Optical waveguide spectroscopy was used under non-contact conditions to analyze the visible absorption spectrum of poly(ethylene oxide) (M w = 150) modified cytochrome c (PEO150-cyt.c) adsorbed on an indium tin oxide (ITO) glass electrode. The redox reactions of the adsorbed PEO150-cyt.c were dynamically measured in situ in PEO oligomers (M w = 200). It was confirmed that PEO modification of cyt.c is effective in maintaining the redox activity of the cyt.c after adsorption on the ITO glass electrode. The PEO150-cyt.c adsorbed on the electrode re-dissolved gradually in PEO oligomer. Against this, PEO1000-cyt.c having longer PEO chains (M w = 1000) was found to be adsorbed stably on the electrode. This paper reports a novel enzyme‐immobilization method for the direct electron transfer (DET) reaction of ascorbate oxidase from Acremonium sp. HI‐25 (ASOM). ASOM was adsorbed onto a gold electrode modified with a self‐assembled monolayer (SAM) of alkanethiol derivatives and immobilized by a cationic polymer membrane and anionic ω‐carboxyalkanethiol combined system. The redox responses of the immobilized ASOM were investigated by cyclic voltammetry. We found that the DET reaction of ASOM was facilitated by this novel immobilization. On the other hand, the redox responses of poly(ethylene oxide) (PEO)‐modified ASOMs were also investigated. ASOM was modified with two types of PEO which possess straight chain‐shaped (PEO2000) or comb‐shaped conformation (PM67). As a result, the DET reactions of PEO‐modified ASOMs were also facilitated by this immobilization method. We concluded that this immobilization method is effective for promoting the DET reaction of ASOMs.  相似文献

The redox reaction of poly(ethylene oxide) (PEO)-modified hemoglobin (PEO–Hb) was analyzed in PEO oligomers with cyclic voltammetry. The PEO–Hb was made soluble in PEO with molecular weight of 200 (PEO200) containing 0.5 M KCI. Quasi-reversible redox signals of PEO–Hb were obtained by using an indium tin oxide (ITO) glass working electrode. PEO–Hb, cast on the ITO electrode, also showed the redox response in PEO with molecular weight of 400 (PEO400). The peak current of PEO–Hb on the ITO electrode gradually increased during potential cycling. The effect of the scan rate on the quantity of electricity (Q) was analyzed after the peak current reached a constant value. The constant Q value was observed at the scan rate ranging from 30 to 500 mV/sec. The number of reactive PEO–Hb molecules was estimated from this constant Q-value. It was suggested that the electron transfer was carried out at the first layer of the PEO–Hb which was in direct contact with the ITO electrode. The quantity of electricity of PEO–Hb increased when the ITO electrode was first washed in an aqueous medium with ultrasonicator. This strongly suggested that the more effective surface area of the ITO electrode turned to be covered with PEO–Hb when the microporous region of the ITO particles was more hydrated.  相似文献

Well‐defined poly(L ‐lactide)‐b‐poly(ethylene oxide) (PLLA‐b‐PEO) copolymers with different branch arms were synthesized via the controlled ring‐opening polymerization of L ‐lactide followed by a coupling reaction with carboxyl‐terminated poly(ethylene oxide) (PEO); these copolymers included both star‐shaped copolymers having four arms (4sPLLA‐b‐PEO) and six arms (6sPLLA‐b‐PEO) and linear analogues having one arm (LPLLA‐b‐PEO) and two arms (2LPLLA‐b‐PEO). The maximal melting point, cold‐crystallization temperature, and degree of crystallinity (Xc) of the poly(L ‐lactide) (PLLA) block within PLLA‐b‐PEO decreased as the branch arm number increased, whereas Xc of the PEO block within the copolymers inversely increased. This was mainly attributed to the relatively decreasing arm length ratio of PLLA to PEO, which resulted in various PLLA crystallization effects restricting the PEO block. These results