The H‐phosphonate bioconjugates of bile acids, conjugated with various alcohols and nucleosides, were obtained in one pot by a tandem transesterification with diphenyl phosphite (DPP). The synthesis of cholic acid derived phosphoramide from the corresponding H‐phosphonate was also demonstrated. The structures of these novel conjugates were confirmed on the basis of IR,31P NMR, 1H NMR, and mass spectra. The synthesized bile acid conjugates were mixtures of diastereoisomers due to the chirality of the phosphorus. © 2008 Wiley Periodicals, Inc. Two novel water‐soluble indocyanine dyes were synthesized. Their chemical structures were confirmed by 1H NMR, IR, MS and UV‐vis techniques. The absorptions of the prepared dyes in different solvents were measured. Two dyes exhibited negative solvatochromism, with a blue shift of the absorption and an emission maximum in (bovine serum albumin) protonic solvent relative to non protonic solvent. The fluorescence properties of the dyes in both free state and BSA were investigated. Significant enhancement of the fluorescence intensity for the Dye2 was observed in the presence of BSA. Dye2 could be proposed as fluorescent dyes for BSA detection in aqueous solution.  相似文献

The binding interaction of a Schiff base compound containing a 1,2,4‐triazole ring [4‐(4‐chlorobenzyl‐ideneamino)‐5‐methyl‐1,2,4‐triazole‐3‐thiol, CTT] with bovine serum albumin (BSA) was studied by spectroscopy methods including fluorescence and circular dichroism spectrum under simulative physiological conditions. Fluorescence investigation revealed that the fluorescence quenching of BSA was induced by the formation of a relative stable CTT‐BSA complex. The corresponding binding constants (Ka) between CTT and BSA at three different temperatures were calculated according to the modified Stern‐Volmer equation. The enthalpy change (ΔH) and entropy change (ΔS) were calculated to be −15.78 kJ·mol−1 and 49.23 J·mol−1·K−1, respectively, which suggested that hydrophobic forces and hydrogen bond played major roles in stabilizing the CTT‐BSA complex. Site marker competitive experiments indicated that the binding of CTT to BSA primarily took place in sub‐domain IIIA (site II) of BSA. The binding distance (r) between CTT and the tryptophan residue of BSA was obtained to be 4.3 nm based on F?rster theory of non‐radioactive energy transfer. The conformational investigation revealed that the presence of CTT decreased the α‐helix content of BSA (from 58.62% to 54.66%) and induced the slight unfolding of the polypeptides of protein, which confirmed some micro‐environmental and conformational changes of BSA molecules.  相似文献

Two small molecules named PI‐DPP and NI‐DPP with a DPP core as the central strong acceptor unit and phthalimide/naphthalimide as the terminal weak acceptor were designed and synthesized. The effects of terminal phthalimide/naphthalimide units on the thermal behavior, optical and electrochemical properties, as well as the photovoltaic performance of these two materials were systematically studied. Cyclic voltammetry revealed that the lowest unoccupied molecular orbitals (LUMO) (~ ‐3.6 eV) of both molecules were intermediate to common electron donor (P3HT) and acceptor (PCBM). This indicated that PI‐DPP and NI‐DPP may uniquely serve as electron donor when blended with PCBM, and as electron acceptor when blended with P3HT, where sufficient driving forces between DPPs and PCBM, as well as between P3HT and DPPs should be created for exciton dissociation. Using as electron donor materials, PI‐DPP and NI‐DPP devices exhibited low power conversion efficiencies (PCEs) of 0.90% and 0.76% by blending with PCBM, respectively. And a preliminary evaluation of the potential of the NI‐DPP as electron acceptor material was carried out using P3HT as a donor material, and P3HT: NI‐DPP device showed a PCE of 0.6%, with an open circuit voltage (V OC) of 0.7 V, a short circuit current density (J SC) of 1.91 mA•cm‐2, and a fill factor (FF) of 45%.  相似文献

