The β-galactosidase from bovine testes (EC 3.2.1.23) promotes the transfer of a galactose unit to glucose or galactose-containing residues in manifold derivatives, establishing β1→3 linkages.The synthesis of several potentially biologically important oligosaccharides β-d-Galp-(1→3)-α-d-Glcp-(1→2)-β-d-Fruf2, β-d-Galp-(1→3)-β-d-Galp-(1→4)-α,β-d-Glcp4, β-d-Galp-(1→3)-α-d-Glcp-(1→4)-d-Glcp-ol/Manp-ol 6, β-d-Galp-(1→3)-α-d-Glcp-(1→6)-β-d-Fruf8, β-d-Galp-(1→3)-α-d-Glcp-(1→6)-[α-d-Glcp-(1↔2)]-β-d-Fruf10, α-d-Galp-(1→6)-[β-d-Galp-(1→3)]-α-d-Glcp-(1↔2)-β-d-Fruf12, β-d-Galp-(1→3)-α-d-Glcp-(1↔2)-β-d-Fruf-(1↔2)-β-d-Fruf14 has been reached in yields between 7 and 44% by implementation of this specific enzyme. In addition, we found that it is feasible to gain high yields without an enzyme-specific buffer and even making upscaled preparation on a gram scale.  相似文献

Two new 24-hydroxylated asterosaponins,sodium(20R,24S)-6α-O-(4-O-sodiumsulfato-β-D-quinovopyranosyl)-5α-cholest-9(11)-en-3β,24-diol 3-sulfate(1) and sodium(20R,24S)-6α-O-[3-O-methyl-β-D-quinovopyranosyl-(1→2)-β-D-xylopyranosyl-(1→3)-β-D-glucopyranosyl]-5α-cholest-9(11)-en-3β,24-diol 3-sulfate(2),were isolated from the starfish Culcita novaeguineae.Their structures were elucidated by extensive spectral analysis and chemical evidences.  相似文献

The first total synthesis of ganglio-series gangliosides GQ1b, GT1b and GD1b, which contain α-sialyl-(2→8)-α-sialic acid residue in the structure, will be described. Glycosylation of 2-(trimethylsilyl)ethyl O-(2-acetamido-6-O-benzyl-2-deoxy-3,4-O-iso-propylidene-β- -galactopyranosyl)-(1→4)-O-(2,6-di-O-benzyl-β- -galactopyranosyl)-(1→4)-2,3,6-tri-O-benzyl-β-D-glucopyranoside (7) with methyl [phenyl 5-acetamido-8-O-(5-acetamido-4,7,8,9-tetra-O-acetyl-3,5-dideoxy- -glycero-α- -galacto-2-nonulopyranosylono-1′,9-lactone)-4,7-di-O-acetyl-3,5-dideoxy-2-thio- -glycero- -galacto-2-nonulopyranosid]onate (8) using N-iodosuccinimide (NIS)-trifluoromethanesulfonic acid (TfOH) in acetonitrile gave the protected GD2 pentasaccharide 9, which was converted into the pentasaccharide acceptor 10 by de-O-isopropylidenation. Glycosylation of 10 with methyl thioglycoside derivatives 18, 26, 34 by use of dimethyl(methylthio)sulfonium triflate (DMTST) gave the protected ganglioside oligosaccharides 19, 27 and 35, respectively. Compounds 9, 19, 27 and 35 were transformed into the corresponding α-trichloroacetimidates 13, 22, 30 and 38, via reductive removal of benzyl groups, O-acetylation, selective removal of 2-(trimethylsilyl)ethyl group, and treatment of trichloroacetonitrile. Condensation of the imidates 13, 22, 30 and 38 with (2S,3R,4E)-2-azido-3-O-benzoyl-4-octadecene-1,3-diol (14) gave the corresponding β-glycosides 15, 23, 31 and 39, which were converted, via selective reduction of azido group, coupling with octadecanoic acid, de-O-acylation, and saponification of methyl esters and lactone groups, into the corresponding gangliosides GD2 (17), GD1b (25), GT1b (33) and GQ1b (41).  相似文献