indicated that both the PLLA and PEO blocks within the block copolymers mutually influenced each other, and the crystallization of both the PLLA and PEO blocks within the PLLA‐b‐PEO copolymers could be adjusted through both the branch arm number and the arm length of each block. Moreover, the spherulitic growth rate (G) decreased as the branch arm number increased: G6sPLLA‐b‐PEO < G4sPLLA‐b‐PEO < G2LPLLA‐b‐PEO < GLPLLA‐b‐PEO. © 2006 Wiley Periodicals, Inc. A novel biosensor was developed by entrapping cytochrome c (Cyt c) in thin films of the room temperature ionic liquid (RTIL) containing nanocomposites of poly(diallyldimethylammonium chloride)‐graphene nanosheets‐gold nanoparticles (PDDA‐Gp‐AuNPs) at a 11‐mercaptoundecanoic acid‐6‐mercapto‐1‐hexanol modified gold electrode. The synthesized PDDA‐Gp‐AuNPs hybrid nanocomposites were characterized by UV‐vis spectroscopy, Raman spectroscopy, scanning electron microscopy and atomic force microscopy. The PDDA‐Gp‐AuNPs nanocomposites could increase the effective surface of the electrode, enhance the fixed amount of Cyt c on the electrode surface, promote the electron transfer and facilitate the catalytic activity of Cyt c. The RTIL could provide a biocompatible microenvironment to keep Cyt c biological activities, act as an effective mediator to immobilize a large number of Cyt c on the electrode and have good conductivity to improve electron transfer. Therefore, the resultant electrode exhibited good electrochemical performance and electrocatalytic activity. It could be used for electrochemical detection of H2O2 with rapid response, high sensitivity, wide linear range and low detection limit, as well as good stability, repeatability and selectivity. Here we report a facile method for the preparation of a PEO113‐b‐P4VP93 brush on gold surface with a grafting density as high as 1.32 chains/nm2; the P4VP blocks were physically adsorbed on gold surface forming an inner layer while the PEO blocks stretched towards the solution forming PEO brush. PEO113‐b‐P4VP93 micelles with P4VP core and PEO shell formed in methanol/water mixed solvents were used as the precursor. By adsorbing PEO113‐b‐P4VP93 micelles from pure water, in which the density of the micelles is the largest, maximum amount of the micelles was adsorbed onto gold surface, and the adsorbed micelles existed as individual domains on the surface. To prepare the polymer brush with a density as high as possible, we annealed the adsorbed micelles by methanol/water mixed solvent at the volume fraction of methanol (VF) of 20%, which was the proper proportion at which the core‐forming P4VP chains began to be flexible but the integrity of the micelles was remained. At this volume fraction, almost all the adsorbed micelles originally existing as individual domains were transformed into a dense polymer brush.  相似文献

A simple method for constructing gold nanoparticle‐modified electrodes with three‐dimensional nanostructures is demonstrated. The electrodes were prepared by casting citrate‐reduced AuNPs onto polycrystalline gold electrodes. The resultant electrodes had a large surface area‐to‐volume ratio, adequate for high protein loading and conferring high stability. The gold nanoparticle electrodes were covered with a self‐assembled monolayer of 11‐mercaptoundecanoic acid for electrostatic immobilization of cytochrome c (cyt c). At the electrode, direct, reversible electron transfer from cyt c was observed with remarkable stability. Moreover, an extremely high surface coverage of electrochemically active cyt c, 167 fully packed monolayers, was obtained through use of the electrode.  相似文献