The interaction between CdTe quantum dots (QDs) and bovine serum albumin (BSA) was systematically investigated by fluorescence, UV‐vis absorption and circular dichroism (CD) spectroscopy under physiological conditions. The experimental results showed that the fluorescence of BSA could be quenched by CdTe QDs with a static quenching mechanism, indicating that CdTe QDs could react with BSA. The quenching constants according to the modified Stern‐Volmer equation were obtained as 1.710×106, 1.291×106 and 1.010×106 L·mol?1 at 298, 304, and 310 K, respectively. ΔH, ΔS and ΔG for CdTe QDs‐BSA system were calculated to be ?33.68 kJ·mol?1, 6.254 J·mol?1·K?1 and ?35.54 kJ·mol?1 (298 K), respectively, showing that electrostatic interaction in the system played a major role. According to F?rster theory, the distance between Trp‐214 in BSA and CdTe QDs was given as 2.18 nm. The UV‐vis, synchronous fluorescence and CD spectra confirmed further that the conformations of BSA after addition of CdTe QDs have been changed.  相似文献

Six novel asymmetrical monomethine cyanine dyes were synthesized via the condensation reaction of 1‐butyl‐2‐(methylthio)benzo[c,d]indol‐1‐ium iodide and various 1,5‐substituted indolenine salts under basic conditions. The dyes were characterized using UV–vis spectroscopy, fluorescence, 1H NMR, 13C NMR, and mass spectrometry; furthermore, the purity of these compounds was observed using LC/ELSD/MS.  相似文献

A series of water‐soluble red‐emitting distyryl‐borondipyrromethene (BODIPY) dyes were designed and synthesized by using three complementary approaches aimed at introducing water‐solubilizing groups on opposite faces of the fluorescent core to reduce or completely suppress self‐aggregation. An additional carboxylic acid functional group was introduced at the pseudo‐meso position of the BODIPY scaffold for conjugation to amine‐containing biomolecules/biopolymers. The optical properties of these dyes were evaluated under simulated physiological conditions (i.e., phosphate‐buffered saline (PBS), pH 7.5) or in pure water. The emission wavelength (λmax) of these labels was found in the 640–660 nm range with quantum yields from modest to unprecedentedly high values (4 to 38 %). The bioconjugation of these distyryl‐BODIPY dyes with bovine serum albumin (BSA) and the monoclonal antibody (mAb) 12A5 was successfully performed under mild aqueous conditions.  相似文献

A novel π‐conjugated triad and a polymer incorporating indolo[3,2‐b]‐carbazole (ICZ) and 4,4‐difluoro‐4‐bora‐3a,4a‐diaza‐s‐indacene (BODIPY) were synthesized via a Sonogashira coupling. Compared to the parent BODIPY the absorption and fluorescence spectrum were for both compounds broader and redshifted. The redshift of the fluorescence and the decrease of the fluorescence quantum yield and decay time upon increasing solvent polarity were attributed to the formation of a partial charge‐transfer state. Upon excitation in the ICZ absorption band the ICZ fluorescence was quenched in both compounds mainly due to energy transfer to the BODIPY moiety. In a similar ICZ–π–DPP polymer (where DPP is diketopyrrolopyrrole), a smaller redshift of the absorption and fluorescence spectra compared to the parent DPP was observed. A less efficient quenching of the ICZ fluorescence in the ICZ–π–DPP polymer could be related to the unfavorable orientation of the transition dipoles of ICZ and DPP. The rate constant for energy transfer was for all compounds an order of magnitude smaller than predicted by Förster theory. While in a solid film of the triad a further redshift of the absorption maximum of nearly 100 nm was observed, no such shift was observed for the ICZ–π–BODIPY polymer.  相似文献

Fluorescence spectroscopy, Fourier transform infrared (FT‐IR) spectroscopy, circular dichroism (CD) and FT‐Raman spectroscopy were employed to analyze the binding of the asiatic acid (AA) to bovine serum albumin (BSA) under simulative physiological conditions. Fluorescence data revealed that the fluorescence quenching of BSA by AA was the result of the formation of BSA‐AA complex. The fluorescence quenching mechanism of BSA by AA was a static quenching procedure. According to the Van′t Hoff equation, the thermodynamic parameters enthalpy change (ΔH0) and entropy change (ΔS0) for the reaction were evaluated to be ?12.55 kJ·mol?1 and 67.08 kJ·mol?1, respectively, indicating that hydrophobic and electrostatic interactions played a major role in stabilizing the complex. The influence of AA on the conformation of BSA has also been analyzed on the basis of FT‐IR, CD and FT‐Raman spectra.  相似文献

