This paper discusses the requirement for, and presents an analytical procedure for, the determination of four steroid hormones and a conjugated steroid (estrone-3-sulfate) in wastewaters. The method utilizes LC/MS/MS following solid phase extraction and a two stage clean-up procedure, achieving limits of detection of 0.2 ng l(-1) for estriol, 17beta-estradiol and 17 alpha-ethinylestradiol, and 0.1 ng l(-1) for estrone and the conjugate. The approach demonstrates that using appropriate clean-up and deuterated internal standards, the impact of matrix effects on ionization can be overcome to reliably determine estrogens at environmentally relevant concentrations. The robustness of the method was demonstrated by achieving recoveries of >83% for all steroids in settled sewage and final effluent samples with relative standard deviations of 0.5-12%.  相似文献

An improved LC-electrospray ionization MS method was established for four estrogens (17beta-estradiol (E2), estriol (E3), estrone (E1), and ethynyl estradiol (EE)) in environmental water. Almost complete separation of all estrogens was achieved on a phenyl column with methanol/water as the mobile phase. Quantification was achieved in the negative ionization mode using selected ion monitoring. The instrumental detection limits were 20-30 ng/l for the four analytes. In Milli-Q spiked water, the recoveries of the four estrogens were 72-81%, which was similar to those found for river water spiked with the corresponding deuterated estrogens. The detection limits for the four estrogens in river water were 0.1-0.2 ng/l. The method was used to detect residual estrogens in the Tonghui River, which receives water from a municipal sewage treatment plant in Beijing; E1 (1.1 ng/l) and E2 (0.2 ng/l) were detected.  相似文献

A method for the quantitative determination of three macrolides, five sulfonamides, ranitidine, omeprazole and trimethoprim in sewage sludge samples was developed by using pressurized liquid extraction and high-performance liquid chromatography-electrospray ionization mass spectrometry. The extraction solvent and such operational parameters as temperature, pressure, extraction time and purge time were optimized in pressurized liquid extraction. The experimental conditions were: an extraction solvent of water (pH 3):methanol (1:1, v/v), a temperature of 80 degrees C, a pressure of 1500 psi, a sample weight of 5 g, an extraction time of 5 min, one cycle, a flush volume of 60% and a purge time of 120 s. All recoveries were over 74%, except those for ranitidine whose value was 54%. The repeatability and reproducibility between days expressed as relative standard deviation (n = 3) were lower than 11% and 15%, respectively. The limit of detection values ranged from 2 to 11 microg/kg dry weight (d.w.). The method was applied to determine the pharmaceuticals in sewage sludge from two domestic sewage treatment plants. Roxithromycin and tylosin were determined in the samples and tylosin showed the highest value (4.0 mg/kg d.w.).  相似文献

A multi-residue analytical method has been developed for the determination of various classes of selected endocrine disruptors. This method allows the simultaneous extraction and quantification of different estrogens (estradiol, estrone, estriol, estradiol-17-glucuronide, estradiol diacetate, estrone-3-sulfate, ethynyl estradiol and diethylstilbestrol), pesticides (atrazine, simazine, desethylatrazine, isoproturon and diuron), and bisphenol A in natural waters. In the method developed, 500 ml of water are preconcentrated on LiChrolut RP-18 cartridges. Further analysis is carried out by liquid chromatography-mass spectrometry (LC-MS) using atmospheric pressure chemical ionisation (APCI) in the positive ion mode for determination of pesticides and electrospray in the negative ionisation mode for determination of estrogens and bisphenol A. Recoveries for most compounds were between 90 and 119%, except for bisphenol A (81%) and diethylstilbestrol (70%), with relative standard deviations below 20%. Limits of detection ranged between 2 and 15 ng/l. The method was used to study the occurrence of the selected pollutants in surface and groundwater used for abstraction of drinking water in a waterworks and to evaluate the removal efficiency of the different water treatments applied. Water samples from the river, the aquifer, and after each treatment stage (sand filtration, ozonation, activated carbon filtration and post-chlorination) were taken monthly from February to August of 2002. The presence in river water of atrazine, simazine, diuron and bisphenol A were relatively frequent at concentrations usually below 0.1 microg/l. Lower levels, below 0.02 microg/l, were usual for isoproturon. Estrone-3-sulfate and estrone were detected occasionally in the river. Most of the compounds were completely removed during the water treatment, especially after activated carbon filtration.  相似文献

