Arylboronic acids having a remote azido group were prepared from the corresponding azidosubstituted aryl bromides via lithiation and treatment with trialkyl borates. Preparative yields were achieved when the starting aryl bromides possessed ortho-alkoxy groups, which would stabilize the intermediate aryllithium species. Conventional Suzuki cross-coupling of the arylboronic acids proceeded generally well with retention of azido group; however, sometimes azidomethyl fragment underwent oxidative transformation into a nitrile.  相似文献

We have developed shelf- and air-stable ortho-stannylated aniline reagents that can directly be coupled with alkenyl and aryl halides via Migita-Kosugi-Stille cross-coupling. We report (i) the efficient preparation of o-(tributylstannyl)aniline (2a) and o-(trimethylstannyl)aniline (2b), (ii) the comparison of the reactivities of 2a and 2b with those of related organostannanes in cross-coupling reaction with an alkenyl halide, and (iii) the cross-coupling of 2a and 2b with a series of arylhalides and triflate.  相似文献

1,5-Di(hetero)arylated-pyridin-2(1H)-one derivatives have been readily obtained in good yields starting from 2-fluoro-5-pyridylboronic acid. The sequence comprises three steps: (i) palladium-catalysed Suzuki-Miyaura reaction; (ii) base-catalysed hydrolysis; (iii) copper-catalysed C-N coupling. X-ray crystal structures are reported for selected pyridin-2(1H)-one derivatives. These compounds are of interest as new scaffolds for drug discovery.  相似文献

The air and moisture stable triphenylphosphine adduct of cyclopalladated ferrocenylimine 2 has been successfully used in palladium-catalyzed Suzuki cross-coupling for the synthesis of ortho-substituted biaryls in air. In the presence of 0.05 mol% of 2 as catalyst and 3 equivalent of CsF as base in dioxane at 100 °C, ortho-substituted biaryls were synthesized with moderate to high yields in the reactions of 2-methoxy-1-naphthylboronic acid with aryl halides, and 14 new ortho-substituted biaryls were obtained and characterized.  相似文献

The total synthesis of the opioid modulator pawhuskin C has been accomplished in eight steps from methyl 3,5-dihydroxybenzoate. Sugar-protected 6-halouridine derivatives underwent Suzuki-Miyaura cross-coupling reactions with arylboronic acids in the presence of palladium(II) acetate as a catalyst, triphenylphosphine as a ligand, and sodium carbonate as a base. A modified approach for the synthesis of 3-substituted 2-aryl/heteroaryl imidazo[4,5-b]pyridines utilising palladium catalysed cross-coupling reactions under microwave enhanced conditions has been achieved. utilisation of (A-taphos)2PdCl2-catalysed cross-coupling reactions enables rapid derivatization of this (imidazo[4,5-b]pyridines) pharmaceutically relevant core. This catalytic system is compatible with a broad spectrum of arylboronic acids—electron rich, electron poor, and heteroarylboronic acids.  相似文献

Chiral 3-N-arylsulfonyloxazolidin-2-ones 1a-f, prepared from (l)-amino acids, were reacted with lithium diisopropylamide in anhydrous THF and HMPA. The resulting new, optically active benzisothiazolinone 1,1-dioxides 2a-c and naphthisothiazolinone 1,1-dioxides 2d-f were obtained in good yields.  相似文献

A series of 9-(hetero)arylpurine derivatives has been prepared through N-arylation of 6-chloropurine with boronic acids in the presence of copper(II) acetate. Screening reaction conditions in terms of bases and solvents led to the successful coupling of a series of sterically demanding (hetero)arylboronic acids, never described so far. The coupling products were next readily converted into the target adenine derivatives. The described procedure provides easy access to original fragments for screening applications. Moreover these 9-aryl-6-chloropurine derivatives might be useful as intermediates for the preparation of purine derivatives with potential biological properties.  相似文献

One-pot synthesis of 2-aryl benzothiazoles from gem-dibromomethylarenes using 2-aminoarylthiols is described. Benzothiazoles were obtained in high chemical yields under mild conditions. Simple catalytic systems for cross-coupling reactions of acyl chlorides with arylboronic acids under microwave conditions were tested. Microwave irradiation facilitated the reaction course. Mild reaction conditions afford the symmetrical and unsymmetrical aryl ketones in reasonable to high yields within a short time. A wide range of substrates bearing an electron-donating or an electron-withdrawing substituent on aryl ring of acid chloride as well as on boronic acid were examined and high yields of ketones were produced.  相似文献

Axially dissymmetric P,S-heterodonor ligand L3 synthesized from BINOL is an effective promoter in the palladium(0)-catalyzed Suzuki cross-coupling reaction of phenylboronic acid with aryl bromides and iodide at 60-80 °C. A novel polymer-supported N-heterocyclic carbene (NHC) was prepared from chloromethyl polystyrene (CM PS) resin using a simple procedure, and was used as the ligand for palladium (Pd) catalysts. The activity of dimeric [Pd{C6H2(CH2CH2NH2)-(OMe)2-3,4}(μ-Br)]2 and monomeric [Pd{C6H2(CH2CH2NH2)-(OMe)2-3,4}Br(PPh3)] complexes as efficient, air, and moisture tolerant catalysts was investigated in Stille and Hiyama cross-coupling reactions of various aryl halides. Substituted biaryls were produced in excellent yields in short reaction times using these complexes. The monomeric complex had been demonstrated to be more active than the corresponding dimeric catalyst for the cross-coupling of some of aryl bromides and unreactive aryl chlorides. The combination of homogenous metal catalyst, microwave irradiation, and microwave-active polar solvents gave high yields of products in short reaction times.  相似文献

A rapid, efficient, and facile synthesis of an assortment of C-2 substituted imidazopyrazines has been achieved by utilizing the palladium catalyzed Suzuki cross-coupling of 2-bromo-1H-imidazo[4,5-b]pyrazine with various boronic acids under microwave irradiation. The utilization of(A-~(ta)phos)_2PdCl_2 as a catalyst in combination with Cs F as base and DME-H_2O(4:1) as the solvent system at 100℃ procured the diaryls in acceptable to excellent yields. Prominent features of this developed methodology include short reaction times, fewer side products, and exceptional tolerance to a wide variety of functional groups.  相似文献