At different temperatures, the interactions between imidacloprid (IMI) and bovine serum albumin (BSA) were investigated with a fluorescence quenching spectrum, a synchronous fluorescence spectrum, a three-dimensional fluorescence spectrum and an ultraviolet-visible spectrum. The average values of bonding constants (KLB: 3.424 × 10^4 L,mol^-1), thermodynamic parameters (△H: 5.188 kJ,mol^-1, △G^(○—):-26.36 kJ,mol^-1, △S: 103.9 J,K^-1,mol^-1) and the numbers of bonding sites (n: 1.156) could be obtained through Stern-Volmer, Lineweaver-Burk and ther- modynamic equations. It was shown that the fluorescence of BSA could be quenched for its reactions with IMI to form a certain kind of new compound. The quenching belonged to a static fluorescence quenching, with a non-radiation energy transfer happening within a single molecule. The thermodynamic parameters agree with △H〉 0, △S〉0 and△G^(○-)〈0, suggesting that the binding power between IMI and BSA should be mainly a hydrophobic interaction.  相似文献

The ring‐opening polymerizations (ROPs) of εcaprolactone (ε‐CL) and L ‐lactide (LLA) have been studied using the organocatalysts of diphenyl phosphate (DPP) and 4‐dimethylaminopyridine (DMAP). The “dual activation” property of DPP and the “bifunctional activation” property of DPP/DMAP were confirmed by the NMR measurement for ε‐CL and its chain‐end model of poly(ε‐caprolactone) and for LLA and its chain‐end model of poly(L ‐lactide) (PLLA), respectively. The molar ratio of DPP/DMAP was optimized as 1/2 for the ROP of LLA leading to the well‐defined PLLA, such as the molecular weight determined from 1H NMR measurement of 19,200 g mol?1 and the narrow polydispersity of 1.10. Additionally, functional initiators were utilized for producing the end‐functionalized PLLAs. The DPP‐catalyzed ROPs of ε‐CL and its analogue cyclic monomers and then the DPP/DMAP‐catalyzed ROP of LLA produced block copolymers. © 2014 Wiley Periodicals, Inc. J. Polym. Sci., Part A: Polym. 在模拟生理条件下,采用荧光光谱法、圆二色光谱法和红外光谱法研究了花椒油素(XT)与牛血清白蛋白(BSA)的相互作用。结果表明花椒油素与牛血清白蛋白之间发生动态和静态联合猝灭,二者间的的猝灭常数(K)在286, 298和310 K分别为3.31 × 105, 到2.03 × 105 和 0.94 × 105 L∙mol-1. 热力学参数表明, 花椒油素与牛血清白蛋白间以疏水作用力为主。圆二色光谱和红外光谱法表明加入花椒油素后,牛血清白蛋白的二级结构发生了变化,其中α-螺旋减少了3.9%。另外,我们还研究了共存离子对两者结合的影响。  相似文献

The binding of kaempferol‐3,7‐αL‐rhamnopyranoside (KRR) with bovine serum albumin (BSA) was investigated by different spectroscopic methods under simulative physiological conditions. Analysis of ?uorescence quenching data of BSA by KRR at different temperatures using Stern‐Volmer methods revealed the formation of a ground state KRR‐BSA complex with moderate binding constant of the order 104 L·mol?1. The existence of some metal ions could weaken the binding of KRR on BSA. The changes in the van't Hoff enthalpy (ΔH0) and entropy (ΔS0) of the interaction were estimated to be ?26.53 kJ·mol?1 and 3.33 J·mol?1·K?1 and both hydrophobic and electrostatic forces contributed to stabilizing the BSA‐KRR complex. According to the F?ster theory of non‐radiation energy transfer, the distance r between the donor (BSA) and the acceptor (KRR) was obtained (r=2.83 nm). Site marker competitive experiments showed that KRR could bind to Site I of BSA. In addition, synchronous fluorescence, UV‐Vis absorption and circular dichroism (CD) results indicated that the KRR binding could cause conformational changes of BSA.  相似文献

Two diketopyrrolopyrrole (DPP)‐based donor–acceptor (D–A) conjugated molecules, DPP‐F and DPP‐2F, which contain E‐(1,2‐difluorovinyl) moieties, are reported. The LUMO energies of DPP‐F and DPP‐2F were estimated to be ?3.49 and ?3.70 eV, respectively, based on their redox potentials and absorption spectral data; these values were clearly lowered because of the incorporation of electron‐withdrawing E‐(1,2‐difluorovinyl) moieties. Organic field‐effect transistors (OFETs) with thin films of DPP‐F and DPP‐2F were successfully fabricated with conventional techniques. Based on the respective transfer and output characteristics measured in an inert atmosphere, thin films of DPP‐2F display ambipolar semiconducting behavior with hole and electron mobilities reaching 0.42 and 0.80 cm2 V?1 s?1, respectively. The as‐prepared OFET of DPP‐2F already shows high hole and electron mobilities that are not influenced remarkably by thermal annealing. For thin films of DPP‐F, only p‐type semiconducting behavior was observed in both an inert atmosphere and air, and the hole mobility increased to 0.1 cm2 V?1 s?1 after thermal annealing. XRD and AFM studies were performed with thin films of DPP‐F and DPP‐2F after annealing at different temperatures.  相似文献

