Despite the rapid development of fluorescence detection modalities for disease diagnosis, novel fluorescent molecules and probes still face with tremendous pressure to transform before employing such fluorescent tools in the clinic. Impressively, the fluorescent probes based on the traditional fluorescent dye are expected to accelerate the transformation process. Herein, methylene blue is requisitioned to design the GSH responsive probe MB-SS-CPT elaborately. The as-synthesized MB-SS-CPT provides a dramatic optical advantage for GSH detection in vitro, cell fluorescence imaging, in vivo imaging, and antitumor therapy.  相似文献

In the context of the circular economy, the huge amounts of biomass waste should be converted into value-added materials and energy to diminish pollution, atmospheric CO2 levels and costly waste disposal. Biological imaging usually uses expensive and toxic chemicals e.g., organic dyes, semiconductor quantum dots, calling for safer, greener, cheaper fluorescent probes for biological imaging in vitro and in vivo. In these regards, carbon quantum dots (CQDs)-based fluorescent probes using biomass waste as a precursor may have much higher potential. Here we transformed the biomass waste of peach leaves into value-added fluorescent CQDs through a low-cost and green one-step hydrothermal process. The obtained CQDs show excitation-dependent photoluminescence properties with a fluorescence lifetime of 5.96 ns and a quantum yield of 7.71% without any passivation. In addition, the CQDs have a fine size of 1.9 nm with good hydrophilicity and high fluorescent stability over pH 4.0–11.0 range. Fluorescence imaging of in vitro cell cultures and in vivo with zebrafish show that CQDs possess ultra-low toxicity and remarkable performance for biological imaging. Even when CQDs present at a concentration as high as 500 µg/mL, the organism can still maintain more than 90% activity both in vitro and in vivo, and present bright fluorescence. The cheaper, greener, ultra-low toxicity CQDs developed in this work is a potential candidate for biological imaging in vitro and in vivo.  相似文献

Increasing evidence has highlighted the endogenous production of formaldehyde (FA) in a variety of fundamental biological processes and its involvement in many disease conditions ranging from cancer to neurodegeneration. To examine the physiological and pathological relevance and functions of FA, fluorescent probes for FA imaging in live biological samples are of great significance. Herein we report a systematic investigation of 2-aza-Cope reactions between homoallylamines and FA for identification of a highly efficient 2-aza-Cope reaction moiety and development of fluorescent probes for imaging FA in living systems. By screening a set of N-substituted homoallylamines and comparing them to previously reported homoallylamine structures for reaction with FA, we found that N-p-methoxybenzyl homoallylamine exhibited an optimal 2-aza-Cope reactivity to FA. Theoretical calculations were then performed to demonstrate that the N-substituent on homoallylamine greatly affects the condensation with FA, which is more likely the rate-determining step. Moreover, the newly identified optimal N-p-methoxybenzyl homoallylamine moiety with a self-immolative β-elimination linker was generally utilized to construct a series of fluorescent probes with varying excitation/emission wavelengths for sensitive and selective detection of FA in aqueous solutions and live cells. Among these probes, the near-infrared probe FFP706 has been well demonstrated to enable direct fluorescence visualization of steady-state endogenous FA in live mouse brain tissues and elevated FA levels in a mouse model of breast cancer. This study provides the optimal aza-Cope reaction moiety for FA probe development and new chemical tools for fluorescence imaging and biological investigation of FA in living systems.

Endogenous gaseous signaling molecules including nitric oxide (NO), carbon monoxide (CO) and hydrogen sulfide (H2S) have been demonstrated to perform significant physiological and pharmacological functions and are associated with various diseases in biological systems. In order to obtain a deeper insight into their roles and mechanisms of action, it is desirable to develop novel techniques for effectively detecting gaseous signaling molecules. Small-molecule fluorescent probes have been proven to be a powerful approach for the detection and imaging of biological messengers by virtue of their non-invasiveness, high selectivity, and real-time in situ detection capability. Based on the intrinsic properties of gaseous signaling molecules, numerous fluorescent probes have been constructed to satisfy various demands. In this perspective, we summarize the recent advances in the field of fluorescent probes for the detection of NO, CO and H2S and illustrate the design strategies and application examples of these probes. Moreover, we also emphasize the challenges and development directions of gasotransmitter-responsive fluorescent probes, hoping to provide a general implication for future research.