Macrocyclic molecular brushes c‐PHEMA‐g‐(PS‐b‐PEO) consisting of macrocyclic poly(2‐hydroxylethyl methacrylate) (c‐PHEMA) as backbone and polystyrene‐b‐poly(ethylene oxide) (PS‐b‐PEO) amphiphilic block copolymers as side chains were synthesized by the combination of atom transfer radical polymerization (ATRP), click chemistry, and single‐electron transfer nitroxide radical coupling (SET‐NRC). First, a linear α‐alkyne‐ω‐azido heterodifunctional PHEMA (l‐HC?C‐PHEMA‐N3) was prepared by ATRP of HEMA using 3‐(trimethylsilyl)propargyl 2‐bromoisobutyrate as initiator, and then chlorine end groups were transformed to ? N3 group by nucleophilic substitution reaction in DMF in the presence of an excess of NaN3. The 3‐trimethylsilyl groups could be removed in the presence of tetrabutylammonium fluoride, and the product was cyclized by “click” chemistry in high dilution conditions. The hydroxyl groups on c‐PHEMA were transferred into bromine groups by esterification with 2‐bromoisobutyryl bromide and then initiate the ATRP of styrene. The formed macrocyclic molecular brushes c‐PHEMA‐g‐PS were coupled with the TEMPO‐PEO to afford the target macrocyclic molecular brushes c‐PHEMA‐g‐(PS‐b‐PEO) by SET‐NRC, and the efficiency is as high as 80~85%. The effects of the lamellar growth direction, extinction rings, and spherulitic boundaries of poly(butylene succinate) (PBSU) on the spherulitic growth of poly(ethylene oxide) (PEO) were investigated in miscible blends of the two crystalline polymers. In the crystallization process from a homogeneous melt, PBSU first developed volume‐filling spherulites, and then PEO spherulites nucleated and grew inside the PBSU spherulites. The lamellar growth direction of PEO was identical with that of PBSU even when the PBSU content was about 5 wt %. PEO, which intrinsically does not exhibit banded spherulites, showed apparent extinction rings inside the banded spherulites of PBSU. The growth rate of a PEO spherulite, GPEO, was influenced not only by the blend composition and the crystallization temperature of PEO, but also by the growth direction with respect to PBSU lamellae, the boundaries of PBSU spherulites, and the crystallization temperature of PBSU, TPBSU. The value of GPEO first increased with decreasing TPBSU when a PEO spherulite grew inside a single PBSU spherulite. Then, GPEO decreased when TPBSU was further decreased and a PEO spherulite grew through many tiny PBSU spherulites. The crystallization behaviors and morphology of asymmetric crystalline–crystalline diblock copolymers poly(ethylene oxide‐lactide) (PEO‐b‐PLLA) were investigated using differential scanning calorimetry (DSC), wide angle X‐ray diffraction (WAXD), and microscopic techniques (polarized optical microscopy (POM) and atomic force microscopy (AFM)). Both blocks of PEO5b‐PLLA16 can be crystallized, which was confirmed by WAXD, while PEO block in PEO5b‐PLLA30 is difficult to crystallize because of the confinement induced by the high glass transition temperature and crystallization of PLLA block with the microphase separation of the block copolymer. Comparing with the crystallization and morphology of PLLA homopolymer and differences between the two copolymers, we studied the influence of PEO block and microphase separation on the crystallization and morphology of PLLA block. The boundary temperature (Tb) was observed, which distinguishes the crystallization into high‐ and low‐temperature ranges, the growth rate and morphology were quite different between the ranges. Crystalline morphologies including banded spherulite, dendritic crystal, and dense branching in PEO5b‐PLLA16 copolymer were formed. The typical morphology of dendritic crystals including two different sectors were observed in PEO5b‐PLLA30 copolymer, which can be explained by secondary nucleation, chain growth direction, and phase separation between the two blocks during the crystallization process. Lozenge‐shaped crystals of PLLA with screw dislocation were also observed employing AFM, but the crystalline morphology of PEO block was not observed using microscopy techniques because of its small size. © 2008 Wiley