We report a column-switching liquid chromatography (LC) tandem mass spectrometry (MS/MS) method for highly sensitive determination of both free estrogens (estrone, estradiol, and estriol) and their conjugates (estrone-3-sulfate, estradiol-3-sulfate, estriol-3-sulfate, estrone-3-glucuronide, estradiol-3-glucuronide, estriol-16-glucuronide, and estriol-3-glucuronide) in river water. This technique combines reversed phase (RP) chromatographic separation of the dansyl chloride derivatized free estrogens and hydrophilic interaction liquid chromatographic (HILIC) separation of the estrogen conjugates with multiple reaction monitoring (MRM). Using this new method, sensitivity increases 100- to 1000-fold for free estrogens and 2- to 10-fold for estrogen conjugates over RPLC-MS/MS alone. Method detection limits (MDL) range from 0.038 to 6.9 ng L−1 with accuracy of 68-105% and precision of 1.7-17%. We successfully used this method to analyze river water samples collected from the North Saskatchewan River at the same location and detected trace concentrations of estrone (0.042 ng L−1) and estrone-3-sulfate (0.84 ng L−1), demonstrating the application of this method for environmental analysis.  相似文献

Although there are existing methods for determining estrogen in human bodily fluids including blood plasma and serum, very little information is available regarding estrogen levels in human cerebrospinal fluid (CSF), which is critical to assess in studies of neuroprotective functions and diffusion of neuroprotective estrogens across the blood–brain barrier. To address this problem, a liquid chromatography with tandem mass spectrometry method for the simultaneous quantification of four endogenous estrogens (estrone, 17α‐estradiol, 17β‐estradiol, and estriol) in human CSF was developed. An aliquot (300 μL) of human CSF was bulk derivatized using dansyl chloride in the sample and 10 μL was directly injected onto a restricted‐access media trap column for protein removal. No off‐line sample extraction or cleanup was needed. The limits of detection of estrone, 17α‐estradiol, 17β‐estradiol, and estriol were 17, 28, 13, and 30 pg/mL, respectively, which is in the parts‐per‐trillion regime. The method was then applied to human CSF collected from ischemic trauma patients. Endogenous estrogens were detected and quantified, demonstrating the effectiveness of this method.  相似文献

A simple and selective high‐performance liquid chromatography method coupled with fluorescence detection was developed for the simultaneous measurement of trace levels of four estrogens (estrone, estradiol, estriol and 17α‐ethynyl estradiol) in environmental matrices. For feces samples, solid–liquid extraction was applied with a 1:1 v/v mixture of acetonitrile and ethyl acetate as the extraction solvent. For liquid samples (e.g., leachate and groundwater), hydrophobic/lipophilic balanced automated solid‐phase extraction disks were selected due to their high recoveries compared to conventional C18 disks. Chromatographic separations were performed on a reversed‐phase C18 column gradient‐eluted with a 45:55 v/v mixture of acetonitrile and water. The detection limits were down to 1.1 × 10?2 (estrone), 4.11 × 10?4 (estradiol), 5.2 × 10?3 (estriol) and 7.18 × 10?3 μg/L (17α‐ethynyl estradiol) at excitation/emission wavelengths of 288/310 nm, with recoveries in the range of 96.9 ± 3.2–105.4 ± 3.2% (n = 3). The method was successfully applied to determine estrogens in feces and water samples collected at livestock farms and a major river in Northeast China. We observed relatively high abundance and widespread distribution of all four estrogens in our sample collections, implying the urgency for a comprehensive and intricate investigation of estrogenic fate and contamination in our researched area.  相似文献

A method for the simultaneous determination of 16 pharmaceutical compounds in three types of sewage sludge (primary, secondary and anaerobically digested dehydrated sludge), compost and sediment samples is described. Pharmaceutical compounds evaluated were nonsteroidal anti‐inflammatory drugs (acetaminophen, diclofenac, ibuprofen, ketoprofen, naproxen and salicylic acid), antibiotics (sulfamethoxazole and trimethoprim), an anti‐epileptic drug (carbamazepine), a β‐blocker (propranolol), a nervous stimulant (caffeine), estrogens (17α‐ethinylestradiol, 17β‐estradiol, estriol and estrone) and lipid regulators (clofibric acid, metabolite of clofibrate and gemfibrozil). The method is based on the ultrasonic‐assisted extraction, clean‐up by SPE and analytical determination by HPLC with diode array and fluorescence detectors. The best extraction recoveries were achieved in a three‐step extraction procedure with methanol and acetone as extraction solvents. Extraction recoveries of several pharmaceutical compounds as caffeine were highly dependent on the type of sample evaluated. The applicability of the method was tested by analyzing primary, secondary and anaerobically digested dehydrated sludge, compost and sediment samples from Seville (Southern Spain). Ten of the sixteen pharmaceutical compounds were detected in sludge samples and five in compost and sediment samples. The highest concentration levels were recorded for ibuprofen in sewage samples, whereas salicylic acid and 17α‐ethinylestradiol were detected in all of the samples analyzed.  相似文献

