本文利用2-氨基喹啉和2-氨基苯骈咪唑作为亲核试剂,与5-(双甲硫基亚甲基)丙二酸亚异丙酯(1)、5-(甲硫基亚烃基)丙二酸亚异丙酯(3)反应,开发出3-取代-(1H)-嘧啶-[1,2-a]喹啉-1-酮(5)和2-取代嘧啶[1,2-a]苯骈咪唑-4-(10H)-酮(6)通用的简便合成法。  相似文献

A method for the synthesis of 3-substituted benzo[g]quinolin-4-ones by the condensation of 1,2,3,4-tetrahydrobenzo[g]quinolin-4-one with aromatic aldehydes in an alkaline medium has been developed. It has been found that the first stage of the reaction is the formation of the corresponding benzylidene derivative, which then isomerizes into the more stable benzyl derivative. The reactions of 4-amino derivatives of imidazo[4.5-c]pyridin-2-ones with -bromomethylketones have been studied. Depending on the nature of the reagents and the reaction conditions, either 5-acylmethyl salts of the starting amine or 2-substituted imidazo[4.5-c]imidazo[1,2-a]pyridin-8-ones can be obtained. An efficient method for the synthesis of novel benzo[b]pyrimido[4',5':5,4]thieno[2,3e]-[1,6]naphthyridine-8-one derivatives via Pictet–Spengler cyclization is reported. The reaction of 4-(3-aminopyrimido[4,5-d]thieno-2-yl)quinoline-2-ones, which could be obtained from Thorpe–Ziegler isomerization of 4-bromomethylquinoline-2-ones and 5-cyano-1,6-dihydro-4-methyl-2-phenyl-6-thioxopyrimidine,with aromatic aldehydes in the presence of BF_3·OEt_2 gives pyrimidothieno[1,6]naphthyridines in good yields.  相似文献

This report describes a type of tautomerization reaction that proceeds via isomerization of π-bonds across the azulene moieties of tricyclic benzo[cd]azulen-3-ones. The reaction mechanism shows similarities to an elimination reaction that was recently developed in our group. Furthermore, the facile four-step syntheses of the benzo[cd]azulen-3-ones, the starting materials for the tautomerization reactions, and computational analyses of the tautomerization reaction are included.  相似文献

[reaction: see text] Benzopyranoimidazolones could virtually exist in four tautomeric forms, namely N3-H, N1-H, coumarin O-H, and C2-H. Experimental evidence reported thus far has been unable to lead to a unique statement about the preferred tautomeric forms in solution. In this work, tautomeric equilibria for a series of 2-substituted [1]benzopyrano[3,4-d]imidazol-4(3H)-ones were investigated by DFT calculations, in both gas phase and solution. The influence of the solvent was included in the calculations by the CPCM solvent model. 13C chemical shifts of all tautomers were computed at different levels of theory and then compared with experiments to assign the preferred tautomers. Theoretical findings were then compared to dynamic 1H NMR experiments results.  相似文献

3-Aryl-1,2,4-triazole-5-thiones react with dimethyl acetylenedicarboxylate and methyl 3-phenyl-propynoate to afford the corresponding 5-substituted 2-aryl-7H-[1,2,4]triazolo[3,2-b][1,3]thiazin-7-ones. The one-pot synthesis of benzo[f]quinolin-3-ones and benzo[a]phenanthridein-5-ones was achieved by the inter- and intramolecular photoannulation of 6-chloropyridin-2-ones and 3-chloroisoquinolin-1-ones with phenylacetylene or tethered phenylacetylene. The reactions were proceeded by photoaddition of 6-chloropyridin-2-ones and 3-chloroisoquinolin-1-ones to phenylacetylene to give the chlorine-substituted stilbenoids, and then 6π electrocyclization of the stilbenoids and oxidation aromatization to afford the polycyclic products.  相似文献

Summary The reaction of 3,4-methylenedioxycinnamic acid (1) with thionyl chloride resulted in the formation of 7-chlorothieno[2,3-f]-1,3-benzodioxole-6-carbonyl chloride (2) and cinnamoyl chloride (3). Subsequent reaction of the former withp-substituted anilines led to the formation of 7-chloro-N-(p-substituted phenyl)-thieno[2,3-f]-1,3-benzodioxole-6-carboxamides (4a–c) which on photocyclization afforded 2-substituted [1,3]dioxolo[5,6][1]benzothieno[2,3-c]quinolin-6(5H)-ones (5a–c) in fairly good yields and high purity. A palladium-catalyzed [4+1] cycloaddition of prop-2-yn-1-ones with double isocyanides is developed herein.The transformation worked well to produce a series of 2-amino-4-cyanofurans with high efficiency and a broad reaction scope.Based on mechanism studies,it is believed that the palladiumcatalyzed [4+1] imidoylative cycloaddition of prop-2-yn-1-ones was concerted.Treated with a ryl amine and H_2 O,the [4+1] cycloaddition of prop-2-yn-1-ones with double isocyanides provided 2-amino-4-amidylpyrroles efficiently.  相似文献

We have studied the reaction of 2,3-dihydro-4H-[1,3]thiazino[3,2-a]benzimidazol-4-one and 2-aryl-2,3-dihydro-4H-[1,3]thiazino[3,2-a]benzimidazol-4-ones with amines, alkylating reagents, and hydrogen peroxide. A series of novel 3-substituted imidazo[4,5-c]quinolin-4(5H)-ones (2a-w) was prepared by the reaction of imidazo[4,5-c]quinolin-4(5H)-ones (6) with several electrophiles under basic conditions. The bronchodilatory activity of these compounds was evaluated on the basis of their protective effects against antigen-induced contraction (the Schultz-Dale reaction) of guinea-pig trachea (in vitro) and antigen inhalation-induced bronchospasm in passively sensitized guinea-pigs (in vivo). Although correlations between in vitro and in vivo activities were not clear, short alkyl chains such as the methyl and ethyl groups at the 3-position were important for potent activity, especially in vivo. Substituents at the 5-position were more tolerant of the activity than those at the 3-position. 5-Ethyl-3-methyl-3H-imidazo[4,5-c]quinolin-4(5H)-one (21) exhibits the most potent bronchodilatory activity among our tested compounds and is at least 5-fold more active than theophylline in vivo.  相似文献