This perspective article aims to introduce the design principles and recognition strategies of small-molecule fluorescent probes which are applied for the detection of gas signaling molecules including NO, CO and H2S in biological systems.  相似文献

Nassarius reticulatus (whelk) imposex levels and organotin body burden (b.b.) were surveyed along the Portuguese coast, from Vila Praia de Âncora (northern limit) to Lagos (southern limit), between May and August 2003. The percentage of females affected with imposex (%I), the relative penis length index (RPLI), the vas deferens sequence index (VDSI) and the degree of female oviduct convolution index (AOS) were used to assess the level of imposex at each site. These imposex indices were determined for 23 sampling stations throughout the coast and were in the range 0.0–100%, 0.0–90%, 0.0–5.0 and 0.0–1.3 respectively. Sterile females (i.e. females carrying aborted egg capsules inside the capsule gland) were found inside the harbours of Viana do Castelo (8.5%) and Aveiro (3.7%). Organotin compounds were assessed at 10 sampling sites spread along the coast. Tributyltin (TBT) b.b. in females varied between 39 and 1679 ng g?1 (as tin) dry weight, and dibutyltin (DBT) and monobutyltin (MBT) varied in the ranges 23–1084 ng g?1 (as tin) d and 18–939 ng g?1 dry wt respectively. Among the butyltins, the major fraction corresponded to TBT (47.4%), followed by DBT (27.6%) and MBT (25.0%), which indicates recent TBT inputs. Triphenyltin (TPT) levels ranged from <5 to 21 ng g?1 (as tin), and, when quantifiable, represented on average 10% of that of TBT. TPT was the dominant phenyltin and was detected in 60% of the sampling stations. The imposex was significantly correlated to ln (TBT) (Spearman r = 0.918, p < 0.001 for RPLI; r = 0.864, p < 0.001 for VDSI; r = 0.828, p < 0.01 for AOS). The higher levels of imposex and TBT contamination occurred inside or close to harbours, which we identified as ‘hotspots’ of pollution along the coast. Comparing the results obtained in the current work with those reported in a similar survey in 2000, imposex and TBT b.b. varied locally but did not reveal any global trend in the variation of TBT pollution along the Portuguese coast over the 3 year period. Recent reports from our laboratory on the occurrence of methylbutyltins in marine sediments and seawater suggest that these compounds are formed in the environment by the methylation of both tributyltin (TBT) and that­of its degradation products, i.e. dibutyltin and monobutyltin, to give MenBu(4?n)Sn for which n = 1, 2 and 3 respectively. We investigated the possibility of inducing methylation of TBT in seawater–sediment mixtures in experiments carried out in vitro using environmental materials collected from a yacht marina in Msida, Malta. Three water–sediment mixtures, which were shown to contain TBT, dibutyltin and monobutyltin but no other organotins, were spiked with tributyltin chloride (90 mg in 100 ml sea‐water/100 ml sediment); to one mixture was added sodium acetate and to another methanol, to act as possible additional carbon sources, and all mixtures were allowed to stand at 25 °C in stoppered clear‐glass bottles in diffused light for a maximum of 315 days. Speciation and quantification of organotins was performed using aqueous phase boroethylation with simultaneous solvent extraction followed by gas chromatography with flame photometric detection. The atmosphere inside the bottles quickly became reducing with abundant presence of H2S, and after an induction period of about 112 days, and only in the reaction mixture containing methanol, methyltributyltin (MeBu3Sn) was observed in both sediment (maximum concentration 0.87 µgSn g?1) and overlying water (maximum concentration 6.0 µgSn l?1). The minimum conversion yield of TBT into MeBu3Sn was estimated to be 0.3%. MeBu3Sn has a significantly lower affinity for sediment than TBT and, therefore, is more mobile in the marine environment, possibly also migrating into the atmosphere to generate a hitherto unsuspected flux of organotin into that phase. Butyltin concentrations have been measured at eight freshwater sites (rivers and lakes) in the Norfolk Broads, UK, during 1986 and 1987. Tributyltin (TBT) was found in water samples from seven of the sites. Wherever TBT was present, dibutyltin and (usually) monobutyltin occurred. Levels of TBT exceeded 100 ng dm?3 in open stretches of both the Rivers Bure and Yare in 1986 and 1987. The highest concentration of TBT recorded for Wroxham Broad (a shallow lake) was 898 ng dm?3. Values of up to 3.26 μg dm?3 were measured in water samples taken from a marina. A depth profile for Wroxham Broad showed TBT to be uniformly distributed throughout the shallow water column. The results are discussed in relation to toxicity of TBT to freshwater organisms. Laboratory measurements of the degradation of TBT in freshwater from a marina gave a half-life of six days at 20°C in the light.  相似文献

