A procedure for the determination of six derivatives of 2-hydroxybenzophenone, used as UV absorbers, in water samples is presented. Compounds were first concentrated using a solid-phase extraction (SPE) cartridge and then selectively determined by liquid chromatography-tandem mass spectrometry (LC-MS/MS) using electrospray ionization (ESI). The effect of different parameters on the performance of concentration and determination steps is discussed. The highly polar and acidic 2-hydroxy-4-methoxybenzophenone-5-sulfonic acid (BP-4) required the use of ammonium acetate as modifier during desorption of SPE cartridges and also to improve the performance of its separation in the LC column. Under optimized conditions, the proposed method provided limits of quantification from less than 1 to 32 ng L−1, depending on the compound and the type of water sample. Recoveries from the SPE step (83-105%) remained unaffected by the nature of the matrix; however, the efficiency of electrospray ionization was compound and sample dependant. Real sample analysis reflected the presence of three of the six investigated species (BP-4, 2-hydroxy-4-methoxybenzophenone, BP-3, and 2,4-dihydroxybenzophenone, BP-1) in the aquatic environment, particularly in raw wastewater samples. In this latter matrix, BP-4 was the compound presenting the highest concentrations; moreover, it was poorly removed in sewage treatment plants and consequently it also appeared in river water.  相似文献

This paper describes a liquid chromatography-electrospray-tandem mass spectrometry (LC-ESI-MS/MS) for the qualitative and quantitative analysis of three isoflavone aglycones (glycitein, daidzein and genistein) in human serum. Positive ion mode was used for the detection of these compounds and selective reaction monitoring (SRM) was employed for quantitative measurement. The SRM transitions monitored were as 285.0  242.0, 270.0 for glycitein, 255.0  137.0, 153.0, 181.0, 199.0 for daidzein and 271.0  153.0, 215.0 for genistein. d3-Daidzein was used as an internal standard for quantitative measurement. The linearity was good from 0.5 to 500 ng/ml. The detection limit based on a signal-to-noise ratio of three was 0.27, 0.38 and 0.29 ng/ml for glycitein, daidzein and genistein, respectively. A newly developed solid phase extraction (SPE) procedure was developed for sample pre-treatment. Good recovery, 92.3-103.2%, for three isoflavone aglycones were obtained. This newly developed method was successfully applied to evaluate isoflavone pharmacokinetic in human serum after oral administration.  相似文献

建立了测定室内灰尘样品中39种多氯联苯(PCBs)的分析方法。样品经吸尘器采集、正己烷-二氯甲烷(1:1,v/v)超声萃取、浓缩后,利用气相色谱-三重四极杆质谱法(GC-MS/MS)在选择反应监测(SRM)模式下测定。结果表明,39种PCBs在30 min内得到了很好的分离,在0.1~100 μg/L范围内线性关系良好,相关系数为0.9910~0.9999,方法的加标回收率为57.2%~120.3%,日内测定的相对标准偏差(RSD)为0.3%~24.7%,日间测定的RSD为0.6%~29.9%,检出限(信噪比为3)为0.0003~0.2080 ng/g。本方法灵敏度高、准确度和精密度好,简便快速,溶剂消耗量少,适用于灰尘中多种多氯联苯的同时测定。  相似文献

有机氯农药(OCPs)和多氯联苯(PCBs)是两类重要的持久性有机污染物,可在环境介质中长期存在,并通过多种途径进入人体,导致人体的高暴露风险。OCPs和PCBs对人体存在诸多健康危害,精准定量人体内OCPs和PCBs的暴露水平是健康效应评价的关键。该研究基于固相萃取-气相色谱-串联质谱联用技术(SPE-GC-MS/MS)建立了同时检测100 μL血清中35种OCPs和PCBs的分析方法。血清样品经尿素沉淀蛋白后,采用Oasis® HLB小柱净化,正己烷-二氯甲烷混合溶液(1∶1, v/v)洗脱,氮吹近干,正己烷定容,多反应监测(MRM)模式检测,内标法定量分析。结果表明,OCPs和PCBs在0.05~50.0 ng/mL范围内线性关系良好,检出限在1.2~71.4 ng/L之间。35种目标分析物的加标回收率在72.6%~142%之间,相对标准偏差小于25%。利用所建立的方法检测了武汉市普通人群血清样本中OCPs和PCBs的浓度水平,结果表明武汉市普通人群广泛暴露于OCPs和PCBs,且以OCPs为主。有8种OCPs和7种PCBs检出率高于50%,其中p,p'-滴滴伊、p,p'-滴滴滴和甲氧滴滴涕检出率达100%,非类二噁英PCBs是PCBs的主要成分。血清中OCPs浓度随年龄增长呈升高趋势,在60岁以上存在性别差异;不同性别、年龄人群血清中PCBs浓度无统计学差异。该方法样本用量少,操作简便,具有较高的准确度和精密度,适用于环境健康研究中大量人群血清样本中痕量OCPs和PCBs的生物监测。  相似文献