Caged rhodamine dyes (Rhodamines NN) of five basic colors were synthesized and used as “hidden” markers in subdiffractional and conventional light microscopy. These masked fluorophores with a 2‐diazo‐1‐indanone group can be irreversibly photoactivated, either by irradiation with UV‐ or violet light (one‐photon process), or by exposure to intense red light (λ~750 nm; two‐photon mode). All dyes possess a very small 2‐diazoketone caging group incorporated into the 2‐diazo‐1‐indanone residue with a quaternary carbon atom (C‐3) and a spiro‐9H‐xanthene fragment. Initially they are non‐colored (pale yellow), non‐fluorescent, and absorb at λ=330–350 nm (molar extinction coefficient (ε)≈104 M?1 cm?1) with a band edge that extends to about λ=440 nm. The absorption and emission bands of the uncaged derivatives are tunable over a wide range (λ=511–633 and 525–653 nm, respectively). The unmasked dyes are highly colored and fluorescent (ε= 3–8×104 M?1 cm?1 and fluorescence quantum yields (?)=40–85 % in the unbound state and in methanol). By stepwise and orthogonal protection of carboxylic and sulfonic acid groups a highly water‐soluble caged red‐emitting dye with two sulfonic acid residues was prepared. Rhodamines NN were decorated with amino‐reactive N‐hydroxysuccinimidyl ester groups, applied in aqueous buffers, easily conjugated with proteins, and readily photoactivated (uncaged) with λ=375–420 nm light or intense red light (λ=775 nm). Protein conjugates with optimal degrees of labeling (3–6) were prepared and uncaged with λ=405 nm light in aqueous buffer solutions (?=20–38 %). The photochemical cleavage of the masking group generates only molecular nitrogen. Some 10–40 % of the non‐fluorescent (dark) byproducts are also formed. However, they have low absorbance and do not quench the fluorescence of the uncaged dyes. Photoactivation of the individual molecules of Rhodamines NN (e.g., due to reversible or irreversible transition to a “dark” non‐emitting state or photobleaching) provides multicolor images with subdiffractional optical resolution. The applicability of these novel caged fluorophores in super‐resolution optical microscopy is exemplified.  相似文献

We describe multimodal imaging probes for gastrin-releasing peptide receptor (GRPR)-specific targeting suited for positron emission tomography and optical imaging (PET/OI), consisting of PESIN (PEG3-BBN7-14) dimers connected to multimodal imaging subunits. These multimodal agents comprise a fluorescent dye for OI and the chelator ((1,4,7-triazacyclononane-4,7-diyl)diacetic acid-1-glutaric acid) (NODA-GA) for PET radiometal isotope labelling. Special focus was put on the influence of the used dyes on the properties of the whole bioconjugates. For this, several compounds with different fluorescent dyes and non-dye carrying subunits were synthesized and investigated. As fluorescent dyes, dansyl, NBD, derivatives of fluorescein, coumarin and rhodamine as well as three pyrilium-based dyes were employed. Considerable influence of the charge of the colored unit on hydrophilicity as well as in vitro target receptor binding was observed and classified. High radiochemical yields and purities were found during radiolabeling of the multimodal imaging subunits as well as their GRPR-specific bioconjugates with 68Ga. Examinations of the photophysical properties of both molecule species displayed no loss or alteration of fluorescence characteristics.  相似文献