The presence of compounds of tributyltin (TBT), dibutyltin (DBT) and monobutyltin (MBT) was determined in outdoor settled dust collected from several sites on the island of Malta, mainly from flat rooftops of school buildings. The dust was separated into three size fractions with diameters (µm) > 250, 125–250 and < 125, and the two finer fractions were analysed for butyltins using extraction with glacial acetic acid followed by derivatization/solvent extraction with sodium tetraethylborate in the presence of iso‐octane and quantitation by gas chromatography with flame photometric detection. The presence of TBT, DBT and MBT was established in most of the samples and TBT concentrations varied from non‐detectable (<5 ng Sn g?1) to highs of 15.5 and 18.7 µg Sn g?1 in Senglea and Marsaxlokk. TBT was generally found at concentrations significantly higher than reported hitherto in house dust collected from European homes. The geographical distribution of total organotins in both dust fractions suggests that TBT originates mainly from antifouling marine paint residues which contaminate the urban environment when ships' hulls are sand‐ or hydro‐blasted during maintenance and repair at the drydocks facility in Grand Harbour. Other significant sources of TBT are located at Marsaxlokk fishing port and Wied i??urrieq creek, both hosting sizeable communities of fishermen and leisure boating. The data also suggest that the municipal solid waste landfill at Maghtab is an inland source of butyltins. We suggest that dust containing harmful butyltins could possibly be ingested to expose humans to a risk which is probably of concern especially for young children living close to the hotspots of contamination. We show that the conversion of a known intercalating dye (i.e., thiazole orange) into a bivalent protein binder could lead to the realization of a novel class of ‘turn-on’ fluorescent molecular probes that detect proteins with high affinity, selectivity, and a high signal-to-noise (S/N) ratio. The feasibility of the approach is demonstrated with monomolecular probes that light-up in the presence of three different proteins: acetylcholinesterase (AChE), glutathione-s-transferase (GST), or avidin (Av) at low concentrations and with minimal background signal. The way by which such probes can be used to detect individual protein isoforms and be applied in inhibitor screening, cell imaging, and biomarker detection is described.  相似文献

Fluorescent probes that can selectively detect tumour lesions have great potential for fluorescence imaging-guided surgery. Here, we established a library-based approach for efficient screening of probes for tumour-selective imaging based on discovery of biomarker enzymes. We constructed a combinatorial fluorescent probe library for aminopeptidases and proteases, which is composed of 380 probes with various substrate moieties. Using this probe library, we performed lysate-based in vitro screening and/or direct imaging-based ex vivo screening of freshly resected clinical specimens from lung or gastric cancer patients, and found promising probes for tumour-selective visualization. Further, we identified two target enzymes as novel biomarker enzymes for discriminating between tumour and non-tumour tissues. This library-based approach is expected to be an efficient tool to develop tumour-imaging probes and to discover new biomarker enzyme activities for various tumours and other diseases.

Efficient methodology to develop tumor-imaging fluorescent probes based on screening with our newly constructed probe library for aminopeptidase/protease (380 probes) and clinical samples has been established.  相似文献