Periodicals, Inc. Horse heart cytochrome c (cyt c) was adsorbed on the binary self-assembled monolayers (SAMs) composed of thioctic acid (T-COOH) and thioctic amide (T-NH2) at gold electrodes via electrostatic interaction. The cyt c adsorbed on the modified gold electrode exhibited well-defined reversible electrochemical behavior in 10 mM phosphate buffer solution (PBS, pH 7.0). The surface concentration (Γ) of electroactive species, cyt c, on the binary SAMs was higher than that in single-component SAMs of T-COOH, and reached a maximum value of 9.2 × 10−12 mol cm−2 when the ratio of T-COOH to T-NH2 in adsorption solution was of 3:2, and the formal potential (E0=(Epa+Epc)/2) of cyt c was −0.032 V (vs. Ag|AgCl (3 M NaCl)) in a 10 mM PBS. The interaction between cyt c and the binary SAMs made the E0 shift negatively when compared with that of cyt c in solution (+0.258 V vs. NHE, i.e., +0.058 V vs. Ag|AgCl (3 M NaCl)). The fractional coverage of bound cyt c was a 0.64 theoretical monolayer. The standard electron transfer rate constant of cyt c immobilized on the binary SAMs was also higher than that on single-component SAMs of T-COOH, and the maximum value of 15.8 ± 0.6 s−1 was obtained when the ratio of T-COOH to T-NH2 in adsorption solution was at 3:2. The results suggest that the electrode modified with the binary SAMs functions better than the electrode modified with single-component SAMs of T-COOH.  相似文献

Mesoporous silica particles were grafted with thermoresponsive poly(ethyleneoxide‐b‐N‐vinylcaprolactam), PEO‐b‐PVCL. N‐vinylcaprolactam was first polymerized on particle surfaces using surface initiated atom transfer radical polymerization (SI‐ATRP) and then, the poly(ethyleneoxide) blocks were attached to the PVCL chain ends with click chemistry. The sizes, thermoresponsiviness, and colloidal stability of SiO2‐PVCL and SiO2‐PVCL‐b‐PEO particles and their aqueous dispersions were studied by scanning electron microscopy, turbidimetry, dynamic light scattering, zeta sizer, and microcalorimetry. The phase separation temperature of the PEO‐b‐PVCL grafted particles did not considerably differ from that of the SiO2‐PVCL particles. The zeta potential of the grafted particles was close to zero at room temperature but decreased strongly upon heating. The decrease is related to the collapse of the PVCL blocks and correspondingly, the exposure of the silica surface toward the aqueous phase. The colloidal stability of the particles could be enhanced by adding PEO blocks to the chain ends of the PVCL grafts. © 2013 Wiley Periodicals, Inc. J. Polym. Sci., Part A: Polym. Direct electron transfer (DET) of bilirubin oxidase from Myrothecium verrucaria (BOD) was established on promoter‐modified gold electrodes. The electrochemical behavior of the enzyme in solution was studied by means of cyclic voltammetry evaluating the biocatalytic reduction of dioxygen. The reaction of BOD at Au electrodes was shown to be efficient only at low pH. In addition, a novel interaction between BOD and cytochrome c (cyt.c) was found. It was shown that BOD efficiently accepts cyt.c as an electron donor in both cases when cyt.c is in solution and electrostatically adsorbed. The results suggest that cyt.c can play the role of a mediator facilitating electron transfer in a pH range where no DET could be observed between the enzyme and the electrode. For the interaction between cyt.c and BOD in solution the reaction kinetics has been studied electrochemically and spectrophotometrically.  相似文献