The continuous chemical investigation on the ethyl acetate (EtOAc) soluble fraction of the MeOH extract afforded two new lanostane triterpenoid derivatives including one with a rearranged lanostane skeleton. They were identified as 3,4-seco-8-(14→13R)abeo-17,13-friedo-9β-lanosta-4(28), 7,14(30),24-tetraen-26,23-olide-23-hydroxy-3-oic acid (1) and 7,14-mariesa- dien-3oL-hydroxy-25-methoxy-26-oic acid (2). Structural determination of these compounds were carried out by the spectral studies especially by the two digital (2D)-NMR and high-resolution MS experiences.  相似文献

A series of substituted benzoyl modified β-cyclodextrins, including mono-6-O-(p-methylbenzoyl)-β-CD (1), mono-6-O-(m-methylbenzoyl)-β-CD (2), mono-6-O-(o-methylbenzoyl)-β-CD (3), mono-6-O-(p-methoxylbenzoyl)-β-CD (4), mono-6-O-(m-methoxylbenzoyl)-β-CD (5), mono-6-O-(o-methoxylbenzoyl)-β-CD (6), mono-6-O-(m, p-dimethoxylbenzoyl)]-β-CD (7), mono-6-O-(o,m-dimethoxylbenzoyl)-β-CD (8), and mono-(6-O-benzoyl)-β-CD (9) were synthesized and their inclusion properties were studied by using fluorescence spectroscopy. The binding constants (Ka) of the modified β-CD derivatives with 2-p-toluidinylnaphthalene-6-sulfonate (TNS) were determined on the basis of the fluorescence spectroscopy. The effect of types and location of substituted groups of the benzene ring of the modified β-cyclodextrins on the binding property was discussed. Results indicated that the substituents had significant influences on the binding abilities of modified β-cyclodextrins.  相似文献

Eight known glycosides of hederagenin and the new triterpene glycoside 3-O-β-D-xylopyranosyl-(1→3)-O-α-L-rhamnopyranosyl-(1→2)-O-α-L-arabinopyranosyl-28-O-α-L-rhamnopyranosyl-(1→4)-O-6-O-acetyl-β-D-glucopyranosyl-(1→6)-O-β-D-glucopyranosyl ester of hederagenin were isolated by chromatographic methods from leaves of Kalopanax septemlobum var. maximowichii introduced to Crimea. The new dipyridyl ligands N,N′-(methylenedi-p-phenylene)bis(pyridine-4-carboxamide), L1, and N,N′-(methylenedi-p-phenylene)bis(pyridine-3-carboxamide), L2, incorporating amide spacers have been synthesized and reacted with metal salts to give complexes of the types [Cu(L1)2X2] (X = Cl, 1 and X = Br, 2), {[Cu(L1)2(DMF)](NO3)2}, 3, {[Ag2(L1)2](SO4)}, 4, and {[Cu(L2)(DMSO)2(NO3)](NO3)}, 5. All compounds have been characterized by spectroscopic methods and their structures determined by X-ray crystallography.Complexes 1, 2 and 3 form 1-D double-stranded polymeric chains showing rhombic molecular squares with approximate dimensions of 16.95 × 19.13 Å2 for 1, 17.03 × 19.06 Å2 for 2 and 16.66 × 19.94 Å2 for 3. Complex 4 forms infinite 1-D zigzag polymeric chains, which are interlinked through a series of Ag–O interactions to form wavy 1-D ladder like chains, and complex 5 forms 1-D sinusoidal chains. While the L1 ligands in complexes 1, 2 and 3 adopt the cis conformation and that in complex 4 adopts trans conformation, the L2 ligand in complex 5 adopts the trans-anti conformation. The ligand conformations also differ in the dihedral angles between the pyridyl and phenyl rings. All complexes exhibit emissions which may be tentatively assigned as intraligand (IL) π → π* transition.  相似文献