The application of fluorescent probes for in vivo retinal imaging is of great importance, which could provide direct and crucial imaging evidence for a better understanding of common eye diseases. Herein, a group of bright organic luminogens with typical electron-donating (D) and electron-accepting (A) structures (abbreviated as LDs-BDM, LDs-BTM, and LDs-BHM) was synthesized through a simple single-step reaction. They were found to be efficient solid-state emitters with high fluorescence quantum yields of above 70% (e.g., 83.7% for LDs-BTM). Their light-emission properties could be tuned by the modulation of π-conjugation effect with methoxy groups at different substituent positions. Their resulting fluorescent nanoparticles (NPs) were demonstrated as specific lipid droplets (LDs) targeting probes with high brightness, good biocompatibility, and satisfactory photostability. LDs-BTM NPs with a large two-photon absorption cross section (σ2 = 249 GM) were further utilized as ultrabright two-photon fluorescence (2PF) nanoprobes for in vivo retina imaging of live zebrafish by NIR excitation at an ultralow concentration (0.5 µmol/L). Integrated histological structures at the tissue level and corresponding fine details at the cellular level of the embryonic retina of live zebrafish were clearly demonstrated. This is the first report of using ultrabright LDs-targeting nanoprobes to accurately measure fine details in the retina with 2PF microscopic technique. These good results are anticipated to open up a new avenue in the development of efficient 2PF emitters for non-invasive bioimaging of living animals.  相似文献

The ban on the use of tributyltin (TBT) is promoting an increasing use of copper as an active biocide in antifouling paints, with consequent rising levels of this metal in the environment. This study assesses the acute toxicity of copper and tributyltin to the larvae of the mollusc gastropod Nassarius reticulatus. Recently hatched veligers were exposed to nominal TBT‐Sn concentrations of 0.9, 1.4, 1.9, 2.8, 3.8, 4.7 and 5.6 µg l?1 and nominal copper concentrations of 9.4, 23.4, 46.9, 70.3, 93.8, 117.2, 140.6 and 164.1 µg l?1 for up to 96 h, under static conditions (17 ± 1 °C and 33 ± 1 psu). The percentage of larval mortality was determined for each organometal/metal concentration and exposure time (1, 24, 48, 72 and 96 h). Both TBT and copper had a highly significant effect on larvae survival (p < 0.001) for all times of exposure, except for the first hour in the particular case of TBT. The lowest observed effect concentration for TBT‐Sn decreased over time from 3.8 µg l?1 at 24 h to 1.9 µg l?1 at 96 h, whereas for copper it remained constant over time (46.9 µg l?1). The median lethal concentration (LC50) for TBT‐Sn decreased from 4.87 µg l?1 at 24 h to 1.78 µg l?1 at 96 h, and the LC50 for copper decreased from 83.08 µg l?1 at 24 h to 58.84 µg l?1 at 96 h. TBT is far more toxic to N. reticulatus larvae than copper. However, owing to the higher copper environmental concentrations, the risk factors of the two biocides may approach each other. Butyltin (tributyltin TBT; dibutyltin DBT and monobutyltin MBT) speciation was measured in the liver of beluga whales from the St Lawrence Estuary and Hudson Strait (northern Quebec). Using GC–MS, liver samples were analysed from 21 beluga whales found dead, stranded along the shores of the St Lawrence during the period 1995–1998. In all cases, including a neonate specimen, the liver was contaminated with butyltin compounds with concentrations in the range 0.04–2.1 mg Sn kg−1 on a dry weight basis. Liver samples of five beluga whales from Hudson Strait obtained in the summer of 1998 were also analysed. For these animals, hepatic butyltin concentrations were consistently below the detection limit (<0.5 ng Sn g−1 for MBT and <0.2 ng Sn g−1 for DBT and TBT). Compared with published data on the contamination by TBT of the marine mammals of the St Lawrence in 1988, these contemporary results clearly indicate that the level of contamination of the beluga whales in this coastal marine ecosystem has not decreased ten years after regulating the use of TBT‐based antifoulants on small craft. Nucella lapillus imposex—superimposition of male characters onto prosobranch (a subclass of gastropod molluscs) females—and organotin female body burden were surveyed on the Portuguese coast, from Vila Praia de Âncora (northern limit) to Praia da Luz (southern limit), at 17 sampling stations, between May and August 2003. The vas deferens sequence index (VDSI), the relative penis size index (RPSI), the percentage of females affected with imposex (%I) and the percentage of sterile females (%S) were used to assess the level of imposex at each site. VDSI, RPSI and %I were 0.20–4.04, 0.0–42.2% and 16.7–100.0%, respectively. Sterile females were found at stations 2 (6.2%), 5 (4.0%) and 7 (5.0%). Tributyltin (TBT) and dibutyltin (DBT) female body burdens were 23–138 and <10–62 ng Sn/g dry weight, respectively. TBT female body burden was significantly correlated with RPSI and VDSI [Spearman rank order linear correlation: RPSI vs TBT body burden (b.b.) r = 0.71, p < 0.01; VDSI vs logTBT body burden r = 0.71, p < 0.01]. Imposex and TBT b.b. were highest at sites located in the proximity of harbours, where TBT leaching from antifouling paints is more intense owing to the high concentration of ships and dockyard activities. Organic phosphate biomolecules (OPBs) are indispensable components of eukaryotes and prokaryotes, such as acting as the fundamental components of cell membranes and important substrates for nucleic acids. They play pivotal roles in various biological processes, such as energy conservation, metabolism, and signal modulation. Due to the difficulty of detection caused by variety OPBs, investigation of their respective physiological effects in organisms has been restrained by the lack of efficient tools. Many small fluorescent probes have been employed for selective detection and monitoring of OPBs in vitro or in vivo due to the advantages of tailored properties, biodegradability and in situ high temporal and spatial resolution imaging. In this review, we summarize the recent advances in fluorescent probes for OPBs, such as nucleotides, NAD(P)H, FAD/FMN and PS. Importantly, we describe their identification mechanisms in detail and discuss the general strategies for these OPBs probe designs, which provide new insights and ideas for the future probe designs.  相似文献