Single-wall carbon nanotubes(SWNTs) modified gold electrodes were prepared by using two different methods.The electrochemical behavior of cytochrome c on the modified gold electrodes was investigated.The first kind of SWNT-modified electrode (noted as SWNT/Au electrode)was prepared by the adsorption of carboxylterminated SWNTs from DMF dispersion on the gold electrode.The oxidatively processed SWNT tips were covalently modified by coupling with amines (AET) to form amide linkage.Via Au-S chemical bonding,the self-assembled monolayer of thiol-unctionalized nanotubes on gold surface was fabricated so as to prepare the others SWNT-modified electrode (noted as SWNT/AET/Au electrode).It was shown from cyclic voltammetry cxperiments that cytochrome c exhibited direct electrochemical responses on the both electrodes, but only the current of controlled diffusion existed on the SWNT/Au electrode while both the currents of controlled diffusion and adsorption of cytochrome c occurred on the SWNT/AET/Au electrode.Photoelastic Modulation Infared Reflection Absorpthion Spectroscopy (PEM-IRRAS) and Quartz Crystal Microbalance (QCM) were employed to verify the adsorption of SWNTs on the gold electrodes.The results proved that SWNTs could enhance the direct electron transfer proecss between the electrodes and redox proteins.  相似文献

A surface‐active p‐vinyl benzyloxy‐ω‐hydroxy‐poly(ethylene oxide) macromonomer containing 22 pendant structural units of ethylene oxide (St–PEO22) was synthesized with an initiation method. Because of its solubility in a large variety of solvents, the free‐radical copolymerization with electron‐acceptor N‐phenylmaleimide (NPMI) was performed at 60 °C in benzene and tetrahydrofuran (THF) as isotropic media and in a water–THF mixture or water as a heterogeneous medium. Oil‐soluble 2,2′‐azobisisobutyronitrile and water‐soluble 4,4′‐azobis(4‐cyanovaleric acid) were used as the initiators at fixed concentrations. Two different St–PEO22/NPMI comonomer ratios (1/1 and 3/7) at a fixed total comonomer concentration in the polymerization system were used. The structures, compositions, and microstructure peculiarities of the obtained alternating, amphiphilic, comblike copolymers were determined by NMR analysis. For the copolymers synthesized in hydrophilic media, differential scanning calorimetry showed, near the endothermic peak attributed to the melting of the poly(ethylene oxide) side chains, the presence of a second peak due to the partially ordered phase that could exist between the crystalline state and the isotropic melt. Cytochrome c was immobilized on a mixed-thiol (mercaptoundecanoic acid/mercaptoundecanol) modified gold electrode (MUA:MU/cyt c electrode). Characterization of the cyt c electrode showed a quasi-reversible, electrochemical redox behavior with a formal potential of −13±5 mV (versus Ag/AgCl) for the surface adsorbed protein and 3±5 mV for covalently immobilized cyt c. The heterogeneous electron transfer rate constants were determined to be about 70 and 40 s−1 for both states of the protein, respectively. They were found to be significantly higher than those of pure MUA-modified cyt c electrodes (MUA/cyt c electrodes). The interaction of superoxide radicals (O2) with the (MUA:MU)/cyt c electrode was characterized and used for an amperometric O2 detection. The influence of H2O2 and uric acid on the sensor signal was investigated. The sensitivity of the (MUA:MU)/cyt c electrode to O2 was significantly improved compared with that of the MUA/cyt c electrode. Based on a kinetic model for the superoxide detection system, a new calibration method was established. This simple and fast method used the spontaneous dismutation of KO2 and was compared with the enzymatic superoxide generation system using xanthine oxidase.  相似文献

Core/shell wormlike polymer brushes with densely grafted poly(ϵ‐caprolactone)‐b‐poly(ethylene oxide) (PCL‐b‐PEO) are synthesized via grafting an alkynyl terminated PCL‐b‐PEO (ay‐PCL17b‐PEO113) onto a well‐defined azido functionalized polymethacrylate (PGA940) and are evaluated preliminarily as a single molecular cylindrical vehicle for drug delivery. Water soluble molecular worms of ca. 230 nm are obtained and then the anticancer drug doxorubicin (DOX) is loaded into its PCL core by hydrophobic interaction. Compared with spherical micelles from linear PCL17b‐PEO113, the brushes demonstrate a lower loading efficiency but a faster release rate of DOX. Confocal laser scanning microscopy measurements show that DOX‐loaded cylindrical molecular brushes can easily enter into HeLa and HepG2 cells in 1 h.  相似文献

