dichloride ( 5g ) in the presence of triethylamine at 45‐65 °C. Their ir, 1H, 13C, 31P nmr and mass spectral data are discussed. The compounds were screened for antifungal activity against Curvularia lunata and Aspergillus niger and for antibacterial activity against Bacillus subtilis and Escherichia coli. Most of these compounds exhibited moderate activity in the assays.  相似文献

Regioselective reactions of morpholine‐1‐carbothioic acid (2‐phenyl‐3H‐quinazolin‐4‐ylidene) amide ( 1 ) with electrophiles and nucleophiles were studied. The compound ( 1 ) reacts with alkyl halides in basic medium to afford S‐substituted isothiourea derivatives, with amines to give 1,1‐disubstituted‐3‐(2‐phenyl‐3H‐quinazolin‐4‐ylidene) thioureas and l‐substituted‐3‐(2‐phenyl‐quinazolin‐4‐yl) thioureas via transami‐nation reaction. The reaction of ( 1 ) with amines in the presence of H2O2 provided N4‐disubstituted‐N'4‐(2‐phenylquinazolin‐4‐yl)morpholin‐4‐carboximidamide via oxidative desulfurization. Estimation of reactivity sites on ( 1 ) was supported using the ab initio (HF/6‐31G**) quantum chemistry calculations. The ir, 1H nmr, 13C nmr, mass spectroscopy and x‐ray identified the isolated products.  相似文献

A series of eleven new 2‐methylthio‐3H‐7‐[(o‐; m‐ and p‐substituted) phenoxy]‐4‐(p‐substituted‐phenyl)‐[1,5]benzodiazepines, which have potentially useful pharmacological activities, has been synthesized by condensing the 4‐[(o‐; m‐ and p‐R1)phenoxy]‐1,2‐phenylendiamines with 3,3‐dimercapto‐1‐(p‐R2‐phenyl)‐2‐propen‐1‐one. Afterward the lH‐[1,5]benzodiazepine‐2‐thiones obtained were treated with sodium hydride and methyl iodide. The structure of all products was corroborated by ir, 1H nmr, 13C nmr and ms.  相似文献

A new synthesis to obtain eleven novel derivatives of 11‐[(om‐ and p‐substituted)‐phenyl]‐8‐chloro‐3,3‐dimethyl‐2,3,4,5,10,11‐hexahydro‐1H‐dibenzo[b,e][1,4]diazepin‐1‐ones with possible pharmacological activity in the central nervous system in two efficient steps has been developed. The final products were obtained by condensation and cyclization between 3‐[4‐chloro‐1,2‐phenylenediamine]‐5,5‐dimethyl‐2‐cyclohexenone with (om‐ and p‐substituted)benzaldehyde. The synthesis of new pyrazolo[4,3‐c]β‐carbolines ( 8a,b ) is achieved by condensation of the appropriate aldehyde with 3‐(4‐amino‐1,3‐dimethylpyrazol‐5‐yl)indole ( 4 ) under Pictet‐Spengler reaction conditions. Regioselective cyclization occurred at the usual indole C‐2 position as evidenced from the 1H‐and 13C nmr spectra of 8a,b which lack the pyrrolic H‐2 signal, present in 4 (δ 7.26, 1H, d, Jch‐NH = 2‐5 Hz).  相似文献

Novel 6‐substituted 2,10‐dichloro‐4,8‐dinitrodibenzo[d,g][1,3,6,2]dioxathiaphosphocin‐6‐oxides 4 were synthesized by reacting 5,5′‐dichloro‐3,3′‐dinitro‐2,2′‐dihydroxydiphenyl sulfide ( 2 ) with different aryl phosphorodichloridates, trichloromethylphosphonic dichloride and O‐2‐chloroethyl phosphoryldichloride (3) in the presence of triethylamine at 55–60°. Some of these compounds are prepared by reacting the monochloride, 2,6,10‐trichloro‐4,8‐dinitrodibenzo[d,g][1,3,6,2]dioxathiaphosphoein‐6‐oxide ( 5 ) in situ with substituted phenols and thiols. 5 is prepared by condensing 2 with phosphorus oxychloride. The 1H nmr chemical shifts of the dibenzodioxathiaphosphocin moiety indicates the presence of more than one conformer in solution. However the presence of more than one conformer in each example cannot be entirely eliminated. Interestingly 4d on oxidation to 12‐sulphone by H2O2 in acetic acid medium yielded only 12‐sulphoxide 6a . The ir, 1H, 13C, 31P nmr and mass spectral data are discussed. Some of these compounds were screened for antifungal activity against Curvularia lunata and Aspergillus niger and antibacterial activity on Bacillus subtilis and Klebsiella pneumoniae. Naphtho[1,2‐b][1]benzothiophene‐6‐carboxylic acids, 6H‐benzo[b]naphtho[2,3‐d]thiopyran‐6‐ones and 6H‐benzo[b]naphtho[2,3‐d]pyran‐6‐ones were synthesized in one step by the photocyclization reaction of 3‐aryl‐2‐([1]benzothien‐3‐yl)propenoic acids. The photocyclization reaction did not occur when the 3‐aryl group contained the electron‐withdrawing nitro group. The assignment of the 1H and 13C nmr spectra of 6H‐benzo[b]naphtho[2,3‐d]thiopyran‐6‐one and 6H‐benzo[b]naphtho[2,3‐d]pyran‐6‐one by two‐dimensional nmr methods is described. The difference between the chemical shift values of H12 for these two compounds is attributed to different molecular geometries.  相似文献