A multiresidue method for determining 22 polychlorinated biphenyls (PCBs) in air has been developed and validated by gas chromatography (GC) coupled to tandem mass spectrometry (MS/MS) using a triple quadrupole analyzer (QqQ). The method was validated in terms of both steps of sampling and analysis. The sampling method, which is based on active sampling using polyurethane foam (PUF) as adsorbent, was validated by generating standard atmospheres. The retention capacity of this sampling sorbent allows up to 5 m3 of air to be sampled without any breakthrough for most compounds. Two solvent extraction methods were compared: sonication and Soxhlet extraction with a mixture of n-hexane:diethyl ether (95:5 v/v). Both extraction methods yielded similar results, but the first one required less solvent and time. The method exhibited good accuracy (80.3–99.8%), precision (2.2–15.2%) and lower limits that allowed quantification and confirmation at levels as low as 0.008 ng/m3. Finally, the method was applied to the analysis of PCBs in the air in areas near to a municipal solid-waste landfill and directly above the refuse in the landfill, where it indicatedd the presence of some of the target compounds. Traditionally, the determination of hydroxylated polychlorinated biphenyls (OH-PCBs) has been carried out by gas chromatography (GC). However, the gas chromatographic behavior and sensitivity of this type of hydroxylated compounds are not always satisfactory, hence a prior derivatization of the OH-PCBs must be performed. Therefore, the development of liquid chromatographic methods should prove to be a very interesting task aimed at dealing with the instrumental determination of OH-PCBs. Taking into account that octadecylsilane stationary phases are not the most adequate for the separation of isobaric compounds, an amide-type column has been tested. For the development of the method, the Response Surface Methodology was used, based on a Box-Wilson Central Composite experimental design. The initial content of methanol in the mobile phase, the gradient time, and the concentration and the pH value of the buffer were chosen as relevant experimental parameters. A global optimum was obtained by selecting the elution time, the sensitivity and the overall resolution as responses to optimize. The developed method for liquid chromatography presented a very good resolution and sensitivity, and a reasonably short analysis time. In addition, a retention study was conducted in order to survey the different interactions that take place in the separation process, showing that hydrogen bonding is the main interaction between OH-PCBs and the amide-type stationary phase. However, a substantial contribution of dispersion forces was present in methanol contents in the mobile phase below 65%. Besides, the pH value of the mobile phase was found to be the most important parameter to control the hydrogen bond forces and therefore, to regulate the OH-PCBs separation.  相似文献

建立了固相萃取-高效液相色谱-串联质谱法(HPLC-MS/MS)同时测定畜禽粪便中四环素类化合物(四环素、金霉素、土霉素)、喹诺酮类化合物(诺氟沙星、环丙沙星、洛美沙星)和磺胺二甲嘧啶7种抗生素的检测方法。样品中的抗生素用含有甲醇、乙酸和水(6:3:1, 体积比)的混合溶液提取后,经HLB固相萃取小柱纯化富集,采用Symmetry C18色谱柱分离,0.3%甲酸水溶液和0.3%甲酸乙腈溶液作为流动相进行梯度洗脱,电喷雾正离子(ESI+)模式电离,多反应监测(MRM)模式检测,外标法定量。结果表明,四环素类化合物和喹诺酮类化合物在50~1000 μg/L、磺胺二甲嘧啶在5~100 μg/L的范围内具有良好线性。3倍信噪比下,四环素类化合物、喹诺酮类化合物和磺胺二甲嘧啶的检出限分别为0.25~7.18、0.15~3.16和0.04 μg/kg。在猪粪和鸡粪样品中添加0.1~10 μg/g水平的四环素类化合物、喹诺酮类化合物和磺胺二甲嘧啶,其平均添加回收率为40%~124%,相对标准偏差(RSD)为3.0%~9.5%。采用该方法对部分养殖场的猪粪和鸡粪进行了检测,结果表明,四环素类化合物均有不同程度的检出,喹诺酮类化合物和磺胺二甲嘧啶有部分检出。该方法具有灵敏度和准确度高的特点,可满足畜禽粪便中四环素类化合物、喹诺酮类化合物及磺胺二甲嘧啶的检测。  相似文献