A simple, rapid and sensitive method for the determination of five estrogens, estrone, 17beta-estradiol, estriol, ethynyl estradiol, and diethylstilbestrol, was developed using a fully automated method consisting of in-tube solid-phase microextraction (SPME) coupled with liquid chromatography-tandem mass spectrometry (LC/MS/MS). These estrogens were separated within 8 min by HPLC using an XDB-C8 column and 0.01% ammonia/acetonitrile (60/40, v/v) at a flow rate of 0.2 mL/min. Electrospray ionization conditions in the negative ion mode were optimized for MS/MS detection of the estrogens. The optimum in-tube SPME conditions were 20 draw/eject cycles of 40 microL of sample using a Supel-Q PLOT capillary column as an extraction device. The extracted compounds were easily desorbed from the capillary by passage of the mobile phase, and no carryover was observed. Using the in-tube SPME LC/MS/MS method, good linearity of the calibration curve (r > or = 0.9996) was obtained in the concentration range from 10 to 200 pg/mL for all compounds examined. The limits of detection (S/N= 3) of the five estrogens examined ranged from 2.7 to 11.7 pg/mL. The in-tube SPME method showed 34-90-fold higher sensitivity than the direct injection method (5 microL injection). This method was applied successfully to the analysis of environmental water samples without any other pretreatment and interference peaks. Several surface water and wastewater samples were collected from the area around Asahi River, and estriol was detected at 35.7 pg/mL in the effluent of a sewage treatment plant. The recoveries of estrogens spiked into river waters were above 86%, except for estriol, and the relative standard deviations were below 0.9-8.8%.  相似文献

A procedure for the determination of six natural and synthetic estrogens (diethylstilbestrol, estrone, 17beta-estradiol, mestranol, 17alpha-ethinylestradiol and estriol) in water samples is described. Samples, up to 2000 ml, were concentrated using Oasis HLB solid-phase extraction cartridges. Analytes were derivatized with N-methyl-N-(trimethylsilyl)trifluoroacetamide and determined by GC-MS or GC-MS-MS. The reactivity of several silylation reagents versus aliphatic and aromatic hydroxyl groups contained in the structure of the selected analytes was evaluated. Influence of parameters such as sample pH, nature of the water samples and derivatization conditions on the performance of the whole analytical procedure was systematically studied. Under optimal conditions, quantification limits between 1 and 3 ng/l were achieved for the determination of the considered estrogens in sewage water.  相似文献

An analytical method has been developed for the determination of five naturally occurring estrogens (estradiol, estriol, estrone, genistein, daidzein), one synthetic hormone (ethynylestradiol) and three xenoestrogens (4-nonylphenol (NP), 4-tert-octylphenol (4-tert-OP), bisphenol A (BPA)) in coastal marine waters. The procedure includes a solid-phase extraction of approx. fifty litres of water samples on the solid-phase copolymer Oasis HLB followed by a clean-up on silica. Twenty-five percent aliquots were used for the analytical determination of the analytes using high performance liquid chromatography coupled with electrospray-ionisation tandem mass spectrometry (HPLC-ESI-MS/MS). Calculated extraction recoveries between 52 (4-tert-octylphenol) and 91% (nonylphenol) were obtained for the method developed. Matrix interferences occurring during electrospray ionisation were quantified by spiking the extracts prior to the measurements. Method detection limits ranged from 0.02 (estrone) to 1 ng L(-1) (estriol). The method was applied to determine environmental estrogens in coastal waters of the Baltic Sea. The analyses showed the presence of five compounds at levels between 0.10 (estrone) and 17 ng L(-1) (ethynylestradiol).  相似文献

A simplified, in-line solid-phase extraction and clean-up process is presented for rapid extraction and analysis of steroid estrogens and nonylphenols by gas chromatography with tandem mass spectrometry. Isotope dilution with multiple deuterated standards has been developed, evaluated, and verified using this method against standard addition for the analysis of estrone, estradiol, estriol, ethinylestradiol, and 4-nonylphenols in septic, soil, and groundwater matrices. The accuracy and precision of isotope dilution was comparable to standard addition yet was able to quantify analytes with the use of fewer samples by including [(2)H(4)]estradiol, [(2)H(4)]ethinylestradiol, [(2)H(4)]nonylphenol, and estradiol-17-acetate as internal standards. Specific details and validation of the method are presented as are data showing the occurrence of estrogens and nonylphenols in an on-site wastewater treatment (septic) system with advanced stages of pretreatment and recirculation to an office building for non-potable reuse.  相似文献