A new triterpenoid saponin, 3-O-[(6′-butyryl)-β-D-glucopyranosyl]-28-O-[α-L-rhamnopyranosyl-(1→4)-β-D-glucopyranosyl-(1→6)-β-D-glucopyranosyl] oleanolic acid, as well as three known triterpenoid saponins were isolated from the rhizomes of Anemone flaccida. Treatment of β-monosubstituted vinylic sulfoxides 1 with trifluoroacetic anhydride in dichloromethane gave excellent yields of 1,2-bis(trifluoroacetoxy)thioethers 6. Mildly basic methanolysis of 2-alkyl-substituted 6 gave α-hydroxyaldehydes 11 as monomer-dimer mixtures; similar treatment of the 2-aryl analogues afforded aryl (hydroxymethyl) ketones 12. Compounds 11 underwent Wittig reactions with methoxycarbonylmethylenetriphenylphosphorane to give high yields of γ-hydroxy-α,β-unsaturated esters 13, predominantly as the E-isomers. β-Monosubstituted vinylic sulfoxides 1 possessing a β-aryl group, and β-disubstituted vinylic sulfoxides 3 reacted with trifluoromethanesulfonic anhydride-sodium acetate in acetic anhydride to give 2-(phenylsulfenyl)acylals 14. These gave 2-phenylsulfenyl aldehydes 15 upon basic methanolysis, and the corresponding primary alcohols 16 on reduction with sodium borohydride. Reaction of both geometric isomers of enantiomerically pure vinylic sulfoxide 1o with TFAA gave racemic 6o as a mixture of diastereomers. Reaction of optically pure (E)- and (Z)-1p with trifluoromethanesulfonic anhydride-sodium acetate in acetic anhydride gave acylal 19 in 10.5 and 23% e.e., respectively.  相似文献

Dicyclohexylcarbodiimide-promoted self-condensation of 2,3:4,5-di-O-isopropylidene-d-galactonic acid (3) led to the macrocyclic oligomeric cyclo[(2,3:4,5-di-O-isopropylidene-(1→6)-d-galactonate)2] (4) and cyclo[(2,3:4,5-di-O-isopropylidene-(1→6)-d-galactonate)3] (5), having, respectively, 14- and 21-membered rings. The macrocycles 4 and 5 were also synthesized by cyclization of the respective linear dimer 11 and trimer 14 ω-hydroxy acids precursors prepared by stepwise additions of 3. Compounds 4 and 5 are biomaterials that may be described as macrolactone-cyclodextrins.  相似文献