The spatial and temporal behaviours of the organotin compounds (OTCs) (butyl- and phenyltin) were investigated in the Manko and Okukubi protected estuarine ecosystems on Okinawa Island, Japan from February to October 2006. Butyltin compounds (BTCs) were frequently detected in all seasons, while phenyltin (PhTs) were found in winter and early spring. In Manko estuary, the total mean concentrations of BTCs and PhTs were 22.78?±?30.85, (mean?±?SD, n?=?53) and 0.08?±?0.27?ng(Sn)?L?1, respectively. In Okukubi estuary, BTCs and PhTs were 12.58?±?23.96 and 0.47?±?1.67 (n?=?55) ng(Sn)?L?1, respectively. The Manko sediments can be classified as lightly contaminated, while the Okukubi sediments were uncontaminated with tributyltin (TBT). The mean levels of TBT shown in Manko estuary exceeded the threshold level and represent an ecotoxicological risk to sensitive aquatic life. Generally, the present study reports the occurrence and continuous input of OTCs in the protected estuaries, even 16 years after legal restriction of TBT usage in coastal waters was implemented by the Japanese Environmental Authorities.  相似文献

Fluorescent imaging has been expanded, as a non-invasive diagnostic modality for cancers, in recent years. Fluorescent probes in the near-infrared window can provide high sensitivity, resolution, and signal-to-noise ratio, without the use of ionizing radiation. Some fluorescent compounds with low molecular weight, such as rhodamine B (RhB) and indocyanine green (ICG), have been used in fluorescent imaging to improve imaging contrast and sensitivity; however, since these probes are excreted from the body quickly, they possess significant restrictions for imaging. To find a potential solution to this, this work investigated the synthesis and properties of novel macromolecular fluorescent compounds. Herein, water-soluble dextran fluorescent compounds (SD-Dextran-RhB) were prepared by the attachment of RhB and sulfadiazine (SD) derivatives to dextran carrier. These fluorescent compounds were then characterized through IR, 1H NMR, 13C NMR, UV, GPC, and other methods. Assays of their cellular uptake and cell cytotoxicity and fluorescent imaging were also performed. Through this study, it was found that SD-Dextran-RhB is sensitive to acidic conditions and possesses low cell cytotoxicities compared to normal 293 cells and HepG2 and HeLa tumor cells. Moreover, SD-Dextran-RhB demonstrated good fluorescent imaging in HepG2 and HeLa cells. Therefore, SD-Dextran-RhB is suitable to be potentially applied as a probe in the fluorescent imaging of tumors.  相似文献

Increasingly serious microbial infections call for the development of new simpler methods for the precise diagnosis and specific inhibition of such pathogens. In this work, a peptide mineralized Au cluster probe was applied as a new simplified strategy to both recognize and inhibit a single bacteria species of Staphylococcus aureus (S. aureus) simultaneously. The probes are composed of peptides and Au clusters. Moreover, the peptides specifically target S. aureus cells and the Au clusters provide fluorescent imaging and have an antibacterial effect. These new probes enable the simultaneous specific detection and effective destruction S. aureus cells in situ.  相似文献