Glutathione (GSH) tagged with a ferrocene (Fc) label at its C‐terminal was synthesized via coupling ferrocenyl amine to glutathione using o‐(benzotriazol‐1‐yl)‐N,N,N′,N′‐tetramethyluronium (HBTU)/1‐hydroxybenzotrizole (HOBt). The presence of Fc yielded well defined voltammetric signals, rendering the Fc‐tagged GSH (GSH‐Fc) suitable for electrochemical studies of GSH binding to other biological species. The interaction of GSH‐Fc with bovine serum albumin (BSA) was investigated, and a binding ratio of 1.41±0.06 (GSH‐Fc/BSA) and an affinity constant Ka of 6.53±2.01×106 M?1 were determined. These results compare well with those measured by fluorescence using untagged GSH, suggesting that the attachment of Fc to GSH does not significantly perturb the GSH structure and binding behavior. By contrasting the binding behavior to several compounds that are known to conjugate to different domains of BSA, the voltammetric study confirmed that GSH‐Fc binds at subdomain IIA of BSA with high affinity. The versatility of GSH‐Fc for studying GSH binding to surface‐confined proteins was also demonstrated with the GSH binding to electroinactive Zn‐metallothionein (Zn7‐MT) through hydrogen binding at the region between the Zn7‐MT α and β domains.  相似文献

This article describes a series of nine complexes of boron difluoride with 2′‐hydroxychacone derivatives. These dyes were synthesized very simply and exhibited intense NIR emission in the solid state. Complexation with boron was shown to impart very strong donor–acceptor character into the excited state of these dyes, which further shifted their emission towards the NIR region (up to 855 nm for dye 5 b , which contained the strongly donating triphenylamine group). Strikingly, these optical features were obtained for crystalline solids, which are characterized by high molecular order and tight packing, two features that are conventionally believed to be detrimental to luminescence in organic crystals. Remarkably, the emission of light from the π‐stacked molecules did not occur at the expense of the emission quantum yield. Indeed, in the case of pyrene‐containing dye 4 , for example, a fluorescence quantum yield of about 15 % with a fluorescence emission maximum at 755 nm were obtained in the solid state. Moreover, dye 3 a and acetonaphthone‐based compounds 1 b , 2 b , and 3 b showed no evidence of degradation as solutions in CH2Cl2 that contained EtOH. In particular, solutions of brightly fluorescent compound 3 a (brightness: ε×Φf=45 000 M ?1 cm?1) could be stored for long periods without any detectable changes in its optical properties. All together, these new dyes possess a set of very interesting properties that make them promising solid‐state NIR fluorophores for applications in materials science.  相似文献

The modular assembly of boronic acids with Schiff‐base ligands enabled the construction of innovative fluorescent dyes [boronic acid salicylidenehydrazone (BASHY)] with suitable structural and photophysical properties for live cell bioimaging applications. This reaction enabled the straightforward synthesis (yields up to 99 %) of structurally diverse and photostable dyes that exhibit a polarity‐sensitive green‐to‐yellow emission with high quantum yields of up to 0.6 in nonpolar environments. These dyes displayed a high brightness (up to 54 000 m ?1 cm?1). The promising structural and fluorescence properties of BASHY dyes fostered the preparation of non‐cytotoxic, stable, and highly fluorescent poly(lactide‐co‐glycolide) nanoparticles that were effectively internalized by dendritic cells. The dyes were also shown to selectively stain lipid droplets in HeLa cells, without inducing any appreciable cytotoxicity or competing plasma membrane labeling; this confirmed their potential as fluorescent stains.  相似文献

An electrochemical biosensor was developed for the determination of Escherichia coli (E. coli) in water. For this purpose, silver‐gold core‐shell (Ag@Au) bioconjugates and anti‐E. coli modified PS‐microwells were designed in a sandwich‐type format in order to obtain higher sensitivity and selectivity. Ag@Au bimetallic nanoparticles were synthesized by co‐reduction method. The core‐shell formation was analyzed by using UV‐Vis spectroscopy and transmission electron microscopy. Biotin labeled anti‐E. coli antibodies were coupled with Ag@Au nanoparticles to form bioconjugates. The electrochemical immunosensor was prepared by immobilizing anti‐E. coli on polystyrene (PS)‐microwells via chemical bonding. These modified microwells were identified with X‐ray photoelectron spectroscopy and surface enhanced Raman spectroscopy. E. coli was sandwiched between Ag@Au bioconjugates and anti‐E. coli on PS‐microwells at different concentrations. The relationship between the E. coli concentration and stripping current of gold ions (Au3+) were investigated by square wave anodic stripping voltammetry at pencil graphite electrode. The proposed method can provide some advantages such as lower detection limit and shorter detection time. The electrochemical response for the immunosensor was linear with the concentration of the E. coli in the range of 101 and 105 cfu/mL with a limit of detection 3 cfu/mL. The procedure maintains good sensitivity and repeatability and also offers utility in the fields of environmental monitoring and clinical diagnosis.  相似文献