The sponges Stelleta clarella Tethya aurantia, Lissodendoryx noxiosa, Haliclona permollis and Haliclona sp. were examined for steroids. All sponges contained C27-C29, Δ5, mono and diunsaturated sterols. In addition, the sponge Tethya aurantia contained Z - 24 - propylidene - cholest - 5- en -3β-ol (19) and the 5α,8α-peroxides of cholesta - 5,7 - dien - 3β - ol, ergosterol, ergosta - 5,7,24(28) - trien - 3β - ol and 24ξ - ethyl - cholesta - 5,7 - dien - 3β - ol (29, 30, 31 and 32). The sponge Stelleta clarella also contained 24 - nor - cholesta - 4,22 - dien - 3 - one (21), cholesta - 4,22 - dien - 3 - one (22), 24ξ - methyl - cholesta - 4,22 - dien - 3 - one (24), ergosta - 4,24(28) - dien - 3 - one (25), (E) - stigmasta - 4,24(28) - dien - 3 - one (28), 5α - cholestanol (5), 5α - ergostanol (7) and 5α - poriferastanol (9) The possible biosynthetic significance of these hitherto undescribed peroxides and enones from marine sources is discussed. Five triterpenoid saponins were isolated from the stems of Hedera colchica K. Koch (Araliaceae). Two of them are new natural substances. HCS-A (1): 3-O-α-L-arabinopyranoside, 28-O-α-L-rhamnopyranosyl-(1→ 4)-O-β-D-glucopyranosyl-(1→6)-O-β-D-glucopyranosyl-arjunolic acid. HCSt-B (2):3-O-β-D-xylopyranoside, 28-O-α-L-rhamnopyranosyl-(1→4)-O-β-D-glucopyranosyl-(1→6)-O-β-D-glucopyranosyl-hederagenin. A derivative of arjunolic acid is described for the first time in the Araliaceae family. A new polyhydroxysteroidal glycoside,(20R,24R)-16-O-(4-O-methyl-2-acetamido-2-deoxy-β-D-galactopyranosyl)-24-ethyl- 5α-cholest-8(14)-en-3β,6β,7β,16α-tetrol(anthenoside A,1),was isolated from the starfish Anthenea chinensis.Its structure was elucidated by extensive spectral analysis and chemical evidences.1 exhibited significant cytotoxicity against human tumor K-562, BEL-7402 and U87MG cells.  相似文献

The rational development of multinuclear arene ruthenium complexes (arene = p-cymene, hexamethylbenzene) from generation 1 (G1) and generation 2 (G2) of 4-iminopyridyl based poly(propyleneimine) dendrimer scaffolds of the type, DAB-(NH2)n (n = 4 or 8, DAB = diaminobutane) has been accomplished in order to exploit the ‘enhanced permeability and retention’ (EPR) effect that allows large molecules to selectively enter cancer cells. Four compounds were synthesised, i.e. [{(p-cymene)RuCl2}4G1] (1), [{(hexamethylbenzene)RuCl2}4G1] (2), [{(p-cymene)RuCl2}8G2] (3), and [{(hexamethylbenzene)RuCl2}8G2] (4), by first reacting DAB-(NH2)n with 4-pyridinecarboxaldehyde and subsequently metallating the iminopyridyl dendrimers with [(p-cymene)RuCl2]2 or [(hexamethylbenzene)RuCl2]2. The related mononuclear complexes [(p-cymene)RuCl2(L)] (5) and [(hexamethylbenzene)RuCl2(L)] (6) were obtained in a similar manner from N-(pyridin-4-ylmethylene)propan-1-amine (L). The molecular structure of 5 has been determined by X-ray diffraction analysis and the in vitro anticancer activities of the mono-, tetra- and octanuclear complexes 1–6 studied on the A2780 human ovarian carcinoma cell line showing a close correlation between the size of the compound and cytotoxicity.  相似文献

The ground- and excited-state structures for a series of Os(II) diimine complexes [Os(NN)(CO)2I2] (NN = 2,2′-bipyridine (bpy) (1), 4,4′-di-tert-butyl-2,2′-bipyridine (dbubpy) (2), and 4,4′-dichlorine-2,2′-bipyridine (dclbpy) (3)) were optimized by the MP2 and CIS methods, respectively. The spectroscopic properties in dichloromethane solution were predicted at the time-dependent density functional theory (TD-DFT, B3LYP) level associated with the PCM solvent effect model. It was shown that the lowest-energy absorptions at 488, 469 and 539 nm for 13, respectively, were attributed to the admixture of the [dxy (Os) → π*(bpy)] (metal-to-ligand charge transfer, MLCT) and [p(I) → π*(bpy)] (interligand charge transfer, LLCT) transitions; their lowest-energy phosphorescent emissions at 610, 537 and 687 nm also have the 3MLCT/3LLCT transition characters. These results agree well with the experimental reports. The present investigation revealed that the variation of the substituents from H → t-Bu → Cl on the bipyridine ligand changes the emission energies by altering the energy level of HOMO and LUMO but does not change the transition natures.  相似文献