Benzophenones (BPs) are a family of compounds widely used to protect the skin and hair from UV irradiation. Despite human exposure to BPs through dermal application of products containing sunscreen agents and the increasing evidence that BPs are able to interfere with endocrine systems, few studies have examined the occurrence of BPs in humans. In this work, we propose a new liquid chromatography-tandem mass spectrometry (LC-MS/MS) method to determine six BPs, namely, benzophenone-1 (BP-1), benzophenone-2 (BP-2), benzophenone-3 (BP-3), benzophenone-6 (BP-6), benzophenone-8 (BP-8) and 4-hydroxybenzophenone (4-OH-BP) in human placental tissue samples. The method involves an extraction step of the analytes from the samples using ethyl acetate, followed by a clean-up step using centrifugation prior to their quantification by LC-MS/MS using an atmospheric pressure chemical ionization (APCI) interface in the positive mode. Benzophenone-d10 (BP-d10) was used as surrogate. Found detection limits (LOD) ranged from 0.07 to 0.3 ng g−1 and quantification limits (LOQ) from 0.3 to 1.0 ng g−1, while inter- and intra-day variability was under 5%. The method was validated using standard addition calibration and a recovery assay. Recovery rates for spiked samples ranged from 98 to 104%. This method was satisfactorily applied for the determination of BPs in 16 placental tissue samples collected from women who live in Granada (Spain).  相似文献

Endocrine disruptors are a group of organic compounds widely used, which are ubiquitous in the environment and in biological samples. The main effect of these compounds is associated with their ability to mimic or block the action of natural hormones in living organisms, including humans. Parabens (esters of p-hydroxybenzoic acid) belong to this group of compounds. In this work, we propose a new liquid chromatography-tandem mass spectrometry (LC-MS/MS) method to asses the presence of parabens most commonly used in industrial applications (methyl-, ethyl-, propyl- and butyl-paraben) in samples of human placental tissue. The method involves the extraction of the analytes from the samples using ethyl acetate, followed by a clean-up step using centrifugation prior to their quantification by LC-MS/MS using an atmospheric pressure chemical ionization (APCI) interface in the negative mode. Deuterated bisphenol A (BPA-d16) was used as surrogate. Found detection limits (LOD) ranged from 0.03 to 0.06 ng g−1 and quantification limits (LOQ) from 0.1 to 0.2 ng g−1, while inter- and intra-day variability was under 13.8%. The method was validated using standard addition calibration and a spike recovery assay. Recovery rates for spiked samples ranged from 82% to 108%. This method was satisfactorily applied for the determination of parabens in 50 placental tissue samples collected from women who live in the province of Granada (Spain).  相似文献

建立了固相萃取-高效液相色谱-串联质谱(HPLC-MS/MS)同时检测人体尿液中N-乙酰基-S-(3,4-二羟基丁基)-L-半胱氨酸(DHBMA)、N-乙酰基-S-(3-羟基丙基)半胱氨酸(3-HPMA)、N-乙酰基-S-(2-氰乙基)-L-2-氨基-3-巯基羧酸(CEMA)和苯巯基尿酸(SPMA)的检测方法。冰冻的人体24 h尿液在室温下解冻,混合均匀后离心过滤,经C18固相萃取小柱净化富集后在多反应监测模式下采用HPLC-MS/MS进行定量分析。在3个添加水平下,尿液中DHBMA、3-HPMA、CEMA和SPMA的加标回收率分别为105.6%~124.4%、102.7%~106.5%、103.2%~103.9%和101.7%~104.3%,相对标准偏差为2.6%~7.7%。以不低于3倍的信噪比估算DHBMA、3-HPMA、CEMA和SPMA的检出限分别为0.062、0.031、0.020和0.003 μg/L。应用该方法检测了37例吸烟和非吸烟者的24 h尿液样本,结果发现吸烟者尿液中3-HPMA、SPMA和CEMA的平均含量比非吸烟者高3到6倍。  相似文献