Complex amphiphilic polymers were synthesized via core‐first polymerization followed by alkylation‐based grafting of poly(ethylene oxide) (PEO). Inimer 1‐(4′‐(bromomethyl)benzyloxy)‐2,3,5,6‐tetrafluoro‐4‐vinylbenzene was synthesized and subjected to atom transfer radical self‐condensing vinyl polymerization to afford hyperbranched fluoropolymer (HBFP) as the hydrophobic core component with a number‐averaged molecular weight of 29 kDa and polydispersity index of 2.1. The alkyl halide chain ends on the HBFP were allowed to undergo reaction with monomethoxy‐terminated poly(ethylene oxide) amine (PEOx‐NH2) at different grafting numbers and PEO chain lengths to afford PEO‐functionalized HBFPs [(PEOx)y‐HBFPs], with x = 15 while y = 16, 22, or 29, x = 44 while y = 16, and x = 112 while y = 16. The amphiphilic, grafted block copolymers were found to aggregate in aqueous solution to give micelles with number‐averaged diameters (Dav) of 12–28 nm, as measured by transmission electron microscopy (TEM). An increase of the PEO:HBFP ratio, by increase in either the grafting densities (y values) or the chain lengths (x values), led to decreased TEM‐measured diameters. These complex, amphiphilic (PEOx)y‐HBFPs, with tunable sizes, might find potential applications as nanoscopic biomedical devices, such as drug delivery vehicles and 19F magnetic resonance imaging agents. © 2010 Wiley Periodicals, Inc. This report describes a gold(III)‐catalyzed efficient general route to densely substituted chiral 3‐formyl furans under extremely mild conditions from suitably protected 5‐(1‐alkynyl)‐2,3‐dihydropyran‐4‐one using H2O as a nucleophile. The reaction proceeds through the initial formation of an activated alkyne–gold(III) complex intermediate, followed by either a domino nucleophilic attack/anti‐endo‐dig cyclization, or the formation of a cyclic oxonium ion with subsequent attack by H2O. To confirm the proposed mechanistic pathway, we employed MeOH as a nucleophile instead of H2O to result in a substituted furo[3,2‐c]pyran derivative, as anticipated. The similar furo[3,2‐c]pyran skeleton with a hybrid carbohydrate–furan derivative has also been achieved through pyridinium dichromate (PDC) oxidation of a substituted chiral 3‐formyl furan. The corresponding protected 5‐(1‐alkynyl)‐2,3‐dihydropyran‐4‐one can be synthesized from the monosaccharides (both hexoses and pentose) following oxidation, iodination, and Sonogashira coupling sequences. Furthermore, to demonstrate the potentiality of chiral 3‐formyl furan derivatives, a TiBr4‐catalyzed reaction of these derivatives has been shown to offer efficient access to 1,5‐dicarbonyl compounds, which on treatment with NH4OAc in slightly acidic conditions afforded substituted furo[3,2‐c]pyridine.  相似文献

The electrochemical behavior of cytochrome c (cyt‐c) that was electrostatically immobilized onto a self‐assembled monolayer (SAM) of captopril (capt) on a gold electrode has been investigated. Cyclic voltammetry, scanning electrochemical microscopy (SECM) and electrochemical impedance spectroscopy were employed to evaluate the blocking property of the capt SAM. SECM was used to measure the bimolecular electron transfer (ET) kinetics (kBI) between a solution‐based redox probe and the immobilized protein. In addition, the tunneling ET between the immobilized protein and the underlying gold electrode was calculated. A kBI value of (5.0±0.6)×108 mol?1 cm3 s?1 for the bimolecular ET and a standard tunneling rate constant (k0) of 46.4±0.2 s?1 for the tunneling ET have been obtained.  相似文献