The title compound, C18H18N4OS2, was prepared by reaction of S,S‐diethyl 2‐thenoylimidodithiocarbonate with 5‐amino‐3‐(4‐methylphenyl)‐1H‐pyrazole using microwave irradiation under solvent‐free conditions. In the molecule, the thiophene unit is disordered over two sets of atomic sites, with occupancies of 0.814 (4) and 0.186 (4), and the bonded distances provide evidence for polarization in the acylthiourea fragment and for aromatic type delocalization in the pyrazole ring. An intramolecular N—H...O hydrogen bond is present, forming an S(6) motif, and molecules are linked by N—H...O and N—H...N hydrogen bonds to form a ribbon in which centrosymmetric R22(4) rings, built from N—H...O hydrogen bonds and flanked by inversion‐related pairs of S(6) rings, alternate with centrosymmetric R22(6) rings built from N—H...N hydrogen bonds.  相似文献

Guanine radicals are important reactive intermediates in DNA damage. Hydroxyl radical (HO.) has long been believed to react with 2′‐deoxyguanosine (dG) generating 2′‐deoxyguanosin‐N1‐yl radical (dG(N1‐H).) via addition to the nucleobase π‐system and subsequent dehydration. This basic tenet was challenged by an alternative mechanism, in which the major reaction of HO. with dG was proposed to involve hydrogen atom abstraction from the N2‐amine. The 2′‐deoxyguanosin‐N2‐yl radical (dG(N2‐H).) formed was proposed to rapidly tautomerize to dG(N1‐H).. We report the first independent generation of dG(N2‐H). in high yield via photolysis of 1 . dG(N2‐H). is directly observed upon nanosecond laser flash photolysis (LFP) of 1 . The absorption spectrum of dG(N2‐H). is corroborated by DFT studies, and anti‐ and syn‐dG(N2‐H). are resolved for the first time. The LFP experiments showed no evidence for tautomerization of dG(N2‐H). to dG(N1‐H). within hundreds of microseconds. This observation suggests that the generation of dG(N1‐H). via dG(N2‐H). following hydrogen atom abstraction from dG is unlikely to be a major pathway when HO. reacts with dG.  相似文献

Details of the structures of two conformational polymorphs of the title compound, C12H17N2OS+·Cl, are reported. In form (I) (space group P), the two N—H groups of the cation are in a trans conformation, while in form (II) (space group P21/c), they are in a cis arrangement. This results in different packing and hydrogen‐bond arrangements in the two forms, both of which have extended chains lying along the a direction. In form (I), these chains are composed of centrosymmetric R42(18) (N—H...Cl and O—H...Cl) hydrogen‐bonded rings and R22(18) (N—H...O) hydrogen‐bonded rings. In form (II), the chains are formed by centrosymmetric R42(18) (N—H...Cl and O—H...Cl) hydrogen‐bonded rings and by R42(12) (N—H...Cl) hydrogen‐bonded rings.  相似文献

β‐Carbolines ( 1–5 ) undergo electrophilic aromatic substitution with N‐bromosuccinimide under different experimental conditions. Although 6‐bromo‐nor‐harmane ( la ) obtained by bromination of nor‐harmane ( 1 ) was isolated and fully characterized sometime ago, the other bromoderivatives of nor‐harmane ( 1b‐1e ) and harmane ( 2a‐2e ) were partially described as part of the reaction mixtures. The preparation and subsequent isolation, purification and full characterization of 1b, 1c, 1d, 1e, 2a, 2b, 2c, 2d, 2e are reported (mp, R f, 1H‐nmr, 13C‐nmr and ms) together with the preparation, isolation and charaterization, for the first time, of the bromoderivatives obtained from harmine ( 3a‐3e ), harmol ( 4a, 4b ) and 7‐acetylharmol ( 5a‐5c ). As brominating reagent N‐bromosuccinimide and N‐bromosuccinimide‐silica gel in dichloromethane and in chloroform as well as the β‐carboline ‐ N‐bomosuccinimide solid mixture have been used and their uses have been compared. Semiempirical AMI and PM3 calculations have been performed in order to predict reactivity in terms of the energies of HOMO, HOMO‐LUMO difference and in terms of the charge density of β‐carbolines ( 1–5 ) and bromo‐β‐carbolines ( 1a‐1e, 2a‐2e, 3a‐3e, 4a, 4b, 5a, 5b and 5c ) (Scheme 1).