建立了饲料和卧床土中雌三醇、雌二醇、雌酮、双酚A和己烯雌酚5种环境雌激素的固相萃取结合高效液相色谱-串联质谱(HPLC-MS/MS)测定方法。对色谱流动相、质谱条件、固相萃取柱等影响因素进行了优化,得到的最优化条件为:样品经乙腈提取后,用固相萃取柱(NH2-SPE)进行富集,采用Acquity UPLCTM HSS T3色谱柱分离,以乙腈-甲醇(4:1, v/v)与0.01%氨水为流动相,采用梯度洗脱,在负离子模式下进行MS/MS测定。在该优化条件下,5种环境雌激素的检出限(以信噪比为3计)为0.06~0.22 μg/kg,回收率为81.70%~102.20%,相对标准偏差小于10.00%。该方法用于饲料和卧床土中的5种环境雌激素残留量的测定具有简便、快速、灵敏的特点。  相似文献

A rapid, selective, sensitive, and reproducible method for the analysis of environmental estrogens, either natural or synthetic, present in samples from sewage treatment works (STW) has been developed. Isolation of these compounds from STWs was performed by applying a simple extraction procedure using an ENVI-CARB cartridge (a graphitized non porous carbon black) as the solid-phase extraction (SPE) system. Liquid chromatography coupled with atmospheric pressure chemical ionization (APCI) and tandem mass spectrometry was used for detection. For the multiple reaction monitoring (MRM) mode, the [M + H](+) ion of estrone and the [M + H-H(2)O](+) ions of 17beta-estradiol, estriol and 17alpha-ethynylestradiol were selected as the precursors for collisionally induced dissociation (CID). The average recoveries from sewage final effluent samples ranged from 84 to 93% for low levels, and from 89 to 95% for high levels. The precision of the method ranged from 11 to 8% for low level and from 9 to 7% for high level samples. The lower level of quantitation for these estrogens in STW samples was determined at 0.5 ng/L for 17beta-estradiol and 17alpha-ethynylestradiol, and 1 ng/L for estrone and estriol, based on 1-L aliquots of sewage treatment works water, using the optimum tuning parameters for each individual selected precursor ion/product ion transition. Compared to a previous gas chromatography/mass spectrometry (GC/MS) method for the analysis of ten STW samples, this method was shown to provide higher sensitivity and lower time consumption.  相似文献

Estrone, 17β‐estradiol and 17α‐ethinylestradiol are increasingly recognised as important micropollutants to be monitored in wastewater treatment plants. These estrogens are retained onto sludge due to their high adsorption and since they are largely used in land applications, the monitoring of these chemicals in sludge samples is of great importance. This study describes a method for the determination of estrone and 17α‐ethinylestradiol in fresh sludge samples. After spiking fresh digested sludge with estrone and 17α‐ethinylestradiol and maintaining in contact during 5, 30 and 60 min, the freeze‐dried samples were subjected to ultrasonic liquid extraction, with methanol and acetone, and analysed by high‐performance liquid chromatography with fluorescence detection. The average recoveries obtained for estrone and 17α‐ethinylestradiol using the different contact times were 103 ± 3 and 97 ± 4%, respectively. Fresh sludge samples from one waste water treatment plant located in Portugal were analysed and estrone was detected in primary fresh sludge, anaerobic digested sludge and dehydrated sludge at a concentration in the range of 1–4.8 μg/g. The method here developed does not require any sample clean‐up, being fast and simple, reliable and inexpensive, making possible its application for monitoring the contamination of sludge with these estrogens.  相似文献