The different parameters affecting the ionisation and fragmentation of selected polychlorinated biphenyls (PCBs) in an IT detector working in the MS/MS mode, ITD(MS/MS), have been optimised for maximum selectivity and sensibility. The low LODs (in the range 0.03-0.3 microg/L), the satisfactory repeatability (RSDs in general below 11%) and reproducibility (RSDs below 17%) obtained when analysing standard solutions ensured proper determination of the PCBs studied at the concentrations typically found in food samples. Foodstuffs naturally contaminated with varying levels of PCBs have been analysed using the optimised GC-ITD(MS/MS) method. The results obtained compared favourably with those found using more conventional detectors, such as (micro-)electron capture detection (for ortho-PCBs) and high-resolution MS (for non-ortho-PCBs), as well as with the consensus PCB levels established for these particular samples via an international interlaboratory exercise. 建立了液相色谱-串联质谱(LC-MS/MS)测定食品中二甲基黄(DMY)的分析方法。样品经乙酸乙酯提取,二甲基黄专用固相萃取小柱(ProElut DMY SPE)净化,XDB-C18色谱柱(50 mm×4.6 mm,1.8μm)分离,并以5mmol/L乙酸铵水溶液(含0.1%(v/v)甲酸)-乙腈(含0.1%(v/v)甲酸)为流动相,梯度洗脱,电喷雾正离子模式(ESI~+)电离,多反应监测模式(MRM)检测,内标法定量。结果表明,DMY在0~50μg/L范围内线性关系良好,相关系数(r~2)均大于0.999。方法的检出限(LOD,S/N3)和定量限(LOQ,S/N10)分别为2μg/kg和10μg/kg。不同食品基质中,DMY在10、20和100μg/kg的添加水平下的平均加标回收率为93.3%~98.9%,相对标准偏差为1.6%~3.9%(n=6)。该方法有效补偿了液相色谱-串联质谱检测过程中的离子化抑制效应,灵敏度和准确度高,适用于腐乳、辣椒酱、禽蛋、豆干、糖果和火腿中DMY的测定。  相似文献

建立了超高效液相色谱-串联质谱(UPLC-Ms/Ms)快速检测豆芽中10种喹诺酮类抗生素(QNs)的方法。样品用乙腈-0.1%甲酸水溶液提取后,经PRiME HLB快速净化,氮吹浓缩后,用电喷雾离子源正离子-多反应监测(MRM)模式串联质谱进行检测。在0.25~100μg/L范围内,10种目标物的线性关系良好,其相关系数均大于0.997。方法对两种豆芽基质进行前处理并测定,检出限为1.0μg/kg和2.0μg/kg,定量限为3.0μg/kg和6.0μg/kg。回收率实验结果表明:10种目标物回收率范围为78.1%~106.4%,RSD<8.1%。方法能有效地避免杂质的干扰。  相似文献

建立了液相色谱-串联质谱技术同时检测水产品中15种喹诺酮类药物(氟罗沙星、氧氟沙星、依诺沙星、诺氟沙星、环丙沙星、恩诺沙星、洛关沙星、单诺沙星、奥比沙星、双氟沙星、沙拉沙星、司帕沙星、口恶喹酸、萘啶酸、氟甲喹)残留量的方法.试样中残留的喹诺酮类药物采用乙腈提取,提取液经正已烷液液分配脱脂后,以强阳离子固相萃取小柱净化,液相色谱.串联质谱法测定.对液/质分离条件与样品前处理条件进行了优化,并对喹诺酮类药物在分析过程的稳定性进行了研究.15种喹诺酮类药物在1.0~100 μg/L范围内线性关系良好,相关系数为0.9924~0.9992.在0.002~0.04 mg/kg浓度范围内,平均加标回收率在79.9%~93.8%;相对标准偏差为4.8%~14.6%.方法可满足水产品中喹诺酮类药物多残留检测与确证的需要.  相似文献