The development of a simple and sensitive analytical approach that combines multiple monolithic fiber solid‐phase microextraction with liquid desorption followed by high‐performance liquid chromatography with diode array detection is proposed for the determination of trace levels of seven steroid sex hormones (estriol, 17β‐estradiol, testosterone, ethinylestradiol, estrone, progesterone and mestranol) in water and urine matrices. To extract the target analytes effectively, multiple monolithic fiber solid‐phase microextraction based on a polymeric ionic liquid was used to concentrate hormones. Several key extraction parameters including desorption solvent, extraction and desorption time, pH value and ionic strength in sample matrix were investigated in detail. Under the optimal experimental conditions, the limits of detection were found to be in the range of 0.027–0.12 μg/L. The linear range was 0.10–200 μg/L for 17β‐estradiol, 0.25–200 μg/L estriol, ethinylestradiol and estrone, and 0.50–200 μg/L for the other hormones. Satisfactory linearities were achieved for analytes with the correlation coefficients above 0.99. Acceptable method reproducibility was achieved by evaluating the repeatability and intermediate precision with relative standard deviations of both less than 8%. The enrichment factors ranged from 54‐ to 74‐fold. Finally, the proposed method was successfully applied to the analysis of steroid sex hormones in environmental water samples and human urines with spiking recoveries ranged from 75.6 to 116%.  相似文献

Abstract

We report a sensitive high performance liquid chromatography (HPLC) method for determination of free and conjugated estrogens (estrone, estradiol and estriol) by a fluorescent pre-labeling regent, 2-(4-carboxyphenyl)-5,6-dimethylbenzimidazole, with modification of previous work. The modified method was also tried, in preliminary work, for diagnosis of the in-vitro fertilization embryo transfer (IVF-ET) process. The reagent volumes were changed to one-tenth, derivatization conditions were changed to mild conditions at 40 C, and a solid-phase extraction process by SEP-PAK could be omitted after restudy of reaction conditions. As a result analysis time could be shorted within 40 min. The proposed HPLC method was applied to monitoring of free and conjugated estrogens in the patients who attend in-vitro fertilization embryo transfer (IVF-ET). The subsequent increase of free and conjugated estrone, estradiol and estriol was observed with the progress of follicle growth following ovulation stage in the IVF-ET process. We tried to plot estrogens for assist of clinical diagnosis of IVF-ET. The free estrone: 200-600 pg/ml, estradiol: 200-600 pg/ml and estriol: 100-300 pg/ml, conjugate estrone: 1000-5000 pg/ml, estradiol: 3000-8000 pg/ml and estriol: 2000-7000 pg/ml) in the patients without hormone disease were observed before human chorionic gonadotropin stimulation (hCG) on IVF-ET process. It was expected that free estrogen values, especially E1 and E3 could be use as validation products for diagnosis of hormone disease in IVF-ET process.  相似文献

An automated, environmentally friendly, simple, selective, and sensitive method was developed for the determination of ten primary aliphatic amines in sewage sludge at μg/kg dry weight (d.w.). The procedure involves a pressurized hot water extraction (PHWE) of the analytes from the solid matrix, followed by a fully automated on‐fiber derivatization with 2,3,4,5‐pentafluorobenzaldehyde (PFBAY) and headspace solid‐phase microextraction (HS‐SPME) and subsequent gas chromatography ion‐trap tandem mass spectrometry (GC‐IT‐MS‐MS) analysis. The limits of detection (LODs) of the method were between 0.5 and 45 μg/kg (d.w.) for all compounds except for ethyl‐, isopropyl‐, and amylamine, whose LODs were 70, 109, and 116 μg/kg (d.w.), respectively. The limits of quantification (LOQs) were between 10 and 350 μg/kg (d.w.). Repeatability and intermediate precision, expressed as RSD(%) (n=3), were lower than 18 and 21%, respectively. The method developed enabled to determine primary aliphatic amines in sludge from various urban and industrial sewage treatment plants as well as from a potable treatment plant. Most of the primary aliphatic amines were found in the sewage sludge samples analyzed corresponding to the maximum concentrations to the samples from the urban plant: for instance, isobutylamine and methylamine were found at 7728 and 12 536 μg/kg (d.w.), respectively. Amylamine was detected only in few samples but always at concentrations lower than its LOQ.  相似文献

建立了雌酮、雌二醇、雌三醇、己烯雌酚、己烷雌酚和炔雌醇6种雌激素在生物体中的HPLC-MS/MS分析方法.采用加速溶剂萃取、固相萃取技术进行提取、富集及净化,有效降低了基质的干扰.以甲醇-0.1%氨水溶液为流动相,以C18色谱柱进行分离,质谱采用电喷雾负离子扫描模式,6种雌激素的回收率为88%~104%,相对标准偏差在1.3%~8.3%之间.雌酮、雌二醇、雌三醇在生物体中的方法检出限0.35ng/g;己烯雌酚、己烷雌酚、17α-乙炔基雌二醇在生物体中的方法检出限为0.13ng/g.方法适用于生物体内雌激素的分析和检测.  相似文献