Polychlorinated biphenyls (PCBs) and polychlorinated naphthalenes (PCNs) are listed as persistent organic pollutants (POPs) under the Stockholm Convention. Because they have similar physical and chemical properties, they are coeluted and are usually analyzed separately by different gas chromatography high-resolution mass spectrometry (GC-HRMS) methods. In this study, a novel method was developed for simultaneous analysis of six indicator PCBs, 12 dioxin-like PCBs, and 16 PCNs using isotope dilution comprehensive two-dimensional gas chromatography with high-resolution time-of-flight mass spectrometry (GC × GC-HRTOF-MS). The method parameters, including the type of GC column, oven temperature program, and modulation period, were systematically optimized. Complete separation of all target analytes and the matrix was achieved with a DB-XLB column in the first dimension and a BPX-70 column in the second dimension. The isotope dilution method was used for quantification of the PCBs and PCNs by GC × GC-HRTOF-MS. The method showed good linearity from 5 to 500 pg μL−1 for all the target compounds. The instrumental limit of detection ranged from 0.03 to 0.3 pg μL−1 for the 18 PCB congeners and from 0.09 to 0.6 pg μL−1 for the 16 PCN congeners. Repeatability for triplicate injections was always lower than 20%. The method was successfully applied to the determination of 18 PCBs present at 0.9–2054 pg g−1 and 16 PCNs present at 0.2–15.7 pg g−1 in three species of fish. The GC × GC-HRTOF-MS results agreed with those obtained by GC-HRMS. The GC × GC-HRTOF-MS method proved to be a sensitive and accurate technique for simultaneous analysis of the selected PCBs and PCNs. With the excellent chromatographic separation offered by GC × GC and accurate mass measurements offered by HRTOF-MS, this method allowed identification of non-target contaminants in the fish samples, including organochlorine pesticides and polycyclic aromatic hydrocarbons.  相似文献

建立了采用固相萃取-液相色谱-串联质谱同时测定麦冬、玄参、延胡索中7种甲氧基丙烯酸酯类杀菌剂残留量的方法。样品经乙酸乙酯提取,氨基固相萃取柱净化后,经C18色谱柱分离,以甲醇和1.0‰甲酸溶液为流动相梯度洗脱,电喷雾正离子模式离子化,选择反应监测方式监测,串联质谱测定,外标法定量。结果显示,各目标物在1或2个数量级浓度范围内的线性关系良好,相关系数均大于0.996。醚菌胺、啶氧菌酯和肟菌酯的定量限为2 μg/kg,嘧菌酯和吡唑醚菌酯的定量限为4 μg/kg, Z-苯氧菌胺和醚菌酯的定量限为10 μg/kg。各杀菌剂的回收率为60.4%~110%,相对标准偏差为1.2%~17%。该方法能满足麦冬、玄参、延胡索中7种目标杀菌剂残留量的检测。  相似文献

建立了一种以13C6-磺胺二甲基嘧啶为内标快速测定肠衣中17种磺胺类药物残留量的方法.样品经过V(乙腈)∶V(乙酸乙酯)=1∶1 混合溶剂提取后,离心,上清液在旋转蒸发仪上蒸发至干,体积分数30%甲醇溶液定容,正己烷除去脂肪,经过滤膜后,进高效液相色谱-串联四极杆质谱进行分析,内标法定量.方法的回收率在80%~100%之间,相对标准偏差在15.4%以下,定量限为1.0 μg/kg(磺胺脒为2.0 μg/kg),检出限在0.4 μg/kg以下(磺胺脒为1.0 μg/kg).方法适合于大批量样品的快速检测.  相似文献

建立了分散固相萃取-液相色谱-串联质谱同时快速测定化妆品中81种非法添加糖皮质激素(GCs)的分析方法。样品用水分散后加乙腈超声提取,经十八烷基键合硅胶(C18)和N-丙基乙二胺(PSA)净化,待测物选用具有多重色谱保留模式的Poroshell 120 PFP色谱柱(100 mm×2.1 mm,2.7μm)分离,以0.2%(v/v)乙酸水溶液-乙腈为流动相梯度洗脱,在电喷雾正离子模式下以动态多反应监测方式测定,内标法定量。81种待测物在各自的浓度范围内线性关系良好,相关系数均大于0.99,在3个不同的添加水平下,平均回收率为68.8%~105.3%,RSD为2.9%~13.1%(n=6),方法的检出限(S/N≥3)和定量限(S/N≥10)分别为0.002~0.006μg/g和0.005~0.020μg/g。筛查了137个化妆品样品,发现16个阳性样品,含量为16.9~158μg/g。结果表明,该法简便快速,灵敏可靠,适用于化妆品中81种GCs的同时快速定性定量筛查分析。  相似文献

建立了利用固相萃取-液相色谱-串联质谱同时测定水稻中17种细胞分裂素含量的分析方法。水稻样品经冷冻研磨,用甲醇-水(80 : 20, v/v)溶液浸提,经聚合物阳离子交换树脂(PCX)纯化,采用ZORBAX Extend-C18色谱柱,以甲醇和5 mmol/L甲酸铵水溶液为流动相进行梯度洗脱。采用电喷雾正离子模式电离,选择反应监测模式扫描,外标法定量。优化了色谱分离条件,研究了不同提取溶剂对17种细胞分裂素的提取效率,并考察了PCX固相萃取柱的纯化效果。结果表明,17种细胞分裂素在线性范围内的相关系数(r)均大于0.9984,方法检出限为0.01~0.05 ng/g。水稻根、茎和叶基质在0.2、1和5 ng/g 3个添加水平的平均回收率为60.2%~125.4%(n=6),相对标准偏差(RSD)为5.4%~29.7%。在水稻样品中检出5种细胞分裂素,其含量为0.02~0.93 ng/g。本方法纯化效果好、灵敏度高,可用于水稻中多种痕量细胞分裂素的同时分析。  相似文献

建立了基于通过型固相萃取小柱净化的超高效液相色谱-三重四极杆质谱联用(UPLC-MS/MS)同时快速准确测定牛蛙中9种雌激素(雌三醇(E3)、17β-雌二醇(β-E)、17α-雌二醇(α-E)、17α-炔二雌醇(EE2)、雌酮(EI)、己烯雌酚(DES)、己二烯雌酚(DE)、己烷雌酚(HEX)、醋酸双烯雌酚(DD))残留的检测方法。样品经乙腈提取,经PRiME HLB固相萃取柱净化,Waters Acquity UPLC BEH C_(18)柱(100 mm×2.1 mm,1.7μm)分离,以0.5 mmol/L氟化铵水溶液-乙腈体系为流动相梯度洗脱,流速为0.3 mL/min,采用电喷雾正负离子切换模式(ESI^(+)/ESI^(-))和多反应监测(MRM)扫描方式检测,基质匹配外标法定量分析。该研究优化了液相色谱条件,相比于乙酸铵水溶液-乙腈体系和氨水溶液-乙腈体系,0.5 mmol/L氟化铵水溶液-乙腈体系作为流动相时9种雌激素普遍具有更佳的灵敏度。相比于甲醇和乙酸乙酯,乙腈作为提取溶剂时9种雌激素的提取率提高15%~40%。考察了HLB、C_(18)、Silica、PRiME HLB共4种不同类型的固相萃取小柱的基质净化效应,结果表明,PRiME HLB柱具有更好的基质净化能力。经PRiME HLB净化后,所有化合物的回收率均在70%~125%之间。DD的回收率从47%提高到74%,DES的回收率从180%降低到123%,有效减弱了基质效应。在最佳的实验条件下,E3、β-E、α-E、EI、DE、HEX、DD的线性范围为0.5~100.0μg/L,EE2和DES的线性范围为1.0~100.0μg/L,9种雌激素在各自的线性范围内均有良好的线性关系,相关系数为0.9953~0.9994,方法检出限为0.17~0.33μg/kg,方法定量限为0.5~1.0μg/kg,在2.0、10.0、80.0μg/kg 3个加标水平下,9种雌激素的加标回收率为65.1%~128.2%,相对标准偏差为1.9%~17.6%。该方法操作简便、快速、灵敏,重复性好,可用于大批量样品的同时快速准确检测。  相似文献
